수침과정 전후의 한국산 검정콩의 지질성분 변화에 관한 연구 (Changes of Lipid Composition of Korean Black Soybean before and after Soaking)

  • 수침이 진행되는 동안 한국산 검정콩의 수분 흡수는 수침 초기에 상당히 빠른 속도로 진행되었고 균일하게 완전히 수화(hydration)되는 시간은 약22시간 정도였다. 검정콩의 총지질중 중성지질은 89.1%, 당지질은 1.5%, 인지질은 9.4%였으며, 수침 후에도 그 조성은 비슷하였다. 그리고 중성지질 성분은 triglycerides(92.1%)가 대부분이었고 그 외 sterol esters와 hydrocarbons (3.0%), diglycerides (2.8%), free fatty acids (1.5%), free sterols (0.3%), monoglycerides (0.3%)순으로 함유되어 있었고, 수침 후에도 그 조성은 크게 변화하지 않았다. 당지질 성분은 esterified steryl glycosides(43.6%), steryl glycosides (26.6%), digalactosyl diglycerides(14.5%) 등이 주성분이었고 이러한 성분들은 수침으로 인해 약간 변화하였다. 인지질 성분은 phosphatidyl choline(41.6%) 및 phosphatidyl ethanolamine(39.5%)등이 가장 않았으며 이러한 성분들은 수침 과정 중 다소 감소하였고 phosphatidyl serine, phosphatidyl inositol, lysophosphatidyl choline등의 성분들은 약간 증가하였다. 총지질의 지방산 조성은 linoleic acid (51.1%), oleic acid (23.1%), palmitic acid(11.8%)등이 주성분으로 구성되어 있었고 중성지질에서는 이와 유사한 경향이었으나, 당지질과 인지질에서는 palmitic, linoleic acid등의 함량이 많았다. 수침 과정중 총지질, 중성지질의 지방산 조성 변화는 거의 없으나, 당지질의 지방산 조성은 미량씩의 증감의 변화가 있었고 인지질의 지방산 조성은 palmitic, oleic acid 등은 증가에 비해 linoleic, linolenic acid 등은 감소하였다.

한국산 마지질의 분획 정량과 지방산조성 (Quantitative Fractionation of Total Lipids and Their Fatty Acid Composition in Korean Yam Tubers.)

  • 한국산 마(긴마, 단마)에서 총지질을 추출하고 이의 각종 구성 지질을 SCC(silicic acid column chromatography)에 의해 분획 정량하고, 이들의 조성을 TLC에 의해 분별한 후 각 획분의 지방산 조성을 GC에 의하여 분석하였다. 마의 건조물 중 총지질의 함량을 보면, 긴마, 단마는 각각 1.70%, 1.31%이었고 정제 총지질은 각각 1.35%, 0.99%였다. 또한 정제 총지질을 SCC로 중성지질, 당지질 및 인지질로 분획 정량한 결과, 긴마는 각각 43.33%, 39.60%, 17.07%, 단마는 각각 43.94%, 34.74%, 21.32%로 중성지질이 가장 많았다. 마의 중성지질을 구성하는 주요성분은 triglyceride와 sterol ester이며 당지질의 구성성분은 monogalactosyl diglyceride, trigalactosyl dialyceride이었다. 그리고 인지질의 주요 구성성분은 phosphatidyl choline, phosphatidyl ethanolamine이었다. 총 지질을 구성하는 주된 지방산을 보면, 마는 linoleic acid와 palmitic acid가 가장 많은 부분을 차지하였고 또한, 총지질 중의 중성지질, 당지질 및 인지질을 구성하는 주된 지방산 조성은 총지질을 구성하는 지방간 조성의 pattern과 거의 같았으며, 다만 당지질 경우 linolenic acid가 중성 및 인지질에서 보다는 구성성분으로 다소 많았다.

