Lipid and fatty acid compositions of the bound lipids in Panax quinquefolium (Korea, Japan and China), Panax quinquefolium (America, Canada) and Panax notoginseng (China) were studied by means of silicic acid column chromatography, thin-layer chromatography and gas-liquid chromatography. The could lipid contents in various ginsengs were 0.29 to 0.48%, in which neutral lipid fractions were 63.6 to 67.3%, glycolipid fractions 21.9 to 25.7% and phospholipid fractions 7.7 to 12.4%. The content compositions of neutral lipid fractions were lower and those of glycolipid and phospholipid fractions were higher in the bound lipids than in the free lipids from the various ginseng. The major components were fatty acids, diglycerides and free sterols in neutral lipid fractions, monogalactosyl diglyceride, sterol glucoside and esterified steryl g1ycoside in glycolipid fractions and phosphatidyl glycerol, phosphatidyl ethanolamine and phosphatidic acid in phosphoipid fractions. Seventeen fatty acids were analyzed in the four bound lipid fractions from the various ginsengs and main fatty acids were linoleic acid, palmitic acid and oleic acid. Total saturated fatty acid and palmitic acid contents were higher and total unsaturated fatty acid and linoleic acid contents lower in the total bound lipids than in the total free lipids from the various ginsengs.

The contents and compositions of neutral lipids and glyclipids were some different among various ginsengs, whereas phospholipids showed relatively similar compositions in the contents. Seventeen fatty acids were analyzed in the four free lipid fractions from the various ginsengs and the main fatty acids were linoleic acid, palmitic acid, oleic acid and linolenic acid. It was found that the amounts of some fatty acids were different among the various ginsengs, but the fatty acid patterns of these ginsengs were on the whole similar.
The rhizoma of Polygonatum odoratum (Liliaceae) promotes the production of body fluid and relives dryness symptoms and has hypoglycemic effect. In order to evaluate the prevention of lipid peroxidative efficacy of P. odoratum, its extracts (Et$_2$O and MeOH ex.) and its fractions ($H_2O$, 20% MeOH, 40% MeOH, 60% MeOH and 100% MeOH fr.) were measured by TBARS assay on rat liver S9 and human erythrocyte ghost membrane. The order of anti-lipid peroxidative effect was as follows: Et$_2$O ex. >40% MeOH fr. >60% MeOH fr.>100% MeOH fr. Based on these results, we conclude that membrane lipid peroxidation is inhibited in vitro by addition of P. odoratum ether extract and its gradient MeOH fractions excepts $H_2O$ fr., and which have significant hepatoprotective activity in $CCl_4$-treated rats.
Objectives: Atriplex gmelinii C. A. Meyer is a halophyte belonging to the Chenopodiaceae family, and its young leaves and stems are used as fodder for livestock. The aim of the present study was to investigate the effects of A. gmelinii extract and its solvent fractions on lipid accumulation during adipogenesis of 3T3-L1 preadipocytes. Methods: The samples of A. gmelinii were separately extracted using methylene chloride and methanol. Subsequently, they were combined to formulate the initial extract, which was then partitioned based on polarity to prepare solvent fractions. Oil Red O staining was employed to measure lipid accumulation during the differentiation of 3T3-L1 preadipocytes. To verify cytotoxicity in 3T3-L1 cells, MTT assays were conducted. The expression levels of transcription factors in 3T3-L1 preadipocytes were measured through Western blotting analysis. Results: At 50 ㎍/mL, treatment of A. gmelinii extract and its solvent fractions during the differentiation of 3T3-L1 preadipocytes significantly diminished lipid accumulation with no noteworthy cytotoxicity on cell viability. Additionally, when investigating the biochemical pathways that underlie the prevention of lipid accumulation using solvent fractions, it was found that the n-BuOH and n-hexane fractions significantly decreased the expression of key transcription factors involved in the generation of fat, such as peroxisome proliferator-activated receptor γ (PPARγ), CCAAT/enhancer binding protein α (C/EBPα), and sterol regulatory element-binding protein-1c (SREBP1c). Conclusions: These findings indicate that A. gmelinii can effectively reduce the accumulation of fat in 3T3-L1 adipocytes, making it a potentially valuable material for mitigating and preventing obesity.
