Effect of Dietary Streptococcus faecium on the Performances and the Changes of Intestinal Microflora of Broiler Chicks (Streptococcus faecium의 급여가 육계의 성장과 장내 세균총 변화에 미치는 영향)

  • Effect of Streptococcus faecium(SF) and an antibiotic, Colistin(Col), supplemented to diets singly or in combination, on the performances and changes of intestinal population of microflora of broiler chicks studied. A total of 252, day-old chicks(Arbor Acre) of mixed sex(M:F=1:1) were alloted into six groups. A diet with no Col and SF was referred as a control diet. The basal diets were added with two levels of SF, 0.04 and 0.08%, singly or in combination with Col 10ppm Another diet was prepared by adding only Col 10 ppm. Numbers of the microorganism in diets added with SF 0.04% and 0.08% were 7$\times$10$^{4}$ and 1.4$\times$10$^{5}$ /g diet respectively The diets consisting of corn and soybean meal as major ingredients were fed for a period of seven weeks . During the feeding trial, fresh excreta were sampled at the end of every week in a sterilized condition to count microbial changes from each dietary group. Microbial changes of large intestine were also measured from nine birds sacrificed at the end of the 4th and 7th weeks each time per dietary group. Excreta from all the groups were also collected quantitatively at the end of 3rd and 6th weeks to measure digestibility of the diets, At the end of 7th week, nine birds from each group were also sacrificed to measure weight changes of gastrointestinal tracts . Average body weight gains of broilers fed the diets added with SF 0.08% (2.37kg) or SF 0. 08%+col 10ppm(2.34kg) were significantly larger than that of the control(2.18kg). The weight gains of the other groups were not statistically different from that of the control Feed/gain ratios of the supplemental groups were better than that of control (P<0.05) except that of birds fed the diet added only with SF 0.04%. Digestibilities of nutrients such as dry matter, crude protein, crude fat and total carbohydrates were not altered by the consumption of the diets added with SF and/or Col throughout the whole feeding period. As expected, the numbers of Streptococci in the excreta from birds fed diets added with SF increased significantly with a statistical difference between groups with SF 0.04% and SF 0.08% most of the time. However. addition of Colistin to the diets supplemented with SF did not give any effects on the number of the microorganism. Numbers of coliforms in the excreta were apparently reduced by feeding the diets added with SF and/or Col(P<0.05). There were, however, no additive effects observed between the two feed additives in this regard when supplementing Col to the SF diets. Distributions of intestinal microflora exhibited exactly the same pattern as those of the excreta. Length of small intestine of the birds fed diets added with SF 0.08% with or without Col 10 ppm became significantly longer with a range of about 10% than those of the birds fed diets without SF. However, the empty weight of the small inestine of the former group was lighter than that of control These changes resulted in a significant reduction in weight/unit length of the intestine of the birds fed diets supplemented with Col and SF singly or in combination. In overall conclusion, diet added with SF 0.08% appeared most effective in improving broiler performances. Colistin added at a level of 10ppm was not beneficial at all in itself or in combination with SF in terms of broiler performances or changes of intestinal microflora population. The efficacy of SF and Col could be attributed to the changes of wall thickness of the small intestine.

Phylogenetic and expression analysis of the angiopoietin-like gene family and their role in lipid metabolism in pigs

