Analysis of Chemical and Physical Characteristics of Log Woods for Oak Mushroom Production Depending on Cultivation Periods and Steam Explosion Treatment (표고버섯 골목의 사용연수에 따른 화학적, 물리적 성상 및 폭쇄처리 후 변화 관찰)

  • In order to investigate the ability of log wood for oak mushroom production as a source of an alternative energy, both chemical and physical characteristics of log wood were investigated according to the cultivation periods. Also, both chemical and physical characteristics of material that treated by steam explosion were investigated to confirm the pretreatment effect by remaining enzyme as a control. The contents of ash, water-, alkali- and organic soluble extracts have been increased after the inoculation. It appeard that holocellulose contents substantially decreased and the contents of lignin as another main component of wood remained constant after the inoculation. However this result implied that indeed, a sufficient amount of lignin has been degraded paritially by enzymes of oak mushroom Lentinus edodes if we consider that the amount of holocelulose was substantially reduced. It also indicated that the degree of degradation gradually progressed but crystallinity decreased after the inoculation. The contents of water-, alkali- and organic soluble extracts have been increased by steam explosion. Holocellulose contents increased within narrow limits and lignin contents remained constant. However the contents of holocellulose and lignin have been decreased by steam explosion, considering that the amount of other extractives was relatively increased. The degree of crystallinity and lignin contents reduction by steam explosion was almost similar to the result obtained by increasing cultivation periods. According to the results, log woods for mushroom production have a potential as material for developing alternative energy.

Changes in Contents and Composition of Insoluble Dietary Fiber during Buckwheat Germination (메밀 발아 중 불용성 식이섬유 함량과 조성의 변화)

  • To provide the basic information of buckwheat insoluble dietary fiber (IDF) and efficient material for the buckwheat processed foodstuffs, buckwheat (Fagopyrum egculentum Moench) was germinated at 1$0^{\circ}C$ for 7 days and the contents and composition of the insoluble dietary fiber were examined at 24 hour intervals. The NDF and ADF contents in ungerminated seeds were 20.27%, 18.95% on dry weight basis, respectively. During the germination period, the NDF and ADF contents increased gradually. The cellulose, hemicellulose and lignin contents in ungerminated seeds were 11.10%, 1.32% and 7.85%, respectively. During the germination period, the cellulose and lignin contents increased. However, there was no significant change in the hemicellulose contents. The composition of cellulose and hemicellulose in the NDF decreased with germination time, whereas that of lignin increased. The IDF contents obtained by Prosky method were higher than the NDF contents obtained by Van Soest method. However, the IDF and NDF contents were closely correlated(r=0.9785, p<0.01). The cellulose(Xl), hemicellulose(X2), lignin(X3) and soluble dietary fiber (SDF) (BL) showed the significant regression equation(p<0.001) with the root length(Y). The multiple regression equation was Y = -12.1306 + 0.9213xl - 0.1615$\times$2 + 0.1071$\times$3 + 0.7397$\times$4 and R2(coefficient of determination) was 0.942.

  • A study on the effect of the biodegradability of the composting bulking agent in the swine manurecomposting was carried out in a batch system. The purpose of this study is to prove the effect of the biodegradability of the composting-bulking agent on the efficiency of the composting. In this study, it is the lignins: Klason-Lignin in the volatile solid that the index of the biodegradability of the composting-material mixes which are pig manure-rice straw pig manure-sawdusts pig manuremixture of rice and ricestraw (2:1) pig manure-mixture of rice and sawdust (1:1). It was carried out in the same condition (moisture contents, air supply rate, C/N ratio, initial input weight, porosity-structure) except the biodegradability of the raw material mixes. One of the results from this study is that the biodegradability of the bulking agent in the sense of the VS lignin content is not an insignificant factor in composting reaction. The less contents of the lignin in VS, the more efficiencies of the cornposting reaction in use of these parameters for the degree of the reaction: temperature, the trends of the ash contents, the change pattern of the C/N ratio. Under some assumptions, it is able to induce rough model on the relation of the VS lignin contents with the efficiency of the degradability. In this model, the biodegradability of the bulking agent is not an insignificant factor however, it is flexible within some degrees of range.

