The Effectiveness of a Prolonged-speech Treatment Program for School-age Children with Stuttering (학령기 말더듬 아동의 첫음연장기법을 이용한 치료프로그램 효과 연구)

  • The purpose of this study was to know the effectiveness of prolonged-speech treatment program on school-age children with stuttering. Two male and One female subjects participated in this study. The speech of 3 subjects in the treatment was assessed on frequency of stuttering, stuttering Pattern, degree of severity in stuttering. This Program was taken from Ryan's the step of traditional therapy Program and prolonged-speech technique program. and then, modified in accordance with the purpose of this study. The treatment program were consisted of Four stages. The results of this study were as follows: First, 3 subjects can speak with greatly reduced stuttering frequency after treatment Second, in the stuttering pattern, all subjects were changed from part-word repetition in stuttering into a prolongation in stuttering. And also, all subjects showed similar effect in the maintenance.

Model Construction of Sexual Adjustment of Patients with Spinal Cord Injury (척수장애인의 성적적응과 관련요인에 관한 모형구축)

  • The purpose of this study was to construct model of sexual adjustment in people with spinal cord injury and to determine factors that relate to sexual adjustment using methodological triangulation. A total of 134 persons who were registered members of spinal cord injury organization and admitted rehabilitation unit in the hospital were included in the study. Participants answered questionnaire concerning importance of life events, sexual concern, sexual adjustment. Qualitative data were collected through semi-structured indepth interviews from 10 individuals with spinal cord injury who were previously included in the quantitative study. Constant compatative method was used to analyze the data. The results were as follows : 1) With respect to eleven other areas of life, sex life ranked the sixth and economic status ranked the highest in terms of importance. However social life ranked the lowest among the 11areas. 2) Among seven topics related to sexuality were methods and techniques to achieve sexual satisfaction, and helping a partner cope emotionally with limitation on sexual dysfunction was the second greastest. 3) The mean score for sexual adjustment was 19.47 which can be considered. 4) A process on how individuals with spinal cord injury adjust to their changed sexual life immerged from the qualitative data. It includs 4 stages: 'stage of loss' 'stage of endeavoring' 'stage of effort' and 'stage of adjustment'. Categories showing context for the action/interaction strategies were 'steadiness' and 'rediscovery as a sexual being'. There were three factors which may stimulate the adjustment process while the others may interrupt it. Those factors included personal matters, family matters and social matters. The individuals may follow each stage step by step but may go back to the previous step depending on the outcomes of their adjustment. 5) There were three factors which may stimulate the adjustment process while the others may interrupt it. Polymorphism and Expression of Isoflavone Synthase Genes from Soybean Cultivars

  • Isoflavones are synthesized by isoflavone synthases via the phenylpropanoid pathway in legumes. We have cloned two isoflavone synthase genes, IFS1 and IFS2, from a total of 18 soybean cultivars. The amino acid residues of the proteins that differed between cultivars were dispersed over the entire coding region. However, amino acid sequence variation did not occur in conserved domains such as the ERR triad region, except that one conserved amino acid was changed in the IFS2 protein of the GS12 cultivar ($R_{374}G$) and the IFS1 proteins of the 99M06 and Soja99s65 cultivars ($A_{109}T$, $F_{105}I$). In three cultivars (99M06, 99M116, and Simheukpi), most of amino acid changes were such that the difference between the amino acid sequences of IFS1 and IFS2 was reduced. The expression profiles of three enzymes that convert naringenin to the isoflavone, genistein, chalcone isomerase (CHI), isoflavone synthase (IFS) and flavanone 3-hydroxylase (F3H) were examined. In general, IFS mRNA was more abundant in etiolated seedlings than mature plants whereas the levels of CHI and F3H mRNAs were similar in the two stages. During seed development, IFS was expressed a little later than CHI and F3H but expression of these three genes was barely detectable, if at all, during later seed hardening. In addition, we found that the levels of CHI, F3H, and IFS mRNAs were under circadian control. We also showed that IFS was induced by wounding and by application of methyl jasmonate to etiolated soybean seedlings.

Potential functional roles of follistatin on bovine somatic cell nuclear transfer embryos

  • To demonstrate that follistatin treatment enhances the efficiency of nuclear transfer (SCNT), cell allocation and preimplantational development were determined in bovine SCNT embryos in the present study. Treatment of activated SCNT embryos with 10 ng/ml follistatin significantly increased the proportion of blastocyst development compared to untreated SCNT embryos. In addition, an increase in trophectoderm (TE) cell numbers and relatively higher proportion of TE cells to total cells were observed, but the number of inner cell mass (ICM) cell and total cell numbers were not changed (P < 0.05). No significant effect of other doses of follistatin was observed for the above endpoints. However, treatment with 1 and 10 ng/ml follistatin reduced the proportion of nuclear transfer blastocysts with an ICM ratio of > 60% relative to untreated nuclear transfer blastocysts at Day 7. No significant effect of follistatin treatment on proportions of nuclear transfer blastocysts with ICM ratio of 20-40% or 40-60% was observed. Taken together, these results suggested that follistatin can be used to increase developmental competence of SCNT embryos in terms of cell allocation, particularly TE cells, during preimplantation stages, subsequently enhancing placentation and birth of live offspring.

