Genetical identification and morphological description of the larvae and juveniles of Porocottus leptosomus (Pisces: Cottidae) from Korea

  • The larvae and juveniles of Porocottus leptosomus belonging to the family Cottidae were collected (n = 95, 3.9-16.5 mm in body length, BL) from Busan, Korea, in March 2015. The larvae and juvenile were identified using DNA barcoding as P. leptosomus, and their morphological description was presented in detail. The yolk-sac larvae (3.9-5.6 mm BL) body was slightly compressed, the head was large, the eye was round and large, and the anus was before the middle of the body. The preflexion larvae (5.2-10.0 mm BL) body length drastically increased; caudal fin rays began to occur. The flexion larvae (9.4-11.8 mm BL) notochord flexion started; dorsal, pectoral, and anal fin rays began to occur; pelvic fin buds are seen; they possessed a pair of parietal spine; and a pair of supraocular cirri was first to develop. At 12 mm BL, the notochord was completely flexed. The larva stage (3.9-12.6 mm SL) had the stellate melanophores in the head, isthmus, gut, and tail (along to the ventral midline). During the juvenile stage (11.4-16.5 mm BL), melanophores covered the head and began to form five black bands on the side of the body. The larvae of P. leptosomus spent pelagic life, but moved to the bottom during the juvenile stage. The larvae and juveniles of P. leptosomus differ from other cottid larval fishes by body shape, melanophore head pattern, and spine development. P. leptosomus can be distinguished from Porocottus allisi by morphological development and the occurrence of larval fish: preopercular spine development, melanophore pattern, and caudal fin development.

Early Osteological Development of Larvae and Juveniles in Red Spotted Grouper, Epinephelus akaara (Pisces: Serranidae)

  • We observed the osteological development of larval and juvenile red spotted grouper (Epinephelus akaara) in order to generate data for the assessment of skeletal deformities and to inform phylogenetic systematics research. Larvae and juveniles were obtained from a aquafarm in Muan-gun, Jeolla-namdo Province, Korea. The average water temperature at the time of breeding was $23.0^{\circ}C$ and average water salinity was 33.0 psu. Freshly hatched fish larvae had not undergone any ossification, but ossification of the parasphenoid bone, which forms the base of the cranium, occurred as the juveniles reached an average body length (BL) of 2.49 mm. At the same time, ossification of the preopercle and opercle occurred in the operculum, and ossification of the maxilla, which forms the upper jaw, and the dentary bones, which form the lower jaw, began. In addition, ossification of the vertebra occurred by formation of 7 vertebral centra and the neural spine in the abdominal vertebra. When the juveniles reached an average (BL) of 5.22 mm, ossification of the nasal, lateral ethmoid, and alisphenoid bones occurred in the cranium; ossification of the endopterygoid and metapterygoid bones began in the palatine region; and ossification of the hypohyal and interhyal bones occurred in the hyoid arch. At an average (BL) of 20.9 mm, ossification of the basisphenoid bone in the cranium and the suborbital bone in the orbital region occurred. Ossification of the vertebra then occurred by the formation of long pairs of ribs from the third to the ninth abdominal vertebrae, completing osteological development.

영산강 하구역 주변해역의 난자치어 분포 (Distribution of Fish Eggs, Larvae and Juveniles around the Youngsan River Estuary)

  • Distribution of ichthyoplanktons and its relation to environmental factors (temperature, salinity, $chlorophyll\;\alpha$) around the Youngsan River Estuary of Mokpo, southwest coast of Korea were investigated based on the samples collected bimonthly from February to December, 2001. Unidentified fish eggs were most abundant $1,345\;eggs/100m^3$ in August, while eggs of Engraulis japonicus were most abundant $63\;eggs/100m^3$ in June. A total of forty species of fish larvae and juveniles belonging to the twenty families under six orders were caught, with six gobiids, four icefishes, four croakers and three herrings being represented. Fish larvae and juveniles peaked $17,757\;inds./100m^3$ in August, which consisted of $91.02\%$ of gobiids, $3.69\%$ of gunnels and $3.67\%$ of icefishes, and they were most abundant $7,467\;Inds./100m^3$ at St. 7. Cluster analysis based on the individuals of fish larvae and juveniles showed that six months were classified into two groups at the dissimilarity 70, the first was composed of June and August and the second was February and December and April and October were not clustered with any other months. In August when species diversity index was the highest, eight stations were classified into two groups at the dissimilarity 70, the first was composed of St. 1, 3, 4, 5 and 8 and the second was St. 6, 7 and 2. As a result from correspondence analysis, there is no effect of the environmental factors with a density of fish eggs and larvae, but correlation could be found between a density of fish eggs and $chlorophyll\;\alpha.$

