  • The pressure-volume curve parameters were investigated to elucidate the effects of shading treatment on the water relations of the one year old seedlings of Betula platyphylla var. japonica, Betula schmidtii, Zelkova serrata, Acer mono and Prunes sargentii subjected to five levels of artificial shading treatments. The osmotic potentials at full turgor(${\phi}_{{\pi}o}$) measured under full sunlight changed with species and growing season in the ranges of -1.04~-1.27MPa, -1.03~-1.48MPa, -0.94~-1.44MPa in first year treatment, and -0.90~-1.37MPa, -1.05~-1.79MPa, -0.99~-1.30MPa in second year treatment in June, July, and September, respectively. The osmotic potentials at full turgor increased with increment of shading level in the ranges of -0.90~-1.79MPa in full sunlight and -0.58~-1.23MPa in nearly full shading level(E) through the growing seasons in all the species studied. The osmotic potentials at turgor loss point(${\phi}_{{\pi}p}$) measured in full sunlight changed in the ranges of -1.64~-2.11MPa, -1.67~-2.15MPa, -1.47~-2.11MPa, and -1.45~-2.04MPa, -1.30~-2.00MPa, -1.28~-2.33MPa in June, July, and September of first and second years, respectively. Most of ${\phi}_{{\pi}p}$ measurements were lower within about 0.5MPa in comparison with those of ${\phi}_{{\pi}o}$. The measurements of ${\phi}_{{\pi}p}$ also increased with increment of shading level, and the differences in ${\phi}_{{\pi}p}$ among shading levels were generally greater than those in ${\phi}_{{\pi}o}$ by species and by growing season. Most of the osmotic potentials at turgor loss point as like as at full turgor were lowered in July than in June and September. The measurements of relative water content at turgor lass point(RWCp) in full sunlight were in the similar ranges of 81~88%, 71~86%, 75~84%, and 82~87, 72~84%, 76~86% in June, July, and September of first and second years, respectively. The RWCp were a little higher in A. mono and P. sargentii than in B. platyphylla var. japonica, B. schmidtii, and Z. serrata. The RWCp also decreased from 71~88% in full sunlight to 48~77% in nearly full shading treatment with increment of shading level. Even if there were some exceptions by species or by growing season, the shading effects on the changes in some P-V parameters were distinctly observed in the present study. The change in P-V parameters following shading treatment may be presumably inferred on the changes in solute accumulation, membrane elasticity, symplasmic water volume, and so on. But much more experiments should be necessarily continued for getting detailed informations on the physiological mechanism of shading effects relating to the changes in P-V parameters.

Eliminatory Effect of Mixed Extract of Hovenia Dulcis Thunb and Gastrodia Elata on Ethanol-Induced Hangover in Rats (랫드에서 헛개나무와 천마 혼합 추출물의 에탄올 섭취에 의한 숙취 제거 효과)

  • To investigate an eliminatory effect of mixed extract of Hovenia dulcis Thunb, Gastrodia elata and Alnus japonica etc., on the ethanol-induced hangover, 12 hr-fasted male Sprague-Dawley rats weighing 220±20 g were given mixed extract (10 mL/kg, p.o.) and administered ethanol at a dose of 3 g/kg bw (25% in distilled water) orally 30 min postdosing. Blood was collected from caudal artery at 0.25, 0.5, 1, 2, 4, 6, 8, 10, and 12 hr and then the animals were sacrificed at 24 hr after the ethanol treatment. From 0 to 12 hr, the administration of mixed extract significantly decreased the area under the serum alcohol concentrations-vs.-time curves by 21 % compared with control group. In these experiments, liver function indices, such as alanine aminotransferase, aspartate aminotransferase and sorbitol dehydrogenase activities, showed unaltered results in all treated groups compared with the normal group. These results suggest that oral intake of the mixed extract may be effective on elimination of ethanol-induced hangover.

Growth and Photosynthetic Responses of Cuttings of a Hybrid Larch (Larix gmelinii var. japonica x L. kaempferi) to Elevated Ozone and/or Carbon Dioxide

  • We studied the effects of elevated ozone ([$O_3$]) and $CO_2$ concentrations ([$CO_2$]) on the growth and photosynthesis of the hybrid larch $F_1(F_1)$ and on its parents (the Dahurian larch and Japanese larch). $F_1$ is a promising species for timber production in northeast Asia. Seedlings of the three species were grown in 16 open top chambers and were exposed to two levels of $O_3$ (<10 ppb and 60 ppb for 7 h per day) in combination with two levels of $CO_2$ (ambient and 600 ppm for daytime) over an entire growing season. Ozone reduced the growth as measured by height and diameter, and reduced the needle dry mass and net photosynthetic rate of $F_1$, but had almost no effect on the Dahurian larch or Japanese larch. There was a significant increase in whole-plant dry mass induced by elevated [$CO_2$] in $F_1$ but not in the other two species. Photosynthetic acclimation to elevated [$CO_2$] was observed in all species. The net photosynthetic rate measured at the growing [$CO_2$] (i.e. 380 ppm for ambient treatment and 600 ppm for elevated $CO_2$ treatment) was nevertheless greater in the seedlings of all species grown at elevated [$CO_2$]. The high [$CO_2$] partly compensated for the reduction of stem diameter growth of $F_1$ at high [$O_3$]; no similar trend was found in the other growth and photosynthetic parameters, or in the other species.

