마늘성분의 산화방지작용에 관한 연구 - 제1보 전자공여능 및 과산화지질생성억제효과에 미치는 영향 (Studies on antioxidative action of Garlic components isolated from garlic - Part 1. Effects of garlic components on electron donating ability and inhibitory effect of lipoperoxide formation)

  • In order to study antioxidative acton of garlic, alliin, scordinin, garlic oil, ethanol fraction and non-kaolin fraction which have been discovered from garlic until now were extracted and isolated, and each fraction was utilized as the experimental materials. Antioxidative action of each fraction was compared through the in vitro and in vivo experiments. Electron donatingability on $\alpha$, $\alpha$-diphenyl-$\beta$picrylhydra-$\chi$yl, the inhibitory effect of lipoperoxide formation by TBA and peroxide value were measured and analyzed. RESULTS : 1. When observed antioxidative ability by EDA value, ethanol fraction of garlic components showed the strongest reaction as 15.25. 2. In vitro experiment with TBA value, garlic oil, alliin and ethanol fracton showed distinctive effect on inhibitory effect of lipoperoxide formation. 3. Comparing with the inhibitory effect of lipoperoxide formation with TBA value in vivo, the ethanol fraction was the most effective in the blood or liver by intraperitoneal administration, whereas the ethanol fraction in the blood and non-kaolin fraction in the liver was most effective each other by orally administration. 4. In vitro experiment with peroxide value, garlic oil was distinctive effect on the inhivitory effect of lipoperoxide formation, which was a similar to the trend of TBA value in vitro. 5. Examining the induction time for the first period of lipoperoxide formation in vitro, garlix oil, ethanol fraction and alliin were effective, which was a similar to the trend of TBA value and peroxide value in vitro.

  • In Order to study antioxidative action of water soluble fractions which had been steamed according to time as 15, 30, 60 and 120 minutes, they were compared through the inhibitory effect of lipoperoxide formation by TBA, peroxide value and induction time for the first period of lipoperoxide formation. Results are obtained as follows: 1. In vivo experiment with TBA value, water soluble fractions showed aninhibitory effect of lipoperoxide formation. 2. Comparing with the inhibitory effect of lipoperoxide formation with TBA value in vivo, water soluble fractions which had been steamed for 30 and 60 minutes and those from fresh garlic proved effective in the blood by intraperitoneal administration. But in the liver all of water soluble fractions showed distinctive effect as in the case of fresh garlic. 3. Effect of water soluble fractions which had been steamed for 15, 30 and 60 minutes showed a distinctive effect, water soluble fraction which had been steamed for 120 minutes and that from fresh garlic also showed effective in the blood as compared with control. 4. In oral administration, water soluble fraction of fresh garlic was the most effective in the liver. 5. In vitro experiment with peroxide value, water soluble fractions which had been steamed for 60 and 120 minutes were effective, all of water soluble fractions from steamed garlic were more effective than fresh garlic. 6. In Examining the induction time for the first period of lipoperoxide formation in vitro, water soluble fractions steamed for 30, 60 and 120 minutes were effective. Other fractions were also more effective than control.

인삼(人蔘)사포닌의 항산화작용(抗酸化作用)에 대한 지용성(脂溶性) 비타민의 첨가효과(添加效果) (Study on the Additional Effect of Fat-Soluble Vitamins to Antioxidative Action of Ginseng Saponin)

  • 1. DPPH에 대한 전자공여능은 vitamin A와 D가 인삼사포닌 단독보다 높아 지용성 비타민 첨가에 의한 상승효과가 인정되었다. 2. 과산화지질 생성초기의 유도기간은 vitamin A, D를 첨가했을 때 대조군이나 인삼 사포닌보다 효과적으로 유도기간을 연장시킴을 알 수 있었다. 3. in vitro에서 과산화지질 생성억제작용을 TBA value로 조사한 결과 vitamin E>A>D의 순으로 지용성 비타민의 첨가효과가 인정되었다. 4. in vivo실험에서 인삼 사포닌 및 지용성 비타민 첨가 투여에 의한 rat의 체중 및 간장, 신장의 장기중량 변화는 지용성 비타민의 첨가가 체중증가에 효과적임을 알 수 있었고, 간장은 비타민 첨가군에서 다소 중량증가가 인정되었으나 신장의 중량변화에는 큰 유의성을 발견 할 수 없었다. 5. in vivo실험에서 인삼 사포닌 및 지용성 비타민 첨가 투여에 의한 rat의 혈액에서의 과산화지질 생성억제작용은 vitamin E>D>A의 순으로 나타났고, 간장에서는 vitamin A와 E첨가군에서 효과적임이 인정되었다.