  • Dragonet fish (Callionymidae), living in benthic upper 900 m of all subtropical, tropical and temperate oceans, comprises 200 species in 20 genera worldwide, of which 18 species in 6 genera occur in Korea. Although dragonet fish plays an important role in linking between top predators and lower trophic levels, there are only few studies about their early life history. Herein, we present molecular and morphological data on larvae and juveniles of Bathycallionymus kaianus (Günther, 1880) collected from the Korean waters. During 2016 to 2018, one preflexion larvae [2.69 in total length (TL)], three flexion larvae (3.65 and 4.77 mm TL), six postflexion larvae (6.07 and 7.94 mm TL), and three juveniles (10.81 and 12.26 mm TL) were collected in the East Sea, Korea Strait, and Jeju Island using Bongo net. Of them, 13 individuals were identified through molecular markers (COI or 16S rRNA) and morphologically described. The larvae of B. kaianus are well distinguished from other species of Callionymidae in melanophore distribution, body shape and development of preopercular spine. It was very similar to larvae of two Repomucenus species, R. valenciennei and R. virgis, but was clearly distinguished in melanophore distribution, preopercular spine development, and head shape.

Ontogenetic comparison of larvae and juveniles of Diaphus garmani and Benthosema pterotum (Myctophidae, Pisces) collected from Korea

  • During June 2017, we collected two postflexion larvae (6.01 and 7.56 mm in standard length [SL]) and two juveniles (7.72 and 9.62 mm SL) belonging to Myctophidae in the waters of Jejudo Island. Those four individuals were identified as Diaphus garmani, which had not been reported in Korea. They were distinguished from Benthosema pterotum by melanophores in the abdominal cavity (absent in D. garmani vs. present in B. pterotum) and the development of photophores (developed in D. garmani vs. rudimentary in B. pterotum) when shorter than 10.0 mm SL. Analysis of 16S rRNA sequences showed that the sequences of four individuals matched those of adult D. garmani (Kimura 2-parameter distance: 0.6-0.8%). This is the first record of larvae and juveniles of D. garmani in Korean waters, and we propose a new Korean name, Gar-ma-ni-sat-bi-neul-chi.

Egg Development of the Ussurian Bullhead Fish, Leiocassis ussuriensis (Pisces: Bagridae) and Morphological Development of Its Larvae and Juveniles

  • This study was examined the ovogenesis of Ussurian bullhead, Leiocassis ussuriensis and the morphological development of its larvae and juveniles and to use the results as basic information for the preservation of species and resource enhancement. For artificial egg collection, human chorionic gonadotropin (HCG) was injected at a rate of 10 IU per gram of fish weight. During breeding period, water temperature maintained at $24.5{\sim}26.5^{\circ}C$ (mean $25.0{\pm}0.05^{\circ}C$). The process of ovogenesis reached the two-cell stage in 50 minutes after fertilization. In 73 hours of fertilization the movement of the embryoid body became active state and the larvae began to hatch from the tail through the oolemma. Length of prelarvae were 6.33~6.50 mm long (mean $6.40{\pm}0.06mm$) just after hatching having yolk with their mouth not opened. After thirty eight days of hatching, juveniles were 30.6~32.5 mm long (mean $31.5{\pm}0.65mm$). The color was dark yellowish brown throughout the entire body, and the number of caudal fin rays developed to thirty six perfectly.

  • 이번 연구에 사용된 문치가자미 어미 (female : 전장 25.1~30.4 cm, male : 전장 24.5~28.5 cm)는 1999년 12월 20일 전라남도 여수시 돌산 군내리 활어위판장에서 어미를 구입하여 전라남도 수산종합관 사육동으로 운반하여 건도법으로 인공 수정시켰고, 부화한 자치어는 사육수조에서 사육하면서 자치어의 형태발달과정을 관찰하였다. 부화 직후 자어의 전장은 3.13~3.42 mm (평균 3.31 mm, n = 10)로 눈은 황갈색을 띠며 입과 항문은 열려 있지 않았다. 부화 후 3~6일째의 자어는 전장이 3.35~4.61 mm (평균 3.45 mm, n = 10)로 입과 항문이 열리고, 난황이 거의 흡수되어 입이 커지면서 섭이활동이 활발하였다. 부화 후 25일째의 자어는 전장 5.47~5.91 mm (평균 5.96 mm, n = 10)로 척색말단이 위로 굽어지기 시작하고, 꼬리지느러미즐기의 원기가 형성되기 시작하였고, 등지느러미의 원기가 출현하였다. 부화 후 35일째의 자어는 전장 6.83~7.60 mm (평균 7.38 mm, n = 10)로 등지느러미 및 뒷지느러미에 줄기가 출현하고, 왼쪽 눈이 조금 이동하였다. 부화 후 55일의 치어는 전장 9.38~11.73 mm (평균 10.03 mm, n = 10)로 눈은 완전히 머리부분의 오른쪽으로 이동하여 체형이나 반문이 성어와 완전히 닮아 있었고, 저서생활로 이행하였다 (D. 68~70 : A. 50~52 : P. 11: V. 6 : C. 18~19).