The Antioxidant Activity of Cnidii Fructus and Torilis Fructus in Leydig cells (Leydig Cell의 항산화에 미치는 벌사상자와 사상자의 비교연구)

  • Objectives : The purpose of this study was to estimate the antioxidant activity of water extract of Cnidii Fructus (CF) and Torilis Fructus (TF) in Leydig cells. Methods : Free radical scavenging activity of CF and TF against 2,2-diphenyl-1-picrylhydrazyl (DPPH) was determined spectrophotometrically. We investigated the effect of CF and TF in Leydig cells by MTT assay. The protective effects of CF and TF against hydrogen peroxide-induced oxidative stress in Leydig cells. Superoxide dismutase (SOD), and catalase activity assays were performed in Leydig cells. Results : The results showed that CF scavenged DPPH radical in a dose-dependent manner by up to 81.2%, TF scavenged DPPH radical in a dose-dependent manner by up to 63.8%. CF showed cell viability as 121.0, 132.7, 126.6% in 5, 10, $100{\mu}g/ml$ concentrations. TF showed cell viability as 127.5, 111.8% in 5, $100{\mu}g/ml$ concentraions, respectively. The hydrogen peroxide-induced cytotoxicity of Leydig cells were protected to 86.3% by CF at concentration of $10{\mu}g/ml$ and protected to 83.5% by TF at concentration of $100{\mu}g/ml$. Both CF and TF at all concentrations, SOD activity was not significantly changed. Catalase activity was significantly increased at 10, $100{\mu}g/ml$ concentrations of CF, respectively. TF's catalase activity showed no significant difference from that of the control. Conclusions : These results suggest that CF, as an antioxidant, protects Leydig cells in hydrogen peroxide-induced oxidative stress. know that "Kwangjebikeup" played a role in settlement and spreading of foreign knowledge to civilians.

Effects of Plant Growth Regulators on Growth of Rice Seedling for Machine Transplanting (식물생장조절제 처리가 벼기계이앙 어린모 생육에 미치는 영향)

  • The infant seedling of rice(8-to 10-day-old) has often suffered from submergence just after transplanting due to the too short seedling height, 5 to 8cm, This experiment was conducted to elucidate the effect of gibberellic acid($GA_3 : 25% soluble liquid) with the application methods of foliar spray and seed-soaking on the elongation of seedling height of infant rice seedling in machine transplanting, We used a japonica rice variety, Hwaseongbyeo, and the seeding rate was 200g per seed tray(30$\times60$\times3cm). Generally,$GA_3 and Metalaxyl increased the elongation of seedling height of infant seedling while Tetracycle decreased the seedling height, To increase the seedling height of infant seedling, the foliar spary application of $GA_3 was better than seed-soaking treatment. The optimum concentration of $GA_3 for the foliar spray was about l0ppm and the optimum application time of $GA_3 was 3 to 4 days after sowing.

  • Gelatinization properties of heat-moisture treated Olchal(japonica) and Hankangchalbyeo(j x indica) waxy rice starches were compared. The blue value, light transmittance of starch suspension and viscosity in sodium hydroxide solution were similar between two starches. Olchal starch showed lower water binding capacity, swelling power md peak viscosity by amylograph than Hankangchalbyeo starch. Upon heat-moisture treatments all above parameters were decreased. The critical sodium hydroxide concentration for gelatinization was increased by treatments. The initial pasting temperature of Olchal starch was higher than that of Hankarlgchalbyeo. All amylograph reference points increased by the treatments, except the maximum viscosity or 2l% moisture-treated Olchal starch. Starches treated at 18% moisture level showd the highest value of consistency index. The activation energy of consistency index for gelatinized Olchal starch was lower than that for Hankangchalbyeo starch. The heat-moisture treatments had no effect on activation energy.

Effects of Cool-Season Turfgrass Overseeding onto Zoysiagrass (들잔디 위의 한지형 잔디 덧파종 효과)

  • This study was carried out to find out the effect of cool-season turfgrass overseeding onto zoysiagrass(Zoysia japonica L.) for extending green color and solving traffic problem during dormant periods. Overseeding rates of cool-season turfgrasses were $10g/m^2$ of Kentucky bluegrass(KB1), $20g/m^2$of Kentucky b1uegrass(KB2), $60g/m^2$ of perennial ryegrass(PR), $80g/m^2$ of Tall fescue(TF), and $10g/m^2$ of Kentucky bluegrass + $10g/m^2$ of perennial ryegrass mixture(KB+PR) onto 2m${\times}$2m(width${\times}$length) zoysiagrass plots with 3 replications.1 to 9-sca1e of visual quality and visual color were measured after treatment, Consequently cool-season turfgrass overseeding gave a good performances of extending the green periods and enhancing the turf grass quality. KB1 and KB2 were found to be excellent performances for visual quality and visual color compared to PR, TF, KB+PR and control. KB2 showed better turf performances than KB1 within Kentucky bluegrass overseedings. Turfgrass color was kept up for 10 months by overseeding of cool-season turfgrasses(KB1, KB2, PR, TP, KB+PR) compared to about 5~6 months by zoysiagrass(Control). The visual quality and visual color of PR and TF showed good performances during a green-up period of spring, but decreasing tendencies through the summer.Percent diseases data revealed also higher ratings for PR and TF compared to for KB1 and KB2.

