Screening of Hyaluronidase Inhibitory and Free Radical Scavenging Activity in vitro of Traditional Herbal Medicine Extracts (생약재 추출물의 hyaluronidase 저해 및 라디칼 소거 활성 검색)

  • For the screening of anti-inflammation and antioxidative activities, ethanolic extract of 40 species of traditional herbal medicines were examined their hyaluronidase inhibitory effect and radical scavenging activity in vitro. From the result of the hyaluronidase inhibitory activity using a Morgan-Elson assay, Astragali Radix, Eucommia Cortex, Schizandrae Fructus, Scutellaria Radix, Acanthopanacis Cortex, Chaenomelis Fructus, Amomum xanthioides Wallich and Moutan Radicis Cortex showed more than 50% hyaluronidase inhibitory effects at the concentration of 10 mg/mL. In the various solvent fractions (n-hexane, chloroform, ethyl acetate, water) prepared from ethanolic extracts, the ethyl acetate fraction of all extracts tested showed strong activity. Antioxidative activity was evaluated by assaying electron-donating ability to DPPH free radical and scavenging of hydroxyl radical (ㆍOH) generated through Fenton reaction, respectively. Rubus coreanus Miq, Moutan Radicis Cortex, Paeoniae Radix Alba, Plantaginis Semen and Sorbus commixta Hedl. showed high activity more than 90%, yet similar activity to $\alpha$-tocopherol and BHA at the concentration of 1 mg/mL in electron donating activity. The scavenging effects of ethanolic extracts on hydroxyl radical were investigated using a 2-deoxyribose oxidation method and tested all extracts showed significant radical scavenging activity. The experiment was also performed to examine whether herbal medicines having significant lipid peroxidation inhibitory activity, Schizandrae-Fructus is the strongest inhibitory activity in both linoleic acid and liposome peroxidation.

The Antioxidant Activity in Extracts of Symphyocladia latiuscula (보라우무 (Symphyocladia latiuscula) 추출물의 항산화활성)

  • The antioxidant activity of Symphyocladia latiuscula was determined by measuring lipid peroxide produced when a mouse liver homogenate was exposed to the air at $37^{\circ}C$, using 2-thiobarbituric acid (TBA) and radical scavenging effect on 1,1 - diphenyl - 2 - picrylhydrazyl (DPPH) radical, and free radical generation inhibition by $ACF_2$(Hepatocyte). The methanol extract of S. latiuscula showed high antioxidant activity. And the methanol extract was fractionated with several solvents. With regard their fractions, the antioxidant activity were in the order of dichloromethane > hexane > butanol > ethyl acetate > water fraction. The dichloromethane fraction showed the strongest radical scavenging activity ($50\%$ inhibitory concentration[$IC_{50}$]=3,14 $\mu$g/ml), and strong inhibitory effect on the lipid peroxidation of the mouse liver homogenate, which was compared with lascorbic acid, inhibition effect was stronger than Lascorbic acid. The methanol extract of S. latiuscula and its dichlromethane soluble fraction also inhibited over $50\%$ at concentration of 0.2 mg/ml and 0.1 mg/ml on free radical generation of hepatocyte ($AC_2F$). While the water fraction was inactive in all the assay for antioxidant activity.

Chemical Composition of Maize Germs Separated in the Manufacture of Cornstarch (옥수수전분 제조공정의 부산물인 maize germ의 지질 및 아미노산 조성)

  • This study was conducted to research the industrial utilizaion of the maize germs separated in the manufacture of the cornstarch. The maize germs contained crude protein (25.3%) and crude fat (19.2%), with the 512 Kcal/100g of determined food energy values. It was found to have the valuable amounts of essential minerals, such as the content of calcium and phosphorus were low (53.0, 12.3mg%), while those of potassium (495.4mg%), magnesium (95.0mg%), zinc (48.3ppm) and manganese (45.2ppm) were plentiful. It was noted that the iodine value was 44.0, saponification value 292.3, acid value 1.5 and peroxide value about 3.3. The lipid fractions obtained by silicic acid column chromatography were mainly composed of neutral lipid, whereas, the contents of glycolipid and phospholipid were scarcely. The major fatty acids in total lipids were linoleic $(60{\sim}62.5%)$, oleic $(20{\sim}22.5%)$ and palmitic $(10{\sim}13.5%)$ acids. The content of the unsaturated fatty acids was more predominant than that of the saturated fatty acids. The content of the insoluble proteinous nitrogen (32.5%) was the most abundant. whereas, the contents of soluble proteinous nitrogen (12.7%) and peptide proteinous nitrogen (3.3%) were low. The major amino acids of the maized germs were glutamic, glycine, alanine and aspartic. It has been identified by the SDS-disc electrophoresis that the protein of maized germs had $5{\sim}6$ bands. Compared with standard substances, the molecular weight of the main protein was estimated to be $14,500{\sim}24,500$.

