This paper describes an algorithm that can efficiently analyze the ECG signal using frame knowledge representation technique. Input to the analysis process is a set of significant points which have been extracted from an original sampled signal(lead II) by the syntactic peak recognition algorithm. The hierarchical property of ECG signal is represented by hierarchical AND/OR graph. The semantic information and constraints of the ECG signal are desctibed by frame. As the control mechanism for labeling points, the search mechanism with the mixed paradigms of data-driven and model driven hypothesis formation, scoring function, hypothesis modification network and instance inheritance are used. We used the CSE database in order to evaluate the performance of the proposed algorithm.
For a large corpus of time-aligned data, HMM based approaches are most widely used for automatic segmentation, providing a consistent and accurate phone labeling scheme. There are two methods for training in HMM. Flat starting method has a property that human interference is minimized but it has low accuracy. Bootstrap method has a high accuracy, but it has a defect that manual segmentation is required In this paper, a new algorithm is proposed to minimize manual work and to improve the performance of automatic segmentation. At first phase, voiced, unvoiced and silence classification is performed for each speech data frame. At second phase, the phoneme sequence is aligned dynamically to the voiced/unvoiced/silence sequence according to the acoustic phonetic rules. Finally, using these segmented speech data as a bootstrap, phoneme model parameters based on HMM are trained. For the performance test, hand labeled ETRI speech DB was used. The experiment results showed that our algorithm achieved 10% improvement of segmentation accuracy within 20 ms tolerable error range. Especially for the unvoiced consonants, it showed 30% improvement.
Several herbs including Artemisia are known to possess conceptive property. In the present study, mouse spermatozoa were incubated with ethanol extract of Artemisia vulgaris leaves. The effect of extract on acrosome exocytosis was studied by labeling spermatozoa with fluorescein isothiocyanate (FITC) peanut agglutinin and by staining with Coomassie blue. Viability and membrane integrity were studied by Trypan-blue staining and hypo-osmotic swelling test. Artemisia extract at very low concentration caused precocious acrosome reaction and loss of sperm viability. Acrosome reaction increased remarkably from 22.63% to 88.42% with increasing extract concentration from 0 to 2,000 ㎍/mL. However, the viability loss of spermatozoa was increased from 11.71% in control to 63.73% in samples treated, evaluated by Trypan-blue staining method. Membrane damage caused by the extract, evaluated by hypo-osmotic swelling test was even low, ranging from 2.27% to only 24.23%. These results indicate that Artemisia extract might block fertilization by causing precocious acrosome exocytosis in spermatozoa. A direct contraceptive effect was tested by injecting the plant extract into the vagina of female mice and then allowing them to mate with normal males. The treated female mice delivered significantly fewer litters in comparison to the control.

Label-free optical diffraction tomography (ODT), an imaging technology that does not require fluorescent labeling or other pre-processing, can overcome the limitations of conventional cell imaging technologies, such as fluorescence and electron microscopy. In this study, we used ODT to characterize the cellular organelles of three different stem cells-namely, human liver derived stem cell, human umbilical cord matrix derived mesenchymal stem cell, and human induced pluripotent stem cell-based on their refractive index and volume of organelles. The physical property of each stem cell was compared with that of fibroblast. Based on our findings, the characteristic physical properties of specific stem cells can be quantitatively distinguished based on their refractive index and volume of cellular organelles. Altogether, the method employed herein could aid in the distinction of living stem cells from normal cells without the use of fluorescence or specific biomarkers.
This paper describes the development of $^{99m}Tc$ tricarbonyl cysteine as potential renal function diagnostic radiopharmaceutical and evaluation of its biological characteristics using experimental animals. l-Cysteine was labeled efficiently with $^{99m}Tc$ tricarbonyl precursor $([^{99m}Tc(CO)_3(H_2O)_3)]^{+})$ under 30 min heating at ${75^{\circ}C}$. Labeling yield and stability were analyzed by high performance liquid chromatography (HPLC). The biodistribution property of $^{99m}Tc$ tricarbonyl cysteine in mice and its dynamic imaging profiles in rabbits were carried out. To investigate the excretion mechanism of $^{99m}Tc$ tricarbonyl cysteine, tubular transport inhibition test with probenecid was adopted. $^{99m}Tc$ tricarbonyl cysteine was obtained with a high labeling yield under the moderate condition. The results of biodistribution experiments of $^{99m}Tc$ tricarbonyl cysteine in ICR mice at 3 and 90 min provided that $^{99m}Tc$ tricarbonyl cysteine was very highly accumulated in the kidney and bladder, thereby almost 99% of $^{99m}Tc$ tricarbonyl cysteine was excreted within 90 min post injection. The same results were confirmed by the whole body dynamic images for 30 minutes and static images in rabbits at given time intervals after injection. Renogram of $^{99m}Tc$ tricarbonyl cysteine in rabbits showed that its $T_{max}$ and $T_{1/2}$ of $^{99m}Tc$ tricarbonyl cysteine were $2.33{\pm}0.56$ and $4.30{\pm}0.79$ min, respectively. The $T_{max}$ of $^{99m}Tc$ tricarbonyl cysteine with probenecid pretreatment was $2.30{\pm}0.17$ min, whereas $T_{1/2}$ of that with probenecid pretreatment was $17.0{\pm}32.47$ min. $T_{1/2}$ of $^{99m}Tc$ tricarbonyl cysteine with probenecid pretreatment was significantly different, as compared to the result without probenecid (p<0.0001). The results showed that the excretion of $^{99m}Tc$ tricarbonyl cysteine was extremely affected by probenecid. Therefore, $^{99m}Tc$ tricarbonyl cysteine was rapidly excreted from the kidney principally by the tubular secretion.

