Objectives : The objective of present study was to investigate the vasorelaxant effects of 20 Korean native plants on isolated rat thoracic aorta precontracted with phenylephrine (PE). Methods : Dried 20 plant materials were extracted 3 times with water, ethanol, or methanol for 3h in the reflux apparatus at 70 ± 5℃. Male SD rats were anesthetized by ether inhalation, and their aorta rings were isolated and placed in 10 ㎖ Krebs Henseleit (KH) buffer. While using an isolated organ-chamber technique, the aorta rings were maintained by bubbling with a gas mixture of 95% O2-5% CO2 at 37℃. Changes in isometric tension of aorta rings were recorded via isometric transducers connected to a Powerlab Data Acquisition System. Results : Among the 20 native plants, Chrysanthemum indicum L. flower, Nelumbo nucifera Gaertn. rhizome, Schisandra chinensis (Turcz.) Baill. fruit, Anthriscus sylvestris (L.) Hoffm. root, Corydalis turtschaninovii Besser tuber, Corydalis decumbens (Thunb.) Pers. tuber, and Dolichos lablab L. seed showed significant vasorelaxant effect on the contraction of aorta rings induced by PE. In contrast, Mertensia maritima subsp. asiatica Takeda whole plant, Ajuga decumbens Thunb. whole plant, Trigonotis peduncularis (Trevis.) Benth. ex Baker & S.Moore whole plant, Dioscorea quinquelobate Thunb. rhizome, Allium microdictyon Prokh aerial part, Momordica charantia L. fruit, Carthamus tinctorius L. flower, and Clematis terniflora DC. root constricted more the aorta rings precontracted by PE Conclusion : These results suggest that the possibility as useful herbal resources for the development of functional foods or medicines for hypertension treatment.
Purpose: The vascular smooth muscle cells (VSMCs) in mature animals have implicated to play a major role in the progression of cardiovascular diseases such as atherosclerosis. This study aimed at optimizing the protocol in culturing primary VSMCs (pVSMCs) from rat thoracic aorta and investigating the effect of cellular zinc (Zn) deficiency on cell proliferation of the isolated pVSMCs. Methods: The thoracic aorta from 7-month-old Sprague Dawley rats was isolated, minced and digested by the enzymatic process of collagenase I and elastase, and then inoculated with the culture Dulbecco Modified Eagle Medium (DMEM) at 37℃ in an incubator. The primary cell culture morphology was observed using phase-contrast microscopy and cellular Zn was depleted using Chelex-100 resin (extracellular zinc depletion only) or 3 µM N,N,N',N'-tetrakis(2-pyridinylmethyl)-1,2-ethanediamine (TPEN) (extracellular and intracellular zinc depletion). Western blot analysis was used for the detection of SM22α and calponin as smooth muscle cell marker proteins and von Willebrand factor as endothelial cell marker protein to detect the culture purity. Cell proliferation by Zn depletion (1 day) was measured by MTT assay. Results: A primary culture protocol for pVSMCs from rat thoracic aorta was developed and optimized. Isolated cultures exhibited hill and valley morphology as the major characteristics of pVSMCs and expressed the smooth muscle cell protein markers, SM22α and calponin, while the endothelial marker von Willebrand factor was hardly detected. Zn deprivation for 1 day culture decreased rat primary vascular smooth muscle cell proliferation and this pattern was more prominent under severe Zn depletion (3 µM TPEN), while less prominent under mild Zn depletion (Chelexing). Conclusion: Our results suggest that cellular Zn deprivation decreased pVSMC proliferation and this may be involved in phenotypic modulation of pVSMC in the aorta.

Objective : To examine the inhibition of 5-Hydroxytryptamine(5-HT) induced vasocontraction of the water extract of Acanthopanax senticosus Harms roots (ASR) on rat thoracic Aorta and mesenteric artery with and without endothelium. Methods : Segments of thoracic aorta and mesenteric artery obtained from rats immediately after delivery were mounted in organ baths superfused on a polygraph. Results : We found that the thoracic aorta segments responded to the water extract of ASR with a dose-dependent and concentration-dependent vasorelaxation. 5-HT produced a concentration-dependent contraction of the thoracic aorta and mesenteric artery. At high concentrations of ASR, the inhibition responses were 93.7% (Jang-su), 93.5% (Heok-ryong-kang-sung), 92.8% (Mt. Back-doo), and 83.5% (Yeon-byun) of the maximum 5-HT induced contraction. At high concentrations of ASR, the relaxational response at thoracic aorta and mesenteric artery wi1h endothelium were 95.2% and 94.6%; without endothelium were 93.5% and 92.5% of 1he maximum 5-HT induced contraction. Conclusions : In conclusion, the effect of water extract of ASR had potent inhibition at 5-HT and the effect of ASR in isolated thoracic aorta and mesenteric artery showed dose-dependent inhibition but endothelium-independent response.

Methods : Segments of thoracic aorta and mesenteric artery obtained from rats immediately after delivery were mounted in organ baths superfused on a polygraph. Results : We found that the thoracic aorta segments responded to the water extract of genus Panax with a dose-dependent vasorelaxation. At $10^{-5}m$ 5-hydroxytrptamine (5-HT), the maximal contraction force were 94.9% of the maximum KCl-response. At $10^{-5}m$ 5-HT - induced contraction, The contractile response of thoracic aortic rings were inhibited by 54.7%, 36.3% and 31.3% after addition of the high concentration (100 mg/ml) of water extract of Panax ginseng, Panax japonicus and Panax quinquefolium. The contractile response of mesenteric arteries were inhibited by 88.3%, 87.7%, and 70.3% after addition of the high concentration (100 mg/ml) of water extract of Panax ginseng, Panax japonicus and Panax quinquefolium. Conclusions : In conclusion, water extract of genus Panax - induced relaxation in the isolated rat thoracic aorta and mesenteric artery were composed of endothelium - independent relaxation and dose - dependent relaxation.