Lipid and fatty acid compositions of free lipids and bound lipids from fresh ginseng, red ginseng and white ginseng were studied by means of silicic acid column chromatography, thin-layer chromatography and gas-liquid chromatography. Free lipid and bound lipid contents in those three samples were 1.21 to 1.45% and 0.32 to 0.45%. Neutral lipid fractions in free lipids from the samples were 76.6 to 79.7%, while glycolipid and phospholipid fractions were 11.6 to 14.7% and 8.5 to 8.7%, respectively. The major lipids were triglycerides, sterol esters and hydrocarbons, diglycerides and free sterols in neutral lipids, sterol glucoside, monogalactosyl diglyceride, esterified steryl glycoside, digalactosyl diglyceride in glycolipids and phosphatidyl ethanolamine, phosphatidyl glycerol, phosphatidyl choline and phosphatidyl inositol in phospholipids. Fourteen kinds of even numbered and four kinds of odd numbered fatty acids were identified in the four lipid fractions (TL, NL, GL and PL) by GLC, and the main fatty acids were linoleic acid, palmitic acid, oleic acid and linolenic acid.
The comparative activities of acetone, ethanol, and aqueous fractions extracted from fruit powder of Cudrania tricuspidata by different temperature were tested by in vitro experimental models; peroxidation of linoleic acid and autooxidation of rat hepatic and renal microsomes by using thiobarbituric acid (TBA) for assay of free malondialdehyde production, and scavenging activities of free radicals by DPPH (α, α'-diphenyl-β-picrylhydrazyl). In DPPH method, acetone fraction extracted at 30℃ showed the highest free radical scavenging activities and acetone fractions extracted at 30℃ and 60℃ and ethanol fraction extracted at 30℃ showed stronger than BHT (butylated hydroxitoluene) although used ten-fold lower concentrations. In thiocyanate method used linoleic acid an inhibitory effects of all fractions showed higher than control treatment. TBA method used linoleic acid showed the highest antioxidative activity in acetone fraction extracted at 30℃ and 60℃. an inhibition activity against lipid peroxidation in hepatic microsomes of rats showed the highest at acetone faction extracted at acetone fraction among extracted fractions was shown to be the most potent antioxidative properties and this action was more potent in fractions extracted at 30℃ than those extracted at 60℃.
대두식품들로부터 추출한 지질분획들이 사람의 K562 백혈구암세포, mouse의 Yak1 백혈구암세포 및 S180 고형암세포에 미치는 세포독성 효과를 MTT assay법으로 조사하였다. 대두, 비지 및 된장의 총지질을 chloroform/methanol(2 : 1) 및 물포화 butanol로 추출하고 20배 용량의 acetone을 가하여 아세톤상층액(AS 획분) 및 아세톤침전물(AP 획분)을 얻었다 된장지질의 AS 획분 및 AP 획분이 실험에 사용한 K562, Yac1 및 S180암세포에 가장 강한 세포독성을 일으켰으며 비지 의 AS 및 AP 획분 역시 된장보다는 약하지만 세포독성을 나타냈으나 대두의 지질획분들은 세포독성효과를 나타내지 않았다. 된장지질의 AS 및 AP분획의 지질조성을 보면 대두 및 비지에 비하여 많은 양의 유리지방산이 함유되어 있었다. 또한 지질획 분들을 0.4 N KOH/methanol로 약알카리 분해하고 silica gel TLC로 분석한 결과, 된장의 지질획분에서는 대두 및 비지에서 보이지 않았던 알카리에 안정하고 ninhydrin에 양성인 물질들이 여러개 나타났으며 이들은 유리 sphingoid base 및 psycusin으로 추정되었다.