한국인 성인 남성에서 혈청 비타민 E 및 과산화지질과 혈청지질과의 상관성 분석 (Relation of Serum Vitamin E and Lipoperoxide Levels with Serum Lipid Status in Korean Men)

  • Secrum lipid and vitamin E levels were determined and smoking, alcohol drinking and exercise habits were asked in 357 healthy male subjects aged 49.4$\pm$6.7 years in Taegu. Average serum levels of total cholesterol, HDL-cholesterol and triglyceride(TG) were 189$\pm$43mg/이, 42$\pm$13mg/dl and 136$\pm$73mg/이, respectively. Serum level of lipid peroxide measured as thiobarbituric acid reactive substances(TBARS) of the subjects was 2.01$\pm$0.73MDA nmoles/ml and that of $\alpha$-tocopherol was 9.53$\pm$3.14ug/ml. The correlation coefficients between $\alpha$-tocopherol and serum lipids were 0.3631 for triglyceride, 0.2993 for cholesterol, and 0.3025 for total lipid. Heavy smokers who smoked more than 20 cigarettes a day had higher levels of TG and TBARS than those who smoke less. Alcohol drinkers had higher levels of TG and TBARS than nondrinkers. Vitamin E level(per ml serum)was significantly higher in the heavy smokers and drinkers, which was reflected by the higher level of serum lipid. When the level of $\alpha$-tocopherol was expressed as $\alpha$-tocopherol/triglyceride, it was negatively correlated with serum TBARS level and was lower in the heavy smokers than in the moderate smokers. It is concluded that vitamin E level as $\alpha$-tocopherol/triglyceride would be better compared to $\alpha$-tocopherol/total cholesterol or $\alpha$-tocopherol/total lipid for the evaluation of vitamin E status in Korean men.

대구지역 중년 남성의 혈청 지질과 혈청 과산화지질의 관련 인자 연구 (Serum Lipid and Lipoperoxide levels and Their Related Factors in Middle-Aged Men in Teagu)

  • The present study was aimed to examine various factors related to serum total cholesterol, triglyceride, atherogenic index, lipoperoxides and vitamin E in 174 middle-aged men residing in Taegu. Body mass index was positively correlated with serum triglyceride, and waist-hip circumference ratio was positively correlated with serum total cholesterol, triglyceride, atherogenic index and vitamin E concentration. Total cholesterol, triglyceride and atherogenic index of the subjects who smoked more than 20 cigarettes per day were significantly higher than those with smoking less or non-smokers. drinking alcohol more than 5 times per week showed higher atherogenic index than drinking less frequently of non-drinking. Subjects doing exercise had higher HDL-cholesterol level than those with no exercise. The intakes of all nutrients were above 90% of RDA and the subjects with good food habit had lower of total cholesterol, triglyceride, atherogenic index and lipoperoxides. The study indicates that avoiding heavy smoking and drinking as well as obesity and keeping regular exercise, good food habit and diversity of foods will reduce risk of cardiovascular diseases in middle-aged men.

  • This study was undertaken to elucidate the role of L-carnitine on cardiotoxicity induced by doxorubicin in mice. We designed to investigate both lipoperoxidation and histopathology in experimental group. The results obtained from prior with concurrent treatment of L-carnitine showed as follows; 1) Combination of L-carnitine showed significant inhibition of lipoperoxide in liver than heart. 2) By means of electron microscopy, we obtained histological evidence that doxorubicin-induced cardiomyopathy, is prevented by L-carnitine.