Leaf extracts of Cassia alata L (akapulko), traditionally used for treatment of a variety of diseases, were evaluated for their potential antitumor properties in vitro. MTT assays were used to examine the cytotoxic effects of crude extracts on five human cancer cell lines, namely MCF-7, derived from a breast carcinoma, SK-BR-3, another breast carcinoma, T24 a bladder carcinoma, Col 2, a colorectal carcinoma, and A549, a nonsmall cell lung adenocarcinoma. Hexane extracts showed remarkable cytotoxicity against MCF-7, T24, and Col 2 in a dose-dependent manner. This observation was confirmed by morphological investigation using light microscopy. Further bioassay-directed fractionation of the cytotoxic extract led to the isolation of a TLC-pure isolate labeled as f6l. Isolate f6l was further evaluated using MTT assay and morphological and biochemical investigations, which likewise showed selectivity to MCF-7, T24, and Col 2 cells with $IC_{50}$ values of 16, 17, and 17 ${\mu}g/ml$, respectively. Isolate f6l, however, showed no cytotoxicity towards the non-cancer Chinese hamster ovarian cell line (CHO-AA8). Cytochemical investigation using DAPI staining and biochemical investigation using terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL)-a method used to detect DNA fragmentation-together with caspase assay, demonstrated apoptotic cell death. Spectral characterization of isolate f6l revealed that it contained polyunsaturated fatty acid esters. Considering the cytotoxicity profile and its mode of action, f6l might represent a new promising compound with potential for development as an anticancer drug with low or no toxicity to non-cancer cells used in this study.
Most angiogenesis assays are performed using endothelial cells. However, blood vessels are composed of two cell types: endothelial cells and pericytes. Thus, co-culture of two vascular cells should be employed to evaluate angiogenic properties. Here, we developed an in vitro 3-dimensional angiogenesis assay system using spheroids formed by two human vascular precursors: endothelial colony forming cells (ECFCs) and mesenchymal stem cells (MSCs). ECFCs, MSCs, or ECFCs+MSCs were cultured to form spheroids. Sprout formation from each spheroid was observed for 24 h by real-time cell recorder. Sprout number and length were higher in ECFC+MSC spheroids than ECFC-only spheroids. No sprouts were observed in MSC-only spheroids. Sprout formation by ECFC spheroids was increased by treatment with vascular endothelial growth factor (VEGF) or combination of VEGF and fibroblast growth factor-2 (FGF-2). Interestingly, there was no further increase in sprout formation by ECFC+MSC spheroids in response to VEGF or VEGF+FGF-2, suggesting that MSCs stimulate sprout formation by ECFCs. Immuno-fluorescent labeling technique revealed that MSCs surrounded ECFC-mediated sprout structures. We tested vatalanib, VEGF inhibitor, using ECFC and ECFC+MSC spheroids. Vatalanib significantly inhibited sprout formation in both spheroids. Of note, the $IC_{50}$ of vatalanib in ECFC+MSC spheroids at 24 h was $4.0{\pm}0.40{\mu}M$, which are more correlated with the data of previous animal studies when compared with ECFC spheroids ($0.2{\pm}0.03{\mu}M$). These results suggest that ECFC+MSC spheroids generate physiologically relevant sprout structures composed of two types of vascular cells, and will be an effective pre-clinical in vitro assay model to evaluate pro- or anti-angiogenic property.
최근에는 슈퍼-픽셀 (super-pixel)은 컴퓨터 발전 응용에 널리 사용되고 있다. 슈퍼 픽셀 알고리즘은 픽셀을 지각적으로 실행이 가능한 영역으로 변환하여 그리드 픽셀의 경직된 특징을 줄일 수 있다. 특히, 슈퍼 픽셀은 깊이 추정, 골격 작업, 바디 라벨링 및 기능 국소화 등에 사용된다. 그러나 이러한 작업을 수행하기 위해 우수한 슈퍼 픽셀 파티션을 생성하는 것은 쉽지 않다. 특히 슈퍼 픽셀은 비합, 지속, 폐쇄, 지각 불변과 같은 형태 측면을 고려할 때보다 의미있는 특징을 만족시키지는 못한다. 본 논문에서는 단순 선형 반복 클러스터링과 퍼지 클러스터링 개념을 결합한 고급 알고리즘을 제안한다. 단순 선형 반복 클러스터링 기술은 이미지 경계, 속도, 메모리 효율이 기존 방법보다 높다. 그것은 형태 측면의 맥락에서 슈퍼 픽셀 형태에 대해 양호하게 작거나 규칙적인 특성을 제안하는 것은 아니다. 퍼지 유사성 측정은 제한된 크기와 이웃을 고려하여 합리적인 그래프를 제공한다. 보다 작고 규칙적인 픽셀을 얻으며 부분적으로 관련된 특징을 추출 할 수 있다. 시뮬레이션은 퍼지 유사성 기반 슈퍼 픽셀 생성은 사람의 이미지를 분해하는 방식으로 자연적 특징을 대표적으로 나타낸다.
