Life Cycle of Heterotrophic Dinoflagellate Cryptoperidiniopsis brodyi (Dinophyceae) (Cryptoperidiniopsis brodyi (Dinophyceae)의 생활사)

  • Pfiesteriaand Pfiesteria-like organisms were reported to be linked to major fish kills(involving well over a billion fish) in North Carolina and Maryland estuaries on the U.S. east coast during the 1990s. Occurrences of these species have been recently reported from Korean waters including Chinhae Bay and the coast of Yeosu. In this study, the life cycle of Cryptoperidiniopsis brodyi and Pfiesteria piscicida were examined using DAPI staining. Their excystment and growth were stimulated directly by the addition of prey cells such as Rhodiminas salina. Amoeboid stages in C. brodyi and P. piscicida were never observed in culture, even after addition of filter-sterile fish mucus and tissue. The dominant life cycle stages consisted of motile flagellated zoospores and cysts. A typical dinoflagellate life cycle was demonstrated by direct observation and DAPI staining.

  • As over 70% of the total life cycle cost (LCC) of a product is committed at the early design stage, designers are in an important position to substantially reduce the LCC of the products they design by giving due to life cycle implications of their design decisions. During early design stages, there may be competing concepts with dramatic differences. In addition, the detailed information is scarce and decisions must be made quickly. Thus, both the overhead in developing parametric LCC models fur a wide range of concepts, and the lack of detailed information make the application of traditional LCC models impractical. A different approach is needed, because a traditional LCC method is to be incorporated in the very early design stages. This paper explores an approximate method for providing the preliminary LCC, Learning algorithms trained to use the known characteristics of existing products might allow the LCC of new products to be approximated quickly during the conceptual design phase without the overhead of defining new LCC models. Artificial neural networks are trained to generalize product attributes and LCC data from pre-existing LCC studies. Then the product designers query the trained artificial model with new high-level product attribute data to quickly obtain an LCC for a new product concept. Foundations fur the learning LCC approach are established, and then an application is provided.

Teaching Korean Living through the Method of Currere (쿠레레(Currere) 방법을 활용한 한국의 실생활문화 교육방안)

  • This paper aims to propose teaching plans based on individual learner's experience and from a cross-cultural viewpoint by applying Currere as a method of Korean life-culture education. Current Korean culture education programs are not systematic, and are being executed without differentiation from language education. Thus, this study proposes Korean life-culture teaching plans based on the method of Currere. We suggested Currere teaching-learning stages and strategies for Korean language learners by adapting Pinar's and Jeong Seong-a's method of Currere. The Currere teaching-learning stages consisted of introduction, regression, progression, theme analysis, data analysis, group discussion, and integration. Although the method of Currere is implemented through autobiographic writing, this study added the strategies of 'personal experience telling' and 'schema utilization and background knowledge provision'. In order to enhance the applicability of Currere to Korean life-culture education, we suggested teaching plans for the theme of "How to dispose rubbish" as an example. We suggested detailed teaching plans that teachers can apply in the actual classroom. We expect that these teaching plans may be applied to actual classes so that Korean culture education may not be limited to the acquisition of knowledge, but be linked to their real life.

  • Accelerated life test is a viable method for identifying failure modes, incorporating design changes on an on-going basis during the early stages of automotive development program. The information from tests at high stress levels is extrapolated to obtain estimate of life at normal stress levels. This paper presents a practical method for accelerated life test to achieve a specified accuracy in estimating life at a design stress. Recommended and optimum plan are presented and the plans are illustrated with a simulated test data for the automotive power element example.

Studies on the organic acids metabolism in chlorella cells. (Chlorella의 유기산대사에 관한 연구)

  • Using the synchronous culture method and the manometric technique, changes in respiratory activities, utilization of some organic acids (succinate, malate, lactate and acetate etc.) and its effect on glucose metabolism in Chlorella cells at different growing stages were measured. 1) Endogenous respiration of the cells was not active at growing stage and was almost constant throughout the early ripening, maturing and division stages. 2) Lactate was utilized as respiratory substrate better than other organic acids tested. Exogenous respiration of glucose was most active at growing and maturing stages and was decreased strikingly at division stage. 3) Succinate and citrate inhibited endogenous and glucose respiration of the cells throughout the all life cycle. 4) Malate and acetate were utilized in the cells at early growing and division stages better, and malate enhanced the glucose respiration while in case of acetate it was depressed. 5) Calcium ion inhibited not only permeability of respiratory substrate but endogenous respiration itself.

