마디풀(Polygonum aviculare L.) 성분의 지질과산화억제 및 간보호에 미치는 효과 (Anti-lipid Peroxidation and Liver Protective Effects of Polygonum aviculare L.)

  • The ethylacetate fraction of the overground portion of Polygonum aviculare L. exhibited the anti-lipid peroxidation and the liver protective effect in intoxicated rats. Through silica gel chromatography of the ethylacetate fraction monitered by bioassay, two flavonoids, avicularin and juglanin were isolated as active components. Avicularin and juglanin remarkablely inhibited the lipid peroxidation of rat liver induced by 50% ethanol. Especially avicularin exhibited the stronger anti-lipid peroxidation effect than juglanin. Avicularin as a main principle of Polygonum aviculare L. significantly exhibited liver protective activities by decreasing s-GOT and s-LDH levels which represent for the hepatotoxicity induced by $CCl_4$ in rats. In addition, avicularin significantly decreased not only s-LDH but also s-bilirubin levels in intoxicated rat induced by ${\alpha}-naphthylisothiocyanate\;(ANIT)$. These results suggest that avicularin has the protective effects against the hepatoxicity induced by $CCl_4$ and ANIT in rats.

애기땅빈대의 화학적 성분 (Chemical constituents from the whole plants of Euphorbia supina Rafin)

  • Eight compounds were isolated from the whole plants of Euphorbia supina (Euphorbiaceae) through repeated silica gel, YMC gel and Sephadex LH-20 column chromatography. Their chemical structures were elucidated as 7-hydroxy-6-methoxycoumarin (scopoletin) (1), p-hydroxybenzaldehyde (2), methyl gallate (3), gallic acid (4), quercetin (5), quercetin $3-O-{\alpha}-L-arabinofuranoside$ (avicularin) (6), kaempferol $3-O-{\alpha}-L-arabinofuranoside$ (juglanin) (7) and kaempferol $3-O-{\beta}-D-glucopyranoside$ (astragaline) (8) by spectroscopic (NMR and MS) analysis.

  • 일본잎갈나무 낙엽(8.5 kg)을 채취하여 95% 에탄올 용액으로 추출하고 농축한 후 분획깔때기로 헥산, 메틸렌클로라이드, 에틸아세테이트 및 수용성으로 순차 추출하여 동결건조하였다. 에틸아세테이트용성과 수용성 분획에 대하여 칼럼크로마토그래피를 실시하였고, 충진물질로는 Sephadex LH-20을, 용리용매로는 메탄올 수용액 및 에탄올-헥산 혼합용액을 사용하였다. 단리된 화합물들은 TLC로 확인한 후 NMR 스펙트럼을 사용하여 구조규명을 하였고 FAB-MS와 EI-MS 스펙트럼으로 분자량을 측정하여 7개의 flavonoid 화합물인 (+)-catechin, (-)-epicatechin, 2"-O-rhamnosylvitexin, juglanin, afzelin, laricitrin-3-O-${\beta}$-D-glucopyranoside 및 isoquercitrin과 neolignan 화합물인 cedrusin을 단리하였다.

  • Phytochemical investigation of the MeOH extract of the aerial parts of Geranium eriostemon resulted in the isolation of one triterpene, three furofuran lignans, one syringic acid and four flavonoids. Their chemical structures were characterized by spectroscopic methods to be oleanolic acid (1), (-)-kobusin (2), (-)-eudesmin (3), (+)-magnolin (4), syringic acid (5), quercetin (6), juglanin (7), juglalin (8), and hyperin (9). All compounds (1 - 9) were isolated for the first time from this plant source and the compounds 2 - 4 were reported first from the genus Geranium. Compounds 4 - 6 exhibited moderate cytotoxicity against four human cancer cell lines in vitro using a SRB bioassay.

  • Eight compounds were isolated from the EtOAc soluble fraction of Euphorbia supina MeOH extract as the radical scavengers for antioxidant activity. Their structures were identified as kaempferol (1), quercetin (2), juglanin (3), avicularin (4), astragalin (5), isoquercitrin (6), hyperin (7), and nicotiflorin (8) by spectroscopic analysis. The antioxidant activity was evaluated by the ORAC (oxygen radical absorbance capacity) assay, which measures scavenging activity against peroxy radicals induced by 2,2'-azobis (2-methylpropionamidine) dihydrochloride, and the ORAC value is expressed as relative trolox equivalent. Compounds 4, 6, and 7 exhibited potent antioxidant activity, whereas the other compounds showed weaker activity than trolox.

Corilagin with Inhibitory Activity against NO Production from Euphorbia supina

  • Bioactivity-guided investigation on whole plant of Euphorbia supina, using LPS-induced Raw264.7 cells, led to the identificaiton of a tannin which was not reported from this plant along with four known constituents (quercetin, astragalin, juglanin and methyl gallate). The structure of the tannin was determined as corilagin by the interpretation of NMR (1D and 2D) and MS spectroscopic data. All the isolates were tested for the inhibitory activity against NO production in LPS-induced Raw264.7 cells. Among the tested isolates, corilagin was found to be the most active compound.

