The effect of various supplies of lead singly and in combination with aluminium on growth and chlorophyll biosynthesis was investigated in 7-day-old Vigna anguluris seedlings. Expose to 50 $\mu$N Pb or more drastically reduced root elongation rate. Significant depressions in root growth was observed within 1 day and no recovery of growth was seen over the duration of treatment period. Root elongation decreased depending on the Pb concentrations. Root growth inhibition was stronger than shoot growth inhibition. The initiation of lateral roots appeared to be more sensitive to Pb than the growth of main roots. Inhibition of root and shoot elongation by Pb was lessened by combined exposure of Pb and Al, suggesting that the presence of AA reverse the inhibitory effect of Pb alone. With the histochemical sodium rhodizonate method the rate of Pb uptake was dependent on the Pb concentration and exposure time of the roots to Pb salts. Pb was first deposited on the root surface and then translocated radially in the root cap cells. During a longer Pb administration (up to 72 h) Pb penetration was nonuniform, with accumulation within the cortex or endodermis. There was drastic reduction in chlorophyll content by Pb. The Pb inhibition of chlorophyll synthesis was concentration dependent. 5-Aminolevulinic acid dehydratase (ALAD) activity exhibited distinct inhibition from control. Reduction in chlorophyll content was accompanied by proportional changes in ALAD activity. Chlorophyll content and ALAD activity were less affected by combined exposure of Pb and Al, suggesting that Al has a protective effect against the inhibiting action of Pb on photosynthetic activity.

The in vitro plantlets of cassava (Manihot esculenta Crantz cv. MColl 22) could be regenerated from nodal explant cultures in a liquid MS basal medium containing 0.01 mg/L zeatin for 2 weeks. The plantlets of 1.5∼2.5 cm in shoot length were transplanted to a glass bottle containing fine sand and acclimated under non-sterile conditions after excising their intact roots by: 1) prune leaving roots base of 1∼1.5 cm; 2) complete removal of roots; and 3) cutting off the rooting zone. The majority of in vitro-formed intact roots continued growth after transferred to soil, and all of the damaged roots stopped further growth. The plantlets with excised roots began to develop new roots within 7∼10 days after being transferred to a glass bottle, and a few of the pruned roots developed lateral roots from the remaining portion. Pruning and removal of in vitro roots resulted in a high survival rate (>87%), and did not significantly affect ex vitro root regeneration and acclimation, but the plantlets in which the rooting zone had been cut-off showed 73% survival rate. Pruning or removal of in vitro roots before transfer of plantlets is recommended for useful method of commercial micropropagation because of easier handling and high survival rate of plantlets.
Study was performed to know the effects of Panax ginseng C.A. Meyer hairy root due to $^{60}$ Co ${\gamma}$-ray irradiation. We irradiated the hairy roots under the various $^{60}$ Co ${\gamma}$-ray; 0.5~4 Krad. The growth of hairy roots is inhibited over 3 Krad treatment. The lateral roots are used as a cell line after removing the apical meristem of hairy roots irradiated below 2 Krad. We selected 206 hairy root cell lines having various different growth rates and forms, and incubated in the 1/2 Murashige & Skoog(MS) medium in the absence of hormone. We selected 10 out of 206 showing superior growth. Among those, ${\gamma}$-GHR 70 and ${\gamma}$-GHR 94 showed higher growth; 34.5, 44.7%, respectively. We observed shapable, sizable characteristics according to the width of the primary roots, the process formation of the lateral roots, and the growth of lateral roots. The discriminable cell line showed that primary root is thinner, and has a vigorous growth. 8 out of 10 had much more contents than control in the aspect of the ginsenoside. ${\gamma}$-GHR 59 and ${\gamma}$-GHR 94 showed higher contents; 19, 16.9%, respectively. Therefore, we selected ${\gamma}$-GHR 70, ${\gamma}$-GHR 94 as a superior cell line in the aspect of ginsenoside contents, and growth among those irradiated by ${\gamma}$-ray. According to content of ginsenoside, Rb$_2$ effective in anticancer has 7.5% of ${\gamma}$-GHR 59. Rc, also effective in anticancer showed 16.2% content increasement of ${\gamma}$-GHR 69. It is thought that those lines will be effective in manufacturing ginsenoside. Gene analysis (VNTRP) related to the mutation is in progress.

