A Novel Strategy for Thermostability Improvement of Trypsin Based on N-Glycosylation within the Ω-Loop Region

  • The Ω-loop is a nonregular and flexible structure that plays an important role in molecular recognition, protein folding, and thermostability. In the present study, molecular dynamics simulation was carried out to assess the molecular stability and flexibility profile of the porcine trypsin structures. Two Ω-Loops (fragment 57-67 and fragment 78-91) were confirmed to represent the flexible region. Subsequently, glycosylation site-directed mutations (A73S, N84S, and R104S) were introduced within the Ω-loop region and its wing chain based on its potential N-glycosylation sites (Asn-Xaa-Ser/Thr consensus sequences) and structure information to improve the thermostability of trypsin. The result demonstrated that the half-life of the N84S mutant at 50℃ increased by 177.89 min when compared with that of the wild-type enzyme. Furthermore, the significant increase in the thermal stability of the N84S mutant has also been proven by an increase in the Tm values determined by circular dichroism. Additionally, the optimum temperatures of the wild-type enzyme and the N84S mutant were 75℃ and 80℃, respectively. In conclusion, we obtained the thermostability-improved enzyme N84S mutant, and the strategy used to design this mutant based on its structural information and N-linked glycosylation modification could be applied to engineer other enzymes to meet the needs of the biotechnological industry.

  • In this study, To subject the constructed at N-kindergarten in G-city, the position is closed window and opened window was measured using a measuring instrument for radon. The measured results indicate that the measurement was carried out in concentrations of radon gas measured at N-kindergarten is low than United States in the radon concentration in air public 4pCi called radon gas baseline maximum allowable concentrations. As a result, radon exposure is not a problem, but when the accumulation radon gas in the lungs, get damaged same lung cancer. Be defensive of kindergarten windows open for ventilation and dust removal be possible to reduce the exposure.

Reverse Baby-step 2k-ary Adult-step Method for 𝜙((n) Decryption of Asymmetric-key RSA (비대칭키 RSA의 𝜙(n) 해독을 위한 역 아기걸음- 2k-ary 성인걸음법)

  • When the public key e and the composite number n=pq are disclosed but not the private key d in an asymmetric-key RSA, message decryption is carried out by obtaining ${\phi}(n)=(p-1)(q-1)=n+1-(p+q)$ and subsequently computing $d=e^{-1}(mod{\phi}(n))$. The most commonly used decryption algorithm is integer factorization of n/p=q or $a^2{\equiv}b^2$(mod n), a=(p+q)/2, b=(q-p)/2. But many of the RSA numbers remain unfactorable. This paper therefore applies baby-step giant-step discrete logarithm and $2^k$-ary modular exponentiation to directly obtain ${\phi}(n)$. The proposed algorithm performs a reverse baby-step and $2^k$-ary adult-step. As a results, it reduces the execution time of basic adult-step to $1/2^k$ times and the memory $m={\lceil}\sqrt{n}{\rceil}$ to l, $a^l$ > n, hence obtaining ${\phi}(n)$ by executing within l times.

Development of CNT-dispersed Si3N4 Ceramics by Adding Lower Temperature Sintering Aids

  • The study to give electrical conductivity by dispersing carbon nanotubes (CNT) into silicon nitride ($Si_3N_4$) ceramics has been carried out in recent years. However, the density and the strength of $Si_3N_4$ ceramics were degraded and CNTs disappeared after firing at high temperatures because CNTs prevent $Si_3N_4$ from densification and there is a possibility that CNTs react with $Si_3N_4$ or $SiO_2$. In order to suppress the reaction and the disappearance of CNTs, lower temperature densification is needed. In this study, $HfO_2$ and $TiO_2$ was added to $Si_3N_4-Y_2O_3-Al_2O_3$-AlN system to fabricate CNT-dispersed $Si_3N_4$ ceramics at lower temperatures. $HfO_2$ promotes the densification of $Si_3N_4$ and prevents CNT from disappearance. As a result, the sample by adding $HfO_2$ and $TiO_2$ fired at lower temperatures showed higher electrical conductivity and higher bending strength. It was also shown that the mechanical and electrical properties depended on the quantity of the added CNTs.

Soil Properties of Quercus variabilis Forest on Youngha Valley in Mt. Worak National Park

  • Soil properties of Quercus variabilis forest on Youngha valley at Mt. Worak National Park were studied as a part of Korea National Long-Term Ecological Research. Soil sampling was carried out along the 50 cm soil depth with 10cm intervals at every quarter from May 2005 through July 2006. Fresh soil was used for $NH_4{^+}-N,\;NO_3{^-}-N$, and soil water content determination. Remaining soils were air dried in the shade, and then used for determination of soil pH, T-N, T-P and exchangeable cation. Average soil organic matter in top soil was $8.5{\pm}1.2%$ and decreased with soil depth. Bulk density of top soil was $0.82{\pm}0.07g/cm^3 $and increased with soil depth. Soil organic matter and bulk density showed a negative linear correlation ($R^2=0.8464$). Soil pH in top soil and subsoil was similar. T-N, $NH_4{^+}-N,\;NO_3{^-}-N$ and T-P in top soil were $1.9{\pm}0.5mg/g,\;7.3{\pm}1.0mg/kg,\;2.0{\pm}0.4mg/kg\;and\;0.2{\pm}0.05mg/g$, respectively. $K^+,\;Ca^{2+}\;and\;Mg^{2+}$ in top soil were $84.6{\pm}24.4,\;408.8{\pm}137.8\;and\;93.4{\pm}23.0mg/kg$, respectively. They decreased with soil depth. Amounts of organic matter, T-N, $NH_4{^+}-N,\;NO_3{^-}-N$, T-P, $K^+,\;Ca^{2+}\;and\;Mg^{2+}$ in 50 cm soil depth were 250.9, 3.45, 0.025, 0.003, 0.639, 0.181, 0.845 and 0.302 ton $ha^{-1}\;50cm-depth^{-1}$, respectively.

