Effects of Culture Conditions on Organogenesis in Gladiolus 'Topaz' Callus (글라디올러스 'Topaz' 캘러스의 기관형성에 미치는 배양 조건의 영향)

  • This study was carried out to establish improved techniques on organogenesis from callus culture of Gladiolus. Organogenesis from the callus was effective in the half strength of MS solid medium without 2,4-D at 15 $^{\circ}C$ under 24 hours of daylength. Formation of adventitious root was most effective in the liquid shaking culture, and adventitious shoot induction was effective in the liquid stationary culture. From these results, we could find optimal culture conditions for redifferentiation from callus, in addition, liquid shaking culture revealed as more useful when compared with that of solid culture method for the redifferentiation of callus in Gladiolus `Topaz'.

  • This study was performed to enhance the proliferation rate of Gladiolus 'Topaz' callus. The callus was induced from the cermet tissue explants on MS solid medium with 10 mg/L 2,4-D. In the case of liquid shaking culture, proliferation of the callus was effective in MS medium with 0.05 mg/L 2,4-D at 2$0^{\circ}C$ under 16 hours daylength and in a 100 mL Erlenmeyer flask containing 20 mL of the liquid medium and at 75 rpm in rotation speed of the horizontal shaking culture. Furthermore the callus was also able to be subcultured in the same liquid medium.

  • Effects of shaking, medium consistency and anther density on polyhaploid production in two wheat cultivars, Pavon and Chris, were studied using a modified 85D12 medium. Pavon produced more calli in shaking and more albino plants tban Chris. However, Chris produced threefold more green plants than Pavon in non-shaking treatment. More calli and green plants were derived from non-shaking treatment than those from shaking treatment. Anthers were cultured on both liquid and semi-solid 85D12 media, using two anther densities, 48 and 96 anthers per plate. Although Pavon generally produced more calli and albino plants than Chris, Chris produced more green plants than Pavon. More green plants were derived from semi-solid medium than those from liquid medium. A factor that may affect plant regeneration from anthers is the length of time on initiation medium. Most of the calli for both genotypes were transferred during the first two time periods. Fertility, as measured by seed set, was determined for all surviving regenerated plants. About 24% of Chris and Pavon anther-derived green plants in the experiment of medium consistency and anther density produced seed.

Improvement of Liquid Culture Methods of Paecilomyces japonica (눈꽃동충하초균의 액체배양법 개선)

  • This study was carried out to improve the liquid culture methods of Paecilomyces japonica. The results show that the size of granular mycelium is smaller when the shaking speed is increased. Especially, the granular mycelium is the smallest at the shaking speed of 150rpm under the photoperiod of 12L-12D. Dry weight of mycelium was averagely 1.216 g in the Sikworm larva (SL) medium, and the weight was 2 times heavier than in the Potato dextrose (PD) medium. By adding 6 g of 6 mmbeads in the SL medium, the dry weight is increased to 1.332 g. The optimal addition of silkworm larval powder to the culture medium for gest harvest was 1.360$\pm$0.67 g in dry weight.

Extraction of β-glucosidase from Bagasse Fermented by Mixed Culture under Solid State Fermentation

  • Various parameters such as solvent selection, concentration, solid/liquid ratio, soaking time, temperature, stationary, shaking conditions, and repeated extractions were investigated in order to determine the optimum extraction conditions of ${\beta}$-glucosidase from bagasse fermented by mixed culture of Aspergillus niger NRC 7A and Aspergillus oryzae NRRL 447. Among various solvents tested, non ionic detergents gave the best results than the inorganic or organic salt solutions and distilled water. The optimum conditions for extraction of ${\beta}$-glucosidase were 30 min soaking time at $40^{\circ}C$ under shaking condition at 150 rpm, with solid/liquid ratio 1:15 (w/v), which yielded $2882.74{\pm}95.52U/g$ fermented culture (g fc) of enzyme activity. With repeated washes under the above optimum conditions, the results showed that enzyme extracted in the $1^{st}$ and $2^{nd}$ washes represents about 90% of the total activity.

  • Liquid shaking culture was conducted to investigate the proper culture conditions for the micropropagation of high quality lily using bulblets (3 mm in diameter) obtained from small scale culture. The combinations of 9% sucrose and 10 mM nitrogen or 6% sucrose and 20 mM $NH_4NO_3$ were effective on the growth and weight of micro-bulbs. However, the number of new bulbs was the highest when 20 to 40 mM $NH_4NO_3$ and 3% sucrose were added to the MS medium. The total fresh weight was increased effectively in MS medium supplemented with BA 0.2 mg/L alone under $60\;{\mu}mol{\cdot}m^2{\cdot}sec^{-1}$ intensity. Also bulblet weight was increased at $60\;{\mu}mol{\cdot}m^2{\cdot}sec^{-1}$ intensity, regardless of BA concentrations (0.2 and 2 mg/L) in the medium. The proper culture period of bulblet was about 2 month in liquid shaking culture.

