쪽 色素에 의한 絹纖維 染色에 관한 硏究 -발효 염색에 대하여- (A Study on the Silk Dyeing With Natural indigo Extracted from Polygoum tinctorium -On the fermentation dyeing-)

  • Colorants were prepared by extraction of natural indigo which was harvested just in the blooming season(in the late of July). 100 g of fresh leaves soaking in 1 ιwater was kept at 3$0^{\circ}C$, 30 hours. A solution of 3g/l calcium hydroxide was added into it to precipitate dye substance and it was freezing-dried into powder form. The fermentation and dyeing conditions were investigated. The results obtained are summarized as follows; K/S value of dyed silk fabrics of fermentation conditions was higher at 95$^{\circ}C$ for 20 min. than at 4$0^{\circ}C$ for 20 hours. Furthermore, K/S value of dyed silk fabric was raised by the addition of 5g/l of glucose and 5g/l of NaOH. K/S value of dyed silk fabric was raised by the addition of 5g/l of glucose and 5g/l of NaOH. K/S value increased as extending of dyeing time when dyed till 2 hours at 3$0^{\circ}C$. K/S value decreased in order of 3$0^{\circ}C$, 4$0^{\circ}C$ and 5$0^{\circ}C$, at the various dyeing temperatures and dyeing concentrations, and colour fastness ranged from 4 to 5 grade in terms of washing, perspiration and light fastness.

조선 후기 왕실 복식의 염색 문화 (The Dyeing Culture of Royal Garments in the Late Joseon Dynasty)

  • This study explores the culture surrounding the dyeing of royal garments in the late Joseon Dynasty. The findings of this study are as follows. First, several dyes were used to color royal garments, such as jicho, honghwa, danmok, simhwang, sambo, goehwa, chija, and namjong. Mordants such as maesil, hwanghoemok, yeohoe, and baekban were also used with the dyes. Second, the Sangeuiwon (尙衣院) was the department in charge of the purchasing of dyes and the entire dyeing practice. It was the Seonhyecheong (宣惠廳) and the Hojo (戶曹) who provided revenue to the Sangeuiwon through a wongong, regular tribute, and a bokjeong (supplementary tribute). Additionally, additional dyes, if found to be insufficient, could be provided by the Hojo. Every year the Hojo provided jicho, honghwa, and danmok to the Sangeuiwon, and sometimes imported namjong from China. Third, royal garments were, in most cases, dyed by the Sangeuiwon's professional dyers and court ladies belonging to the sewing department in each palace. Naenongpo (內農圃) eunuchs were in charge of the indigo crops of each palace. Finally, more dye was used in royal garments than in the clothes of commoners to obtain a deeper shade of color. In addition, dyers tried to achieve a clear and vivid tone in their garments. Silk which absorbed color relatively easily, was dyed inside the palace using an ice vat filled with fresh indigo leaves; however, cotton was difficult to dye and was sent to professional indigo dyers outside the palace.

  • 나주재래종과 새로이 선발된 나주2호의 지역별 재배시험 결과, 초장은 나주2호가 다소 컸으며, 분지수는 나주재래종이 나주2호 보다 많은 경향이었다. 엽면적은 나주2호가 더 넓었으며, 엽장폭비도 나주2호가 더 크게 나타나 둥근 잎 특성의 안정성을 보였다. 두 계통의 지상부 생중과 건중은 큰 차이가 없었으나, 지상부 전체에 대한 엽의 무게 비율은 나주2호가 더 높아 색소수율면에서 유리하였다. 인디고(indigo) 성분과 니람(泥藍) 함량은 나주2호가 나주재래종 보다 높게 나타났으며 실크로 생엽 이용 염색을 한 결과 청색을 더 많이 나타냈다. 따라서 엽 수량이 높고 색소 품질이 좋은 나주2호가 쪽 재배에 유리한 계통으로 판단되었다.