  • Superconducting B $i_2$S $r_2$C $a_{n-l}$C $u_{n}$ $O_{x}$(n=2~4) thin films were prepared by single target DC-magnetron sputtering. And, that was compared with the B $i_2$S $r_2$C $a_{n-l}$C $u_{n}$ $O_{x}$(n=1~3) thin film fabricated by using the ion beam sputtering. Phase intergrowth among n=2-3, 3-4 and 4-5 phases was observed. The molar fraction of each phase in the mixed crystal of the deposited films was determined by x-ray diffraction analyses and investigated as a function of $O_2$ gas pressure during sputtering. We investigated the changes of the superconducting properties by molar fraction of each phase. Also, the thin film surface observation was carried out by atomic force microscope. The images show the average particle size decreases, and the distribution density of particles on the film surface was to increase with lower gas pressures. The fabrication conditions for selective growth of the single n=2, 3 and 4 phases in BiSrC $a_{n-l}$C $u_{n}$ $O_{x}$(n=2~4) thin film are discussed.e discussed.ussed.

  • This paper proposes a method to find out the type and location information of Pauli X, Y, Z errors generated in quantum channels using only the quantum information processing part of the multiple-rate quantum turbo short-block code without external help from the classical information processing part. In order to obtain the location information of the Pauli X,Y error, n-auxiliary qubits and n-CNOT gates were inserted into the C[n,k,2] QSBC-QURC encoder. As a result, the maximum coding rate is limited to about 1/2 as the trade-off characteristics. The location information of the Pauli Z error for C[n,k,2] QSBC-QURC was obtained through the Clifford-based stabilizer measurement. The proposed method inherits all other characteristics of C[n,k,2] QSBC-QURC except for the coding rate.

Morphological study on non-seeded grown AlN single crystals (무종자결정 상에 성장된 AlN 결정의 형태학적 연구)

  • The growth of AlN single crystals of large size and good quality is of prime importance for UV LEDs and power devices applications. However, the crystals having the size of more than 1 inch and high quality have not been reported in the world. In the PVT growth of AlN, the crystal morphology of as grown were important because the preferred orientation of growth of it was evaluated for growth rate increase. In the present study, the AlN single crystals grown by PVT process were evaluated by the side of the growth morphology. Optical microscopic characterization was carried out to observe the shape of the crystals and the growth facets. Furthermore the growth habit of it were discussed by observation of the surface of AlN crystals.

Study on the Characteristics of Noise/Vibration in the Upright Laying Hen House (직립식 산란계사 내의 소음 진동 발생 현황 조사연구)

  • This study was carried out to measure and analyze the characteristics of noise and vibration, and to analyze their effects on the productivity of layers, mechanical troubles, and abnormal wear-out failure of facilities and equipment of the layer house. The measurements of noise and vibration were taken at 13 layer farms nationwide for the operations of feed supplier system, feed distribution system, automatic egg collection system, ventilation system, blot conveyer for layer feces, and fur the case of with and without their operation by a sound level meter and a vibration measuring system in the layer house equipped with upright multi-tier cages. Measurement results showed that normal times were noise(N) 82 dB and vibration(V) 0.2072 cm/s, feed supplier system were 90 dB(N) and 2.8560 cm/s(V), feed distribution system were 90 dB(N) and 2.0222 cm/s(V), automatic egg collection system were 87 dB(N) and 0.1865 cm/s(V), ventilation system 88 dB(N) and 2.5364 cm/s(V), belt conveyer fur layer feces were 88 dB(N) and 0.2387 cm/s(V), and then maximum values of noise and vibration were 90 dB and 2.8560 cm/s, respectively, when feeding systems(feed supplying system and feed distribution system) were operated. Based on these results, an experiment is being conducted to find out the effect of noise and vibration on the productivity of layers in the layer house equipped with upright multi-tier cages.

Statistical Method of Ranking Candidate Genes for the Biomarker

  • Receive operating characteristic (ROC) approach can be employed to rank candidate genes from a microarray experiment, in particular, for the biomarker development with the purpose of population screening of a cancer. In the cancer microarray experiment based on n patients the researcher often wants to compare the tumor tissue with the normal tissue within the same individual using a common reference RNA. Ideally, this experiment produces n pairs of microarray data. However, it is often the case that there are missing values either in the normal or tumor tissue data. Practically, we have $n_1$ pairs of complete observations, $n_2$ "normal only" and $n_3$ "tumor only" data for the microarray. We refer to this data set as a mixed data set. We develop a ROC approach on the mixed data set to rank candidate genes for the biomarker development for the colorectal cancer screening. It turns out that the correlation between two ranks in terms of ROC and t statistics based on the top 50 genes of ROC rank is less than 0.6. This result indicates that employing a right approach of ranking candidate genes for the biomarker development is important for the allocation of resources.