  • Shake mixing has been widely used in cell culture. The mixing performance for shake mixing, however, has not been reported quantitatively. The critical circulating frequency and the power consumption for complete suspension of particles, based on the definition of Zwietering, were measured in a shaking vessel containing a solid-liquid system. The critical suspension frequency was correlated by the equation from Baldi's particle suspension model modified with the physical properties of the particles. Critical suspension frequency was correlated as following ; $$N_{JS}={\frac{0.58\;d{_p}^{0.06}(g{\Delta}{\rho}/{\rho}_L)^{0.004}X^{0.03}}{D^{0.35}d^{0.17}{\upsilon}^{0.04}}}$$ The power consumption at the critical suspension condition in the shaking vessel was less than that in an agitated vessel with impeller.

Inhibition of Lipid Autoxidation by the Extract of the Submerged-liquid Culture of Mushrooms in the Medium Containing Mulberry Tree Powders (뽕나무 첨가 배지에서 배양한 버섯균사체 배양물의 자동산화 억제 효과)

  • Effect of mulberry tree powders on the antioxidant activity of submerged -liquid culture of mushrooms was investigated. Agaricus blazei (AB), Hericicum erinacium (HE), Pleurotus ostreatus (PO), Phellinus linteu (PL) and Paecilomyces japonicus (PJ) were cultured in a shaking incubator (200 rpm, $25^{\circ}C$) for 7 days in culture media: 1) basal medium (BM) and 2) BM+1% mulberry tree powders (BMM). Hot water extracts from the submerged-liquid cultures of mushrooms and BMM were freeze-dried for the measurement of antioxidant activity, of which reaction mixture (25 mL: 10 mL of 0.2 M sodium phosphate buffer, pH 8.0; 4.5 mL distilled water; and 10.5 mL ethanol) contained 275 $\mu$mol linoleic acid and one mg test sample. The reaction mixture was incubated in a shaking incubator (200 rpm, 4$0^{\circ}C$) for 16 days. Peroxide value (POV) was measured for a period of over 16 days, and malonaldehyde (MA) was determined only for samples from the day 16 of incubation. Mycelial weight of mushroom strains cultured in BMM was greater than BM. The antioxidant activities of AB-cultured in BW (AB-BMM) and HE-cultured in BMM (HE-BMM) were superior to those of other mushroom strains-cultured in BMM or BM and of BMM. These results suggest that mulberry tree powders enhance the antioxidant activity of submerged-liquid culture of mushroom strains. The AB-BMM and HE-BMM were the most active cultures.

  • An experiment was performed to investigate the combined effect of preservatives and the synergistic effect of sodium chloride to them on the inhibition of bacterial growth. Escherichia coli and Salmonella typhimurium were cultured with or without shaking in liquid media (pH 6) of tryptone-glucose-yeast extract or tryptic soy broth which contained 0.1% potassium sorbate and/or 0.03% sodium benzoate, equivalent to half of the maximum permissible levels, respectively. The growth of E. coli was more inhibited with one or both of the two preservatives by shaking culture than by non-shaking culture. For S. typhimurium the single treatment of the preservatives did not show inhibitory effect whereas the combined treatment of them showed bacteriostatic effect in shaking culture and a prolongation of lag phase in non-shaking culture. Addition of 2% sodium chloride to either potassium sorbate or potassium sorbate plus sodium benzoate remarkably increased the growth inhibition of E. coli for non-shaking cultivation but no effect observed for shaking cultivation. S. typhimurium was more sensitive to the addition of sodium chloride than E. coli in both shaking and non-shaking culture to show lower viable cell counts than initial numbers.

Pigment Production and Color Diference of Liquid Beni-koji under Submerged Cultural Conditions (액체 홍국의 배양조건에 따른 색소생산과 색조의 변화)

  • Mycelial growth, color difference and productivity of red pigment of beni-koji by Monascus anka KCCM 11832 were examined with respect to it's pigment in submerged culture with various medium and culture conditions. Shaking incubation was more promoted mycelial growth and the production of pigments than that for non-shaking incubation, and red color became ten times deeper. The production of red pigment was the highest when incubated at 25$\circ$C for 7 days in pH 6.0, but mycelial growth was showed the highest at 32.5$\circ$C. The levels of carbon and nitrogen source for maximum red pigment production were 2% rice powder and 0.05% peptone, respectively and the level of peptone for maximum pigment production was lower than that for maximum mycelial growth. Among pigmentation promoting agents tested, MgSO$_{4}$, was found to be suitable for the production of red pigment, and the optimum level was 0.1%.
