척추동물의 Isozyme에 관한 비교연구: III. 한국산 뱀목의 Lactate 및 Malate Dehydrogenase Isozyme

  • MDH and LDH isozymes of various tissues of serveral snakes which belong to Colubridae, Crotalidae and Viperidae were studied. MDH isozymes were appeared as two bands in all species. Four LDH isozymes were found inall species of Colubridae, Crotalidae and Viperidae. LDH isozymes of muscle and liver had fundamentally the same pattern. On the other hand, there were two LDH isozymes in the heart of all species and the pattern of heart LDH isozyme was reverse of the pattern in muscle. The pattern of LDH isozymes of stomach tissue can be thought as the same pattern with that of heart in all species, but in the stomach tissue of Crotalidae and Viperidae there were four LDH isozymes.

  • A cellulosse acetate electrophoretic survey of Korean Anura has revealed the presence of diverse lactate and malate dehydrogenase (LDH and MDH) isozymes. The pattern of LDH and MDH isozymes in the tissues of the central nervous system of the six species of Anura examined are species specific and differ from those of mammals and birds. Both Rana nigromaculata nigromaculata and Rana nigromaculata coreana have two molecular forms of LDH and MDH, respectively, with almost the same pattern. Whole brain homogenate of Rana temporaria shows also a maximum of only two LDH isozymes. Both Bufo bufo asiaticus and Bombina orientalis have five molecular forms of LDH with an entirely different spacing on the zymograms, whereas Rana rugosa has three. Two molecular forms of MDH are present in all animals examined and one band is shown in olfactory lobe and mixture of cerebellum and medulla oblongata of Rana nigromaculata nigromaculata.

  • This study was planned to identify the variety of Italian ryegrass using electrophoresis. Thirty seven varieties of Italian ryegrass were tested by starch gel electrophoresis. The specific electrophoretic zymograms of each variety were observed by Alcohol dehydrogenase(ADH) and Esterase. The results were surnrnerized as follows; 1. AU varieties displayed two band zones by ADH and Rf values were 0.63 and 0.6 (Table 2, Fig. 2). 2. There were five band type for ADH isozyme of 37 varieties classified with isozyme banding pattern. According to the isozyme band type 7, 2, 6, 18 and 4 varieties belong to group, I, II, III, IV, and V, respectively (Table 2). 3. The varieties displayed single band zone for Esterase isozyme and Rf value was 1.00 (Table 2, Fig. 4). 4. According to banding type, Esterase isozyme of 37 varieties classified into 3 groups, 22, 8 and 7 varieties belong to group, I , II, and III, respectively (Table 2).

  • Electrophoretic isozyme patterns from mycelia, primordia, cap and stem of Pleurotus spp. collected in Korea were compared. Primordia, cap and stem of fruitbody showed very similar isozyme patterns but mycelial isozyme patterns were different from those of fruitbody. Isozyme patterns of malate dehydrogenase, acid phosphatase in Pleurotus ostreatus collected from different regions in Korea were similar but those of esterase, peroxidase, leucine amino peptidase and superoxide dismutase were different. Interspecific comparison of esterase isozyme patterns among Pleurotus ostreatus P. cornucopiae and, P. florida was very different and may be valuable subsidiary tool to conventional taxonomic techniques for identifying species of Pleurotus. A dendrogram by similarities of isozyme band pattern was presented.

Esterase Isozyme Banding Pattern in Wild Legume Plants (야생 콩과식물의 Esterase Isozyme Banding Pattern에 관한 연구)

  • Starch gel electrophoresis was used to examine the banding pattern of Esterase isozyme in the leaf, root-nodule and seedling of four wild legume species, Trifolim repense, Glycine soja, Phaseolus nipponensis and Vigna uexillata. The number of band, enzyme activity and migrating rate of esterase isozyme varies depending on the species and tissues of legume plants. The isozyme banding pattern in the cotyledon and radicle of T. repense showed same pattern, however, the number of band were varible among the cotyledon of G. soja, P. nipponensis and V. vexzllata, respectively. Est-1 in the leaf of G. soja, V, vexillata. root-nodule of G. soja and seedling of V. vexillata expreesed the highest enzyme activity. The Est-1 showed the rapidest migrating rate among the isozymes.