율무와 염주의 식이섬유, 아미노산 및 지질 성분의 비교 (Comparative Studies on the Dietary Fiber, Amino Acids and Lipid Components of Yullmoo and Yeomjoo)

  • 현율무, 율무쌀, 율무겨 그리고 현염주를 시료로 하여 일반성분, 총 식이섬유 함량, acid detergent fiber 함량, lignin 함량, 수분 결합력, 아미노산 조성 및 중성지질 조성 등을 살펴보았다. 그 결과, 율무쌀의 총 식이섬유량은 2.70% 였고, 현율무는 3.86% 였으며 현염주는 4.33% 그리고 율무겨는 13.12%였다. Acid detergent fiber 양은 율무쌀이 1.81%로 가장 낮았고 현율무, 현염주는 각각 3.45%, 3.26%로 비슷한 값이었다. 율무겨에서 추출한 총 식이섬유와 acid detergent fiber 의 수분 결합력을 측정하여 비교한 바 총 식이섬유의 수분 결합력은 $2.63{\pm}0.02g\;H_2O/g\;TDF$로 매우 낮았고, acid detergent fiber의 수분 결합력은 $5.89{\pm}0.152g\;H_2O/g\;ADF$였다. 율무쌀, 현율무, 현염주 그리고 율무겨의 아미노산 조성은 glutamic acid 함량이 가장 높았고 다음이 leucine 이었으며, 율무쌀의 경우 leucine 의 chemical score는 189로 매우 높았으며 반면에 lysine의 chemical score는 22-32 사이로 매우 낮아 율무, 염주의 제한 아미노산이 되었다. 지질성분은 중성지질이 80.89%-96.55% 범위로 대부분을 차지했고, 당지질이 2.35%-7.48% 사이로 현염주의 당지질 함량이 가장 낮았다. 인지질은 함량이 매우 낮아서 1.10%-2.12% 범위였다. 각 지질 성분의 지방산 조성은 palmitic acid, oleic acid 그리고 linoleic acid 였으며 현염주를 제외하고는 조성상에 크게 차이가 나지 않았다.

모유영양아와 인공영양아의 지질 대사 (Lipid Metabolism of Korean Breast-Fed Formula-Fed Infants)

  • The difference in lipid metabolism between breast-fed(BF) and formula-fed(FF) infants were studied in 31 Korean infants at 1, 2 and 3 months postpartum. The formulas had more total lipids(TL), triglycerides(TG) and phospholipids and less cholesterol(CHOL) and free fatty acids(FFA) than the breast milk. The milk consumption of the FF infants was significantly greater than that of the BF infants with a wide individual variation. As a result, the FF infants appeared to consume more TL, TG and PL and less CHOL and FFA than the BF infants during 3 months. The lipid contents of the breast milk tended to decrease in due course of lactation, therefore the intake of lipids of BF infants was reduced during the first three month of lactation. The plasma TG and CHOL levels of infants at 3 month were not significantly different between the BF and FF infants, but the PL level of the BF infants was higher then that of the FF infants. The plasma levels of TG and CHOL were not significantly correlated with the intakes of TL, TG, CHOL, PL and FFA, respectively. The PL level, however, was positively correlated with the intakes of CHOL and FFA, respectively and negatively correlated with PL intake as well as 18 : 2w6 fatty acid content of the breast milk or the formulas. The fecal weight and fecal loss of TL of the FF infants were greater than those of the BF infants during 3 months, however, apparent lipids digestibility was not significantly different between the BF and FF infants. The fecal excretions of CHOL and bile acids of FF infants were substantially higher than those of BF infants during 3 months. New sterol balance showed a particularly large difference between the BF and FF infants. The value of the net sterol balance of the BF infants was negative, but that of the FF infants was positive. This study shows that the consumption pattern of various lipid components of BF and FF infants were different as a result of different lipid compositions between breast milk and formula. The significantly lower sterol balance of the BF infants than the FF infants may have been derived from the unique dietary characteristic of breast milk.

