Use of .lambda.gt 11 and antibody probes to isolate genes encoding RNA polymerase subunits from bacillus subtilis

  • A genetic analysis of the complex Bacillus subtilis transcriptional apparatus is essential to understand the function, regulation, and interaction of the transcriptase components during growth and sporulation. This approach in Escherichia coli has uncovered fundamental mechanisms regulating gene expression Cole and Nomura, 1986; Lindahl and Zengel, 1986) and an analysis of the B. subtilis transcriptase will allow comoparison of the E.coli system to another bacterium that has evolved under different selective pressures. To this end we used antibody probes to isolate the alpha, beta, and beta' core subunit genes from a .lambda.gtill expression vector library. To address the question of function ans regulation of the minor sigma factors that confer promoter specifity on the polymerase core (Losick et al., 1986), we used the same approach to isolate the gene for the 37,000 dalton sigma factor, sigma-37.

Isolation and Characterization of New Family Genes of DNA Damage in Fission Yeast

  • The SNF2 family includes proteins from a variety of species with roles I cellular processes such as transcriptional regulation, recombination and various types of DNA repair. Several proteins with unknown function are also included in this family. Here, we report the cloning and characterization of hrp 2+ gene (helicase related gene from S. pombe) which was isolated by PCR amplication using the conserved domain of SNF2 motifs within the ERCC6 gene which encodes a protein involved in DNA excision repair. The hrp2+ gene was isolated by screening with yeast S. pombe genomic library. The isolated cloned contained 6.5 kb insert DNA. Southern blot analysis confirmed that S. pombe chromosome contains the same DNA as hrp2+ gene and this gene exists as a single copy in S. pombe genome. The 4.7 kb transcript of mRNA was identified by Northern blot. To examined the transcriptional regulation of hrp2+ gene, DNA damaging agents were treated. These results indicated that the hrp2+ gene may not be directly involved in DNA replication, but may be involved in damage response pathway.

  • The signaling components of high affinity IgE receptor (Fc RI) were searched by yeast-hybrid screening of the cDNA library constructed from RBL-2H3 cells. The cytoplasmic part of the Fc RI- chain was found to specifically interact with PLC 2, and further comparatives studies were conducted focusing on the differential regulation of two PLC- isoforms through Fc RI. The inhibitors of Src, Syk, and protein kinase C similarly affected the tyrosine phosporylations of PLC 1 and PLC 2 but the inhibitors of PI3-kinase and p42/44 ERK effectively inhibited the activation of PLC 1 but not PLC 2.   • The purpose of this study is to find the ways that will be helpful to improve archival system and to establish National Archives. The contents of this study were focused on comparing the characteristics of archival system in governmental administration,: Executive, Legislature, and Judiciary. They were also focused on analyzing the problems of those current system. This research was basically conducted depending on the detailed articles of the legislation and regulation pertinent to the archives. Two major results of the research are 1) There are much differences among the governmental administrations in structuring and organizing for archival administrations. Archival works of government document are divided primarily according to the period of conservation and it is necessary to establish the regional archives and central management system, and to employ archivists as an expert staff. 2) The principles and methods of archival process, such as transferences, classifications, preservations, access, and destructions are similar to each other. In order to improve and co-ordinate current systems, it is necessary to constitute several councils endowed with consultative and decision-making power.

  • Today. the value of the references comes to be high and the reference database construction is increasing. But. the descriptive regulations of the reference which it presents from scholarly journals different with each other. Also, the researchers probably does not recognize a descriptive regulations of reference. It is likely that the effective construction of reference database is very difficult. So, the purpose of this study analyzes the differences of a descriptive regulations for reference and presents the plan for standardization of a descriptive regulations.

  • This study intends to analyze issues of purchasing process in public libraries by the 2014 revised Publication Industry Promotion Agency of Korea and fixed book price. Furthermore, this study suggests future tasks for restoring the intent of revising the laws and toward a better book purchases. The main contents of this study are as follow: 1) preparing the effective methods for selecting book vendor with evaluating the performance not price competition, 2) establishing concept and scope of local bookstore as book vendor for restoring the intent of the activation policy of local bookstore, 3) establishing the well-defined regulation about benefit of the economic dimension.

A Study on the Archives Management of $Chos\v{o}n-chongdokbu$ (일제 조선총독부의 문서관리에 관한 연구)

  • Government archives are basic source materials in the historical study. Whereas, researches on the archives of Choson-chongdokbu have been very scant. Those archives contain information about both colonism of Japan and independent movement of Korea are comparatively well preserved at Government Archives and Record Services. Therefore, it is necessary to analyze the characteristic of archives and to evaluate the system of archival management of Choson-chongdokbu. Based on the official gazette and archives of Choson-chongdokbu, this study has revealed the division of archival work, the writing and treatment of official document, compilation and preservation, and the procedure of use and destruction. As a result of the research, the followings have been identified; 1. Archival regulations were prepared by the instructions of Choson­chongdokbu at the early days of its establishment. Inspite of several revisions, its basis was maintained until 1945. 2. Each agency had its own archival department instead of central archives. 3. The archival department of Choson-chongdokbu controlled the archival administration of headquarters and attached agencies. But the secret archives were managed by the secretarial department. 4. The writing and treatment of archives were done by the archival regulation made in 1911. 5. Each agency had set more or less different preservation period. 6. There were three ways of using archives, 1) the in-house reading 2) the loan 3) the transcriptions. The archives passed the preservation period were destroyed in accordance with the related departments.

  • Norepoine is N-methylated by the enzyme phenly ethanolamine N-metyltransferase(PNMT)to form epinephrine.this enzyme is larhly restructed to the adrenal medulla where epinephrine in mammalian brain where epinephrine function as a neurotransmitter.It seems clear that central epinephrine is involved in the regulation of cardiovacular function and in several forms of hypertension.However,information about the struture of mammalian epinephrine forming enzyme has been limited until now.But recently we isolate bovine and human PNMT cDNA clone using gtll expression library and sequcde total nucleotide composition.To obtain information about the structrue of the human and rat PNMT proteins and gones and to further define the extent of the evolutionary relationships among the PNMT molecules of these species human and rat genomic DNA clones to PNMT were sequentially isolated and characterized.

Identification of Small GTPases That Phosphorylate IRF3 through TBK1 Activation Using an Active Mutant Library Screen

  • Interferon regulatory factor 3 (IRF3) integrates both immunological and non-immunological inputs to control cell survival and death. Small GTPases are versatile functional switches that lie on the very upstream in signal transduction pathways, of which duration of activation is very transient. The large number of homologous proteins and the requirement for site-directed mutagenesis have hindered attempts to investigate the link between small GTPases and IRF3. Here, we constructed a constitutively active mutant expression library for small GTPase expression using Gibson assembly cloning. Small-scale screening identified multiple GTPases capable of promoting IRF3 phosphorylation. Intriguingly, 27 of 152 GTPases, including ARF1, RHEB, RHEBL1, and RAN, were found to increase IRF3 phosphorylation. Unbiased screening enabled us to investigate the sequence-activity relationship between the GTPases and IRF3. We found that the regulation of IRF3 by small GTPases was dependent on TBK1. Our work reveals the significant contribution of GTPases in IRF3 signaling and the potential role of IRF3 in GTPase function, providing a novel therapeutic approach against diseases with GTPase overexpression or active mutations, such as cancer.
