The Hepatotprotective and Antioxidative Effects of Onion (Allium cepa) Extracts in Rat Hepatocyte Primary Culture (양파(Allium cepa) 추출물의 간보호 및 항산화 효과)

  • The objectives of present study were to investigate the hepatoprotective and antioxidative effects of onion extracts. Primary cultures of rat hepatocytes were incubated with 1.5 mM tert-butyl hydroperoxide(t-BHP), potent oxidizing agent for liver injury for 1 hr in the presence or absence of various concentrations (0, 0.01, 0.05, 0.1 or 0.3 mg/ml) of onion extract. Cytotoxicity and cell viability were determined by measuring glutamic oxaloacetic transaminase(GOT) activity, lactate dehydrogenase(LDH) activity and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) value. Lipid peroxidation was evaluated using thiobarbituric acid reactive substances(TBARS) assay. Effects on antioxidant system were determined by measuring catalase, glutathione peroxidase(GSH-Px), glutathione reductase(GSH-Rd) activities as well as DNA strand breaking assay. Incubation with t-BHP alone increased GOT and LDH activities and TBARS concentration but decreased MTT reduction. Onion extracts at the concentration of 0.05 mg/ml began to decrease GOT and LDH activities induced by 1.5 mM t-BHP. Decreased MTT reduction began to be increased by onion extract at the concentration of 0.01 mg/ml. Onion extracts at the concentration of 0.01 mg/ml began to decrease TBARS concentration induced by t-BHP. Taken together, onion extracts prevented t-BHP-induced hepatocyte injury and lipid peroxidation. Catalase, GSH-Px and GSH-Rd activities of hepatocytes were significantly decreased by 1.5 mM t-BHP for 1 hr incubation. Onion extracts, on the other hand, at the concentration of 0.1 mg/ml began to prevent t-BHP-induced decrease in catalase, GSH-Px and GSH-Rd activities. Onion extracts prevented hydroxyl radical-induced single-strand breakage in dose-dependent manner when plasmid DNA was incubated with various concentrations of onion extracts in the presence of Fenton regents producing hydroxyl radical. These results demonstrate that onion extracts suppressed t-BHP-induced cytoctoxicity, decreased viability and lipid peroxidation and increased GSH-Px, GSH-Rd and catalase activities. Thus hepatoprotective and antioxidant effects of onion extract seem to be due to, at least in part, the increase in antioxidant enzyme activities as well as prevention from hydroxyl radical-induced oxidation, followed by inhibition in lipid peroxidation.

Antioxidative Effect of Plant Food Mixtures in Rat Fed on High Fat-High Cholesterol Diet (고지방고콜레스테롤식이를 섭취한 흰쥐에서 식물성식품의 혼합물 첨가로 인한 체내 항산화능 증가 효과)

  • The previous extensive in vitro studies on the antioxidative activities of a number of Korean grains, vegetables, seaweeds and mushrooms, and the various combinations of these food source exhibited a wide range of antioxidative activities, and four food mixtures composed of 5 kinds of foods (5A, 5B, 5C and 5D) were designed from 16 selective foods showing. high antioxidant effect, in vitro, to find the good combinations for the meal planning. Mixture 5B or 5C contained very high levels of total flavonoid and polyphenol, and ethanol extract from 5A, 5B or 5C showed very strong inhibitory effects against in vitro $Fe^{2+}-induced$ lipid peroxidation and ethanol extract from 5B or 5C showed remarkable DPPH radical scavenging effect and lipid peroxide-protein conjugation inhibition effect. And in vivo study was also carried out with two mixtures (5B, 5C). Powders (P5B, P5C) or ethanol extracts (E5B, E5C) of these mixtures were supplemented to Sprague-Dawley rats fed on high fat $(15\%)-high$ cholesterol $(1\%)$ semipurified diet for 5 weeks. The total antioxidant power in serum was significantly higher in P5B, P5C, E5B and E5C groups than in high fat control group, and $ascorbate-Fe^{2+}-induced$ TBARS was significantly lowered by E5B supplementation in rat liver. In liver tissue, Cu, Zn-SOD activity was significantly higher in P5B and E5B groups than in high fat control group, while catalase or GSH-peroxidase (GPx) activity was not changed by any supplementations. In kidney, Cu, Zn­SOD activity was significantly higher in P5B group than in high fat control group, while GPx activity was not changed by any supplementations. Taken together, mixture 5B and 5C showed very strong antioxidative effects both in vitro and in vivo. Therefore, the ingredient Korean foods of 5B and 5C could be recommended to take a lot together for prevention from age-related chronic diseases.