Effect of Chicory Extract on the Lipid Metabolism and Oxidative Stress in Rats (흰쥐의 지질대사 및 산화적 스트레스에 미치는 치커리 추출물의 영향)

  • This study was conducted to investigate the effect of water-soluble extract from roasted chicory on the lipid metabolism and oxidative stress in male Sprague-Dawley rats. The experimental groups were divided into three groups ; the normal group, the cholesterol group and the chicory group. Roasted chicory extract was supplemented at 5.0% (w/w) level in the cholesterol diet. Concentration of total cholesterol in serum was significantly higher in the cholesterol group than in the normal group, but this increase in the cholesterol group was significantly decreased by the cholesterol diet supplemented with chicory extract. Concentration of HDL-cholesterol in serum was significantly lower in the cholesterol group than in the normal group, but this decrease in the cholesterol group tended to increase in the chicory group. However, concentrations of triglyceride, phospholipid and nonesterified fatty acid in serum were not significantly different among the groups. Concentrations of triglyceride and cholesterol in liver were significantly higher in the cholesterol and chicory groups than in the normal group. Feces weight and the excretion of cholesterol and bile acid into feces were significantly higher in the chicory group than in other groups. Concentrations of thiobarbituric acid reactive substances (TBARS) in homogenates and microsomal fractions of liver were not significantly different among the groups. On the other hand, concentration of 8-hydroxydeoxyguanosine (8-OHdG) as an useful marker of oxidative stress in urine was lower in the chicory group than in other groups. Concentration of serum glucose was signnificantly lower in the cholesterol group than in the normal group, but that of the chicory group was significantly higher than in the normal group. These results demonstrated that dietary chicory extract exerted the decreasing effect of cholesterol level and oxidative stress in cholesteral-fed rats.

Inhibitory Effect of Rumex Crispus L. Fraction on Adipocyte Differentiation in 3T3-L1 Cells (소리쟁이 분획물의 지방세포 분화 억제 효과)

  • The anti-obesity effect of ethanol xtract and their fractions from Rumex Crispus L. on the differentiation of 3T3-L1 pre-adipocytes to adipocytes was investigated by suppressing adipocyte differentiation and lipid accumulation with Oil red O assay, western blot and real-time PCR analysis. Ethyl acetate fraction of Rumex crispus L. significantly inhibited adipocyte differentiation when treated during the adipocyte differentiation process, as assessed by measuring fat accumulation using Oil red O staining. In inducing differentiation of 3T3-L1 preadipocytes in the presence of an adipogenic cocktail, isobutylmethylxanthine (IBMX), dexamethasone- and insulin-along with ethyl acetate fraction residue processing treatment significantly decreased protein expression of obesity-related proteins, such as peroxisome-proliferators-activated-receptor-${\gamma}$ ($PPAR{\gamma}$) and CCAAT enhancer-binding-proteins ${\alpha}$ ($C/EBP{\alpha}$). These results indicate that ethyl acetate fraction of Rumex crispus L. is the most effective candidate for preventing obesity. However further studies will be needed to identify the active compounds that confer the anti-obesity activity of ethyl acetate fraction from Rumex crispus L.

