The production of sweet (su) and super sweet corns (sh2) has been economically feasible in Korea in recent years. Major factors limiting super sweet corn production are low germination and low seedling vigor. Since seed quality is closely related to seed maturity, the optimum harvest time for the seed production of sweet and super sweet corns was studied and the quality of seeds with varying maturities was investigated in 2001 and 2002 cropping seasons. The parents of the sweet corn seeds were Hybrid Early Sunglow and 'Golden Cross Bantam 70' and those of super sweet corn were Xtrasweet 82 and 'Fortune'. Seeds were harvested at 21, 28, 35, 42, 49, and 56 days after silking (DAS). As the seeds developed, seed weight of sweet corn increased and the seed moisture content decreased faster than that of super sweet corn. Germination rates of sweet corn seeds harvested 21 and 28 DAS at $25^{\circ}C$ and emergence rates in the cold soil test were significantly lower than those of seeds harvested after 42 DAS in both years. Although the germination rates of super sweet corn seeds with varying maturities showed similar patterns as sweet corn seeds at $25^{\circ}C$, the emergence rate of super sweet corn seeds in cold soil test continuously increased with seed maturity. This suggests that seed quality of super sweet corn should be tested in a cold soil test to estimate field emergence. As the seeds developed, leakage of total sugars and electrolytes from the both sweet and super sweet corn seeds decreased up to 42 or 49 DAS. The $\alpha-amylase$ activities of both sweet and super sweet corn seeds increased with seed maturity from 21 to 35 or 49 DAS depending on genotype and year. The optimum harvest time for the seed production of sweet corn was 42 DAS and 49 DAS for super sweet corn considering emergence rate and plumule dry weight in the cold soil test, leakage of sugars and electrolytes from the seeds, and $\alpha-amylase$ activity.
In Korea, production of super sweet corn has been economically feasible and is substituting for traditional sweet corn due to better flavor in recent years. Major limiting factors for super sweet corn production are low field emergence and low seedling vigor. The optimum water potential (WP) for the priming of normal and aged seeds of dent, sweet (su) and super sweet (sh2) corns was studied to improve low seed quality. Seeds were primed at 0, -0.3, -0.6, -0.9, and -1.2 MPa of polyethylene glycol (PEG) 8000 solution at $15^{\circ}C$ for 2 days. Priming effects differed depending on the type of corn, seed quality, and WP of PEG solution. Although WP of priming solution did not influence the emergence rate of extremely high quality normal dent corn seeds, it reduced time to $50\%$ emergence (T50) and increased plumule weight. In contrast, the emergence rate of aged field corn was improved by seed priming at 0 MPa and plumule weight and $\alpha-amylase$ activity was enhanced. The optimum WP for both normal and aged sweet and super sweet corn seeds was between -0.3 and -0.6 Mpa. At the optimum WP emergence rate, $\alpha-amylase$ activity, and content of DNA and soluble protein increased, while T50 and leakage of total sugars and electrolytes reduced.
In order to determine the optimum harvest time for the seed production of inbreds and hybrids in silage corn, the ears of sib-pollinated 'KS5', 'KS7rhm', and 'Ga209' and cross-pollinated 'KS5' $\times$ 'KS6' (Suwon19), 'KS7 rhm' $\times$ 'KSl17' (Suwonok), and 'Ga209' $\times$ 'DB544'(Kwanganok) were harvested at the one-week intervals from 4 to 10 weeks after silking. The optimum harvest time for the seed production for 'KS5', 'KS5' $\times$ 'KS6', 'KS7 rhm', and 'KS7rhm' $\times$ 'KS117' was 7 weeks after silking considering both emergence rate and plumule growth in cold test. Although earlier harvested seeds showed similar germination rate as the seeds harvested at the optimum time at $25^{\circ}C$, their emergence rate were lower in cold test. Seed weight and $\alpha$-amylase activity of earlier harvested seeds were lower compared to those of seeds harvested at the optimum time, while leakage of total sugars and electrolytes were higher. However, the later harvested seeds showed lower germination rates at $25^{\circ}C$ and emergence rates in cold test probably due to the lower $\alpha$-amylase activity although they showed increased seed weight and reduced leakage of total sugars and electrolytes. In contrast, the emergence rate of 'Ga209' and 'Ga209' $\times$ 'DB544' in cold test increased up to 10 weeks after silking probably due to the increased seed weight and $\alpha$-amylase activity and reduced sugar and electrolyte leakages during the germination. The cross-pollinated F$_1$ hybrid seeds showed higher germination and emergence rates at $25^{\circ}C$ and in cold test, and higher plumule growth and $\alpha$-amylase activity compared to those of sib-pollinated inbreds.