This study was conducted to examine the effect of dietary iron levels on lipid metabolism, antioxidative and antithrombogenic capacities in 16-month-old rats. Thirty-two Sprague-Dawley male 16-month-old rats weighing 618 $\pm$ 6 g were raised for 10 days with medium-iron diet (35 ppm in diet) and blocked into 4 groups according to their body weights. One of groups was sacrificed to obtain initial data and the rest 3 groups were raised for 3 months with experimental diets containing different levels of iron (5 ppm, 35 ppm, and 350 ppm). Total lipid, triglyceride and total chole-sterol concentrations in plasma and liver, HDL-cholesterol concentration in plasma, fecal total lipid triglyceride and total cholesterol excretions, thiobarbituric acid reactive substances (TBARS) level in plasma LDL + VLDL (low density lipoprotein + very low density lipoprotein) fractions, blood-clotting time and eicosanoids levels in plasma were measured. The results are as follows: Plasma total lipid, triglyceride and total cholesterol concentrations, TBARS level in plasma LDL + VLDL fractions were increased and blood-clotting time tended to be shortened during 3 months of experimental period. Low (5 ppm) iron diet improved lipid metabolism via increasing HDL-cholesterol and fecal choles-terol excretion. High (350 ppm) iron diet decreased plasma total lipid, triglyceride and total cholesterol concentrations as compared to medium (35 ppm) iron diet and lowered body weight and epididymal fat pad weight. On the other hand, TBARS level in plasma LDL + VLDL fractions and blood-clotting time were increased with high iron diet. It is plausible that low iron diet improves lipid metabolism, antioxidative and antithrombogenic capacities in 16-month-old rats.
We studied the effects of the methanolic extracts and fractions from leaves, fruits, stems and roots of Curadrania tricuspidata on the formation of lipid peroxide. The methanol extracts of leaves, fructus and stem of this plants decreased the formation of lipid peroxide in the isolated rat liver. The ethyl acetate fraction of leaves and n-butanol fraction of stems reduced the lipid peroxide formation. It showed that arthocarpesin, one of the constituents isolated from this plant, lowered the formation of lipic peroxide by 13%, 21%, 22% and 25% at the concentration of $10^{-8},\;10^{-6},\;10^{-4}\;and\;10^{-2}mg/ml$, respectively.
The studies were attempted to evaluated the therapeutic effects of various fractions(ether, methanol, butanol) of Caragana chamlagu roots on the hyperlipemia induced by feeding the diet containing 1%, cholesterol and 0.5%, cholic acid in rats, and on the hyperglycemia induced by streptozotocin in rats. Also the preventive effects of these fractions were studies on the liver damage in $CCl_4-intoxicated$ rats. The followings were obtained as the results: 1.The butanol fraction was significantly shown to down the serum lipid level in 1%, cholesterol and 0.5%, cholic acid diet-feeding rats and streptozotocin-induced hyperglycemic rats. Cholesterol level in $CCl_4-intoxicated$ rats was reduced in the case of all pre-treated groups. 2.The serum glucose level of streptozotocin-induced hyperglycemic rats was significantly decreased by the administration of various fractions of C. chamlagu roots, and the lipid-peroxidation of pancreas was significantly decreased in the case of administration of these fractions. 3.The activates of s-GOT and s-GPT were decreased by the administration of various fractions, especially in butanol fraction, of C. chamlagu roots in the $CCl_4-intoxicated$ rats. The liver lipid-peroxidation was decreased by administration of 200mg/kg of these fractions in the $CCl_4-intoxicated$ rats. In histological observation, hepatic cellular necrosis and fatty acid deposit were increased remarkably by $CCl_4-intoxication$, but the pretreatment of various fractions of C. chamlagu roots improved the pathological change of parenchymatous cell necrosis and fatty change around centrilobular area of the control.
