Objectives : This study was performed to investigate the anti-fibrogenic effect of Salvia miltiorrhiza on rat hepatic stellate cells. Materials and Methods : Hepatic stellate cells(HSC-T6) were treated with various concentrations of Salvia miltiorrhiza extract for 24 hours. It was extracted either with distilled water or 50% EtOH. After the treatment, cell viability, proliferation, procollagen levels and the mRNA of the collagen type 1a2 and ASMA were measured Results : The viability and proliferation of the hepatic stellate cells were decreased as concentration increased. The mRNA expression decreased consistently with the volume of the secreted procollagen with the extraction made with distilled water. This indicates the herb has inhibitory effect on fibrogenesis of the liver by regulating one of the fibrosis associated genes in transcription. However it increased in 50% EtOH extraction, which shows that a more stable reaction is expected of the extraction made with distilled water than the extraction made with 50% EtOH. The production of procollagen was decreased by a low-concentration treatment with Salvia miltiorrhiza, but increased by a high concentration. It seemed that the cells were responding to Salvia miltiorrhiza in low- concentration, thus producing smaller amounts of collagen. When the drug was administered in high enough concentration to give direct toxicity to cells, an overproduction of collagen was observed. Conclusion : These results suggest that Salvia miltiorrhiza is a possible candidate for the treatment of cirrhotic patients as well as for patients with chronic hepatitis when extracted with water in the proper concentrations.

The behavior of glycogen and histological changes of hepatic tissues in the liver of rats, aged 6 to 7 weeks, fed 18% casein diet under control, gramoxone and gramoxone + vitamin C(Vt. C) diets has been investigated in a comined histropathological and histochemical studies. Cloudy swelling and fat changes of hepatic cells were observed in the gramoxone group with the duration of feeding time. Fat changes of hepatic cells were observed more obviously than cloudy welling, especially in the hepatic cells of periportal area. The number of Kupffer's cells increased signifciantly in the gramoxone group fed for 4weeks. The cloudy swelling and fat changes decreased obviously in the gramxone + Vt. C group. Glycogen content of heaptic cells tended to increase slightly in the gramoxone group as compared with the control group . moreover, glycogen depositons were higher in the hepatic cells where fat changes were obvious. It seems to be that Vt.C alleviating effects on the gramoxone toxicity in the patterns of glycogen distribution and histological structure of heaptic tissues.

The 15-kDa selenoprotein (Sep15) is a selenoprotein residing in the lumen of the endoplasmic reticulum (ER) and implicated in quality control of protein folding. Herein, we established an inducible RNAi cell line that targets Sep15 mRNA in Chang liver cells. RNAi-induced Sep15 deficiency led to inhibition of cell proliferation, whereas cell growth was resumed after removal of the knockdown inducer. Sep15-deficient cells were arrested at the G1 phase by upregulating p21 and p27, and these cells were also characterized by ER stress. In addition, Sep15 deficiency led to the relocation of focal adhesions to the periphery of the cell basement and to the decrease of the migratory and invasive ability. All these changes were reversible depending on Sep15 status. Rescuing the knockdown state by expressing a silent mutant Sep15 mRNA that is resistant to siRNA also reversed the phenotypic changes. Our results suggest that SEP15 plays important roles in the regulation of the G1 phase during the cell cycle as well as in cell motility in Chang liver cells, and that this selenoprotein offers a novel functional link between the cell cycle and cell motility.

These studies were carried out to detect the presence of mycotoxin producing fungi in various kind of grains and foodstuffs in Korea. The experiments were divided into three parts: bacteriologic, toxicologic and electron microscopic studies. The results were summarized as follows: 1. From the 133 various samples, 426 colonies of fungi were isolated. In 405 of 426 colonies, it was possible to identify 17 genera. Among the identified strains, the predominant genera were Penicillium, Aspergillus and Alternaria. 2. In cytotoxicity test, 20 strains showed mild to severe toxic effects in mice and 24 strains showed toxic effects on HeLa cells among the 107 strains of experiments. 3. In electron microscopic studies of liver cells from animal which had been treated with toxin like substances, the liver cells showed the cytoplasmic changes: dilatation of endoplasmic reticulum, swelling of mitochondria, disappearance of mitochondrial cristae, increased number of lipid and glycogen particles. Nucleus and nuclear envelope alterations were also noted. 4. In fine structure of HeLa cells treated with culture filtrates of mycotoxin producing fungi and experimental strains had been noted a certain specific changes induced by culture filtrates. These alterations showed the disappearance of golgi complex, endoplasmic reticulum vacuolization and mitochondrial changes. 5. As a mass screening, the cytotoxicity tests of HeLa cells and histopathologic study of mice liver cells treated with toxin-like substances, employed are feasible to detect mycotoxin producing fungi.