Lipid-lowering and hepatoprotective effects of Vitis vinifera dried seeds on paracetamol-induced hepatotoxicity in rats

  • BACKGROUND/OBJECTIVES: Red grape seeds as functional food are a good source of important bioactive components such as phenolics and antioxidants, which decrease oxidative stress that contributes to the pathogenesis of hepatotoxicity. The current study was conducted in order to evaluate the protective effect of red grape dried seeds (RGDS) on antioxidant properties, lipid metabolism, and liver and kidney functions of rats with paracetamol (750 mg/kg) induced hepatotoxicity. MATERIALS/METHODS: RGDS was added to the basal diet at 5, 10, and 20%. Thirty five adult male rats were assigned to five groups (n = 7) for a six-week feeding period; group (1) normal control, group (2) induced control, groups (3, 4, and 5) fed a diet with RGPS at different levels, 5, 10, and 20%, respectively. At the end of the feeding period, animals' blood and tissues were collected for estimation of serum lipid profile, serum liver, and kidney biomarkers. The protection was measured by detecting lipid peroxidation (LPO), glutathione (GSH), superoxide dismutase (SOD), Catalase (CAT) (in liver tissues), and liver histological examination. RESULTS: The results showed a significant (P < 0.05) decrease in levels of serum cholesterol, triglycerides, low density lipoprotein (LDL-C), and very low density lipoprotein (VLDL-C), with a significant increase in level of high density lipoprotein (HDL-C) for RGDS groups compared to induced control. Rats administered a diet containing RGDS levels produced significant (P < 0.05) hepatoprotection by decreasing the activities of liver enzymes, kidney parameters, and lipid peroxidation, while levels of GSH, SOD, and CAT were increased significantly to near the normal levels. CONCLUSION: The RGDS 20% group was more effective than others against hepatotoxicity of paracetamol, which may be attributed to RGDS total phenols and antioxidant contents, which were 1.438 mg and 1.231 mg, respectively.

Seasonal Variations in Biochemical Components of the Visceral Mass and Adductor Muscle in the Pen Shell, Atrina pectinata

  • Seasonal content changes of the three key nutrients for organisms, protein, lipid and glycogen, were analyzed for a whole year to delineate the seasonal energy strategy in pen shells, Atrina pectinata. Two metabolically important organs, the visceral mass and the posterior adductor muscle, were examined. Protein in the visceral mass rose in April and stayed at the level until June followed by the constant minimum value between August and November. The protein contents in the posterior adductor muscle increased sharply in April and again in July, followed by a gradual decline thereafter. Total lipid contents in the visceral mass gradually increased between January and May, and then slowly decreased until September since which a new weak increase was noticed. Lipid levels in the adductor muscle rapidly dropped in June and July. Glycogen contents in the visceral mass rapidly increased between February and June, followed by a drastic drop in July. This reduced visceral glycogen level was maintained up to September, and a gradual reduction ensued. Glycogen contents in the adductor muscle steadily but markedly increased from April reaching the maximum in August, and then slowly declined thereafter. These results suggest that an accelerated protein and lipid synthesis occurs in the gonad when the pen shell undergoes the ripe stage of gametogenesis, but the levels of these two nutrients decrease on spawning. With this gonadal process, regular protein synthesis and lipid storage in the posterior adductor muscle are temporarily arrested. The most important nutrient reserves that support gonad developmental cycles in a long term seem to be glycogen of the posterior adductor muscle.

지방의 종류가 다른 식이에 L-카르니틴을 첨가했을 때 흰쥐의 혈청과 간의 지질성분과 항산화계에 미치는 영향 (Effect of Dietary L-carnitine Supplementation on Serum and Liver Lipid Composition and Antioxidant Defense System in Rats fed with Different Types of Fat)