  • Objective: The objective of this study was to investigate the phylogenetic and expression analysis of the angiopoietin-like (ANGPTL) gene family and their role in lipid metabolism in pigs. Methods: In this study, the amino acid sequence analysis, phylogenetic analysis, and chromosome adjacent gene analysis were performed to identify the ANGPTL gene family in pigs. According to the body weight data from 60 Jinhua pigs, different tissues of 6 pigs with average body weight were used to determine the expression profile of ANGPTL1-8. The ileum, subcutaneous fat, and liver of 8 pigs with distinct fatness were selected to analyze the gene expression of ANGPTL3, ANGPTL4, and ANGPTL8. Results: The sequence length of ANGPTLs in pigs was between 1,186 and 1,991 bp, and the pig ANGPTL family members shared common features with human homologous genes, including the high similarity of the amino acid sequence and chromosome flanking genes. Amino acid sequence analysis showed that ANGPTL1-7 had a highly conserved domain except for ANGPTL8. Phylogenetic analysis showed that each ANGPTL homologous gene shared a common origin. Quantitative reverse-transcription polymerase chain reaction analysis showed that ANGPTL family members had different expression patterns in different tissues. ANGPTL3 and ANGPTL8 were mainly expressed in the liver, while ANGPTL4 was expressed in many other tissues, such as the intestine and subcutaneous fat. The expression levels of ANGPTL3 in the liver and ANGPTL4 in the liver, intestine and subcutaneous fat of Jinhua pigs with low propensity for adipogenesis were significantly higher than those of high propensity for adipogenesis. Conclusion: These results increase our knowledge about the biological role of the ANGPTL family in this important economic species, it will also help to better understand the role of ANGPTL3, ANGPTL4, and ANGPTL8 in lipid metabolism of pigs, and provide innovative ideas for developing strategies to improve meat quality of pigs.

Utilization of Graded Levels of Finger Millet (Eleusine coracana) in Place of Yellow Maize in Commercial Broiler Chicken Diets

  • An experiment was conducted to study the performance, carcass traits, serum lipid profile and immune competence in commercial broilers (2 to 42 d of age) fed graded levels (25, 50, 75 and 100%) of finger millet (FM) (Elusine coracana) in place (w/w) of yellow maize (YM). Each diet was fed to eight replicates (five female Vencobb broilers/replicate) housed in stainless steel battery brooders. The estimated metabolizable energy content of FM was about 540 kcal less than the YM. FM contained more protein (10.42 vs. 9.05%) and fibre (9.52 vs. 2.24%) compared to YM. Body weight gain, ready to cook yield, relative weights of giblet, liver, intestine and length of intestine at 42 d of age was not affected due to replacing YM with FM. But, the feed efficiency decreased in broilers fed diets containing 75 and 100% FM in place of YM at both 21 and 42 d of age. The amount of fat deposited in abdominal area decreased and the relative weight of gizzard increased with increase in level of FM in the diet. The serum HDL cholesterol at 21 and 42 d of age and serum triglycerides at 42 d of age decreased with increase in level of FM in diet. The relative weight of spleen and antibody titers against sheep red blood cells (SRBC) at 5 d post inoculation (PI) decreased in broilers fed FM at 100% of YM. However, the relative weight of bursa, SRBC titers at 10 d PI, antibody titers against ND virus and mortality were not affected due to incorporation of FM in place of YM in diet. The fat content in thigh muscle and liver decreased, while the protein content in these tissues increased with increase in the level of FM in broiler diet. Based on the results, it may be concluded that YM can be replaced with FM up to 25% on weight basis without affecting weight gain, carcass yields and immunity in commercial broiler diet (up to 42 d of age). Further, inclusion of finger millet reduced the fat deposition in thigh muscle, liver and in abdominal area compared to those fed maize as the principal source of energy.

  • The RLG (relative length of gut) is 1.52 (n=12) in the sablefish, Anoplopoma fimbria. The digestive tract has five or six pyloric caeca in the posterior region of stomach. Morphology of mucosal fold is unbranched type in the esophagus and stomach, but branched type in the intestine. The histological structure of digestive tract can be divided into mucosal layer, submucosal layer, muscular layer and serous membrane in the cross section. In the esophagus, mucosal epithelial layer is a simple, and consists of ciliated columnar epithelia and mucous cells. In the stomach, gastric gland of mucosal epithelial layer is a tubular, and is composed of chief cell, parietal cell and mucin secreting cell, which is columnar and contained secretory granules of red and blue colors in the AB-PAS (pH 2.5) reaction. In the intestine, mucosal epithelial layer is a simple, and consists of ciliated columnar epithelia and goblet cells. The submucosal layer is composed mainly of collagen fibers, and well developed in the esophagus. And the muscular layer of digestive tract is divided into longitudinal and circular muscle layer, and well developed in the stomach. The liver is composed of numerous lobular structure and bile canaliculi. Stainability of hepatocyte cytoplasm was eosinophilic, and nucleus and nucleolus showed basophilic in the H-E stain. The pancreatic tissue was scattered in the fatty tissue near the digestive tract, and acinar gland consisting of numerous exocrine cells. And cytoplasmic stainability of exocrine cell was basophilic, and contained numerous zymogen granules of eosinophilic in the H-E stain.

