목초액이 기내 배양한 풍란(Neofinetia faicata)의 내생물질 함량 및 활성에 미치는 영향 (Effect of Pyroligneous Liquor on the Content and Activity of Endogenous Substances of Neofinetia falcata Cultured in vitro)

  • This experiment was conducted to clarify the effect of pyroligneous liquor on endogenous substances that were GA (gibberelic acid)-like substances, starch, and protein of Neofinetia falcata cultured in vitro. When seedlings of Neofinetia falcata were treated with several concentrations (0, 0.5, 1.0, 2.0, 3.0 ml/L) of pyroligneous liquor, the growth of seedlings was enhanced in 1.0ml/L pyroligneous liquor added media. The activity of GA-like substances was lower in the leaf, but higher in the and root than that of the control. The content of starch in 1.0ml/L pyroligneous liquor was lower in the leaf, but higher in the root than that of the control. And the content of protein in 1.0ml/L pyroligneous liquor was also lower in the leaf, but higher in the root than that of the control.

Proximate, Phytochemical, and In Vitro Antimicrobial Properties of Dried Leaves from Ocimum gratissimum

  • Ocimum gratissimum is a common plant in the tropics and has been used in food and medicine. Its usage in food and medicine could be attributed to its phtyochemical and antimicrobial properties. In this study we investigated the proximate, phytochemical, and antimicrobial attributes of air dried leaves of O. gratissimum. The aqueous extract was found to contain phtyochemicals with alkaloid and saponin present in appreciable amounts. The proximate analysis (crude protein and crude fibre content were 15.075% and 17.365%, respectively) showed that the leaf could be a good source of protein and fibre. The aqueous ethanolic extract of the leaf exhibited activity against a wider range of organisms when compared to the aqueous extract at the investigated concentrations. Aqueous ethanolic extracts of O. gratissimum leaf was active against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Bacillus cereus and the aqueous extract of the leaf was active against P. aeruginosa.

완두 발아시 Polyamine 함량 변화 및 Peroxidase Isozyme 양상 (Peroxidase Isozyme Pattern and Polyamine Contnts in Germinating Peas (Pisum staivum))

  • In germinating pea, contents of enodgenous polyamine in the leaf and stem were determined, and protein content, peroxidase activity and pattern of isozymes were examined in the leaf treated with polyamines. During growth of the pea for 14 days in light condition, the polyamines in leaf and stem showed the highest level at the 5th day, and were decreased rapidly at the 7th day, kept almost constant level since then. The putrescine level was relatively higher than those of spermidine and spermine, and cadaverine was also detected. On the other hand, in the leaf treated with spermine (0.01 mM) protein content increased about 250% than that of the control, the peroxidase activity increased ore than 100% in spermine of 0.01 mM and 0.1 mM. In treating with putrescine of 0.1 mM the pattern of peroxidase isozyme appeared 4 new cathodic bands (pI 4.8, 5.6, 5.9 and 6.8) compared with the control, the clear cathodic bands (pI 5.6, 5.9, 6.4 and 6.6) were also observed in spermine of 0.1 mM. These results suggest that polyamines were important factor in the differentiation of pea at the early stage of germination.

연잎과 연자육 아이스크림 개발 (Development of Ice Cream Prepared Lotus(Nelumbo nucifera Gaertner) Leaf and Seeds)

  • The purpose of this study was to determine the content attributes and sensory characteristics of ice cream flavored with lotus leaf and seeds (Nelumbo nucifera Gaertner). Analysis of freeze dried lotus leaf powder produced the following technical results: Freeze dried lotus leaf powder contained moisture 12.2, crude protein 5.9, crude fat 1.2, crude ash 7.0, crude fiber 13.4g/100mg; raw lotus seed powder contained moisture 65.0, crude protein 8.2, crude fat 0.3, crude ash 1.0, crude fiber 10.8g/100mg; freeze dried lotus seed powder contained moisture 14.0, crude protein 17.1, crude fat 1.9, crude ash 4.0, crude fiber 2.8g/100mg. Lotus seed was processed by 4 methods: freeze dried, roasted, freeze dried peeled inner layer, cooked cut peeled inner layer. Analysis of the color value produced the following technical results: freeze dried lotus leaf powder were L(lightness) 14.5, a(redness) 4.4, b(yellowness) 24.0; freeze dried lotus seed powder L 57.3, a 14.4, b 12.0; roasted lotus seed powder L 52.7, a 22.5, b 11.9; freeze dried lotus seed peeled inner layer L 60.0, a 1.4, b 12.3. Proportional lotus ingredients used for ice cream were 1%, 3%, 5%, 7%, 10% for lotus leaves and 1%, 3%, 5% for lotus seeds. The maximum over run ratio for lotus leaf ice cream was 45.3~56.9% at 20~25 minutes operating time. Over run was significantly decreased by increasing the contents of lotus leaf powder. A maximum over run ratio for lotus seed ice cream was 46.1~54.3% at 20~25 operating time. Over run was not significantly different based on content of lotus leaf versus lotus seed. Sensory evaluation of lotus leaf ice cream produced the following results: the highest score was for color of 4.42 and the lowest score was for sweetness of 3.30. The total mean score significantly decreased by increasing the content of lotus leaf powder. Sensory evaluation of lotus seed ice cream produced the following results: the highest score was for cooked cut peeled inner layer type and the lowest was for freeze dried type 3.86. The sensory scores were not significantly different based on the content of lotus seed. Texture was evaluated highest with a rating of 4.21 and the taste was evaluated lowest with a rating of 3.68 For whole evaluation. This study concluded that lotus leaf ice cream demonstrates relative strength in terms of color and taste, and a relative weakness in terms of over run and sweetness. Suitable content of lotus leaf ranged from 1~5%. The lotus seed processed by the cooked cut peeled inner layer method received a higher rating than dried powder for lotus seed ice cream.