  • The indica ${\times}$ japonica rice (Oryza sativa L.) cultivars released in Japan since 1980 are high-yielding. However, occasionally in these cultivars their high yield potentials cannot be realized, particularly depending on climate conditions. The objectives of this study were to identify the reason for yield instability and the critical yield component factor causing lower grain yield in these high-yielding cultivars. Standard-yield japonica, high-yielding japonica-dominant, and indica-dominant cultivars were grown in western Japan. Rough grain yield (RY) in these high-yielding cultivars ranged from 450 to $980g\;m^{-2}$, and was positively correlated with potential grain yield (PRY). By fluctuations of solar radiation, RY changed with spikelet number (SNO), and SNO was correlated with cumulative radiation during the panicle formation period of 30 days before heading. Even if higher SNO was achieved under higher radiant conditions, RY was lower than PRY. The lower grain-filling in plants bearing higher SNO resulted from a lower filling percentage of spikelets (%F, RY/PRY), and %F was strongly correlated with spikelet fertility (%S) across all cultivars. %S was highly influenced by cumulative radiation per PRY during pollen development and establishment around heading. Inhibition of assimilation by leaf removal lowered %S. Conversely, stem thinning and removal of upper panicles around heading increased %S in spikelets of the lower part of the panicle in which sterility was higher. These results suggest that limitation of assimilate-supply due to low irradiance at the spikelet-formation and flowering stages restrains the potential of the high-yielding cultivars, depending on reduction of SNO and %S, respectively.

The Hospital Life of the Patient with Femoral Neck Fracture (대퇴경부 골절 환자의 입원 생활)

  • Nowerdays, the increase of traffic accidents and old age population make the Femoral Neck Fracture(FNF) patients increase. By the improvement of education and standard of living the patients demand better medical service than before. This study is designed to give practical help for the FNF patients by observing their hospital life and establish practical nursing strategies for the FNF patients. For these purposes the Ethnographic Participant Observation was adopted. By this study is focused on the hospital life patient's view. For this end, the field study adopted orthopedic ward in the C University Hospital with 400 beds in Seoul. The object patients of the study were twelve patients. The patients experienced five stages : Embarrassment, Conflict, Stability, Independent, and Extension Stage. The findings and prepared nursing strategies are stated as follows. First, in the Embarrassment Stage they suffered embarrassment, anxiety, pain, they could not do ordinary things. The patients who accidental fractures had anxiety from unfamiliar tests and from hospitalization itself. They lamented that they could not ordinary things, and do nothing but obeying the hospital, and endure the pain. They recognized the changed environment and resigned themselves to life in the ward. In this stage, full openness by the nurses is needed. Second, the attribute of the Conflict Stage were conflict, fear, curiosity, belief, reflection. When they sign the consentment form, they experience conflicts about the possibility of complication, fear of recovery from anesthesia, curiosity about the operation procedure, post - operation state, reflection on their past life, and promise to care for their family members after discharge and keep their religious life faithfully. And they accepted the operation depending on God, believing in modern medicine, and the surgeon. Asking for their changed informations, they expected positive results from the operation. In this stage, an empathic attitude by the nurses is needed. Third, the attribute of the Stability Stage were relief, gratitude, difficulty with excretion, and pain. When they awoke from anesthesia, they felt relief because of a the end of the operation, but they experienced extreme pain, difficulty of excretion in bed. They accepted the changed environment and expected recovery. In this stage, support by the nurses is needed. Fourth, the attributes of the Independence Stage were freedom, exercise, nurturing, anxiety, and discomfort. When they ambulated and exercised, they experienced freedom. They showed exhibited weakness of the digestive organs and discomfort hospital's space, structure, and facilities, the delay of medical certificate issue the lack of prompt response by the medical agents. They ate nurturious food and felt anxiety on the end of hospital life and returning to their ordinary life. They showed the independence of overcoming their environment by increasing exercise and expected their discharges. In this stage, respect by the nurses is needed for the patients to, overcome their environment and prepare for their independence. Fifth, the attributes of the Extension Stage were pessimism, isolation, dissatisfaction, and pain. Accompanied injury and old age made their ward life extend to over seven weeks. They exhibited weariness, melancholy, skeptisis, general pessimistic feeling, and desperation caused by their isolated life. They experienced the digestive discomfort caused by the prolonged medication and psycological pain caused by long-time hospitalization. As a, result, their dissatisfaction on the human, physical, and systematic environments had been increased. They acquired critical power and sought for something to do spending their time. They expected vaguely about the returning of their ordinary life. In this stage, counseling is needed by the nurse to overcome positively their psychological, social, and physical problems. The process of the FNF patient's ward life starts from the dependent state, when they are hospitalized, and gradually progresses to self-fulfillment in order to keep independent life. As a result, the FNF patients showed "Response in Challenge" or "Adaptation in Conflict" through their experiences of social, physical, and psychological difficulties.