  • The objective of this study is to investigate the efficacy of leaf powder colorants as substitutes for traditional fresh juice extract dyeing. Three kinds of leaf powder colorants were prepared by freeze drying method with or without deep freezing as pre-treatment: one powder colorant from fresh leaf juice with deep freezing; two kinds of powder colorant from fresh leaves with and without deep freezing. Their dyeing properties and storage stabilities were studied and compared with the traditional fresh juice extract dyeing. The presence of indigo in the powder colorants was confirmed by UV/Visible absorption spectra. They showed absorption peak at 602nm which was same with indigo absorption peak. Dyeing was done at low temperature around 6$^{\circ}C$. All three powder colorants produced B colors on silk fabrics, showing similar color to the one dyed traditionally with fresh juice extract. The powder colorants from leaves gave higher color strength than the powder from leaf juice. The powder colorant prepared from leaves with deep freezing was the most stable for long term storage as its color and color strength were not changed after 360 days. So, this was used for further dyeing to study the effects of concentration and repeat dyeing on color strength and colorfastness. Fastnesses to dry cleaning and rubbing were fairly good above 4 rating. Further study is needed to improve light fastness. It was concluded that the leaf powder colorant with deep freezing could be used as a substitute for traditional juice extract dyeing at all seasons.

네트워크 약리학적 분석에 의한 소세포폐암에 대한 청대의 항암기전 연구 (Identifying the Anti-Cancer Effect of Indigo Naturalis in Small Cell Lung Cancer Based on Network Pharmacological Analysis)

  • Lung cancer is the leading cause of cancer-related deaths worldwide. Indigo Naturalis (IN) is a dark blue powder obtained by processing leaves or stems of indigo plants, its anticancer effects have been reported in several studies. However, the pharmacological mechanism of IN in small cell lung cancer (SCLC) is not elucidated. In this study, to investigate the anticancer efficacy of IN for SCLC, we presented potential active ingredients, SCLC-related targets, and pharmacological mechanisms of IN that are expected to have anticancer activity for SCLC using a network pharmacological analysis. The phytochemical compounds of IN have been collected through TCMSP, SymMap, or HPLC documents. The active ingredients of IN such as indirubin, indican, isatin, and tryptanthrin were selected through ADME parameters or literature investigations for each compound. Using the Compounds, Disease-Target associations Databases, 124 common targets of IN and SCLC were obtained. Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG) Pathway enrichment analysis was carried out. GO biological processes are associated with response to xenobiotic stimulus, positive regulation of protein phosphorylation, regulation of mitotic cell cycle, and regulation of apoptotic signaling pathway. KEGG disease pathways included Gastric cancer, Bladder cancer, SCLC, and Melanoma. The main anticancer targets of the IN for SCLC were analyzed in 14 targets, including BCL2, MYC, and TP53. In conclusion, the results of this study based on the network pharmacology of IN can provide important data for the effective prevention and treatment of SCLC.

생쪽잎분말의 염색성 및 저장성(II) - 열풍 및 상온건조방법 - (Dyeing Properties and Storage Stability of Leaf Powder Prepared from Dyer's Knotweed(II) - by Hot Air and Room Temperature Drying Methods -)

  • The objective of this study is to investigate the efficacy of leaf powder colorants as substitutes for traditional indigo dyeing. Leaf powder colorants were prepared by hot air($50^{\circ}C$) and room temperanrre($25^{\circ}C$) drying methods from fresh leaves. The presence of indigo in the leaf powder colorants was confirmed by UV/Visible absorption spectra. All the powder colorants showed broad absorption at 602 nm as same as synthetic indigo. Dyeing was done by reduction method with sodium hydrosulfite and sodium hydroxide. Leaf powder colorants produced blue color on silk fabrics, showing similar color to the one dyed traditionally with fresh juice extract. The powder colorants prepared at room temperature drying were more stable for long term storage than that prepared by hot air drying. Thus, the powder colorants prepared by room temperature drying was reduced and dyed in one-step process without sodium hydroxide in the dyebath for further investigate dyeing properties. K/S value of the fabric dyed without sodium hydroxide was much higher than one dyed with sodium hydroxide. Regardless of the addition of sodium hydroxide, rubbing fastness was fairly good showing above 4 rating. Fastness to dry cleaning and light of the fabrics dyed without sodium hydroxide were mote higher than that dyed in alkaline condition.

