닭의 근섬유(筋纖維) 단백질(蛋白質) 돼지의 백혈구(白血球)에서 추출(抽出)한 lysosomal proteinase에 의해서 분해(分解)될 때 미치는 NaCl과 pH의 영향(影響)에 대해서 연구(硏究)하였다. Leucocyte lysosomal proteinase에 의한 근섬유(筋纖維) 단백질(蛋白質)의 분해(分解)는 다른 pH를 갖는 Tris-maleate buffer에서 partial hydrolysis로 진행(進行)하였다. 분해(分解)는 높은 pH에서 더욱 심화(深化)되었으며, 여기에 NaCl이 첨가(添加)되었을 때 proteinase의 activity는 pH의 고저(高低)에 관계(關係)없이 더욱 증가(增加)함을 보여주었다.
Behcet's disease (BD) is a chronic, multisystemic disorder which is more frequently seen in the Mediterranean basin, Middle East, and Far East. The causes and pathogenesis of BD are unknown although many possibilities are being investigated. The diagnosis of BD is based on clinical manifestations because there are no pathognomonic laboratory tests. So, the purpose of this study was to examine the prevalence of HLA-B51 in patients with BD. We used the whole blood of 33 patients diagnosed with BD at Chosun University Hospital from August 2003 to January 2006. For the HLA-B51 test, we extracted the DNA from the whole blood of 33 BD patients, and we investigated it through the nested PCR method. Data were analyzed using the SPSS/PC 10.0. The frequencies of gender of the 33 cases diagnosed as BD were male 13 (39.4%) and female 20 (60.6%). The frequencies of age group of the 33 cases diagnosed as BD were 20 yrs 8 (24.2%), 30 yrs 12 (36.4%), 40 yrs 8 (24.2%), 50 yrs 1 (3.0%), and 60 yrs and 70 yrs 2 (6.1%), respectively. The frequencies of HLA-B51 of the 33 cases diagnosed as BD were HLA-B51-negative 18 (54.5%) and HLA-B51-positive 15 (45.5%). In conclusion, BD occurred more often in women than men (1: 1.53), and the mean age of the BD patients was 39.8 years old. HLA-B51 was positive in 45.5% of patients with BD, and was statistically significant in age (p<0.05).
Rheumatoid arthritis (RA) is an autoimmune/inflammatory disorder with a complex genetic component. RA is characterized by chronic inflammation of the synovial membrane in the joint, which leads to the progressive destruction of articular cartilage, ligament and bone. Several cytokines such as tumor necrosis $factor-{\alpha}\;TNF-{\alpha}\;and\;interleukin-1{\beta}\;(IL-1{\beta})$ and interleukin-6 (IL-6) have been implicated in the pathological mechanisms of synovial tissue proliferation, joint destruction and programmed cell death in rheumatoid joint. Genome wide screening of subjects suffering from autoimmune diseases especially arthritis revealed linkage to inflammatory molecules like $TNF-{\alpha},\;IL-1{\beta}$ and IL-6, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), nuclear factor-kappaB $(NF-{\kappa}B)$ and human leucocyte antigen/major histocompatibility complex (HLA/MHC) locus. The status of the pharmacological mechanism of herbal drugs in the light of genome wide screening results has been discussed to reinforce the therapeutic potential and the pharmacological basis of the herbal drugs.
In order to compare the diagnostic value of Modified California Mastitis Test (MCMT), Modified Whiteside Test (MWT) and Resazurin Reduction Test (RRT) using the direct microscopic leucocyte count (DMLC) as standard, a total of 739 quarter milk samples were examined. The results obtained were as follows: 1. Of the 739 samples, 24.4% had positive DMLC value (over 500,000 leucocytes per mI.), 32.6% positive MCMT reaction, 34.9% positive RRT reaction and 39.9% positive MWT reaction. 2. The identical ratings of the three mastitis screening tests with DMLC values were 60.7% (MWT), 61. 8% (MCMT) and 72.1% (RRT). 3. The mean reaction values of the predicted mastitis screening tests were $1.09{\pm}0.01$ (MWT), $1.12{\pm}0.06$ (MCMT) and $1.25{\pm}0.40$ (RRT). The efficiency ratings of them were 34.8% (MWT), 49.3% (RRT) and 55.0% (MCMT) respectively.