Effects of Dandelion (Teraxacum platycarpum) with Various Extracting Method on Antioxidative Capacity, Lipid Metabolism in Diet-induced Obese Rats (초음파추출과 열수추출에 의한 민들레의 항산화 및 지질강하 효과)

  • This study was conducted to investigate the effects of dandelion (Teraxacum platycarpum) extracts obtained by only water and with ultrasonification on antioxidative system and lipid metabolism in high cholesterol-fed rats. Five groups of rats were given high cholesterol diets for 8 weeks. The control group received without dandelion extracts and the other four groups received with one of dandelion extracts for 4 weeks respectively ; TP-N-1(100 mg/kg/day of Teraxacum platycarpum water extract), TP-N-2(200 mg/kg/day of Teraxacum platycarpum water extract), TP-S-1(100 mg/kg/day of Teraxacum platycarpum water-ultrasonification extract), TP-S-2(200 mg/kg/day of Teraxacum platycarpum water-ultrasonification extract). The results are summarized as follows; The hepatic and plasma TBARS levels significantly decreased in the dandelion extracts groups compared to those of no treatment group. Especially the group TP-N-2 was comparatively best among those. TP-N-2 groups had significantly higher levels of glutathione peroxidase (GSH-Px) and catalase activities. There was no significant difference between dandelion extracts groups and no treatment group in SOD levels. In plasma triglyceride level, plasma FFA level, TP-S-2 group had significantly lower levels than that of the other groups. In plasma glucose levels, dandelion extracts group were similar to those of normal rats. Plasma total cholesterol levels significantly decreased in the TP-S-2 group compared to those of the other groups. HDL levels were also significantly higher than those of the other groups. Compared with those of no treatment group, dandelion extract groups had significantly higher levels of LDL. In liver total cholesterol level, TP-S-2 groups had significantly lower levels than that of the other groups. Compared with those of no treatment group, dandelion extracts groups had significantly lower levels of liver triglyceride, but especially the TP-S-2 group showed comparatively the best significant effect among those. TBARS, triglyceride, LDL, FFA levels significantly decreased in TP-S-2 groups compared to the other four groups. HDL levels was also significantly higher than the other four groups. According to the above result, it could be suggested that ultrasonic extraction have the upper hand in lipid metabolism and water extraction have the advantage of antioxidative system.

  • The effects of spirulina-added salad dressing on lipid profiles and antioxidant biomarkers such as total glutathionine, TBARS value, carbonyl value, GPx, GR, SOD and paraoxonase activity in plasma or liver of mice were evaluated Sixteen male ICR mice weighing 20$\pm$2 g were divided into two groups and fed low fat ($5\%$ fat) diet (low fat control: LFC) and low fat control plus dressing diet (LFD) for eight weeks. Body weight, tissue weights of liver, heart and kidney, and the distribution of body fat deposition were not significantly different between two groups. Also, the profile of TG, TC, LDL and HDL cholesterol were similar between two groups. The DNA damage was determined using the comet assay (single cell gel assay) with alkaline electrophoresis and quantified by measuring tail length (TL). Spirulina salad dressing consumption resulted in significant decrease in lymphocyte DNA damage expressed by TL (LFC: $28.8{\mu}m$, LFD: $20.3{\mu}m$). Additionally, salad dressing consumption for 8 wks decreased the lipid peroxidation assayed by TBARS to $12.6\%$ compared with the control. The levels of antioxidant vitamins such as $\beta$-carotene were significantly higher in plasma of LFD group than those in LFC group based on HPLC method This study shows that spirulina-added salad dressing exerts degenerative disease-protective effects on oxidative DNA damage and lipid peroxidation possibly via a free radical levels.

