Correlations among Shearing Force, Morphological Characteristic, Chemical Composition, and In situ Digestibility of Alfalfa (Medicago sativa L) Stem

  • Alfalfa (Medicago sativa L) is a high-quality forage for ruminants and the main stem is the dominant morphological component contributing to the forage nutritive value in mature alfalfa forage. Shearing force, a fracturing property of plant stem, is an important indictor of forage value. The objectives of this study were to investigate the effects of morphological characteristic on shearing force, the relationship between shearing force and chemical composition, and the relationship between shearing force and in situ digestibility of alfalfa stem. The results showed that linear density (weight per unit length of stem) was more important than chemical composition in affecting shearing force. There was a positive relationship between lignin content and shearing force (r = 0.78). Correlations were not found between shearing force and other chemical components such as neutral detergent fiber (NDF), acid detergent fiber (ADF), cellulose and hemicelluloses. In situ digestibility (of dry matter and NDF) was related to shearing force. A negative correlation was found between shearing force and dry matter (DM) digestibility (r = -0.70), and there was also a negative correlation between shearing force and NDF digestibility (r = -0.87). When shearing force was standardized for stem diameter or stem linear density, the relationship between shearing force and digestibility was consistent regardless of stem diameter and stem linear density. Shearing force was significantly correlated with lignin content and in situ digestibility (of DM and NDF), and was a more direct indicator for estimating forage nutritive value related to animal performance, so it can be used to predict the forage value of alfalfa.

  • For the preparation of fermented feed with Lyophyllum decastes, optimum cultural conditions for the production of cellulase were $30^{\circ}C$, pH 6.0, 60-70% moisture content and the cultural of 15 days. Among the submaterial added, 30-40% of rice bran and 0.72% of $(NH_4)_2PHO_4$ were effective for the cellulase production and its production increased when rice straw treated with 4% alkaline peroxides. Solid state fermentation of rice straw with Lyophyllum decastes for 40 days removed 19.9% of lignin, and increased the total nitrogen content to 1.6% from 1.1%. As the fermentation proceeded, the in vitro dry matter digestibility of fermented feed was increased and it increased 21.1% after 35 days.

  • To evaluate the fundamental factors in the recovery of biogas from the landfills composed of about 40% of volatile solids, the experiments for the samples from the operating landfill site as well as from the laboratory-scale lysimeter were undertaken. In the test of landfills, the change of moisture content, the content of volatile solids (VS), the ratio of saccharide to ligin(Y) and the estimation of landfills reclaimed and the correlationship between VS and Y were investigated. During the experiments with laboratory-lysimeter, temperature, pH, gas production rate, the composition of gas were measured. The mathematical model derived from the the rate coefficient of gas production(k) were proposed from the results of this investigation. Furthermore, the proposed mathematical model from this study was verified with the obtained values from experiments.

On the Pith-damaged Wood of Larix leptolepis Gorden (낙엽송(落葉松)(Larix leptolepis Gorden)의 심부재(心腐材))

  • This study was carried out to investigate the cause of pith-damaged wood formation, the state of growth, and difference of chemical composition of Japanese Larix(L. leptolepis Gorden). 1. Larix grew normaly to 10 years, and the volume increment was 32.4%. But it was gradually decreased to 19.4% from 11 to 13 years that the heartwood damage was occurred. Therefore, the volume increment was decreased 13%. The decayed wood was formed from fire wound portion of bark by forest fire. The volume loss from pith-dagmaged wood occurred by forest fire was more than 20%. 2. On the ash content, heartwood(0.05%) and pith-damaged wood(0.08%) was lower than sapwood(0.29%). The difference of extractive contents are considered the reason. It is considered that this ash content difference is depend upon the extractive contents among sap, heart and decayed wood parts. Holocellulose contents were from 54.3% to 59.3% and there were no difference among heartwood, pith-damaged wood, and sapwood, On the pentosan contents, heartwood(6.3%) and pith-damaged wood(6.6%) were higher than sapwood. Lignin contents of pith-damaged wood was tower than heartwood (34.4%). This seems to weaken all kinds of wood strength.