This study was performed to investigate the immunological side effects of chloramphenicol (CAP) on olive flounder, Paralichthys olivaceus. To investigate immunological effects on olive flounder, we determined the changes of chemiluminescence (CL) response of flounder kidney-derived leucocyte after the treatment of CAP in vivo and in vitro. The CL activity was significantly decreased in a dose-dependent manner during the treatment of CAP in vitro. Similarly, a dose-dependent reduction of CL response, although not significant, were observed during the treatment of CAP in vivo. The results suggest that CAP reduced the function of flounder phagocytosis in vivo and in vitro, indicating the immunosuppressive ability of CAP.
HRccine was administered subcutaneously to rats for 4 weeks at dose levels of 300, 60 and 12 times the expected clinical dose to evaluate the subacute toxicity. There were no treatment-related effects in clinical signs, body weight changes, food consumption, water consumption, urinalysis and blood biochemistry in any dose groups. In hematological examinations, increase of leucocyte counts and decrease of hemoglobin concentration were observed in the high dose-treated group. However, no treatment-attributable pathological changes were observed in microscopic examinations. The no-effect dose in subacute toxicity study of rats was considered to be 300 times the expected clinical dose.
The methanol extract of Torilis Fructus showed potent anti-edematous effect in carrageenan-induced paw edema in rats. Systematic fractionation of the extract with hexane, chloroform, ethyl acetate and butanol resulted in potent anti-edematous action only in hexane fraction at an oral dose of 237 mg/kg which is corresponding to 4.8 g of raw Fructus. This hexane fraction showed inhibitory effects at 77 mg/kg p.o on vascular permeability in mice, at 8mg/pouch on leucocyte emigration in rats, and at 120 mg/kg p.o. on adjuvant arthritis model in rats. These results demonstrate that hexane fraction of the extract possesses potent anti-inflammatory effect which may support its traditional uses.
Single-cell gel electrophoresis (comet assay) was used to detect DNA single strand break in blood cells from several marine fish species. Three fish species were collected from Georgia coastal area. Mummichog, Fundulus heteroclitus showed higher DNA damage than sea bass, Lateolabrax japonicus and trout, Oncorhynchus masou masou under the same experimental conditions. Mummichogs had more alkaline-labile sites on their DNA than other fish species. The comet assay with mummichog blood cells at pH 12.5 showed a dose-response curve with the increasing concentrations of hydrogen peroxide. While the isolated leucocytes showed no increase of DNA damage after in vitro exposure to 2-methyl-1,4-naphthoquinone (MNQ), erythrocytes showed dose-dependent DNA damage. These results indicate that the comet assay can be applied successfully as a bioassay using erythrocyte for environmental monitoring.
The Forsythiae fructus is described to be used as an antiinflammatory drug, diuretics, antidotes and antibacterials in oriental literatures. In order to investigate the efficacy of Forsythiae viridissima (Oleaceae), the methanol extract and its fraction have been evaluated for the acute toxicity, antiinflammatory, analgesic and spasmolytic action in animals. The methanol extract of Forsythiae fructus was found to have significant antiinflammatory activity in the acute and subacute antiinflammatory model in rats, but have no analgesic action. Furthermore, through fractionation procedure, it was found that the active compounds were easily soluble in chloroform and butanol. It is also noted that the extracts had spasmolytic activities in the rat fundus and uterus and had low acute toxicity in mice.
We herein describe the case of a focal spontaneous spinal epidural abscess who was initially diagnosed to have a free fragment of a lumbar disc. A 71-year-old woman presented with history of low back and right leg pain. Magnetic resonance imaging suggested a peripherally enhancing free fragment extending down from S1 nerve root axilla. Preoperative laboratory investigation showed elevation of c-reactive protein (CRP), erythrocyte sedimentation rate (ESR) levels. She was taken for surgery and a fluctuating mass at the axilla of S1 nerve was found. When the mass was probed with a dissector, a dark yellow, thick pus drained out. Pus cultures were negative. Patients who present with extreme low back plus leg pain and increased leucocyte count, ESR and CRP levels should raise the suspicion of an infection of a vertebral body or spinal epidural space.