오가피 성분의 항돌연변이원성에 관한 연구 (Studies on the Antimutagenic Effect of Acanthopanax sessiliflorum Components)

  • Effects of acanthopanax cortex extracts on glutathion S-transferase (GST), glutathion peroxidase (GSH-px) and superoxide dismutase (SOD) activities related to 7,12-dimethyl-benz(a)anthracene(DMBA) metabolism and on DMBA-induced mutagenicity were investigated in this study. From the comparative study of three extracts, it was found that butanol extract was more potent than other extracts in increment of GST, GSH-px and SOD activities and in inhibitory effects of lipoperoxide formation of liver. Also ether and butanol extracts inhibited DMBA-induced mutagenicity, showing 33% to 36% of inhibition at maximum, when ether and butanol extracts were administered to rats intraperitoneally.

  • In order to study the antioxidative action of the effectient garlic components, peroxidase and superoxide dismutase activity were compared through the in vitro and in vivo experiments. RESULTS : 1. Observing the effects on peroxidase activity of efficient components in vitro, garlic oil, alliin and ethanol fraction showed effective, which was similar to the trend of TBA value, peroxide value and induction time for the first period of lipoperoxide formation in vitro. 2. In vivo experiment with peroxidase activity, the ethanol fraction and garlic oil were effective when intraperitoneally administered as well as orally administered. 3. Considering the superoxide disutase activity in vitro, the garlic oil, alliin and ethanol fraction were effective in efficient components. But non-kaolin fraction inhibited the activity on the contrary. 4. In terms of the efefcts on superoxide dismutase activity in vivo, alliin and garlic oil wee effective in intraperitoneal adminstraton and the ethanol fraction and alliin in oral administration.

  • 홍삼(紅蔘)과 백삼(白蔘) 항산화작용(抗酸化作用)에 의한 노화억제작용(老化抑制作用)을 구명(究明)하기 위하여 80% ethanol extract 및 수포화(水飽和)부탄올로 이행(移行)한 saponin을 시료(試料)로 하여 in vitro 및 in vivo 실험(實驗)을 통하여 홍삼(紅蔘)과 백삼(白蔘)의 노화억제작용(老化抑制作用)을 비교(比較)하였다. DPPH에 대한 전자공여능(電子供與能)(EDA), TBA value 및 peroxide value에 의한 과산화지질생성억제작용(過酸化脂質生成抑制作用), superoxide dismutase 및 peroxidase의 활성측정(活性測定)등으로 홍삼(紅蔘)과 백삼(白蔘)의 노화억제작용(老化抑制作用)을 비교(比較)한 결과(結果), 홍삼(紅蔘)은 장기간(長期間)에 걸친 연속투여(連續投與)에서 백삼(白蔘)보다 과산화지질생성억제작용(過酸化脂質生成抑制作用). superoxide dismutase 및 peroxidase의 활성(活性)을 유의성(有意性)있게 증가(增加)하였다. 생체내실험(生體內實驗)에서 홍삼(紅蔘)이 백삼(白蔘)보다 노화억제작용(老化抑制作用)이 현저한 것은 증삼(蒸參) 및 가공처리중(加工處理中) saponin pattern의 변화(變化)에 기인(起因)하며 백삼(白蔘)의 PT/PD ratio가 0.401인데 반(反)해 홍삼(紅蔘)의 PT/PD ratio가 0.561로서 triol saponin이 증가(增加)되었기 때문이다.

  • Horseradish peroxidase의 열불활성화에 미치는 몇가지 인자의 영향에 대해 실험하였다. lecithin이 존재하는 용액 속에서 peroxidase를 pH 7.0 $70^{\circ}C$에서 처리한 결과 급격한 열불활성화를 보였고 pH 4.0의 용액에서는 $0^{\circ}C$에 짧은 시간 방했을 때도 급격한 불활성화를 나타내었다. linoleic acid는 산소 존재하에서 처리 했을 때가 질소 존재 하의 처리에서 보다 큰 영향을 보였고 oleic acid의 경우는 산소및 질소 존재 하에서 비슷한 영향을 보였다. lecithin과 linoleic acid의 영향에 대한 결과 각각 heme crevis의 개열 내지는 생성된 lipoperoxide에 의한 불활성화라고 추정하였다.