바나나잎 추출물의 미백 개선 효과 (Whitening Effect of Banana Leaf Extract)

  • 바나나잎의 기능성화장품 소재로써 응용 가능성을 규명하고자 바나나잎 추출물이 피부 미백에 미치는 효과를 B16F10 세포주를 활용하여 cell viability, mRNA 발현 및 tyrosinase (Tyr) 활성 저해 실험을 통하여 알아보았다. 10%의 바나나잎 추출물은 tyrosinase 활성을 저해하고, 멜라닌 합성을 65% 감소시키는 것을 확인하였다. 정량적 real-time RT-PCR을 수행한 결과 바나나잎 추출물이 tyrosinase related protein 1 (TRP1) / tyrosinase related protein 2 (TRP2) / tyrosinase mRNA 발현을 각각 20 / 40 / 60% 가량 억제하는 효과를 확인하였다. 또한 바나나잎 추출물이 함유된 크림제형의 임상시험을 수행하여 자외선 조사 직후 바나나잎 추출물이 함유된 크림의 육안평가 결과 유의한 미백효과가 있는 것을 확인하였다. 본 연구 결과는 바나나잎 추출물이 미백 기능성 화장품 소재 뿐 아니라 향후 기전 연구 수행을 통해 다양한 산업화 소재로 개발될 수 있음을 시사한다.

Inhibition of Melanogenesis by Abietatriene from Vitex Trifolia Leaf Oil

  • Vitex trifolia L. has been used traditionally to treat various illnesses, such as inflammation, headache, migraine, and gastrointestinal infections. We analyzed and evaluated the composition of V. trifolia leaf oil. Based on the results, we isolated abietatriene from V. trifolia leaf oil and investigated the effect of V. trifolia leaf oil and its active compound abietatriene on melanogenesis in B16F10 melanoma cells. They significantly decreased melanin contents and melanogenic factors, such as tyrosinase, TRP-1, TRP-2, and MITF dose-dependently in both protein and mRNA levels. Protein and mRNA expressions were determined by Western blot analysis and quantitative real time RT-PCR. Findings indicate that V. trifolia leaf oil and abietatriene reduce melanogenesis by regulating the expression of melanogenic factors. These results suggest that V. trifolia leaf oil and abietatriene could comprise a useful therapeutic agent for treating hyperpigmentation and used as effective skin-whitening agents.

대청 추출물의 항산화 효소 활성 및 항균 효과 (Antioxidant Enzyme Activity and Antimicrobial Activity of Isatis tinctoria Extract)

  • 대청의 이용성 향상을 위한 자료 확보 측면에서 식물체 부위 및 추출 용매별에 따른 항산화 효소 활성 및 항균효과를 조사하였다. APX(Ascorbate Peroxidase) 활성은 줄기의 에탄올 추출물, 잎의 메탄올 추출물, 잎의 증류수의 추출물 순으로 높아 각각 1601.7, 1133.7 및 524.3(Unit/mg protein)을 나타냈다. CAT(Catalase) 활성은 꽃의 에탄올 추출물, 잎의 메탄올 추출물, 꽃의 증류수 추출물 순으로 높았는데, 각각 177.1, 120.8 및 55.4(Unit/mg protein)를 나타냈다. POD(Ascorbate Peroxidase) 활성은 꽃의 에탄올 추출물, 꽃의 메탄올 추출물, 줄기의 증류수 추출물 순으로 높았으며, 각각 27.1, 14.6 및 10.4(Unit/mg protein)를 나타냈다. SOD(superoxide dismutase) 활성은 뿌리의 증류수 추출물, 꽃의 메탄올 추출물, 뿌리의 에탄올 추출물 순으로 높았으며, 각각 90.8, 80.1, 75.5%를 나타냈다. 대청의 꽃 추출물은 Vibrio parahaemolyticus에 대해서만, 뿌리는 Staphy lococcus aureus에 대해서만, 줄기 추출물은 Bacillus subtilis, Escherichia coli, Staphy lococcus aureus에 대해서만 용매에 관계없이 항균활성을 나타냈다. 특히 대청 잎의 증류수 추출물은 Bacillus subtilis, Escherichia coli에 대해 높은 항균활성을 나타내어 저해환 직경이 각각 30.0 및 24.0 mm이었다. 이와 같은 결과는 약용식물로서 대청의 가치가 높음을 시사해 주었다.