단옥수수와 초당옥수수 종자의 활력을 높일 수 있는 저장 방법을 알기 위하여 단옥수수(Early Sunglow ${\times}$ Golden Cross Bantam 70)와 초당옥수수(Xtrasweet 82 ${\times}$ Fortune) 종자를 온도(5 및 $15^{\circ}C$)와 상대습도(70 및 85%)가 다른 조건에서 10개월간 저장한 후 종자 특성의 변화를 보면 다음과 같다. 1. 종자활력은 초당옥수수가 단옥수수보다 더 빨리 저하되었으며, 초당옥수수가 단옥수수보다 더 낮은 저장온도와 상대습도가 요구되었다. 2. 단옥수수는 저장조건과 저장기간에 따른 cold test에서 출아율이 $25^{\circ}C$에서 발아율보다 약간 낮았으나 경향은 같았고, 초당옥수수는 cold test에서 출아율이 $25^{\circ}C$에서 발아율보다 현저히 낮았다. 3. 단옥수수와 초당옥수수 모두 발아율과 저온처리 후 출아율과는 정의 상관이 있었으나 초당옥수수는 관계가 적었다. 그래서 초당옥수수의 포장 출아율을 추정하기 위하여 반드시 저온처리 후 출아율 검정이 필요하였다. 4. 단옥수수는 모든 저장조건에서 5개월간은 종자활력이 저하되지 않았으나, 초당옥수수는 $5^{\circ}C$, RH 70%에서 활력이 유지되었다. 5. 단옥수수에서는 종자활력과 침종시 종자의 당 누출, 전해질 누출 및 ${\alpha}-amylase$ 활성과는 상관이 없었다. 반면 초당옥수수에서는 종자활력과 ${\alpha}-amylase$ 활성과는 정의 상관이, 침종 시 종자의 전해질 누출과는 부의 상관이 있었으나 당 누출과는 상관이 없었다.
단옥수수와 초당옥수수 종자의 활력을 높일 수 있는 탈곡방법을 알기 위하여 단옥수수($Early\;Sunglow{\times}GCB\;70$)와 초당옥수수($Xtrasweet\;82{\times}Fortune$) 종자의 수분함량을 12, 15, 18, 21%로 건조한 후 손과 전기 탈곡기로 탈곡하여 $25^{\circ}C$ 발아율, cold test에서 출아율, 당 및 전해질 누출, ${\alpha}-amylase$ 활성 등 종자특성을 조사한 결과를 요약하면 다음과 같다. 1. 손 탈곡은 종자수분 함량에 관계없이 기계적 상처가 없었으나 기계 탈곡할 때 종자의 상처를 줄일 수 있는 수분함량은 단옥수수는 $15{\sim}21%$, 초당옥수수는 $12{\sim}21%$이었다. 2. Cold test에서 출아율은 종자수분 함량에 관계없이 손 탈곡한 종자가 기계 탈곡한 종자보다 단옥수수는 $6{\sim}14%$, 초당옥수수는 $9{\sim}18%$ 높았다. 3. 단옥수수는 $25^{\circ}C$ 발아율과 cold test 출아율과 정의 상관이 있었으나 초당옥수수는 관계가 없었다. 단옥수수와 초당옥수수의 cold test에서 출아율은 침종 시 당 누출과는 부의 상관, ${\alpha}-amylase$ 활성과는 정의 상관이 있었다. 4. 기계 탈곡 시 $25^{\circ}C$에서 상처립 비율, 발아율, cold test에서 출아율, 당과 전해질 누출량, ${\alpha}-amylase$ 활성을 고려한 적정 종자수분 함량은 단옥수수는 15%, 초당옥수수는 12%이었다.
