Optical Clearing Agent Decreases Scattering of Stratum Corneum and Modulates Physical Properties of Corneocytes by Hydration (광산란 감소 물질에 의한 피부 각질층의 산란 감소 및 수화에 의한 각질세포의 물리적 특성에 대한 연구)

  • The objective of this article is to provide analytical tools for the scattering of stratum corneum (SC) and to check whether the optical clearing agents (OCAs) could be applied in optics affecting the scattering reduction. Dark field images of tape striped corneocyte separates scattered light of the SC from others in vitro. Several optical clearing agents were tested to reduce the scattering. Physical properties of SC such as water contents, keratin configuration and volume after OCAs treatment were investigated by FT-IR and 3D laser microscope. Several reducing sugars, monomeric sugars, sugar alcohol, and hyaluronic acid, which were used as humectants in cosmetic field, also reduced scattering. However, unlike dehydration in optics, water penetrated into the keratin in SC and scattering was decreased at low concentration of OCAs. In that condition, the volume of corneocyte was increased and stiffness seemed to decrease. The analyzing of tape-stripped SC, showed the change of optical and physical properties of corneocyte by optical clearing agents. The hydration of SC layer by optical clearing agents decreased the scattering of corneocyte and thus improved the skin appearance and moisturizing effect, which are important benefits in the cosmetic field and could provide new possibility to develop skin care study targeting at SC.

Efficiency of Essential oil about the Skintroubles induced Surfactants - Palmarosa, Neroli essential oil - (계면활성제 유발된 피부장애의 아로마 에션셜 오일의 유효성 연구 - Palmarosa, Neroli essential oil을 중심으로 -)

  • The effectiveness of Palmarosa and Neroli essential oil on dry skin of rat induced by kitchen detergent are investigated. The experimental groups were divided the control group (C), group treated with surfactant (A1), group treated with Palmarosa (A2), group treated with Neroli (A3), group treated with Palmarosa and Neroli (A4). The protein analysis of all experimental groups was performed with SDS polyacrylamide gel electrophoresis and observation of epidermis and the alteration of mast cell were performed with photomicroscope. According to the protein analysis, the A3 group treated with Neroli essential oil was appeared the most similar with the control group. And then the A4 group treated with Palmarosa + Neroli essential oil was appeared the most similar with the control group. According to the results of morphologic view with keratin layer, the keratin layer's breakaway resulting from Palmarosa essential oil, the keratin layer's restoration resulting from Neroli essential oil was appeared. And then the structure of the epidermal layer was preserved by hyperkeratosis reaction. In photomicrosopic obersevation of mast cell to examine the inflammatory reactions, the increase in size and number of mast cell were showed in A1 group treated with surfactant compared to the control group (C). The number of mast cells definitely decreased in groups (A3, A4) which were treated with Neroli essential oil.

Myoepithelioma Arising From Trachea; A case report (기관의 근상피종 수술치험 1례)

  • Myoepithelioma of the trachea is an extremely rare disease. The neoplasm shows histologic features identical to those described in myoepithelioma of salivary glands. The myoepithelioma cells demonstrate the numerous myofilament, desmosomal cellular attachment, cytoplasmic glycogen and pinocytotic vesicles. In immunohistochemical study, myoepithelioma cells show the positive antibody reactions to actin, keratin, vimentin and S-100 protein. A case of myoepithelioma arising from trachea in the seventy-two-year old male diagnosed by microscopic finding and immunohistochemistry is presented.

  • A pulmonary keratinizing cyst was found incidentally in a 30-week-old female Sprague-Dawley rat. The cyst was lobe of the lung, lined by stratified squamous epithelium and filled with concentric layers of desquamated keratin. The wall consisted of 2-4 layers of well differenciated squamous cells. The adjacent lung tissues were slightly compressed but not otherwise damaged by the cyst. There was no inflammatory reaction or mitotic figures observed in the cyst.

Squamous cell carcinoma of the skin in an Australian shepherd dog (개에서 피부 편평상피세포암의 발생과 치료 예)

  • Squamous cell carcinoma of the skin was diagnosed in an 11-year-old, Australian shepherd dog with a hard mass on the right rump area. On histopathological examination of the tumor showed laminated keratin "pearls" surrounded by proliferated squamous cells, and mitotic figures. The dog was treated by surgical excision and chemotherapy with vinblastine sulfate, cyclophosphamide and prednisolone for 4 weeks. The tumor was effectively treated with a combination of surgery and chemotherapy.motherapy.