The Influence of age on the endothelial modulation of angiotensin II (AII)-induced contractile response was investigated in isolated aortic rings of rats ranging in age from 0.7 to 20 months. Hemoglobin and L-NAME were used to examine whether age-related changes in the EDRF-releasing system were involved in endothelial modulation of All-induced contraction in rat aorta. In all five age groups (0.7, 1.5, 3, 6, 20 months), hemoglobin (10 ${\mu}$M) significantly enhanced All-induced contractile response only in aorta with endothelium intact. L-NAME (10 ${\mu}$M) Produced a significant enhancement in All responses in endothelium-intact aortas from rats aged 0.7 and 1.5 months, but it had no effect in aortas from older rats aged 6 and 20 months. Indomethacin (10 ${\mu}$M) did not affect All-induced contractile responses in both endothelium intact and removed aortas from rats at the age of 0.7 to 20 months. Hemoglobin (10 ${\mu}$M) abolished acetylcholine-induced relaxation response in aortas from young and old rats. L-NAME completely abolished the relaxation in aortas from young (0.7 and 1.5 months), but incompletely in aortas from older (6 and 20 months) rats. The sensitivity of endothelium-dependent relaxation to A23187 increased with age between ages of 0.7 and 6 months, with no further increase noted up to 20 months of age. These results suggest that endothelial modulation of AII-induced contraction in rat aorta might involve age-related alteration in EDRF-releasing system, probably via post-receptor mechanism.

We have examined the relaxative response to the water extract of Rheum undulatum L. (ERU) and water extract of alcohol steamed Rheum undulatum L. (SRU) with isolated thoracic aorta from sprague dawley (SD) rat. Rat thoracic aorta was investigated in vessel segments suspended for isometric tension recording by polygraph. Responses to ERU and SRU were investigated in vessels precontracted with 5-hydroxytryptamine(5-HT). We found that the thoracic aorta segments responded to ERU and SRU with a dose-dependent vasorelaxation : The thoracic aorta segments responded to ERU and SRU with a dose-dependent vasodilation. The amounts of emodin were 0.063%, 0.076% and 0.145% in ERU and SRU, respectable. The 5-HT induced contraction at 10-4M were inhibited by 85.2±4.76% and 84.0±2.91% after addition of the 0.1mg/mL water extract of ERU and SRU. The 5-HT induced contraction at 10/sup -3/M were inhibited by 100% after 10/sup -3/M emodin. In conclusion, vasodilation effect of the water extract of Rheum undulatum L. in rat thoracic aorta was not decreased according to the processing of alcohol steamed Rheum undulatum L.
The present study was conducted to evaluate the effect of KST221085, a newly synthesized antidiabetic agent, on the hearts from streptozotocin (STZ)-induced diabetic rats. In isolated diabetic hearts, left ventricular developed pressure (LVDP), heart rate (HR) and coronary flow rate (CFR) were decreased compared to normal control, indicating cardiovascular dysfunction in diabetic heart. The treatment with 10 $\mu$M KST221085 remarkably improved the diabetes-induced contractile impairment, without any influence on HR. Reduced coronary flow in diabetic heart was also significantly increased by treatment with 10 $\mu$M KST221085. In isolated aorta from diabetic rat, treatment with 10 $\mu$M KST221085 increased endothelium-dependent relaxation, suggesting that KST221085 can improve the impaired endothelial function in diabetic aorta. Our results suggest that KST221085 treatment can improve the cardiovascular dysfunction in STZ-induced diabetic rats.
$\alpha$-Adrenoceptor subtypes in the isolated rat aortic strips were examined by using agonists and antagonists which have varying selectivity for $\alpha_1$- and $\alpha_2$- adrenoceptors. Norepinephrine and phenylephrine produced a similar magnitude of maximum contractions. $pA_2$ values for prazosin and yohimbine were not significantly different using norepinephrine or phenylephrine as the agonist, suggesting a single population of alpha-adrenoceptor. Contractile responses produced by alpha-agonists were antagonized more effectively by prazosin (approximately 1000 fold) than by yohimbine.

The purpose of this study was to investigate the possibility of medicinal plants application as an edible functional food resources. We carried out to develop a traditional functional beverage by using hot-water extraction of 4 medicinal plants(Polygonatun sibiricum, Ophiopogonis radix, Lycii fructus, Schizandriae fructus) and we examined the effects of drink on physiological function in aorta relaxation. Thus, the effect of developed beverage on phenylephrine induced contraction of isolated rat thoracic aorta. Contractile force was measured with force displacement transducer under 1.5g loading tension. Brix, pH and titratable acidity of developed drink were 9.5%, 3.3 and 0.22%. The approximate nutritional composition of beverage was carbohydrate, 5.98%, crude protein, 0.70%, crude fat, 0.20% and crude ash, 0.20%. Developed beverage contained K(4.00 ㎎%), Na(3.68 ㎎%), Ca(2.54 ㎎%), Mg(1.60 ㎎%) and Fe(0.29㎎%). The contraction forces by injection of phenylephrine in isolated thoracic aorta were significantly low in each experimental groups compared with control groups, These results that developed drink with medicinal plants can be used as a functional material to decrease aorta contraction.

Developed beverage contained Na(11.45 mg%), K(6.87 mg%), Ca(2.53 mg%). The contraction forces by injection of phenylephrine in isolated thoracic aorta were significantly law in each beverage treatment groups compared with control group. These results that developed drink with medicinal plants can be used as a functional material to decrease aorta contraction.