About 200 rice varieties for irrigated and rainfed lowland ecosystems were released in the Philippines, which were bred for improving yield under favorable conditions. Root plasticity plays key roles in maintaining crop productivity under abiotic stressed conditions. We hypothesized that some of these varieties possess root plasticity traits in response to water stressed conditions. This study aimed to evaluate the root system development and dry matter production of 14 randomly selected rice varieties (6 irrigated lowland and 9 rainfed varieties) under progressive drought (PDR) and soil moisture fluctuations (SMF) stress conditions. Two experiments were done in rootbox and line source sprinkler systems (LSS). Each of the varieties was subjected to well-watered (WW), PDR and SMF conditions during vegetative stage in rootbox system while the same genotypes were subjected to different intensities of drought stress under LSS. Under rootbox system, PDR and SMF significantly reduced shoot dry matter production in all varieties relative to their WW controls. Among varieties, NSIC Rc238 (irrigated lowland) showed the least reduction in shoot dry weight (SDW) in both PDR (by 11.8%) and SMF (by 26.9%) conditions. Less reductions in SDW of NSICRc238 were partially attributed to the promotion of L-type lateral roots, thus increasing total lateral root length by 24.2% and 30.7% under PDR and SMF, respectively. In LSS, SDW of NSIC Rc238 under mild drought stress (16-21% soil moisture content (SMC) had 31.8% reduction relative to its WW control (${\geq}22%SMC$) and had lower sensitivity drought index. Compared with the IR64 susceptible check and NSIC Rc9 tolerant check, NSIC Rc238 had higher SDW by 90.8% and 38.6%, respectively. Furthermore, no rainfed lowland varieties included in the experiment performed well under different water stress treatments. The results implied that some other irrigated lowland rice varieties may also possess drought dehydration avoidance root plasticity traits under water-stressed growing environments.
Objective : To investigate the potential risk of approach-related complications at different access angles in minimally invasive lateral lumbar interbody fusion. Methods : Eighty-six axial magnetic resonance images were obtained to analyze the risk of approach-related complications. The access corridor were simulated at different access angles and the potential risk of neurovascular structure injury was evaluated when the access corridor touching or overlapping the corresponding structures at each angle. Furthermore, the safe corridor length was measured when the corridor width was 18 and 22 mm. Results : When access angle was $0^{\circ}$, the potential risk of ipsilateral nerve roots injury was 54.7% at L4-L5. When access angle was $45^{\circ}$, the potential risk of abdominal aorta, contralateral nerve roots or central canal injury at L4-L5 was 79.1%, 74.4%, and 30.2%, respectively. The length of the 18 mm-wide access corridor was largest at $0^{\circ}$ and it could reach 44.5 mm at L3-L4 and 46.4 mm at L4-L5. While the length of the 22 mm-wide access corridor was 42.3 mm at L3-L4 and 44.1 mm at L4-L5 at $0^{\circ}$. Conclusion : Changes in the access angle would not only affect the ipsilateral neurovascular structures, but also might adversely influence the contralateral neural elements. It should be also noted to surgeons that alteration of the access angle changed the corridor length.
Root gravitropism was investigated in the pea (Pisum sativum L.) mutant ageotropum lacking normal gravitropic response. Exogenous ethylene treatment inhibited gravitropic response in the normal (wild type) pea rook, but had no significant effect to restore the unresponsiveness in the mutant Neither inhibitors of ethylene biosynthesis nor antagonists of ethylene action were able to bring about the development of gravioopic curvature in the ageotropum roots. Auxin action seems to be normal since asymmetric application of agar blocks containing auxin to the mutant roots caused normal gravitropic response to occur. Endogenous as well as auxin-induced ethylene production in tissue segments of the mutant root was about equal to that of the wild type. However no appreciable lateral transport of labeled auxin was observed in glavistimulated mutant roots whereas typical auxin asymetry was apparent in the wild type roots under the same conditions. It is concluded that the mutant has a defect in either gravity perception or its transduction, but not in the effector system involving auxin action.

We report the tissue-specific distribution of chitinolytic activity in Korean ginseng root and characterize two 31-kDa chitinolytic enzymes. These two enzymes (SBF1 and SBF2) were purified 70- and 81-fold with yields of 0.75 and 1.25%, respectively, and exhibited optimal pH and temperature ranges of 5.0-5.5 and 40-$50^{\circ}C$. With [$^3H$]-chitin as a substrate, $K_m$ and $V_{max}$ values of SBF1 were 4.6 mM and 220 mmol/mg-protein/h, respectively, while those of SBF2 were 7.14 mM and 287 mmol/mg-protein/h. The purified enzymes showed markedly less activity with p-nitrophenyl-N-acetylglucosaminide and fluorescent 4-methylumbelliferyl glycosides of D-N-acetylglucosamine oligomers than with [$^3H$]-chitin. End-product inhibition of both enzymes demonstrated that both are endochitinases with different N-acetylglucosaminidase activity. Furthermore, the $NH_2$-terminal sequence of SBF1 showed a high degree of homology with other plant chitinases whereas the $NH_2$-terminal amino acid of SBF2 was blocked.
Symbiotic associations between land plants and fungi have been known for more than one hundred years. Vesiculararbuscular mycorrhizas (VAM) are the most common symbiosis in flowering plants and can be recognized in almost all plant families. These fungal associations play a very important role in the growth and survival of plant species. However, with respect to the importance and intensity of the VAM, there is great variation among host species. Our knowledge of the VAM fungus-plant association in Araliaceae is very limited. After the first reports of the occurence of VAM in lateral roots of Panax species, mycorrhizal structures are now described as special structures representing the so-called Paris type. In this type, the development of new spores and vesicles is extremely low. This and the type of colonization of the fungus in Panax roots indicates on, one hand, the high intensity of the VAM and, on the other hand, a remarkable dependency for VAM in members of the Panax species. Therefore, it can be easily understood that cultivated Panax plants exhibit a significant uptake of nutrients and this leads to an extremely depleted soil at harvest. Further, the soil is nearly free of the spores of VAM fungi as they germinate each year on the newly developing Panax roots.
This study was conducted to establish an identification method age for cultivated ginseng (Panax ginseng C.A. Meyer) based on counting the stem vestige at rhizome. Weight, diameters of body and lateral roots, and ratios of those diameters were overlapped with those of other ages, so these are not enough as a criterion for identification of ginseng age. Since the stem vestige at rhizome was remained clearly from the 3rd-year age, root age can be the number of stem vestige +2. The number of stem vestige should be counted at the one side of rhizome with abundant vestige for multi stewed plant. It is necessary that the number of year-ring in root is investigated for rhizome damaged plant, but it must be considered that the new year-ring can be shown clearly after around loth of June. All dormant roots had damaged rhizome by some reasons.