Comparative Analysis of the Conserved Functions of Arabidopsis DRL1 and Yeast KTI12

  • Patterning of the polar axis during the early leaf developmental stage is established by cell-to-cell communication between the shoot apical meristem (SAM) and the leaf primordia. In a previous study, we showed that the DRL1 gene, which encodes a homolog of the Elongator-associated protein KTI12 of yeast, acts as a positive regulator of adaxial leaf patterning and shoot meristem activity. To determine the evolutionally conserved functions of DRL1, we performed a comparison of the deduced amino acid sequence of DRL1 and its yeast homolog, KTI12, and found that while overall homology was low, well-conserved domains were presented. DRL1 contained two conserved plant-specific domains. Expression of the DRL1 gene in a yeast KTI12-deficient yeast mutant suppressed the growth retardation phenotype, but did not rescue the caffeine sensitivity, indicating that the role of Arabidopsis Elongator-associated protein is partially conserved with yeast KTI12, but may have changed between yeast and plants in response to caffeine during the course of evolution. In addition, elevated expression of DRL1 gene triggered zymocin sensitivity, while overexpression of KTI12 maintained zymocin resistance, indicating that the function of Arabidopsis DRL1 may not overlap with yeast KTI12 with regards to toxin sensitivity. In this study, expression analysis showed that class-I KNOX genes were downregulated in the shoot apex, and that YAB and KAN were upregulated in leaves of the Arabidopsis drl1- 101 mutant. Our results provide insight into the communication network between the SAM and leaf primordia required for the establishment of leaf polarity by mediating histone acetylation or through other mechanisms.

조사료원이 한국 재래산양의 섭취량과 소화율에 미치는 영향 (The Effect of Roughage Source on Voluntary Feed Intake and Digestibility in Korean Native goats)

  • A study was carried out to evaluate the values of roughages available in Korea on feed intake and digestibility of Korean native goats and consequently to apply its results to the feeding system of Korean goats as a basic information. The results are as follows. 1. The protein contents in Alfalfa and Acacia leaf were 17.6 and 16.3% respectively 11.7 and 6.8% higher than orchardgrass and pine needle. Ether extract tom pine needle was about two times higher than other roughage sources which are 9.2%. 2. Dry matter intake per day in goats fed alfalfa, acacia leaf and orchardgrass was higher (P < 0.05) at 590.3, 543.8 and 496.58 respectively and 217.lg in pine needle. 3. Dry matter intake per basal weight (DM glkg of B$W^{0.75}$ and DM g/kg of BW%) was higher in goats fed acacia leaf at 68.5 and 3.5% respectively than any other treatments goats fed pine needle showed the lowest (P< 0.05) dry matter intake at 28.6 and 1.5% respectively. 4. Dry matter digestibility was highest (PcO.05) in alfalfa fed goats at 61.4%. Dry matter digestibilities in orchardgrass, pine needle and acacia leaf were 58.0, 46.8 and 46.6% respectively. 5. Total digestible nutrients were highest (P< 0.05) in Alfalfa fed goats at 59.5%. Total digestible nutrients in orchardgrass, pine needle and acacia leaf were 54.2, 50.7 and 47.7% respectively. 6. In conclusion, the value of orchardgrass as feed for goats was as excellent as alfalfa and although acacia leaf had a less value than alfalfa in goats, it is considered to be used as a protein supplement in diets for goats due to its high content of protein and excellent palatability. Intake of pine needle in Korean goats in this study was satisfactory and it indicates the possibility of its use as a supplement in diets for goats. However, fkther investigation will be necessary particularly on the deleterious effects of ether extracts of pine needle