Clinical Practice of Blood Transfusion in Orthotopic Organ Transplantation: A Single Institution Experience

  • Background: Orthotopic organ transplantation, a treatment option for irreversible organ dysfunction according to organ failure, severe damaged organ or malignancy in situ, was usually accompanied with massive blood loss thus transfusion was required. We aimed to evaluate the adverse impact of blood transfusion on solid organ transplantation. Materials and Methods: From January, 2009 to December, 2014, patients who received orthotopic organ transplantation at Far Eastern Memorial Hospital medical center were enrolled. Clinical data regarding anemia status and red blood cell (RBC) transfusion before, during and after operation, as well as patient outcomes were collected for further univariate analysis. Results: A total of 105 patients who underwent orthotopic transplantation, including liver, kidney and small intestine were registered. The mean hemoglobin (Hb) level upon admission and before operation were $11.6{\pm}1.8g/dL$ and $11.7{\pm}1.7g/dL$, respectively; and the nadir Hb level post operation and the final Hb level before discharge were $8.3{\pm}1.6g/dL$ and $10.2{\pm}1.6g/dL$, respectively. The median units (interquartile range) of RBC transfusion in pre-operative, peri-operative and post-operative periods were 0 (0-0), 2 (0-12), and 2 (0-6) units, respectively. Furthermore, the median (interquartile range) length of hospital stay (LHS) from admission to discharge and from operation to discharge were 28 (17-44) and 24 (16-37) days, respectively. Both peri-operative and post-operative RBC transfusion were associated with longer LHS from admission to discharge and from operation to discharge. Furthermore, it increased the risk of post-operative septicemia. While peri-operative RBC transfusion elevated the risk of acute graft rejection in patients who received orthotopic transplantation. Conclusions: Worse outcome could be anticipated in those who had received massive RBC transfusion in transplantation operation. Hence, peri-operative RBC transfusion should be avoided as much as possible.

Molecular Cloning, Tissue Distribution and Segmental Ontogenetic Regulation of b0,+ Amino Acid Transporter in Lantang Pigs

  • Cationic amino acid transporter $b^{0,+}AT$ (HGMW-approved gene symbol SLC7A9, solute carrier family 7, member 9) plays a crucial role in amino acid nutrition. In the present study, we describe the cloning and sequencing of porcine $b^{0,+}AT$. Based on the sequence of porcine $b^{0,+}AT$ deposited in the NCBI (National Center for Biotechnological Information), we identified a putative porcine homologue. Using rapid amplification of cDNA ends (RACE), the full-length cDNA encoding porcine $b^{0,+}AT$ was isolated. The porcine $b^{0,+}AT$ cDNA was 1,680 bp long, encoding a 487 amino acid trans-membrane protein. The predicted amino acid sequence was found to have 88.9% and 87.1% identity with human and mouse $b^{0,+}AT$, respectively. Real-time RT-PCR indicated porcine $b^{0,+}AT$ transcripts expressed in heart, kidney, muscle and small intestine. The small intestine had the highest $b^{0,+}AT$ mRNA abundance while the muscle had the lowest (p<0.05). Along the longitudinal axis, the ileum had the highest $b^{0,+}AT$ mRNA abundance while the colon had the lowest (p<0.05). The $b^{0,+}AT$ mRNA level was highest on day 7 and 90 in the duodenum (p<0.05). It increased from day 1 to day 26 in the jejunum (p>0.05) and had the highest abundance on day 60 (p<0.05). There was, however, no difference between day 1, 7, 26, 30, 90 and 150 (p>0.05). The strongest $b^{0,+}AT$ expression appeared on day 7 in the ileum before weaning, and then decreased till day 30 but rose gradually again from day 60 to 150 (p<0.05).