In order to validate the objective efficacy of CBPT on anti-asthma and to develop effective therapeutics for asthma treatments, immunological modulatory mechanism was studied using animal model using OVA-Alum. The results are listed below. When treated with CBPT, survival rate of hFCs at 250 ug/ml was above 90%. AST and ALT, indicators of liver function measurements were in the normal range. Compared to the control group, CBPT treated group showed significant reduction in liver weights at both 400 and 200 mg/kg, and significant decrease of total liver cells at 400 mg/kg. Significant increase in CD4+ and CD8+ cells in DLN was observed in the CBPT treated group. Slight increase in CD3+, CD4+/CD25+ cells were also observed. On the other hand, CBPT significantly reduced the CD3+/CD69+ cell numbers at both concentrations. Slight decrease of CD19+ cells was also observed. CBPT significantly reduced the CD3e+/CD69+, CCR3+ and CD11b+/Gr-1+ cells in lung tissues at both doses. However, significant decrease of CD3e+ and B220+/IgE+ cells was only observed at 400 mg/kg dosed group. The results above strongly suggest the anti-asthmatic effect of CBPT through immunological modulation. By using various concentrations of CBPT, broader clinical applications of CBPT on anti-asthmatic treatment can be developed. The EBM database should provide valuable information in the development of drugs for asthma treatments.
Objective : This study on Sipimikwanjung-tang was undertaken to evaluate its antioxidant capacities and antiperoxidation activities in rat liver tissues. Sipimikwanjung-tang which has been one of the prescriptions in sasang constitutional medicine is usually applied for the therapy of various liver diseases. It is elucidated that Sipimikwanjung-tang has antioxidants on liver tissue of rat and the cytotoxic effects on human hepatoblastoma Hep G2 cells. Methods: Sipimikwanjung-tang extract in antioxidant effects of Hep G2 cells is evaluated by MTT assay, DAPI staining, DNA fragmentation assays and FACS can analysis. Results: Sipimikwanjung-tang induced apoptosis in Hep G2 cells, and induced G1 and G2M arrest of the cell cycle as well as a significant increase in PARP and caspase-3 activity. It induced an increase in $H_2O_2$ generation and the subsequent $NF-{\kappa}B$ activation and also induced cell apoptosis through the caspase-3-dependent pathways in the low concentration of Sipimikwanjung-tang extracts. However, the high dose of Sipimikwanjung-tang extract in Hep G2 cells inhibited $TGF-{\beta}l-induced$ apoptosis via increase in cellular $H_2O_2$, formation and $NF-{\kappa}B$ activation in human hepatoblastoma Hep G2 cells. Conclusion: From this study, the possibility that Sipimikwanjung-tang extracts apply to antioxidant and apoptotic treatment of disease is revealed.

Oxymatrine (OMT) often used in treatment for chronic hepatitis B virus infection in clinic. However, OMT-induced liver injury has been reported. In this study, we aim to investigate the possible mechanism of OMT-induced hepatotoxicity in human normal liver cells (L02). Exposed cells to OMT, the cell viability was decreased and apoptosis rate increased, the intracellular markers of oxidative stress were changed. Simultaneously, OMT altered apoptotic related proteins levels, including Bcl-2, Bax and pro-caspase-8/-9/-3. In addition, OMT enhanced the protein levels of endoplasmic reticulum (ER) stress makers (GRP78/Bip, CHOP, and cleaved-Caspase-4) and phosphorylation of c-Jun N-terminal kinase (p-JNK), as well as the mRNA levels of GRP78/Bip, CHOP, caspase-4, and ER stress sensors (IREI, ATF6, and PERK). Pre-treatment with Z-VAD-fmk, JNK inhibitor SP600125 and N-acetyl-l-cysteine (NAC), a ROS scavenger, partly improved the survival rates and restored OMT-induced cellular damage, and reduced caspase-3 cleavage. SP600125 or NAC reduced OMT-induced p-JNK and NAC significantly lowered caspase-4. Furthermore, 4-PBA, the ER stress inhibitor, weakened inhibitory effect of OMT on cells, on the contrary, TM worsen. 4-PBA also reduced the levels of p-JNK and cleaved-caspase-3 proteins. Therefore, OMT-induced injury in L02 cells was related to ROS mediated p-JNK and ER stress induction. Antioxidant, by inhibition of p-JNK or ER stress, may be a feasible method to alleviate OMT-induced liver injury.
Extrathoracic metastases of thymomas are extremely rare, occurring in less than 2% of cases. We present a case of metastatic thymoma in the liver diagnosed by aspiration biopsy. A fine-needle aspirate from a hepatic mass of a 65-year old man who had a history of malignant thymoma locally invading pericardium was examined. Nests of epithelial cells with few scattered lymphocytes were present. The nuclei of the epithelial cells were round to oval and appeared relatively uniform and regular, with fine chromatin pattern. They had one or two, small but lather conspicuous nucleoli. Unlike previous reports on the findings of the aspiration cytology of thymomas, the characteristic biphasic pattern was not present in this case.
Effects of Ssang Wha Tang (SWT), a blended Chinese traditional medicine, on the pharmacokinetics of sulfobromophthalein (BSP) were studied in the rats. BSP was administered via portal vein to the control and the SWT-treated rats. The in vitro distribution of BSP to blood cells and the hemato-physiological conditions, liver weight, GOT. GPT activity were also examined. The systemic clearance $(CL_s)$ of BSP was increased with the administration of SWT, but no significant differences were observed in the liver weight and in vitro distribution of BSP to blood cells. These results suggest that the intrinsic clearance of free BSP of the liver is increased with the administration of SWT in the rats.
Mitochondria was observed much around the nuclear membrane of liver tissue where the energy metabolism process is active. Testis tissue had a large number of undifferentiated cells, and cristae in Inner membrane of Mitochondria was not observed clearly. Morphological damage occurred first in Inner membrane rather than the outer membrane. The kidney tissue was clearly observed in the form of cristae. Radiation-induced damage occurred at the edges of both ends, and the membrane was observed bursting with the thickness of the outer membrane. Small intestine cells were observed in many mitochondria in the tissues around the villus, where bowel movements were active. Morphological damage occurred with the outer and inner membranes getting tangled. Mitochondria sensitivity to radiation was sensitized in testis and small intestine tissues, and kidney, ovary and liver tissues were found to be resistant.