  • 생쪽잎을 다양한 방법으로 건조, 분말화하여 염색하는 경우 전통적인 색소추출방법에서 얻을 수 있는 색상으로 염색됨을 확인하였다. 특히 급냉/동결건조한 분말은 생즙법의 염색효과에, 상온건조한 분말은 생즙법과 침전법의 염색효과에 더 효율적인 방법으로 나타났다.

  • Dyeings of the most widely used to the synthetic fibers, namely polyamide and polyester, have been carried out using acid dyes and disperse dyes. The above mentioned dye types, and indeed all dyes onto substrates, rely on the reaction properties between the substrates and dye molecules. In terms of fastness properties, however, especially to washings, the satisfactory levels are not present in the results from acid and disperse dyeings. Thus, vat dyeings leaves a feasibility and are an alternative way to overcome the problem. Although attempts and works have been reported in early years, with little if any commercial achievement and success. In this context, to meet increased demands towards a high level of wash fastness from consumers and retailers, an attempt using vat dyes has been carried out in this work.

김치의 선도유지를 위한 천연보존제의 탐색 (Screening of Natural Preservatives to Inhibit Kimchi Fermentation)

  • As a primary step to develop natural preservative for extending the shelf-life of kimchi, the effect of 102 edible plants, 21 antimicrobial agents and related compounds on kimchi fermentation was studied. Among 42 oriental medicinal plants tested, Baical skullcap and Assam indigo were found to be highly effective for maintaining the fresh state of kimchi. Although Bugbane, Red mangolia, Bushy sophora, Szechuan pepper, Chinese quince and Scisandre significantly inhibit the growth of Lactobacilli, their effect was not high enough to be used as raw materials for kimchi preservative. When the effect of 32 herbs and spices was tested, peppermint, cinnamon, lemon balm, clove, hop, rosemary, sage, horseradish and thyme showed high antimicrobial activity against kimchi microorganisms. Among them, the effect of clove ranked top. When it was added to fresh kimchi, initial cfu value ($2.4{\times}10^{6}cfu/g$) changed little even after 2 day's fermentation ($2.6{\times}10^{6}cfu/g$). Sensory test was not a good criteria to evaluate the effect of herbs and spices, since their highly specific flavors affected the taste of kimchies. Twenty eight fruits, vegetables and related plants were tested, but only leaves of pine tree, persimmon and oak leaves showed a significant bactericidal effect, finally contributing to the storage of kimchi. In addition, when 21 natural preservatives and other compounds were added individually to fresh kimchi, nisin and caffeic acid could inhibit fermentation.

Identification of Excision of Ac Transposable Element in P.nigra x maximowiczii Using Agrobacterium-mediated Transformation

  • The Ac (activator) which is one of the well-characterized transposable elements from maize was examined for its transposition possibility to the heterologous plant (P.nigra x maximowiczii) genome via Agrobacterium tumefacience (LBA4404) mediated transformation system. A number of transgenic plants were successfully recovered after 30 weeks by amount reduction from 50 to 15 g/$m\ell$ kanamycin for in vitro selection to minimize phytotoxic effects and to increase callus growth and regeneration efficiency. Among transgenic plants, 62 out of 106 transgenic poplars (58.5%) showed abnormal phenotypes such as severe serrated leaves and light leaf coloration. Indigo staining with X-gluc proved indirectly the restoration of Gus enzyme function and the presence of Ac in poplar genome by PCR. Southern analysis indicated the transposition and existence of Ac element in poplar genomes. In this research, an Agrobacterium-mediated transformation system in poplar species was developed and identified that Ac derived from maize can be excised and trans posed into other poplar genomes.