Effects of Coriandrum sativum L. on Lipid Contents in Streptozotocin-Induced Diabetic Rats (식이내 고수(Coriandrum sativum L.)의 첨가가 당뇨성 흰쥐의 혈장과 간의 지질함량에 미치는 영향)

  • This study was designed to investigate the effects of Coriandrum sativum L. on lipid metabolism in nondiaetic and stretozotocin-induced diabetic.Thirty two mael Sprague-Dawley rats weighting 108$\pm$13 g were divided into four groups as follow: control (C), leaf(L), seed (S) and root (R) groups. For four weeks of experimental periods, the C group were fed basal diet and the L, S and R groups were fed Coriandru sativum L. diets containing 5% dried leaf, seed and root of Coriandrum sativum L, respectively. diet intakes, weight gain and food efficiency were not significantly different among the four groups. The liver and heart weights of L, s and R groups were significantly different amount the control group. The levels of plasma insulin was significantly increased but the plasma glucose levels was not decreased in all Coriandrum group. The levels of plasma lipids was not significantly different among the four groups. the levels of plasma total cholesterol was significantly decreased in R groups. The levels of plasma and liver TBARS values were significantly decreased in R groups. The levels of plasma vitamin E were tendency to decrease in all coriandum groups. These results indicate feeding diet with coriander did not show the hypolipidemic effect but show the antioxidative effect.

Effects of Korean Wheat on LDL Oxidation and Atherosclerosis in Cholesterol-Fed Rabbits (고콜레스테롤혈증 유발 가토를 이용한 우리밀의 LDL산화 및 동맥경화 예방 효과)

  • Numerous studies have suggested the involvement of oxidative processes in the pathogenesis of atherosclerosis and especially of oxidized low density lipoproteins (LDL). We studied the effect of Korean wheat on the oxidizability of LDL and the development of experimental atherosclerosis in rabbits on cholesterol diet. For the approach of the aim, antioxidative activity of wheat extracts against oxidation of LDL was investigated. The antiatherogenic effect of wheat was studied against Newzealand whithe (NZW) rabbits fed containing the wheat powder for 12 weeks. PBS extracts of Korean and imported wheats decreased LDL oxidation induced by copper ion by 62.3%, 52.6% respectively in comparison with control. Liver thiobarbituric acid reactive substance (TBARS ) levels of rabbits were significantly lowered in Korean wheat fed group (0.397$\pm$ 0.029 nmol MDA/mg protein, p<0.05) compared to control (0.496$\pm$0.021 nmol MDA/mg protein) and imported wheat group (0.478$\pm$0.019 nmol MDA/mg protein). TBARS levels of kidney showed no significant difference among treatment groups. The aorta of cholesterol-fed rabbits showed typical atherosclerotic lesions when compared with the controls, but the microscopic differences between groups was not clear. The present study suggests that Korean wheat may have higher antioxidative capacity than imported wheat and have more beneficial effects for the treatment of disease by inhibiting the oxidation of LDL.

Effect of Long Term Buchu (Chinese chives) Diet on Antioxidative System of ICR Mice (장기간의 부추식이가 ICR 마우스의 항산화시스템에 미치는 영향)

  • To evaluate the antioxidative and antiaging effects of buchu in vivo system, 2% or 5% buchu diets were fed to ICR mice for 13 months and lipid peroxidation, protein oxidation, activities of antioxidative enzymes and total glutathione content on liver were measured. Hepatic TBARS contents did not show differences among diet groups, while buchu diet suppressed the protein oxidation significantly. SOD activities of control diet group decreased slowly after 7 month but buchu diet increased its activities steeply for first 3 month and continued to increase twice or three times higher than control diet during 13 month. While GSH-Px activities of control diet group were increased slightly with age, buchu diet increased its activities twice or three times higher than control. While catalase activities of control diet group were almost not changed with age, buchu diet increased its activities in both 2% and 5% diet groups. Total hepatic glutathione contents were gradually increased with age, while buchu diets In-creased its contents remarkably. According to this study, many antioxidative materials and sulfides compounds containing buchu seems to protect antioxidative systems on ICR mice.

Effects of Dietary Supplementation of Pine Needle Powder on Carcass Characteristics and Blood Cholesterol Contents of Broiler Chicken (사료내솔잎분말첨가급여가육계의도체특성및혈액콜레스테롤함량에미치는영향)

  • This study was conducted to investigate the effects of supplementation diets of pine needle powder on performance, proximate composition, carcass characteristics, blood cholesterol concentration, TBARS (thiobarbituric acid reactive substance), WHC (water holding capacity), and shear force of chicken meat. Broiler chicks were fed diets for five weeks containing 0% pine needle powder (Control), 0.3% pine needle powder (T1), 0.6% pine needle powder (T2), and 0.9% pine needle powder (T3). Performance was no significant difference among treatment groups, but mortality was decreased in diets by the supplementation of pine needle powder than that of T1 and control. The moisture, crude protein, crude fat, and crude ash were no significant difference. The liver weight and abdominal fat was significantly decreased by the supplementation of pine needle powder compare to the T1 and control (P<0.05). The total cholesterol and LDL-cholesterol concentration of control was higher than treatment groups, and the HDL-cholesterol concentration and glucose of control was lower than treatment groups (P<0.05). The TBARS value was significantly decreased by the supplementation of pine needle compared to the control (P<0.05). In conclusion, these data indicate that 0.6% pine needle powder supplementation (T2) was most effective in decreasing mortality, total cholesterol and LDL-cholesterol and increasing HDL-cholesterol concentration.