  • The aim of this study was to investigate the effects of L-carnitine on the components of serum and liver and the effects on the anti-oxidant system. For this purpose, five experimental groups were setup. For fat source, perilla oil enough with unsaturated fatty acid and beeftallow enough with saturated fatty acid were supplemented together with L-carnitine to the rats. Five experimental groups kept eight Sprague-Dawley rats respectively, They were co group supplemented with basic diet or AIN-93, PO group supplemented with perilla oil, POC group supplemented with perilla oil and L-carnitine, BT group supplemented with beeftallow, and BTC group supplemented with beeftallow and L- carnitine. The results are. 1) Weight gain, food intake and FER were not different significantly among the experimental groups. 2) Significant difference was observed in serum total lipid(P<0.05), serum triglyceride(P<0.05), serum total cholesterol (P<0.05)and serum LDL cholesterol(P<0.05). Serum total lipid and serum triglyceride were significantly low in the groups supplemented with L-carnitine. Serum total cholesterol showed difference with the supplementation of L-carnitine in BTC only. LDL cholesterol showed no significant difference with the supplementation of L-carnatine, but total values of LDL-cholesterol were high in groups supplemented with beeftallow. 3) Total cholesterol in liver was low in POC group with the supplementation of L-carnitine however, there was no difference in BTC group with the supplementation of L-carnitine. In summary, dietary L-carnitine did not influence the weight gain, food intake and food efficiency ratio among the experimental groups, but had an effect of lowering the serum total lipid and triglyceride significantly in both groups which were supplemented with L-carnitine. The effect of lowering of sew total cholesterol with the supplementation of L-carnitine in beeftallow group(BTC) only. The effect of lowering of liver total cholesterol with the supplementation of L-carnitine in perilla oil group(POC) only.

대두식품 지질추출물의 세포독성 및 지질성분분석 (Cytotoxic Effects and Components of Lipid Fractions from Soybean Products on Cancer Cell Lines)

  • 대두식품들로부터 추출한 지질분획들이 사람의 K562 백혈구암세포, mouse의 Yak1 백혈구암세포 및 S180 고형암세포에 미치는 세포독성 효과를 MTT assay법으로 조사하였다. 대두, 비지 및 된장의 총지질을 chloroform/methanol(2 : 1) 및 물포화 butanol로 추출하고 20배 용량의 acetone을 가하여 아세톤상층액(AS 획분) 및 아세톤침전물(AP 획분)을 얻었다 된장지질의 AS 획분 및 AP 획분이 실험에 사용한 K562, Yac1 및 S180암세포에 가장 강한 세포독성을 일으켰으며 비지 의 AS 및 AP 획분 역시 된장보다는 약하지만 세포독성을 나타냈으나 대두의 지질획분들은 세포독성효과를 나타내지 않았다. 된장지질의 AS 및 AP분획의 지질조성을 보면 대두 및 비지에 비하여 많은 양의 유리지방산이 함유되어 있었다. 또한 지질획 분들을 0.4 N KOH/methanol로 약알카리 분해하고 silica gel TLC로 분석한 결과, 된장의 지질획분에서는 대두 및 비지에서 보이지 않았던 알카리에 안정하고 ninhydrin에 양성인 물질들이 여러개 나타났으며 이들은 유리 sphingoid base 및 psycusin으로 추정되었다.

대두 사포닌의 활성에 관한 기전 연구

  • Saponins are glycosidic compounds present in many plant foods. They are characterized by their ability to lyse cell membranes due to their surface-active properties. Saponins are believed to interact primarily with cholesterol in the cell membrane. In this study, the interaction of soybean(SS) with cell membrane was investigated using erythrocytes as a model. Mechanisms of interaction was also investigated by measuring their binding capacity with different membrane lipid fractions. Throughout the study, gypsophilla saponin(GS) and quillaja saponin(QS) were used to evaluate the membranolytic activity of soybean saponins. All saponins released hemoglobin in a concentration-dependent manner. SS induced 40% hemolysis at the concentration of 400 ppm, however there was no increase in hemoglobin release above 400ppm concentration. 5ppm of GS and 8 ppm of QS hemolyzed 100% of erythrocytes. Isolation of SS fractions by thin layer chromatography revealed that only one non-polar saponin possesses strong hemolytic activity. When saponins were incubated decreased the release of cholesterol. When the hemolytic activity of saponins was measured in the presence of other major membrane lipid components, sphingomyelin significantly reduced the hemolytic activity of SS, while cholesterol reduced the activity of QS. GS showed high affinity to other component(s) in the incubation media as well as lipids. These results suggest that the membranolytic activity of saponins are related to their specific chemical structure, which determines the interaction behavior between saponins and different membrane components, and thereby influence the biological activity.
