Efficient Generation of Spatiotemporal Images for Leukocyte Motion Detection in Microvessels

  • This paper presents an efficient method for generating spatiotemporal images in order to detect leukocyte motion in microvessels. Leveraging the constraint that leukocytes move along the contour line of the blood vessel wall, our proposed method efficiently generates spatiotemporal images for leukocyte motion detection. To that end, translational motion caused by in vivo movement is first removed by a template matching method. Second, the blood vessel region is detected by an automatic threshold selection method in order to binarize temporal variance images. Then, the contour of the blood vessel wall is expressed via B-spline function. Finally, using the detected blood vessel wall's contour as an initial curve, the plasma layer for the most accurate position is determined in order to find the spatial axis via snake, and the spatiotemporal images are generated. Experimental results show that the spatiotemporal images are generated effectively through comparison of each step with three images.

  • Erythrosine used as a colouring agent in drugs, cosmetics and foods in Korea, was examined for its effects on murine immune system and methemoglobin formation. As immunotoxicologic assay parameters, we adopted circulating leukocytes and immunoorgan weights for pathotoxicology, IgM plaque forming cells and arthus reaction for humoral immunity, delayed hypersensitivity reaction of cell mediated immunity and carbon clearacnce for macrophage function. Erythrosine's effects were observed as follows; 1. Ery throsine showed no significant effects on circulating leulocyte counts and relative immunoorgan weight. 2. Erythrosine diminished IgM plaque forming cells. 3. Erythrosine decreased arthus reaction, in the dose dependent manner. 4. Erythrosine had no significant effect on delayed hypersensitivity. 5. Phagocytic and corrected phagocytic index were not affected. 6. Methemoglobin content was similar in the test and control groups.

반사광을 이용한 다채널 임상화학분석기개발 (Development of a multi-channel clinical chemistry analyzer)

  • In this paper we report the device of a multi-channel clinical instrument developed for determi¬nation of the levels of the urinary urobilinogen, glucose, ketone, bilirubin, protein, ascorbic acid, nitrite, pH, occult-blood, specific gravity, and leukocytes semiquantitatively. The test parameters are expressed on the dry test strips as a range of color intensities by chemical reactions. The instrument measures the value of each substance by reading the reflectance light emanated from the test strips. We also designed the reagent strip cassette and loader in order to intercept the outside interference. The loader can be operated semi-automatically. The light source is consisted on light emitting diodes at three specific wavelengths (560 nm, 610 nm, 650 nm). Precision of the system was evaluated by testing a series of commercial control urine samples. Furthermore, the performance of the instrument was compared with two other test methods on the urine samples from 100 persons. Our results showed a good repeatability between tests and a satisfactory agreement between the readings by our instrument and visual evaluation.

Rheumatoid Fibroblast-like Synoviocytes Downregulate Foxp3 Expression by Regulatory T Cells Via GITRL/GITR Interaction

  • Fibroblast-like synoviocytes (FLS) colocalize with leukocyte infiltrates in rheumatoid synovia. Proinflammatory leukocytes are known to amplify inflammation by signaling to FLS, but crosstalk between FLS and regulatory T cells (Tregs) remains uncharacterized. To address this possibility, we cocultured FLS lines derived from arthritic mice with Tregs. FLS that expressed the ligand for glucocorticoid-induced TNF receptor family-related gene (GITR) decreased expression of Foxp3 and GITR in Tregs in a contact-dependent manner. This effect was abolished by blocking antibody to GITR. On the other hand, the Tregs caused the FLS to increase IL-6 production. These results demonstrate that inflamed FLS license Tregs to downregulate Foxp3 expression via the GITRL/GITR interaction while the Tregs induce the FLS to increase their production of IL-6. Our findings suggest that the interaction between FLS and Tregs dampens the anti-inflammatory activity of Tregs and amplifies the proinflammatory activity of FLS, thereby exacerbating inflammatory arthritis.