Habitat Characteristics and Vegetation Structure of the Evergreen Fern in Jejudo, Korea (제주도의 상록양치식물 자생지 환경특성 및 식생구조에 관한 연구)

  • The research was carried out to define the environmental characteristics and vegetation structure of the evergreen fern habitat in Jejudo. The growth conditions of evergreen fern in various habitats was surveyed, including topographical features, vegetation structure, air temperature, humidity, intensity of light, water content and organic matter content in the soil. In the direction of the native habitat, 70% of research area was located in the southeast-facing slopes, 30% was located in southern slopes. The gradient ranged from $0^{\circ}$ to $30^{\circ}$. Temperature ranged from $16^{\circ}C$ to $28^{\circ}C$, and $22.3^{\circ}C$ was the average. Humidity ranged from 20 to 68%, and 36% was the average. In the native habitat, the highest light intensities reached 60,000 to 80,0001ux, but in general ranged from 300 to 40001ux. Water content in the soil ranged from 32% to 59%, organic matter content ranged from 8 to 13%. Within a unit of 25$m^2$, there were tall-tree layer such as Quercus galuca and Castanopsis cuspidata with a covering of rate 40~80%, a sub-tall-tree layer such as Camellia japonica, Staphylea bumalda and Sambucus williamsii with the covering rate of 3~5%, a shrub layer with the covering rate of 5~20%, and a grass layer with the covering rate of 40~95%. This research provides the basic data about the native habitat environment of the evergreen fern plant. Continuous monitoring and accumulation of data is necessary for the use of evergreen fern as vegetation materials.

Analysis of Dry Matter Yield and Feed Value for Selecting of Whole Crop Rice (최적 총체사료벼 품종 선발을 위한 건물수량 및 사료가치 분석)

  • This study was carried out to obtain basic information on variety selection for the utilization of whole crop rices(WCR) at National Institute of Crop Science, RDA, in 2004. Fifteen varieties and elite line were evaluated on feed value such as dry matter yield(DMY), crude protein( CP), acid detergent fiber(ADF), neutral detergent fiber(NDF) and total digestible nutrients(TDN). The dry matter yields were ranged from 13.23 to 17.83 ton per ha, the highest yielding varieties were Sobibyeo(l6.98ton / ha) in Japonica type, SR22060 (17.83 ton / ha) in New plant type, Hangangchalbyeo(I7.66 ton / ha) in Tongil type. Suweon 468 showed the highest value in the RFV and TDN content among the varieties, and Suweon 468, Suweon 498, Suweon 490 and SR22058 were chosen to have the high feed values through cluster analysis. The dry weight(grain) was found to be positively related with percent of the ripened grain, 1,000 grain weight and CPo TDN content was found to be positively related with CP, but negatively related with NDF and ADF. RFV was found to be negatively related with plant height, NDF and ADF. The promising rice varieties for WCR were Suweon 468, Suweon 498, Suweon 490 and SR22058 on the basis of CP, TDN and DMY.

ANTIOXIDATIVE ACTIVITIES OF SOME DIETARY FIBERS DETERMINED BY AN NIR EMISSION SPECTROSCOPY

  • Constituents of several .representative seaweeds, such as wakame Undaria pinnatifida; hijikia Hizikia fusifome; and kombu Laminaria japonica, were found to have fairly large reaction rates determined by quenching experiments of emission spectra in the near-infrared region (1max: 1270 nm) from singlet oxygen (102). Emission spectra of singlet oxygen generated from an aqueous solution of Rose Bengal under irradiation with a green laser (330 nm) were measured by a near-infrared (NIR) emission spectrometer constructed in our laboratory. The quenching experiments were as follows: Intensities of emission spectra were measured in the absence (I0) and in the presence of the seaweed constituents (I): Ratios of I0/I were plotted against every concentration of the quenchers (Stern-Volmer plots) which gives a straight line. The slope of each line gives a kqt value which gives a quenching constant kq value (an antioxidative constant against singlet oxygen) when the t value (half-life time of singlet oxygen in the solvent used) was given. The determined reaction rates are between 103-105 (g/l)-ls-1; the larger ones are as large as that of ascorbic acid, 8.4 ${\times}$ 104 (g/1)-ls-1. Most of these seaweed constituents also showed antioxidative activity against auto-oxidation and superoxide as well as their immunological enhancing activity. These results suggest a possibility that dietary fibers which are supposed to prevent the large-intestine cancer by their physical properties may prevent the cancer, at least in parts, by their chemical, antioxidative activity.