실내수조에서 사육한 참조기 배발생 및 자치어의 형태 (Embryonic Developmen Larvae and Juveniles of the Small Yellow Croaker (Larimichthys polyactis) Reared in Aquarium)

  • On 11 June 1991, eggs from the brood stock of small yellow croaker (Larimichthys polyactis) were artificially fertilized using the standard dry method and were hatched. Each of the fertilized eggs (1.1-1.2 mm in diameter) had an oil globule and was transparent and buoyant. The fertilized eggs hatched in a range of water temperatures $(17.5-20.3^{\circ}C)$ 44 hrs after fertilization. The total lengths of the newly hatched larvae were 3.1-3.3 mm, and these hatchlings had 31 myotomes (10+21). Melanophores and yellow-brown chromatophores were concentrated on the head, at the ventral part of the yolk, and in the middle of the tail. Four days after hatching, the larvae completely absorbed the yolk and became flexions of 5.1-5.5 mm in total length. Fifteen days after hatching, one spine (the anterior tip of the maxillary) appeared in the upper jaw and three spines developed at the upper parts of the eyes and on the posterior part of the head. At this stage, the larvae were approximately 8.3 mm long. Thirty-nine days after hatching, juveniles (1.9-3.4 mm in total length) had a pointed tail fin. By 66 days after hatching, the juvenile fish (about 4.0-6.5 mm in total length) were similar to adult fish in body shape. The larvae of L. polyactis could be distinguished from those of L. croacea by two distinct characteristics: the large number of vertebrae (28-29), and a relatively small bony ridge on the occipital region of the head.

미꾸리 Misgrunus anguillicaudatus (Cypriniformes: Cobitidae)의 난발생 및 자치어 형태발달 (Morphological Development of Eggs, Larvae and Juveniles of the Misgrunus anguillicaudatus (Cypriniformes: Cobitidae))

  • This study was carried out to clarify the egg, larvae and juveniles development of Misgurnus anguillicaudatus, and relationships of M. anguillicaudatus and M. mizolepis, Cobitididae Fishes. The adult fishes were collected in Samsan-cheon, Haenam-gun, Jeollanam-do, Korea and their spawning inducement was carried by ovaprim injections. The egg shape was circular and the size was average 1.12 mm. The eggs were hatched at 61 to 72 h after fertilization. The newly hatched larvae had an average 3.23 mm in total length (TL). At 5 days after hatching, the larvae reached to post larval stage and they were 10.3 mm in TL. At 19 days after hatching, it reached to juvenile stage and was 25.3 mm in TL. The egg size of M. anguillicaudatus was almost same as M. mizolepis but the hatching period of M. anguillicaudatus has taken longer. It was possible for interspecific distinguishability of M. anguillicaudatus and M. mizolepis when their larvae reached to juvenile stage by the development of keel-like ridges.

Osteological Development of the Larvae and Juvenile of Favonigobius gymnauchen (Pisces:Gobiidae)

  • This study was carried out to observe the development of the autonomous skeletal development of the Favonigobius gymnauchen. Total length (TL) of larvae 3 days after hatching (DAH) were mean TL of 3.34 mm, with a line-shaped parasphenoid ossification in the cranium and basioccipital ossification in the back. The 10 DAH larvae had a mean TL of 5.20 mm, with the number of caudal vertebrae increasing to 15. The urostyle and two hypural bones in the lower part also began to ossify. The 23 DAH juveniles had a mean TL of 8.47 mm. The pectoral girdle's skeleton was completed as the scapula and coracoid were ossified. The pelvic girdle also fully supported the ventral fin as its ossification was completed. Favonigobius gymnauchen and Tridentiger obscurus showed similar characteristics in terms of the anus location of hatched larvae, number of myotomes, and melanophore distribution during the morphological development of the larvae and juveniles. However, this study confirmed differences in the development of the vertebrae and urostyle bone.