  • A case of ovarian teratoma is reported in a 11-year-old female Korean Jindo dog. Grossly, the left ovary was markedly enlarged ($5{\times}4{\times}4 cm in size$) and contained several cystic spaces filled with hairs and yellowish, fragile deratinous material on cross section. Histological observation of the hair and keratin containing cystic structures lined by stratified squamous epithelium, mature adipose tissue, and bone and cartilage is compatible with a diagnosis of ovarian teratoma, pyometra was also present in this dog. This is believed to be the first report on canine ovarian teratoma in Korea.

  • Steatocystoma multiplex is an uncommon benign skin disease, which typically manifests as numerous intradermal cysts that can be scattered anywhere on the body. Although usually asymptomatic, it can be significantly disfiguring. One type of steatocystoma multiplex is known to be associated with the autosomal dominant inheritance of a mutation in the gene coding for keratin 17 (KRT17). In such cases, it is often concurrent with other developmental abnormalities of the ectoderm-derived tissues, such as the nails, hair, and teeth. To the best of our knowledge, few cases have been reported of steatocystoma multiplex of the oral and maxillofacial region. This report describes a case of steatocystoma multiplex of both sides of the neck and multiple dental anomalies, with a focus on its clinical, radiological, and histopathological characteristics, as well as the possibility that the patient exhibited the familial type of this condition.

Purification and some Properties of Keratinolytic Protease Produced by Pseudomonas sp. KP-364. (Pseudomonas sp. KP-364가 생산하는 Keratinolytic Pretense의 정제 및 성질)

  • A keratinolytic protease was purified from the culture medium of Pseudomonas sp. KP-364 by use of an assay of the hydrolysis of feather keratin. Membrane ultrafiltration and DEAE-cellulose ion-exchange resin and Sephadex G-150 gel chromatographies were used to purify the enzyme. The specific activity of the purified keratinolytic protease relative to that in the original medium was approximately 72-fold high. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Sephadex G-150 chromatography indicated that the purified keratinolytic protease is monomeric and has a molecular weight of 36 kDa. The optimal pH and temperature of the keratinolytic protease activity were 6.6 and 37 C, respectively, and the keratinolytic protease was relatively stable at pH value from 3.0 to 10.0 at 37 C for 1hour. The keratinolytic protease was inhibited by EDTA and EGTA, indicating that the keratinolytic protease was a kind of metalloprotease that require Li+ for cofactor.

A Distribution of Keratinophilic Fungi Isolated from the Soil of Haeundae Beach in Korea (부산 해운대 백사장에서 분리한 각질친화성 피부사상균의 분포)

  • Keratinophilic fungus (KPF), a type of dermatophytes, is usually present as normal flora on the skin of humans and animals but can produce ring worm-like dermatophytosis by invading the skin in infected individuals. They are distributed worldwide, but their occurrences vary distinctively in accordance with the geographical location and environmental change. Because these fungi grow by degrading keratin, they are abundantly found on the skin, hair, and nails, which are rich in keratin. To investigate the presence of keratinophilic fungi in the soil, we selected a popular beach in South Korea, Haeundae Beach, where numerous people gather each year during the summer holidays. Hundred soil samples were analyzed using the hair-baiting technique, among which, a total of 23 colonies of KPF were identified from 21 soil samples. The identified KPF were Microsporum gypseum (43%), Chrysosporium spp. (35%), Trichophyton ajelloi (13%), and Microsporum cookie (9%). This study confirmed that pathogenic fungi can be found in places crowded by many people. Further research and continuous data collection are needed to confirm the distribution of pathogenic KPF.

Association of the KAP 8.1 Gene Polymorphisms with Fibre Traits in Inner Mongolian Cashmere Goats

  • The objective of this study was to investigate polymorphisms of keratin-associated protein 8.1 (KAP8.1) gene and its effect on fibre traits of Chinese Inner Mongolian Cashmere goats. The fibre traits data investigated were cashmere fibre diameter, combed cashmere weight, cashmere fibre length and guard hair length. Five hundred and forty animals were used to detect polymorphisms in the complete coding sequence of the hircine KAP8.1 gene by means of PCR-SSCP. The results identified six genotypes, AA, BB, CC, AB, AC and BC, coded for by three different alleles A, B and C. Two SNPs in the coding region were confirmed by sequencing, which were T113G and G116C respectively. The relationships between the genotypes and cashmere fibre diameter, combed cashmere weight, cashmere fibre length and guard hair length were analyzed. There were significant differences between the associations of the different genotypes with cashmere weight (p<0.01), cashmere length (p<0.05) and hair length (p<0.01). Cashmere fibre diameter was the only trait that was not associated with the genotypes. The animals of genotype AB and BB had the higher cashmere weight compared with the genotype AA. By further analysis, it appeared that the KAP8.1 genotype effects on fibre traits may be due to a mutation at the 113 locus. These results suggested that polymorphisms in the hircine KAP8.1 gene might be a potential molecular marker for cashmere weight in Cashmere goats.