The Change of Kenaf Fiber Characteristics by the Contents of Noncellulosic Material (비셀룰로오스 함량에 따른 케나프 섬유의 특성변화)

  • The effects of removal of lignin or hemicellulose on the cottonizing and pulping characteristics of kenaf fiber were studied by comparing the conditions of non-cellulosic material contents, fiber lengths and dyeability. And the effects of lignin or hemicellulose on dyeability of the kenaf fiber using CI Direct Green 26 and CI Direct Red 81 were investigated. The results were as follows. The lignin contents decreased and the kenaf fiber became shorter and finer as the reaction time with sodium chlorite increased. The hemicellulose could be removed by treating sodium hydroxide solution to the fiber from which the lignin partly removed. The 80% of hemicellulose could be removed by 5% of sodium hydroxide solution in 5 minutes. But if lignin were not removed at all, hemicellulose could not be removed. The fiber lengths proper for apparel were obtained after treating sodium chlorite for 10-20 minutes and those for pulping were obtained after treating sodium chlorite for 40 minutes. The kenaf fibers from which lignin and hemicellulose partly removed were dyed with CI Direct Green 26 and CI Direct Red 81. Their dyeability increased as the removal rates of lignin increased. The ${\Delta}E$ values of kenaf fiber dyed with CI Direct Green 26 were lower than CI Direct Red 81.

Chemical Composition and Alkaline Pulping of a Stem of Red Pepper (Capsium annuum L.) (고추 줄기의 화학 조성분 및 알칼리 펄프화)

  • Chemical compositions and chemical structure of lignin and alkali cooking condition and fiber length of red pepper were investigated and compared to those of woods. The chemical compositions of red pepper were higher component of extraction than that of wood. The contents of carbon and hydrogen of Klason lignin in red pepper were similar to that of pine and birch wood. On the other hand, the contents of oxygen and nitrogen of Klason lignin in the red pepper were higher than that of wood. The result of nitrobenzene oxidation shows that Klason lignin of red pepper was similar to lignin of softwood. The best alkali cooking condition of red pepper was 0.2%-anthraquinone, active alkali of 20% and liquor ratio of 1:7. The fiber length of red pepper was about 0.47 mm. Therefore, the red pepper fiber will be able to use special purpose of short fiber.

Effect of Gooking on Water Insoluble Dietary Fiber in Vegetables (조리방법에 따른 채소의 불용성 식이섬유 함량 변화에 관한 연구)

  • Recently, interests of dietary fiber associated with critical physiological effects have been rising in Korea physiological effect in the body. In the present study, the contents of fiber components in 15 kinds of vegetables being consumed commonly in Korea were investigated, and the effects of various treatments (cooking and Kimchi fermentation) on fiber were studied. The results are summarized as follows. Fiber contents of vegetables were 11.8∼31.9% of neutral detergent fiber(NDF), total insoluble dietary fiber, 10.9∼25.4% of acid detergent fiber(ADF), 8.8∼23.8% of cellulose, 0.6∼10.6% of hemicellulose and 1.0∼5.2% of lignin, on dry weight basis. Especially, peppers had higher contents of NDF than the other vegetables. In the vegetables used in the present study, it was found that a great portion of NDF, total insoluble dietary fiber, was composed of cellulose because cellulose covered 63% of NDF. 'Cooking increased the NDF, ADF and cellulose contents, and most change was due to the change of cellulose. The values of hemicellulose and lignin showed an Irregular pattern upon cooking. Fermentation slightly increased NDF, ADF and cellulose, while hemicellulose and lignin showed irregular pattern.