한국산 4종 Rubus속 숙과, 미숙과 및 잎의 플라보노이드 함량 분석 (Quantitative Analysis of Flavonoids in the Unripe and Ripe Fruits and the Leaves of Four Korean Rubus species)

  • The five flavonoids, isoqueritrin, juglanin, astragalin and 2-O-trans-p-coumaroyl astragalin were quantitatively analyzed in the leaves, unripe- and ripe fruits of four Korean Rubus species including Rubus crataegifolius, R. pungens var. oldhami, R. parvifolius, and R. coreanus belonging to the Rosaceae family. The HPLC analysis demonstrated that flavonoids are more abundant in the leaves than in the fruits. In the four Rubus species, ripe fruits generally contained similar or more amount of flavonoids than in unripe fruits. In particular, the quantity levels of total flavonoid (49.81${\pm}$0.50 mg/g) and isoquercitrin (35.08${\pm}$0.37 mg/g) were highest in the leaf of R. crataegifolius. In addition, the content of the flavonoids were quite low in the unripe fruits of R. coreanus that is most widely used as Rubi Fructus.

Identification of Antioxidative Constituents from Polygonum aviculare using LC-MS Coupled with DPPH Assay

  • A method for simultaneously identifying antioxidative compounds was developed using time-based LC-MS coupled with DPPH assay regardless of the time consuming process. The methanolic extract of Polygonum aviculare (Polygonaceae) showed significant DPPH radical scavenging activity. Time-based DPPH assay for simultaneous identification of active compounds from the extracts of P. aviculare was used. Major peaks of ethyl acetate fraction of P. aviculare showed high DPPH radical scavenging activity. A simple phenolic compound (1) and six flavonoids (2-7) were isolated from the ethyl acetate fraction of P. aviculare by silica gel and sephadex LH-20 column chromatography. The structures of seven compounds were determined to be protocatechuic acid (1), catechin (2), myricitrin (3), epicatechin-3-O-gallate (4), avicularin (5), quercitrin (6), and juglanin (7) based on the analysis of the $^1H$-NMR, $^{13}C$-NMR and ESI-MS data. All compounds exhibited significant antioxidant activity on DPPH assay and active compounds were well correlated with predicted one.

잣나무 잎 추출물의 항균 및 항산화 활성과 성분분석 (Antibacterial, Antioxidative Activity and Component Analysis of Pinus koraiensis Leaf Extracts)

  • 본 연구에서는 잣나무 잎 추출물의 항균, 항산화 및 성분 분석에 관한 연구를 수행하였다. 피부 상재균에 대한 항균활성 측정결과, Propionibacterium acnes (P. acnes), Staphylococcus aureus (S. aureus), Pityrosporum ovale (P.ovale) 및 Escherichia coli (E. coli)에 대한 ethyl acetate 분획의 MIC는 각각 0.06 %, 0.25 %, 0.13 %, 0.50 %로 나타났으며, P. acnes, P. ovale 및 S. aureus에서 큰 항균활성을 나타내었다. 잣나무 잎 추출물의 free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) 소거활성($FSC_{50}$)은 aglycone 분획에서 $22.93\;{\mu}g/mL$으로 나타났다. Luminol-의존성 화학발광법을 이용한 $Fe^{3+}$-EDTA/$H_2O_2$계에서 생성된 활성산소종(reactive oxygen species, ROS)에 대한 잣나무 잎 추출물의 총 항산화능($OSC_{50}$)은 50 % ethanol extract 분획에서 $0.70\;{\mu}g/mL$, ethyl acetate 분획 및 aglycone 분획이 각각 1.04 및 $1.43\;{\mu}g/mL$으로 매우 큰 항산화능을 보였다. Rose-bengal로 증감된 사람 적혈구의 광용혈 실험에서 추출물의 세포보호 효과를 측정하였다. 잣나무 잎 추출물 분획들은 농도 의존적($5{\sim}50\;{\mu}g/mL$)으로 세포보호 효과를 나타내었다. 잣나무 잎 추출물의 가수분해로 얻은 ethyl acetate 분획은 TLC (PK-4, PK-6)와 HPLC (peak 1, peak 2)에서 2개의 주성분으로 나타났다. 이 성분들은 LC/ESI-MS/MS를 통해서 PK-6는 kaempferol-3-O-glucoside (astragalin)로, PK-4는 kaempferol-3-O-arabinoside (juglanin)로 확인되었다. 이상의 결과들은 잣나무 잎 추출물이 ROS를 소광시키거나 소거함으로써, 그리고 ROS에 대항하여 세포막을 보호함으로써 생체계, 특히 태양 자외선에 노출된 피부에서 항산화제로서 작용할 수 있음을 가리키며, 잣나무 잎 성분 분석, 그리고 피부 상재균에 대한 항균작용으로부터 항산화, 항노화 및 항균성 화장품 소재로서의 응용 가능성이 있음을 확인하였다.