  • 오이의 성발현(性發現) 기구(機構)에 관(關)한 기초(基礎) 자료(資料)를 얻고저 비절성(飛節成) 품종(品種)인 사엽(四葉), 절성(節成) 품종(品種)인 신록다다기, 그 중간형(中間型)인 성호원(聖護院)을 공시(供試)하여 종자(種子)와 최아종자(催芽種子)의 단백질함량(蛋白質含量)의 품종간(品種間) 차이(差異)와 2엽기(葉期)와 4엽기(葉期)의 ethephon(250 ppm)과 gibberellin(100ppm) 처리(處理)에 의한 성발현(性發現) 양상(樣相)과 식물체내(植物體內)의 단백질함량(蛋白質含量), peroxidase 활성(活性) 및 disc polyacrylamide gel 전기영동(電氣泳動)에 의한 단백질(蛋白質) banding pattern과 peroxidase isozyme banding pattern의 소장관계(消長關係)를 조사(調査)하여 다음과 같은 결과(結果)를 얻었다. 1. Ethephon 처리(處理)는 자화(雌花)의 착생(着生)을 약간 증가(增加)시키고, 웅화(雄花)의 착생(着生)을 현저(顯著)히 감소(減少)시켰으며 gibberellin 처리(處理)는 절성종(節成種)인 신록다다기와 성호원(聖護院)의 웅화수(雄花數)를 현저(顯著)히 증가(增加)시켰으나 비절성(飛節成)일 사엽(四葉)의 웅화수(雄花數)는 증가(增加)시키지 못하였다. 2. 종자(種子)와 최아종자(催芽種子)의 단백질함량(蛋白質含量), 단백질(蛋白質) 및 peroxidase의 banding pattern은 품종간(品種間)에는 약간의 차이(差異)가 있었으나 절성(節成)과 비절성간(飛節成間)에는 뚜렷한 차이(差異)를 인정(認定)할 수 없었다. 3. 2 엽기(葉期)와 4 엽기(葉期)의 단백질함량(蛋白質含量)은 잎과 생장점(生長點)에서 모두 처리후(處理後) 일수(日數)가 경과(經過)함에 따라 2엽기(葉期)에는 증가(增加)하으나 4엽기(葉期)에는 오히려 감소(減少)되는 경향(傾向)을 보였다. 그리고 4 엽기(葉期) 생장점(生長點)에 있어서 비절성(飛節成)인 사엽(四葉)은 ethephon 처리(處理)에 의해 단백질함량(蛋白質含量)이 무처리구(無處理區)에 비(比)해 높아졌으나 신록다다기와 성호원(聖護院)은 오히려 낮아졌다. 4. Peroxidase 활성(活性)은 2 엽기(葉期)에는 잎이나 생장점(生長點)에서 모두 일수(日數)가 경과(經過)함에 따라 감소(減少)하였으나 4엽기(葉期)에는 오히려 증가(增加)되는 경향(傾向)이었다. 특(特)히 ethephon 처리구(處理區)는 4엽기(葉期) 생장점(生長點)에서 그 활성(活性)이 현저(顯著)히 높아져 5일(日)째에는 타처리구(他處理區)에 비(比)해 월등(越等)히 높았다. 5. 단백질(蛋白質)의 banding pattern은 2엽기(葉期)에는 잎과 생장점(生長點) 공(共)히 처리후(處理後) 일수(日數)가 경과(經過)함에 따라 3품종(品種)모두 band 수(數)가 감소(減少)되었으나 4엽기(葉期)에는 오히려 band수(數)가 증가(增加)되는 경향(傾向)이었다. 4엽기(葉期) 생장점(生長點)의 단백질(蛋白質) pattern은 비절성(飛節成)인 사엽(四葉)의 ethephon 처리구(處理區)가 절성(節成)인 신록다다기의 무처리구(無處理區)와 같은 양상(樣相)으로 변화(變化)되어 갔다. 6. Peroxidase isozyme banding pattern은 2엽기(葉期) 잎에서 비절성(飛節成)인 사엽(四葉)의 ethephon 처리구(處理區) pattern이 성호원(性護院) 및 신록다닥기의 ethephon 처리ㅜ(處理區) pattern으로 닮아 감을 볼 수 있었고 4엽기(葉期) 생장점(生長點)에서는 처리후(處理後) 일수(日數)가 경과(經過)함에 따라 사엽(四葉) ethephon 처리구(處理區)의 pattern이 성호원(聖護院) 및 신록다다기의 무처리구(無處理區) pattern으로 닮아 감을 볼 수 있었다. 7. 이상(以上)의 결과(結果)로 볼 때 비절성종(飛節成種)은 ethephon에 의해 식물체내(植物體內)의 단백질함량(蛋白質含量), peroxidase 활성(活性) 및 단백질(蛋白質)과 peroxidase isozyme의 pattern이 절성종(節成種)에 가깝게 변화(變化)되어 절성화(節成化)되며, 따라서 이들의 변화(變化)가 오이의 성발현(性發現)에 깊이 관여(關與)하는 것으로 사료(思料)된다.