Effects of maize straw treated with various levels of CaO and moisture on composition, structure, and digestion by in vitro gas production

  • Objective: The objective of this study was to explore the effects of maize straw treated with calcium oxide (CaO) and various moisture, on the composition and molecular structure of the fiber, and gas production by fermentation in an in vitro rumen environment. Methods: The experiment used 4×3 Factorial treatment. Maize straws were treated with 4 concentrations of CaO (0%, 3%, 5%, and 7% of dry straw weight) and 3 moisture contents (40%, 50%, and 60%). Scanning electron microscopy, Fourier transform infrared spectroscopy and X-ray fluorescence spectroscopy were employed to measure the surface texture, secondary molecular structure of carbohydrate, and calcium (Ca) content of the maize straw, respectively. The correlation of secondary molecular structures and fiber components of maize straw were analyzed by CORR procedure of SAS 9.2. In vitro rumen fermentation was performed for 6, 12, 24, 48, and 72 h to measure gas production. Results: Overall, the moisture factor had no obvious effect on the experimental results. Neutral detergent fiber (NDF), acid detergent fiber, acid detergent lignin, hemicellulose and cellulose contents decreased (p<0.05) with increasing concentrations of CaO treatment. Surface and secondary molecular structure of maize straw were affected by various CaO and moisture treatments. NDF had positive correlation (p<0.01) with Cell-H (H, height), Cell-A (A, area), CHO-2-H. Hemicellulose had positive correlation (p<0.01) with Lignin-H, Lignin-A, Cell-H, Cell-A. Ca content of maize straw increased as the concentration of CaO was increased (p<0.01). Gas production was highest in the group treated with 7% CaO. Conclusion: CaO can adhere to the surface of the maize straw, and then improve the digestibility of the maize straw in ruminants by modifying the structure of lignocellulose and facilitating the maize straw for microbial degradation.

  • In the present study, effects of autohydrolysis treatment on the properties of rice hull were investigated. Two temperature condition such as $160^{\circ}C$ and $180^{\circ}C$ and thee treatment time such as 15 min, 30 min and 45 min were applied for autohydrolysis treatment. The pH of extract after autohydrolysis treatment decreased as increasing of the temperature and the treatment time. The ash content and the lignin content of rice hull were not decreased by the autohydrolysis treatment. The structure of rice hull after autohydrolysis treatment became more weak, the better efficiency of the fiberization with PFI-Mill was shown for the autohydrolysis treated rice hull with the higher temperature and the longer treatment time.

Chemical Characteristics of Cell Wall in Pinus thunbergii Parl. Grown with High Salinity (고염분 하에서 성장한 해송 세포벽의 화학 성상)

  • Stems of Pinus thunbergii Parl. grown with high salinity were analyzed for chemical characteristics. Stem of 2 years was rich in soluble compounds and stem of 3 years reduced the amount of the soluble compound. But, the lignin content have not seen a large change. Also, Klason lignins of stem of 2 and 3 years has not changed in nitrogen and hydrogen content. In Klason process, it was significantly increased the carbon concentration due to the hydrolysis of the carbohydrate. In addition, the accumulation of xylan from Pinus thunbergii Parl. with salinity treatment were increased noticeably. Finally, functional group of Pinus thunbergii Parl. with salinity treatment were not changed.

  • We made densified wood pellet by hemp woody core as replacing wood resource. Hemp was separated into the bast fiber and the woody core by hot steaming treatment. The hemp woody core had a similar lignin content and carbohydrate composition with hardwood. Also, the hemp had a low ash content, which resulted in a low ash formation in pellet burning. Heating value of the hemp pellet had a very similar to the pellet made by hardwoods. The hemp woody core can replace hardwood for densified wood pelletmaking.

  • To evaluate short rotation forestry candidates, seven clones of Salix hultenii were investigated as focused on chemical compositions and energy contents. There were significant variations in chemical compositions and energy contents among different clones. Variations in hot-water extractives content were 6.6-16.0% and 19.6-25.4% variation in lignin content. In carbohydrate compositional analysis, xylan contents were 7.5-13.1% with variation among different clones. Higher heating values were 17.69-20.02 MJ/kg.

NUTRITIVE EVALUATION OF SAGO FIBRE

  • Nutrient evaluation of sago fibre showed that the fibre has potential and could be utilized as feed for ruminants. However, as a source of nutrients, it has limitations arising from low intake, digestibility, crude protein and minerals content. The present study showed that the sago fibre is low in crude protein (3.3%) and high in neutral detergent fibre (72.5%) and acid detergent lignin (25.8%) contents. Treatment of sago fibre with urea increased the crude protein content from 3.3 to 16.7%. Both urea and sodium hydroxide treatment decreased the neutral detergent fibre level from 72.5 to 59 and 56.5%, respectively. Rumen degradation of sago fibre by nylon bag showed that both urea and sodium hydroxide treatments increased dry matter and organic matter disappearance of the fibre significantly. In vivo digestibility of 2% urea treated sago fibre was 47.5% and intake of the fibre was 1.57% of body weight of the lamb.