Objectives : The purpose of this study was to apply informations related to herbal medicines to IDMP (Identification of Medicinal Products), an ISO standards related to medicinal products substances, for systematic collection of data through the integration of informations on distribution, manufacturing, and management of herbal medicines. Methods : By analyzing ISO 11238 and ISO/TS 19844, elements that can be used in the information model of herbal medicine were derived from the identification of medicinal products information model on substances. The labeling specified in the safety and quality control regulations for herbal medicines was mapped to the IDMP information model, and ginseng was applied as an example. Results : Herbal medicine corresponded to substance in IDMP. Among the five types of substances specified by IDMP, herbal medicines were expressed as structurally diverse. Scientific name was used as an invariant property of herbal medicine, and the substance level included information about source material and modification, and specifically included information about the scientific name, medicinal part, fraction, and processing. In addition, the specified substance level had information on the constituents, characteristic attributes, manufacturing, and grade of the herbal medicine. Conclusions : It is necessary to establish a code system for identifying herbal medicines. In order to apply the IDMP standards, research on the development of standard terms is required to express the characteristics of herbal medicines. In addition, information for identification of herbal medicines is also required, and information from production to consumption should be systematically accumulated and managed for actual application.
This study was performed to provide scientific information for consumers and to increase the quality and consumption of Sullungtang prepared with bone from Hanwoo. The physico-chemical, sensory, nutritional properties of Sullungtang by different parity were investigated. The extracting extents of blood were significantly higher for heifer among Hanwoo cows with different parity(none, the second or the fourth) and for back bones among different bone portions (round, hind shank, arm, fore shank or back bone) (p〈0.05). For Sullungtang extracted from bones of heifer was higher in collagen contents than those extracted from cows with the second or the fourth parity. Sullungtang extracted from bones of heifer and cows with the second parity had significantly higher contents of condroitin and viscosity than those of cows with the fourth parity, however there was no significant difference between heifer and cows with the second parity. In color, L values were significantly high, and a and b values were significantly low for Sullungtang extracted from bones of heifer when compared to those extracted from the other cows with the second parity. The contents of total nitrogen, sodium, or calcium were higher for Sullungtang extracted from heifer than those from the cows with the second or the fourth parity. In amino acid compositions, the percentages of glycine were highest and proline and glutamic acid were followed fro all Sullungtang samples from Hanwoo. The results showed that the overall quality of Sullungtang significantly decreased as the parity increased for Hanwoo cows. The Sullungtang extracted from bones of heifer had the best sensory scores as well as nutritional quality when compared to those extracted from the cows with parity. Therefore, the labeling and price for cow bones should be differently evaluated by their parity and age in marketing system. 본 연구는 한우뼈로 만든 탕의 품질에 대한 소비자의 요구에 부응하고 한우뼈의 부가가치 향상 및 소비 확대를 위하여 산차별에 따른 이화학적, 관능적 및 영양적 특성을 구명하고자 실시하였다. 산차별에 따른 혈액추출정도는 한우 미경산이 유의적으로 높았으며 (p〈0.05), 부위간 비교에서는 잡뼈가 유의적으로 가장 높았다(p〈0.05). 점도는 미경산과 2산차 간에는 유의성이 없었지만 미경산이 높은 경향을 나타내었다. 콜라겐 함량은 미경산이 유의적으로 높았고(p〈0.05), 콘드로이친황산 함량에서는 미경산이 유의적으로 높았으나(p〈0.05), 2산 차이와 유의성이 인정되지 않았지만 높은 경향을 보였다. 명도에서 미경산 추출물이 높은 경향을 보였고 적색도와 황색도는 미경산이 유의적으로 낮았다(p〈0.05). 관능평가는 미경산이 유의적으로 높게 평가되었다(p〈0.05). 총질소, 나트륨 및 칼슘 함량은 미경산 추출물이 유의적으로 가장 높았다(p〈0.05). 한우암소 추출물의 아미노산 조성중 Glycine이 가장 높았고 다음은 Proline, Glutamic acid 순이었다. 이상의 결과를 종합하여 볼 때 전체적으로 한우 미경산우가 우수하였고 산차가 증가할수록 품질이 떨어지는 것으로 나타나 한우암소뼈를 판매시 산차 및 나이에 따라 가격을 차별화 시키는 것이 바람직할 것으로 사료된다.