Effects of Sodium Alginate and Cellulose on Gastrointestinal Physiology in Rats (Sodium Alginate와 Cellulose가 흰쥐의 소화생리에 미치는 영향)

  • To examine the effects of dietary fibers on gastrointestinal physiology, rats were fed with diets containing 10% sodium alginate,10% cellulose, or fiber-free diets for 5 weeks. The results obtained were as follows: The chronic consumption of sodium alginate induced a significant decrease in body weight gain and feeding efficiency, but a significant increase in length and weight of small intestine. Fecal bulk and weight were higher in fiber-fed group than fiber-free group. The chronic consumption of dietary fiber induced a significant increase in fecal output, resulting in tile decrease of apparent digestibility of protein and lipid. Pancreatic protease activity was lower in fiber-fed group than fiber-free group, whereas pancreatic amylase and lipase activities were not affected. Scanning electron microscopy(SEM) and light microscopy(LM) studies showed small intestine microvilli with numerous ridges and convolutions and goblet cells in fiber-fed groups. As a result of this study, it is concluded that the chronic consumption of dietary fiber decreases apparent digestibility of nutrients and induces morphological and biochemical adaptation of digestive organs.

Molecular Characterization of Metallothionein Gene of the Korean Bitterling Acheilognathus signifer (Cyprinidae) (묵납자루 (Acheilognathus signifer; Cyprinidae) metallothionein 유전자의 클로닝 및 특징 분석)

  • Genetic determinant for metallothionein (MT), a cysteine-rich protein playing essential roles in metal detoxification and homeostasis, was characterized in the Korean bitterling (Acheilognathus signifer, Cyprinidae), an endemic fish species. The full-length A. signifer MT (AsMT) cDNA (551 bp) is composed of a single open-reading frame (ORF) to encode a polypeptide of 60 amino acids containing 20 cysteine residues whose positions are conserved in most cypriniform MTs. At the genomic level, the AsMT (2,593 bp spanning the 5'-flanking region to the 3'-untranslated region) represented a conserved tripartite (three exons interrupted by two introns) structure with AT-rich introns. The upstream regulatory region (-1,914 bp from the ATG initiation codon) of AsMT displayed various sites and motifs for transcription factors involved in the metal-mediated regulation and stress/immune responses. The AsMT transcript was ubiquitously detected in various organs with variable expression levels, where the ovary and intestine showed the highest expression, while the heart and skeletal muscle represented the lowest level. During an exposure to copper (immersion in $0.5\;{\mu}M$ Cu for 48 h), the levels of AsMT transcripts were significantly elevated in the liver (more than 3.5-fold), moderately in the gill, kidney, and spleen (ranging from 1.5- to 2.5-fold), and barely in the brain and intestine. Results of this study could form a useful basis to explore the metal-related stress physiology of this endangered fish species.

Studies on the digestive gland structures of domestic and experimental animals I. Glandular tubule structures in the cecum and colon of piglets (가축 및 실험동물 소화선의 구조에 관한 연구 I. 돼지 맹장 및 결장 장선의 구조에 관하여)