옥수수 종자의 활력을 향상시키는 방법을 모색하기 위하여 마치종 옥수수, 단옥수수, 초당옥수수의 건전종자와 인위적으로 노화처리하여 활력이 낮은 종자를 수분함량이 50, 75, 100, 125%인 vermiculite와 섞어 $25^{\circ}C$에서 24시간 matriconditioning하였다. 처리된 종자를 토양에 파종하여 $25^{\circ}C$와 co]d test(1$0^{\circ}C$에서 7일 처리 후 $25^{\circ}C$에서 7일 처리)조건에서 종자활력을 조사하여 matriconditioning에 알맞은 vermiculite의 수분함량을 구명하였고, 또 침종시 종자의 당과 전해질의 누출량, $\alpha$-amylase 활성, DNA 및 가용성 단백질 함량을 조사하여 mariconditioning할 때 종자의 생리적인 변화를 조사하였으며, 그 결과를 요약하면 다음과 같다. 1. 옥수수의 출아율, 출아속도, 유묘생장은 마치종 > 단옥수수 >초당옥수수의 순으로 작았다. 건전종자가 노화종자보다 출아율, 출아속도, 유묘생장이 좋았고. $\alpha$-amylase 활성, DNA 및 가용성 단백질 함량은 높았으나 당과 전해질의 누출은 적었다. 2. Matriconditioning은 $25^{\circ}C$에서 발아할 때 마치종과 단옥수수의 건전종자의 출아율에는 영향을 미치지 않았으나 활력이 낮았던 초당옥수수 건전종자와 모든 genotype의 노화종자는 matriconditioning할 때 수분함량이 높을수록 출아율이 증가하였다. Cold test에서는 $25^{\circ}C$와 같은 경향이었으나 출아율이 더 낮았다. 3. 출아속도는 단옥수수 건전종자는 $25^{\circ}C$와 cold test에서 모두 matriconditioning의 영향을 받지 않았지만 다른 종자는 $25^{\circ}C$와 cold test에서 모두 matriconditionig 할 때 수분 함량이 높을수록 출아속도가 빨랐다. 4. 유아장과 유아중은 cold test에서 matriconditioning의 영향이 없었던 초당옥수수의 건전종자와 노화종자를 제외한 다른 종자는 모두 $25^{\circ}C$와 cold test에서 수분함량이 높을수록 유아장과 유아중이 증가하였다. 5. 종자의 당 누출량은 마치종 <단옥수수 <초당옥수수의 순으로 많았다. 마치종은 건전종자와 노화종자 모두 당 누출량이 적었다. 단옥수수는 노화종자의 당 누출량이 건전종자보다 다소 많았고, matriconditioning할 때 수분함량이 증가할수록 당누출량은 다소 감소하였다 초당옥수수는 노화종자가 건전종자보다 당 누출량이 현저히 많았으며, 건전종자는 50%, 노화종자는 100%까지 matriconditioning처리할 때 수분 함량이 높을수록 당 누출량은 감소하였다. 6. 종자의 전해질 누출량(침지액의 전기전도도)은 마치종 <단옥수수 <초당옥수수의 순으로 높았고, matriconditioning할때 수분함량이 높을수록 전해질의 누출량은 감소하였다. 7. $\alpha$-amylase활성은 단옥수수 <마치종<초당옥수수의 순으로 높았다. 모든 옥수수 genotype에서 matriconditioning할 때 수분 함량이 높을수록 $\alpha$-amylase활성이 증가하였다. 8. DNA와 수용성 단백질 함량은 초당옥수수 <마치종 <단옥수수의 순으로 높았다 모든 genotype의 건전종자와 노화종자는 matriconditoining할 때 수분함량이 높을수록 DNA와 수용성 단백질 함량이 증가하는 경향이었다.