Effectiveness of stages-matched educational program for cervical cancer screening among adult women in a community (일개 지역사회 여성 주민의 자궁경부암 조기검진 수검에 관한 행동변화단계별 교육 프로그램의 효과)

  • Background: Even though cervical cancer poses a significant global cancer burden, successful implementations for early detection offer an opportunity to prevent deaths and reduce the cancer burden. In Korea, cervical cancer is the most prevalent type of cancer among adult women, but it is one of the few cancers in which a consensus-approved screening test exists for early diagnosis, Pap test, that can be combined with highly efficacious treatment regimens for early-stage disease. Purpose: This study was carried out to identify the cognitive-behavioral factors associated with cervical cancer screening behavior among adult women, aged 40 to 59, and to develop tailored messages and to evaluate the effectiveness of stage-matched educational program. Method: A total of 283 women who aged 40 years or older was recruited in Seoul, from September, 1st to November, 14th, 2003. The intervention group (N=162) and the control group (N=121) were selected from five sub-districts in Seocho-gu, Seoul. Building on the TTM, a quasi-experimental study was conducted to test the effectiveness of stages-matched intervention addressed at the five stages of cervical cancer screening behavior. Women in the intervention group were randomly assigned to one of two conditions, internet or postal services. Results: In our results, 88.9% of participants had received a Pap test at least once in their life-time, and 65.4% had got it in the past two years. With regard to cognitive-behavioral factors, the stages-matched educational program increased attitude and process of change for cervical cancer screening. The percentage changed was the largest in maintenance stage. With regard to delivery methods for tailored messages, the print materials were more effective at increasing screening adherence than the e-mail. Whereas the postal service group showed remarkable the change of behavior stage, the internet service group did not. Also it was not shown any difference of the satisfaction with stages-matched educational program between internet and postal service groups. Conclusion: This study suggested that cervical cancer screening behavior could be changed by tailored messages which had developed with cognitive-behavioral factors. The stages-matched educational program was effective to promote the screening adherence for cervical cancer.

A Study on the Research Support Services of the University Library based on the Research Life Cycle (연구 수명주기 기반 대학도서관의 연구지원서비스 분석 연구)

  • As the research environment, such as Open Science, has recently changed, the scope of research support services conducted by university libraries is also expanding. In particular, there is a movement that needs to develop research support services of university libraries that are suitable for this through a clear understanding of the research life cycle centering on overseas. The purpose of this study is to provide suggestions that university libraries can refer to when developing research support services that can systematically support all phases of the research life cycle. To this end, the research life cycle was divided into five stages, "planning," "project," "publishing," "preservation" and "disseminating" and the research support services provided at each stage were examined around the cases of U.S. and domestic university libraries. Based on the implications obtained from the case, a service plan was proposed for domestic university libraries to systematically support each phase of the research life cycle.

Cloning and Characterization of Porcine Uroplakin II Gene

  • Mammalian urothelium undergoes unique membrane specialization by making the asymmetric unit membrane (AUM) that is covered with the apical cell surface during terminal differentiation. The AUM contains several major integral membrane proteins including uroplakin Ia, Ib, II and III. The genes for uroplakins have been cloned from humans and mice, but not from porcine. In this study, we report the cloning of the UPII genomic DNA, which codes for the full length open reading frame for the uroplakin II protein. The deduced amino acid sequence encodes of a hydrophobic NH$_2$-terminal peptide, a prosequence, and a mature protein. The prosequence contains three potential N-glycosylation sites and a RGRR cleavage site that may be involved in uroplakin II processing and maturation. Northern and immunohistochemistry analyses showed that the porcine UPII gene is only expressed in urothelium and that the protein was specifically localized in urothelial superficial cells. A 2kb of upstream in the promoter sequence contains multiple transcription factor binding sites, including GC-box, SPI, AP2, and GATA-box sites, but not for TATA or CAAT-box sequences. Comparison of the porcine UPII promoter sequence with that of the murine by MEME system presented two conserved motifs, suggesting a cis-acting regulatory role for the conserved sequences. Sequence homology between two species in motif A and B was 79% and 80% respectively, although their relative locations were different. During the gestation, mouse bladder at estrus stages and day 10 after parturition showed higher UPII expression, while showed lower expression at peri-implantation stage. Taken together, our results showed that the porcine UPII gene was expressed highly and specifically in the bladder urothelium and that steroid hormones for implantation changed the expression of UPII in the bladder, although the biological significance of UPII remains to be not determined.