  • The present study was focussed mainly on the morphological changes of the glandular tubules in the large intestine according to age of piglets. Samples were taken from large intesine of 1-, 10-, 20-, 35- and 45-day-old piglets, 2 to 3 piglets in each age group. The intestinal samples were fixed in 10% neutral formalin solution, dehydrated, and then paraffin sections were stained with H-E. The results observed were summarized as follows: 1. The mucosal glands in the cecum and colon tend to be unbranched simple straight tubular glands, or often two or more branched simple stright tubular glands. 2. The number of the longitudinal folds and the number of the crypts per cross section of piglet colons, respectively, were 1-day-old piglets-$3.8{\pm}0.8$, $92.1{\pm}6.9$; 10-day-old piglets-$7.1{\pm}1.1$, $164.2{\pm}10.3$; 20-day-old piglets-$15.2{\pm}0.8$, $178.5{\pm}6.8$; 35-day-old piglets-$19.3{\pm}3.0$, $454.9{\pm}25.3$; 45-day-old piglets-$20.6{\pm}3.1$, $524.6{\pm}37.2$, and the regression equation between age and these two number were $\hat{Y}=0.40X+4.32$ and $\hat{Y}=10.4X+51.52$, respectively. 3. The length and cell number per single side wall of a glandular tubule in the colon section were 1-day-old piglets-$196.3{\pm}7.1{\mu}m$, $40.0{\pm}3.3$; 10-day-old piglets-$236.0{\pm}34.5{\mu}m$, $47.9{\pm}5.3$; 20-day-old piglets-$262.8{\pm}39.6{\mu}m$, $54.3{\pm}9.0$; 35-day-old piglets-$291.75{\pm}48.3{\mu}m$, $56.9{\pm}4.9$; 45-day-old piglets-$364.8{\pm}61.5{\mu}m$, $67.7{\pm}7.4$, respectively, and the regression equation between age and these two data were $\hat{Y}=3.45X+193.8$ and $\hat{Y}=0.56X+41.0$, respectively. 4. The overall percentages of the cell number and length of glandular tubules in piglet colons were the pit and isthmus-$75.3{\pm}11.1%$, $78.8{\pm}12.3%$; gland-$24.7{\pm}5.4%$, $21.2{\pm}5.3%$, respectively. 5. The length and cell number of single side wall of glandular tubules in cecal sections were 1-day-old piglets-$190.3{\pm}31.1{\mu}m$, $37.6{\pm}4.8$; 10-day-old piglets-$235.6{\pm}25.3{\mu}m$, $46.2{\pm}3.6$; 20-day-old piglets-$295.3{\pm}45.6{\mu}m$, $52.0{\pm}6.2$; 35-day-old piglets-$351.3{\pm}28.3{\mu}m$, $60.4{\pm}8.5$; 45-day-old piglets-$366.3{\pm}48.5{\mu}m$, $64.7{\pm}8.2$, respectively, and the regression equation between age and these two data were $\hat{Y}=4.11X+196.6$ and $\hat{Y}=0.60X+38.9$, respectively.

Feeding and Digestion by Postlarvae and Juveniles of the Flounder, Paralichthys olivaceus (넙치, Paralichthys olivaceus 자어 및 치어의 섭식과 소화)

  • There are many problems to be solved for the establishment of more feasible and simplified method of seed production of marine fishes. One of the most important tasks in seed production is to clarify the feeding ecology of larvae and juveniles under. rearing conditions. In the present study, two groups of postlarvae-juveniles of the flounder, Paralichthys olivaceus were used. One ranging 11.38 to 17.08 mm in mean total length was fed with Artemia nauplii at two different water temperatures (19 to $21^{\circ}C$ and 24 to $25^{\circ}C$) and the other ranging 4.39 to 10.64 cm in mean total length was fed with prepared diet. Time required from the start of feeding to satiation was 45 to 55 minutes at 24 to $25^{\circ}C$ and nearly one hour at 19 to $21^{\circ}C$. Mean numbers of Artemia nauplii in digestive tract were 236 to 375 individuals per fish at 24 to $25^{\circ}C$. The amount of food remained in the intestine was $24.8\%$ of the total food in the digestive tract. In juveniles of 4.39 to 10.64 cm in mean total length, amount of prepared diet consumed at satiation was 0.04 to 0.46 g per fish. Digestive tract index in juveniles was rapidly decreased for 24 hours after satiation and kept nearly constant after 48 hours, showing that the time to completely digest the prepared diet in juveniles was estimated to be about 48 hours.