  • The present researches were carried out to investigate the effects of gibberellic acid on the appearance of isozyme patterns of esterase, phosphatase, amylase and peroxidase, also to investigate if there were any differences of the isozyme patterns among root, shoot and seeds of rice plants. It was noticed that the isozyme patterns of the above tested enzymes were differ among the organs, root, shoot and seed. The GA treated plants showed difference of esterase patterns in root from Akibare and the difference in shoot and root from Yushin, phosphatase patterns in root from Akibare. However, the GA did not affect isozyme patterns of amylase or peroxidase. The seed should be the suitable organ to study isozyme patterns for genetics or variety characterization of rice plant.

  • This study was carried out to compare distribution and isozyme pattern of nonspecific esterase, acid phosphatase and alkaline phosphatase on developing sparganum and adult of Spinometra erinacei by using enzyme-histochemical method and electrophoresis The sparganum and adult were recovered from rats and cat that were infected by sparganum. The results obtained were as follows: Nonspecific esterase had a strong activity in the parenchymal musculature of sparganum and adult, but no detectable level in ihe tegument. A total of 7 and 8 nonspecific esterase bands were detectable in sparganum and adult, respectively. Of these bands, band 3 and 4 were major bands in sparganum and adult. Acid phosphatase had a strong activity in the tegument and the epidermal musculature of sparganum, but no detectable level in the parenchymal musculature. A total of 3 bands were detectable in sparganum and adult. Of these bands, band 3 was major band in sparganum and adult. Alkaline phosphatase had a strong activity in the tegument and the epidennal musculature of sparganum and of adult, but no detectable level in the parenchymal musculature. A total of 2 and 4 bands were detectable in sparganum and adult. Of these bands, band 2 was major band in sparganum and adult. Based on the present results isozyme band patterns showed qualitative and quantitative changes in each tissues of sparganum and of adult during the development.

  • A comparative electrophoretic study on protein and several important enzymes of calli induced from stem, intercostal area and minor vein area were conducted in Pinella ternata (Thunb.) Breit. Soluble protein band patterns of calli induced from stem, intercostal area and minor vein area were distinctly different from those of the corresponding plant parts. Esterase isozyme patterns of calli induced from stem, intercostal area and minor vein area were different from those of the corresponding plant parts. Glutamate oxalo-acetate transaminase isozyme patterns of calli grown for 4 weeks induced from 3 plant parts were similar to those of the corresponding plant parts. But a high molecular weight isozyme band appeared in the calli grown for 8 weeks. Alow molecular weight isozyme band disappeared on the peroxidase isozyme patterns of calli grown for 4 weeks appeared on those of the corresponding plant parts.

  • The lactate dehydrogenase isozymes in tissues of eight fresh water fishes were examined by cellulose acetate electrophoresis. The electrophoretic distribution of the isozymes showed clearly species-specific pattern. Various subbands were found more frequently in these fishes than in mammals. The isozyme pattern of muscle seems to tend to be the same with that of brain in these fishes. The fish lactic dehydrogenase suggested in many cases to be consisted of nonrandomly selected hybrids.

  • 연어류의 종간교배 실험과 LDH, MDH, IDH, $\alpha$GODH, ME 등 다섯가지 isozyme에 대한 genetic marker로서의 활용 가능성을 알아보고자 1차적으로 연어, 산천어 및 무지개송어를 이용하여 종간교배를 실시하였고 이들 세종의 isozyme pattern을 비교하였다. 교배실험은 종간교배, 및 allotriploid 구간 등 12구간으로 나누어 실시하였으며 연어 암컷과 산천어 수컷의 교배결과가 초기성장 단계적에서 가장 우수하게 나타났고이들의 allotriploid도 부활율이 28.1%로 가장 우수하였다. Genetic identification을 위한 isozyme loci 분석결과 연어와 산천어는 대부분의 loci에서 거의 찾아볼 수 없었고 무지개송어는 MDH-B와 IDH에서 다형현상을 확인할 수 있었다. 특히 MDH-B loci는 b 유전자의 출현 빈도에서 세 종간의 식별이 가능하였으며, IDH pattern을 산천어와 무지개송어의 비교에 유효한 것으로 나타났다. 이들 두 loci는 hybrid의 genotype 분석시 유용한 marker로 활용할 수 있는 가능성을 보였고, 앞으로의 어류 육종에 좋은 기초 자료로 이용될 것으로 사료된다.