A Study on the Lipid Components of Hazel Nut Oil (개암종실(種實)의 지질성분(脂質成分)에 관한 연구)

  • Physico-chemical characteristics of crude oil extracted from Korean Hazel nut were determined and its proximate composition was also analyzed. The proximate composition of Hazel nut was shown to be moisture 4.0%, crude protein 15.5%, crude fat 64%, nitrogen free extractive 11.7%, crude fiber 2.0% and crude ash 2.5%. The content of crude fat in Corylus sieboldiana was about 3% higher than in Corylus mandshurica.. Physico-chemical characteristics of crude oil found were: specific gravity, $0.916(15/15^{\circ}C)$; refractive index, $1.468(15^{\circ}C)$; saponification value, 184; iodine value, 94.5: acid value, 0.2; and unsaponifiable content, 0.25%. The lipid fractions in the crude oil obtained by silicic acid column chromatography were found to be composed of about 97% neutral lipids and about 3% compound lipids. Among the neutral lipids by TLC, triglycerides were 98% as the major components, free fatty acids and free strols were 0.5% and 1.3%, respectively. Esterified sterols were not found. The predominant fatty acids were oleic $(76{\sim}80%)$, linoleic (15%) and palmitic (5.0%), and the P/S ratio was $1.8{\sim}2.8$.

Peri-implantitis, systemic inflammation, and dyslipidemia: a cross-sectional biochemical study

  • Purpose: The aim of this study was to compare the inflammatory and lipid profile of patients with and without peri-implantitis. Methods: A cross-sectional biochemical study was carried out in which blood samples were collected from 16 patients with peri-implantitis and from 31 subjects with healthy implants. Clinical peri-implant parameters were obtained from all subjects. Levels of tumor necrosis factor-alpha and interleukin-10 (IL-10) were measured in serum. Lipid fractions, glucose and creatinine levels, and complete blood count were also assessed. Results: After controlling for a history of periodontitis, statistically significant differences between peri-implantitis patients and controls were found for total cholesterol (estimated adjusted mean difference, 76.4 mg/dL; 95% confidence interval [CI], 39.6, 113.2 mg/dL; P<0.001), low-density lipoprotein (LDL) cholesterol (estimated adjusted mean difference, 57.7 mg/dL; 95% CI, 23.8, 91.6 mg/dL; P<0.001), white blood cells (WBC) (estimated adjusted mean difference, 2.8×103/µL; 95% CI, 1.6, 4.0×103/µL; P<0.001) and IL-10 (estimated adjusted mean difference, -10.4 pg/mL; 95% CI, -15.8, -5.0 pg/mL; P<0.001). The peri-implant probing pocket depth (PPD) was modestly positively correlated with total cholesterol (r=0.512; P<0.001), LDL cholesterol (r=0.463; P=0.001), and WBC (r=0.519; P<0.001). A moderate negative correlation was observed between IL-10 and PPD (r=0.609; P<0.001). Conclusions: Otherwise healthy individuals with peri-implantitis showed increased low-grade systemic inflammation and dyslipidemia.

Studies on the Development of Food Resources from Waste Seeds IV. Chemical Composition of Red Pepper Seed (폐기종실(廢棄種實)의 식량자원화(食糧資源化)에 관(關)하여 제(第) 4 보(報) : 고추씨의 화학적(化學的) 조성(組成))

  • In order to find out the possibility of utilizing red pepper seed as food resources of fats and proteins, a series of studies were conducted. The red pepper seed contained 27.6% of crude fat and 22.2% of crude protein. The lipid fractions obtained by silicic acid column chromatography were mainly composed of 95.4% neutral lipid, where as compound lipid were 4.6%. Among the neutral lipid separated by thin layer chromatography, triglyceride was 85.6%, sterol ester 4.9%, free fatty acids 3.4%, diglyceride 2.5%, sterol 2.2% and monoglyceride 1.1%, respectively. The predominant fatty acids of red pepper seed oil were linoleic acid (57.1-75.4%), palmitic acid (13.9-21.3%) and oleic acid (8.0-15.1%), especially glycolipid contained 1.7% of linolenic acid and small amount of myristic acid and arachidic acid. The salt soluble protein of red pepper seed was highly dispersible in 0.02M sodium phosphate buffer containing 1.0M $MgSO_4$, and the extractability of seed protein was about 25.0%. Glutamic acid and arginine were major amino acids of red pepper seed protein. The electrophoretic analysis showed 6 bands in seed protein, and the collection rate of the main protein fraction purified by sephadex G-100 and G-200 was about 62.2%. Glutamic acid (19.9%) was major amino acid of the main protein, followed by glycine and alanine. The molecular weight of the main protein was estimated to be 93,000.