The Effect of Black Garlic Extract on Lipid Metabolism in Restraint Stressed Rats (구속스트레스를 가한 흰쥐의 지질 대사에 대한 흑마늘 추출물의 효과)

  • In order to determine the effective concentration, 2, 7, and 14 brix black garlic extracts (BGE) were fed to restraint stressed rats and the lipid metabolism was analyzed. The dietary intake of the 14 brix BGE-fed group was $547.64{\pm}13.99$ g/4 weeks and it was significantly higher than that of the control group. The weight of all organs from the all BGE-fed groups showed no significant differences. While BGE concentration increased, BGE-fed groups tend to have low content of blood glucose. It was the lowest in the 14 brix BGE-fed group ($121.32{\pm}3.50$ mg/dl). Total cholesterol content was $54.54{\pm}1.63{\sim}65.84{\pm}2.61$ mg/dl. Total cholesterol content of the 14 brix BGE-fed group was the lowest and, in addition, lower than the normal group. Triglyceride content of serum showed the same tendency. HDL-cholesterol concentrations of the restraint stressed control (RSC) group were significantly higher than those of 7 and 14 brix BGE-fed groups. LDL and VLDL-cholesterol contents showed contrary results. GOT, GTP, and ALP activity of serum were lower when BGE concentration was higher. It is expected that BGE has a liver protective activity. In the liver, total lipids and triglyceride contents were significantly lower in BGE-fed groups than in the ISC group. However, total cholesterol content showed no significant difference between groups. TBARS content in liver of the 2 brix BGE-fed group was not significantly different than control, while 7 and 14 brix BGE-fed groups were lower than control. Glycogen content in the liver was only significantly lower in the 14 brix BGE-fed group than in the RSC group. Findings indicate that feed of BGE over 7 brix contributes to resolving the restraint-stressed rats by improving the lipid metabolism and liver protective activity.

Effect of a Combined Extract of Orostachys japonicus with Medicinal Plants on the Lipid Composition of the Liver and Kidney from Streptozotocin-induced Diabetic Rats (와송과 생약재 복합물이 Streptozotocin 유발 당뇨쥐의 간장 및 신장 조직의 지질성분에 미치는 영향)

  • This study examined the biological activity and synergistic effects of an extract of Wa-song ($Orostachys$ $japonicus$, OE), a medicinal plant mixture (MPE) and a combination of both at different ratios (1:1, OMPE-1 and 3:1, OMPE-3). Extracts of the medicinal plants mixture were comprised of Baekbokyung, Changchul and Sa-in at the same ratio. The antioxidant activity of the extracts and their complex were tested $in$ $vitro$. The $in$ $vivo$ antioxidant activity was also analyzed by examining the lipid composition in the liver and kidney of streptozotocin (STZ)-induced diabetes rats. The nitric oxide radical scavenging activity was more than 50% in OMPE-3 at a 1,000 ${\mu}g$/mL concentration. Regarding metal ions, such as $Fe^{2+}$ and $Cu^{2+}$, the antioxidant activity of OMPE-1 and OMPE-3 was higher than that of OE and MPE. OMPE-1 and 3 had higher activity on $Cu^{2+}$ ions than $Fe^{2+}$ ions. The ${\alpha}$-glucosidase inhibition activity of the OE extract was higher than that of MPE and OMPE-1 but the relative activity of OMPE-3 was significantly higher than the others. Freeze-dried MPE, OMPE-1 and OMPE-3 were added to the diet at a level of 1% given to STZ induced diabetes rats for 4 weeks. The OMPE-1 and OMPE-3 administered groups showed significant decreases in the total lipid, total cholesterol and triglyceride levels in the liver and kidney. In these groups, the glycogen accumulation level of the liver was increased significantly. The content of thiobarbituric acid reactive substances (TBARS) in the liver and kidney was decreased but the DPPH radical scavenging activity was increased. These results suggest that Wa-song extract exhibits antioxidant and antidiabetic activity, which are enhanced by a complex with a medical plants extract.