  • Lymphatic vessels are routes for leukocyte migration and fluid drainage. In addition to their passive roles in migration of leukocytes, increasing evidence indicates their active roles in immune regulation. Tissue inflammation rapidly induces lymphatic endothelial cell proliferation and chemokine production, thereby resulting in lymphangiogenesis. Furthermore, lymphatic endothelial cells induce T cell tolerance through various mechanisms. In this review, we focus on the current knowledge on how inflammatory cytokines affect lymphangiogenesis and the roles of lymphatic vessels in modulating immune responses.

  • This study was performed to investigate the immunological side effects of chloramphenicol (CAP) on olive flounder, Paralichthys olivaceus. To investigate immunological effects on olive flounder, we determined the changes of chemiluminescence (CL) response of flounder kidney-derived leucocyte after the treatment of CAP in vivo and in vitro. The CL activity was significantly decreased in a dose-dependent manner during the treatment of CAP in vitro. Similarly, a dose-dependent reduction of CL response, although not significant, were observed during the treatment of CAP in vivo. The results suggest that CAP reduced the function of flounder phagocytosis in vivo and in vitro, indicating the immunosuppressive ability of CAP.

Cloning and expression of cDNA for chemokine receptor 9 from Olive flounder, Paralichthys olivaceus

  • Cysteine-cysteine chemokine receptor 9 (CCR9) homologue cDNA was isolated from olive flounder leukocyte cDNA library. Olive flounder CCR9 homologue consisted of 1709 bp encoding 367amino acid residues. When compared with other known CCR peptide sequences, the most conserved region of the olive flounder CCR9 peptide is the seven transmembranes. A phylogenetic analysis based on the deduced amino acid sequence showed the homologous relationship between the olive flounder CCR9 sequence and that of Mouse CCR9. The olive flounder CCR9 gene was predominantly expressed in the Peripheral blood leukocytes (PBLs), kidney, spleen, and gills.

  • The authors have studied histopathologically, based with symptoms, on 51 aged woman who treated in infirmary hospital of dental college of Seoul National University. The results are as follows: 1. Symptoms of patient were swelling of lower left mandible, gingival enlargement, ulceration and severe bleeding. 2. We observed characteristic features on the blood test such as decreased leukocytes with severe decrease of granular leuckocytes and sudden increase of lymphocytes. 3. On the histopathological studies, we observed severe destruction of gingival tissue with few granular leukocytic infiltration, but displaced with massive lymphochtic infiltration.

Cloning and Characterization of the Putative Transferrin Receptor cDNA from the Olive Flounder (Paralichthys olivaceus)

  • A cDNA clone for the olive flounder (Paralichthys olivaceus) transferrin receptor (fTfR) was isolated from a leukocytes cDNA library. The fTfR gene consisted of 2,319 bp encoding 773 amino acid residues. The amino acid sequence alignment of the fTfR showed that their size and hydrophobic profile are similar. In addition, the Tyr-Thr-Arg-Phe (YTRF) motif that is the recognition signal for high-efficiency endocytosis, is conserved very well. This motif is important for functional properties of TfR. The deduced amino acid sequence had $42.4-42.9\%$ identities with the previously reported TfRs of vertebrates. The fTfR was expressed in the blood, kidney, spleen, and liver of healthy olive flounder by the Northern blot hybridization.

  • It is now well established that transient granulocytopenia is common in patients during the initial phase of hemodialysis with new cuprophane membrane, but not with reused nembrane, or some synthetic membranes. In this study, new cuprophane dialyzer was treated with plasma and albumin, respectively, then, peripheral blood leukocytes were counted at the onset and after 15 minute of hemodialysis. In the plasma-treated group, the percent of leukopenia was less (41.3 %) than control group (71.3 %). In the plasmatreated group, the degree of leukopenia was close to that ot'control grou p. The protective factor, therefore is thought to be present in the plasma other than albumin fraction.