  • Substrates used were hardwood-Suwon poplar-(Populus alba${\times}$glandulosa L.) and softwood-pitch pine-(Pinus rigida M.). And these substrates were steam exploded then treated with sodium chlorite at 75$^{\circ}C$ with occasional stirring in order to obtain samples which had different lignin contents and crystallinity. And then this resulting samples incubated with a commercial cellulase derived from Trichoderma ressei. The contents of Klason lignin were decreased as the increasing of the ratio of sodium chlorite in the two species. The effect of hardwood was more effective than that of softwood in the same ratio of sodium chlorite. The minimum contents of Klason lignin were 0.8% and 5.1% respectively. And the crystallinities of cellulose were increased very little as increasing of the ratio of sodium clorite. The hydrolysis extent of the two species were increased as the increasing of delignification. Especially, the hydrolysis extent of hardwood was more higher than that of softwood. The maximum hydrolysis extent were 89.8% and 71.1%, respectively.

Lignin Characterization of Waste Liquor by Modified Pulping Process (개량(改良)펄프화법(化法)으로 제조(製造)된 폐액(廢液)중의 리그닌의 특성(特性))

  • This study was carried out to investigate the characterization of lignins from waste liquors in SP, KP, ASAM, and AS from Pinus densiflora, Quercus mongolica, and Betula ermanii. Spectroscopic study was applied to examine the lignins separated from different pulping process. Lignin contents in waste liqours increased in order of AS, ASAM, KP, and SP. UV spectra of three types of lignin except AS lignin showed similar pattern. IR spectra of AS lignin showed strong C=O absorptions in the range from 1730 to 1750$cm^{-1}$, where as those of KP, SP, and ASAM showed weak stretch in this region. NMR spectra of AS lignin showed strong characteristic chemical shifts of acetoxyl groups of acetylated aliphatic and aromatic hydroxyl groups at 2.0~2.5 ppm. Molecular weight of ASAM lignin from Pinus densiflora determined and found number average molecular weight 1,199, weight average molecular weight 5,458. z average molecular weight 17,242, and viscosity average molecular weight 5,457. It is considered from the results based on spectroscopic study of lignin that waste liquors (in SP, KP, ASAM and AS) from Pinus densiflora, Quercus mongolica, and Betula ermanii can be used for lignin utilization.

  • Chemical composition and contents of carbohydrates, lignin, and extractives in Moso(Phyllostachys pubescens Mazel), Henon(Phyllostachys nigra var. henonis Stapf ex. Rendle), and Timber bamboo(Phyllostachys bambusoides Sieb. et Zucc.) known as economically valuable bamboo species in Korea were analyzed as a function of ages. All of tested bamboo species were composed of fucose, arabinose, rhamnose, galactose, glucose, xylose, and mannose. Glucose was the sugar which showed the highest content followed by xylose. The contents of others were below 3%. Sugar contents of bamboo shoots showed the lowest among the tested bamboo samples, and the constant rates were shown after 50 days of growth. Lignin content increased by the growth of one year, and the increasing rates tended to be constant after the growth of three years. Bamboo shoots have the highest contents of acid-soluble lignin, which turned into insoluble lignin after the growth of 50 days. Bamboo shoots showed the highest content of extractives and tend to be decreased by the growth of 50 days and then the content increased slowly in proportion to the period of bamboo growth. No relationship was found between species and chemical composition of bamboo. However, there were significant difference in chemical composition between bamboo shoots and 50-day old bamboo.

Degradation Pattern of CMC, Xylan, Lignin Components of Rice Straw by Bacillus subtilis DO4 (Bacillus subtilis DO4에 의한 볏짚의 CMC, Xylan 및 Lignin 성분의 분해양상에 관하여)

  • To investigate the biodegradation pattern of rice straw, mainly composed of cellulose, hemicellulose and lignin components, by the isolate stran Bacillus subtilis $DO_4$, the change of cell population was observed on CMC (carboxymethyl cellulose), larch wood xylan and lignosulfonate as a carbon source respectively. Also, the transition pattern of enzyme activities of cellulase and xylanase and lignin contents was measured on rice straw and mixed substrate according to growth. The results in these experiments revealed that xylanase activity was first appeared and cellulase activity in the next, while lignin component was almost not changed through the culture period.