Lipid Composition of Purple Shell and Abalone (피뿔고둥과 전복의 지질조성에 관한 연구)

  • This paper presents the composition of neutral and polar lipids obtained from puple shell, Rapana venosa and the abalone, Haliotis discus hannai. The fatty acid composition and the classification of neutral lipids from two species were determined by gas chromatography (GLC) and thin layer chromatography (TLC). Total lipid contents of samples were $0.5\%$ in purple shell and $0.4\%$ in the abalone. The predominant fatty acids of total lipids were eicosapentaenoic acid ($19.30\%$). eicosenoic acid ($12.10\%$) and palmitic acid ($11.77\%$) in the purple shell, and palmitic acid ($21.29\%$), oleic acid ($14.55\%$) and linoleic acid ($14.21\%$) in the abalone. The lipid composition of non-polar lipid fractions in purple shell and abalone was separated and identified as free sterol, free fatty acid, triglyceride and hydrocarbon & esterified sterol by TLC. The contents of triglyceride from both neutral lipids were shown more abundant than any other subclasses. The main fatty acids of neutral lipids were eicosapentaenoic acid ($18.6\%$), palmitic acid ($14.90\%$) and eicosenoic acid ($14.76\%$) in the purple shell, and palmitic acid ($28.12\%$), oleic acid($20.5\%$) and myristic acid ($12.5\%$) in the abalone. Eicosapentaenoic acid ($17.57\%$), stearic acid ($13.26\%$) and eicosatetraenoic acid ($11.24\%$) were important fatty acids of glycolipid in the purple shell, and myristic acid ($12.75\%$), stearic acid ($12.10\%$) and eicosatetraenoic acid ($10.64\%$) in the abalone. The major fatty acids of phospholipids were eicosapentaenoic acid ($20.18\%$), palmitic acid ($11.26\%$) and eicosenoic acid ($10.90\%$) in the purple shell, and palmitic acid ($21.10\%$), eicosapentaenoic acid ($12.90\%$) and oleic acid($11.13\%$) in the abalone.

Effects of the Feral Peach (Prunus persica Batsch var. davidiana Max) Extract on the Lipid Compositions and Blood Pressure Level in Spontaneously Hypertensive Rats (야생 돌복숭아(Prunus persica Batsch var. davidiana Max.) 추출액이 선천성 고혈압 흰쥐의 지질성분 및 혈압 저하에 미치는 영향)

  • The purpose of this study was to observe the effects of the feeding physiological activity substance in feral peach(Prunus Persica Batsch var. davidiana Max.) extract intake on the improvement of the lipid compositions, apolipoprotein and blood pressure level in spontaneously hypertensive rats(SHR, Wistar strain, male) fed the experimental diets for 33 days. Concentrations of total cholesterol, triglyceride(TG), LDL-cholesterol, free cholesterol and atherosclerotic index in serum were significantly lower in the feral peach extract intake groups[groups 5g% Ex.(basal diet+feral peach 5.0g% extract), 10g% Ex.(basal diet+feral peach 10.0g% extract)] than those in the group Control(basal diet+water). In the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration, feral peach 5.0g%, 10.0g% extract intake groups(group 5g% Ex. and 10g% Ex.) were higher percentage than in the group Control. However, concentrations of total cholesterol and TC in liver and brain were significantly lower in the groups 5g% Ex. and 10g% Ex. than those in the group Control. But the concentrations of apolipoprotein (Apo) A-I and Apo A-II in serum were significantly higher in the feral peach 5.0g% and 10.0g% extract intake groups(5g% Ex. and 10g% Ex.) than in the control group. However, concentrations of Apo C-II, Apo C-III, Apo E and ratio of Apo B to Apo A-I in serum were fairly reduced in the groups 5g% Ex. and 10g% Ex. than in the control group. The levels of systolic and diastolic blood pressure were significantly lower in feral peach 5g% Ex. and 10g% Ex. groups than control group. However, no significance was found in the effect of among the groups(groups 5g% Ex. and 10g% Ex.). From these results, physiological activity substance in feral peach(Prunus persica Batsch var. davidiana Max.) extracts were effective on the improvement of the lipid compositions and cardiovascular heart disease, hypertension in spontaneously hypertensive rats. And particularly, feral peach extracts were more effective as a therapeutic regimen for the control of blood pressure in hypertension.