The Quality Comparison of Grape Vinegar by Two Stages Fermentation with Traditional Grape Vinegar (2단계 발효에 의한 포도식초와 재래식 포도식초의 품질 비교)

  • 편이었다. 미량성분으로는 K이 다량으로 존재하였고, 2단계 발효로 제조된 포도식초는 포도양파식초 보다K, Na, Cu함량이 높게 나타났다. ^x Grape vinegar (A) and grape onion vinegar(B) added (3% of onion juice) were produced through two stages of fermentation(alcohol fermentation and acetic acid fermentation) to increase the grape's use. Grape wine which contained 5.6% alcohol was produced on the 3rd day of first stage. Then through the second stages, grape vinegar, of which total acidity was 5.37% was produced. The quality of (A) and ( B) which was produced through the two stages of fermentation was compared with the traditional grape vinegar (C, D) in the market. The content of sugar in (D) was a little higher such as 5.4 $^{\circ}$Brix than others. That of (A) and (B) was 5.13, 4.98 $^{\circ}$Brix respectively. The content of remaining alcohol in (C) was high such as 0.23% comparatively. But there was no remaining alcohol in (A) and (B). The content of acetic acid was 4.3~5.3% as a major organic acid of vinegars. The content of tartaric acid was 340.0 in (A), 315.7 in (B), 322.6 in (C) and 391.7mg% in (D). The content of lactic acid was distinctly high such as 277.4mg% in (D) There were differences such as 9.2~15.5mg% in the content of total free amino acids among grape vinegars. (D) contained 15.5mg% of total free amino acid and (B) also highly such as 12.0mg%. Potassium was high in grape vinegars. The content of potassium, sodium and copper was higher in (A) than (B).

Spermatogenesis in three Korean shrews and notes on their phylogenetic significance (한국산 땃쥐류 3종의 정장형성 및 이들의 계통 유연관계에 대한 소고찰)

  • The cycle of the seminiferous epithelium and spermiogenesis in three species if the genus Crocidura, the lesser white-toothed shrew, C. suaveolens, the Japanese white-toothed shrew, C. dsinezumi and the big(=Ussuri) white-toothed shrew C. lasiura, in the breeding season were studied with light and electron microscopes. The three species examined are distinguished from each other in the morphology of the seminiferous epithelium and the spermiogenesis, suggesting that these morphological characteristics are useful for the identification of the species. C. dsinezumi and C. lasiura, however, share many characteristics which are not common in C. suaveolens, as follows: In both species, 1) the cycle of the seminiferous epithelium is composed of 10 stages against 11 stages in C. suaveloens; 2) the earliest intermediated type spermatogonia is observed at stage I against stage III in C. suaveolens; 3) the spermatids of step 5 is observed during the stages V-VI against stages V-VII in C. suaveolens; 4) the acrosomal extension occurs during the stages VIII-X against tages IX-XI in C. suaveolens; 5) the condensation of the nucleus occurs simultaneously whereas it begins from the middle and along the nuclear membrane in C. suaveolens; 6) the capitular length in acrosome phase is shorter(about 2/3 of the diameter of the proximal centriole) than in C. suaveolens(longer than the diameter of proximal centriole; 7) length of the post nuclear cap is shorter(less than a half of the nucleus) than in C. suaveolens(about a half on the nucleus). Hudging from the similarities in the spermatogenesis in C. dsinezumi and C. lasiura, the relationship between them seems to be close compared to those with C. suaveolens.

A comprehensive longitudinal study of gut microbiota dynamic changes in laying hens at four growth stages prior to egg production

  • Objective: The poultry industry is a primary source of animal protein worldwide. The gut microbiota of poultry birds, such as chickens and ducks, is critical in maintaining their health, growth, and productivity. This study aimed to identify longitudinal changes in the gut microbiota of laying hens from birth to the pre-laying stage. Methods: From a total of 80 Hy-Line Brown laying hens, birds were selected based on weight at equal intervals to collect feces (n = 20 per growth) and ileal contents (n = 10 per growth) for each growth stage (days 10, 21, 58, and 101). The V4 regions of the 16S rRNA gene were amplified after extracting DNA from feces and ileal contents. Amplicon sequencing was performed using Illumina, followed by analysis. Results: Microbial diversity increased with growth stages, regardless of sampling sites. Microbial community analysis indicated that Firmicutes, Proteobacteria, and Bacteroidetes were the dominant phyla in the feces and ileal. The abundance of Lactobacillus was highest on day 10, and that of Escherichia-shigella was higher on day 21 than those at the other stages at the genus level (for the feces and ileal contents; p<0.05). Furthermore, Turicibacter was the most abundant genus after changing feed (for the feces and ileal contents; p<0.05). The fecal Ruminococcus torques and ileal Lysinibacillus were negatively correlated with the body weights of chickens (p<0.05). Conclusion: The gut microbiota of laying hens changes during the four growth stages, and interactions between microbiota and feed may be present. Our findings provide valuable data for understanding the gut microbiota of laying hens at various growth stages and future applied studies.
