Therapeutic Effects of Bergenin and Acetylbergenin on Galactosamine-induced Hepatotoxicity in Rats (Bergenin 및 Acetylbergenin의 Galactosamine 유발 간독성에 대한 치료효과)

  • The hepatoprotective effects of bergenin and its derivative, acetylbergenin, were evaluated against D-galactosamine-induced liver damage in rats. Bergenin is a C-glucoside of 4-O-methyl gallic acid that has been isolated from the cortex of Mallotus japonicus (Euphorbiaceae). Acetylbergenin was synthesized from acetylation of bergenin to increase lipophilic and physiological activities. Bergenin (50, 100 and 200 mg/kg) and acetylbergenin (25, 50 and 100 mg/kg) were administered orally once daily for successive 5 days after the injection of galactosamine (400 mg/kg, i.p.), respectively. The substantially elevated serum enzyme activities of alanine/aspartate aminotransferase, sorbitol dehydrogenase and ${\gamma}-glutamyltransferase$ due to galactosamine treatment were dose-dependently restored towards normalization by post-treatment with bergenin and acetylbergenin. Bergenin and acetylbergenin also significantly prevented the elevation of hepatic malondialdehyde formation and depletion of reduced glutathione content induced by galactosamine in a dose-dependent manner. In addition, the decreased activities of glutathione S-transferase and glutathione reductase were restored towards normalization. These results suggest that effects of bergenin and acetylbergenin may be related to complex mechanisms that involve prevention of lipid peroxidation and preservation of hepatic glutathione. The results of this study clearly indicate that bergenin and acetylbergenin have potent hepatotherapeutic action against galactosamine-induced hepatotoxicity in rats, and lipophilic acetylbergenin is more active in the antihepatotoxic effects against galactosamine than much less lipophilic bergenin.

  • It was possible to produce W/O/W one step multiple emulsion on the system which satisfied following conditions. 1. 1-5% of hydrophilic liquid surfactant over HLB20 and lipophilic liquid surfactant which has HLB 3∼5 2. Non wax copolymers as oil thickener 3. More than 0.5% of carbomer as aqueous thickener 4. The manufacturing process which neutralize the dispersed carbomer (2.0% in water), after emulsifying. For the selective addition into inner and outer aqueous phase, we melted the glucose in water before emulsifying. Using an Anthrone analysis method, we analyzed the encapsulation yield of glucose in inner water phase. It was possible to raise the water encapsulation yield of the multiple emulsion through the following conditions. 1. Using of anionic hydrophilic surfactant(HLB 40) and lipophilic surfactant (HLB 3∼5) 2. Controlling the ratio of hydrophilic surfactant and lipophilic surfactant 3. Strengthening interface with increase of non wax oil thickener. When the separated adding process of glucose was adopted, approximately 85% of glucose was added selectively within inner aqueous phase.

Effects of Lipophilic Fraction from Korean Red Ginseng on Platelet Aggregation and Blood Coagulation in Rats Fed with Corn Oil and Beef-tallow Diet (옥수수기름 및 우지 식이에 의한 혈소판 응집 반응과 혈액응고에 있어서 홍삼 지용성성분이 미치는 영향)

  • This study was investigated to find the effects of petroleum ether extract (Lipophilic fraction) from Korean red ginseng on platelet aggregation and thrombin time of the plasma in two groups of the experimental rats. One group of rats were fed with 15% corn oil (15%kg-diet) containing a number of 18 : 2 (linoleic acid) or 15% beef-tallow (15%/kg-diet) containing saturated fatty acids for 3 weeks, and were followed by feeding the petroleum ether extract (25 mg/kg-diet) for 3 weeks. The other group of rats (control group) were fed with 15% corn oil or 15% beef-tallow for 6 weeks. The platelet aggregation induced by thrombin and collagen was significantly inhibited and the thrombin time was prolonged in the 15% corn oil plus petroleum ether extract administrated group than in the 15% corn oil administrated group. And the same results were shown in the 15% beef-tallow plus petroleum ether extract administrated group. These results suggest that the petroleum ether extract from Korean red ginseng may have the beneficial effects on the inhibition of the platelet aggregation and the inhibition of blood coagulation induced by dietary fats.

  • In manufacturing emulsion lipstick in the past, heat and emulsifyoil phase made with oil, wax and lipophilic emulsifier with water phase together; and then mold above-mentioned materials, so-called lipophilic emulsified lipstick production method has been used. However it is hard to manufacture products using the above said method and furthermore, weak in moisturizing effect, lip care and stability. In this paper, I will discuss about multi-emulsion lipstick complementing already existing emulsion lipstick in terms of stability, giving moisturing effect of lip, and outstanding protection effectiveness of skin by safely gelatinating the state between amphiphilic lipid and water in stable.

  • Lipophilic Fraction (LF) from Panax ginseng C.A. Meyer strongly inhibited human platelet aggregations induced by thrombin. When platelets were prelabeled with 5-hydroxy[G-$^3$H]-tryptamine (serotonin) and then stimulated by thrombin, LF inhibited the release of serotonin in a dose-dependent manner. From this result, we suggest that LF have antiplatelet and antimigraine functions by inhibiting the release of serotonin.

  • Acyclic polyether dicarboxylic acid have been studied as metal cation carriers in a bulk liquid membrane system. The proton-ionizable ligands feature allows the coupling of a cation transport to reverse proton transport. This feature offers promise for the effective separation and concentration of metal cations with the metal cation transport being driven by a pH gradient. Metal cation transport increased regularly with increasing hydroxide($H^-$) concentration of source phase and with proton($H^+$) concentration of receiving phase. Competitive transport by the acyclic polyether dicarboxylic acids is selective for calcium ion over other alkaline-earth cations.

  • A series of thioureas were synthesized as inhibitors of NO production in lipopolysaccharide-activated macrophages. We investigated the effect of lipophilic moiety and N-substituents of the thioureas on the activity. Phenoxazine and carbazole-containing derivatives revealed higher activity than indole-containing thioureas. The appropriate spacer between lipophilic tail and thiourea head and methyl substituent at N3 position of thiourea brought beneficial effect on the inhibition of NO production. Among prepared compounds, phenoxazine-containing derivative 2a was the most potent with $2.32 {\mu}M$ of $IC_{50}$ value. RT-PCR analysis suggested that the prepared thioureas inhibited NO production through the suppression of iNOS mRNA expression.
