Although corticosteroid have been shown to stabilize lysosomal membranes and prevent release of hydrolytic enzymes, the mechanism of membrane stabilization remains obscure. This study described functional assessment of efficiency of methylprednisolone in GIK solution by using a isolated Rat Heart Model. Isolated rat heart were subjected to a 2-minute period of coronary infusion with a cold GIK or methylprednisolone mixed cold GIK solution immediately before and also at the midpoint of a 60-minute period of hypothermic [$10{\pm}1^{\circ}C$] ischemic arrest. The result of this were as follow: 1.Spontaneous heart beat after ischemic arrest occurred 11 second later after Langendorffs reperfusion in the methylprednisolone mixed GIK group and 14 second later in the control group. 2.The percentage of recoveries of heart rate at 30 minute after postischemic working heart perfusion was 88.6\ulcorner.6% in the methylprednisolone mixed GIK group. This percentage of recovery was not significantly greater than the control group. 3.The percentage of heart function at 30 minute after postischemic working heart perfusion were; peak aortic pressure $90.8{\pm}4.5%$ coronary flow $87.5{\pm}1.45$ and aortic flow $74.9{\pm}11.8%$ in the methylprednisolone mixed GIK group. This percentage of recovery was significantly greater than the control group. [p<0.05]
Objectives: Myocardial reperfusion is the only logical cure for ischemic heart disease. However, ischemic-reperfusion (I/R) injury is one of the underlying factors facilitating and accelerating the apoptosis in the myocardium. This study set to investigate the impact of Teucrium polium (TP) hydro-alcoholic extract on I/R induced apoptosis in the isolated rat heart. Methods: Isolated rat hearts were classified into six groups. The control samples were subjected to 80 min of perfusion with Krebs-Henseleit bicarbonate (KHB) buffer; in control-ischemia group, after primary perfusion (20 min) the hearts were exposed to global ischemia (20 min) and reperfusion (40 min). Pretreated groups were perfused with $500{\mu}M$ of vitamin C and various TP concentrations (0.5, 1, 2 mg/ml) for 20 min, and then the hearts were exposed to ischemia and reperfusion for 20 min and 40 min, respectively. Cardiodynamic parameters including rate pressure product (RPP), heart rate (HR), the maximum up/down rate of left ventricular pressure (${\pm}dp/dt$), left ventricular developed pressure (LVDP), and coronary artery flow (CF) were achieved from Lab Chart software data. The Bax and BCl-2 gene expressions were measured in heart samples. Results: Hearts treated with TP extract and vit C represented a meaningful improvement in cardiac contractile function and CF. The overexpression of Bcl-2, downregulation of Bax, and improvement of apoptotic index (Bax/Bcl-2) were observed in pretreated TP extract and vit C hearts. Conclusion: The TP extract was found to ameliorate the cardiac function in the reperfused myocardium. Also, it can hinder apoptotic pathways causing cardioprotection.
Lampranthus spectabilis is an ornamental plant which belongs to one of the Aizoaceae family as an one-year herb. There are no illustrations on the references that Lampranthus spectabilis has been made use for the purpose of the medicinal plant among the traditional herb doctors and the laymen. However, it has the specific characteristics to be analogous to the Cactaceae family in the botanical classification. Besides, mescaline, the main ingredient of Peyote cactus, demonstrates the sympathomimetic effect as well as the psychotomimetic effect. The authors called our attention to the resemblance to the botanical family between Aisoaceae and Cactaceae, and made a survey whether the alcohol extract of Lampranthus spectabilis might possess any pharmacological effect on the isolated heart of clam or not. The results of the experiment were as follows. 1. Administration of LX to the isolated heart of clam produced an inhibitory effect on the cardiac motility in proportion to its concentration $10^{-4}$, $5{\times}10^{-4}$ and $10^{-3}\;g/ml$ respectively. 2. Administration of LX $5{\times}10^{-4$ and $10^{-3}\;g/ml$ produced the blockade effect on the cardiac motility pretreated with serotonin $10^{-7}$ and $10^{-4}\;g/ml$, as well as with methysergide $10^{-6}$ and $10^{-5}\;g/ml$. 3. Administration of LX $5{\times}10^{-4}\;g/ml$ produced no effect on the cardiac motility pretreated with acetylcholine $10^{-7}\;g/ml$. 4. Administration of LX $5{\times}10^{-4}\;g/ml$ produced the blockade effect on the cardiac motility pretreated with barium chloride $10^{-4}\;g/ml$, as well as with prostaglandin $20\;{\mu}g$. Consequently, it is concluded that the alcohol extract of Lampranthus spectabilis has an effect of depression on the motility of the isolated heart of clam. Its mechanism of action might be due to a direct action on the heart.
In this study, the effects of tauroursodeoxycholic acid (TUDCA) on ischemia/ reperfusion injury were investigated on isolated heart perfusion models. Hezrts were perfused with oxygenated Krebs-henseleit solution (pH 7.4, $37^{\cire}C$) on a Langendorff apparatus. After equilibration, isolated hearts were treated with TUDCA 100 and 200 $\mu\textrm{M}$ or vehicle (0.02% DMSO) for 10 min before the onset of ischemia in single treatment group. In 7 day pretreatment group. TUDCA 50, 100 and 200 mg/kg body weight were given orally for 7 days before operation. After global ischemia (30 min), ischemic hearts were reperfused for 30 min. The physiological (i.e. heart rate, left ventricdular developed pressure, coronary flow, double product, time to contracture formation) and biochemical (lactate dehydrogenase; LDH) parameters were evaluated. In vehicle-treated group, time to contracture formation was 810 sec during ischemia, LVDP was 34.0 mmHg at the endpoint of reperfusion and LDH activity in total reperfusion effluent was 34.3 U/L. Single treatment with TUDCA did not change the postischemic recovery of cardiac function, LDH and time to contractur compared with ischemic control group. TUDCA pretreatment showed the tendency to decrease LDH release and to increase time to contracture and coronary flow. Our findings suggest that TUDCA does not ameliorate ischemia/reperfusion-reduced myocardial damage.

Hearts were perfused with oxygenated Krebs-Henseleit solution (pH 7.4, $37^{\circ}C$) on a Langendroff apparatus. After equilibration, isolated hearts were treated with UDCA 20 to 160 $\mu$M or vehicle (0.04% DMSO) for 10 min before the onset of ischemia. After global ischemia (30 min), ischemic hearts were reperfused and allowed to recover for 30 min. The physiological (i.e. heart rate, left ventricular developed pressure, coronary flow, double product and time to contracture formation) and biochemical (lactate dehydrogenase; LDH) parameters were evaluated. In vehicle-treated group, time to contracture formation was 21.4 min during ischemia, LVDP was 18.5 mmHg at the endpoint or reperfusion and LDH activity in total reperfusion effluent was 54.0 U/L. Cardioprotective effects of UDCA against ischemia/reperfusion consisted of a reduced TTC $(EC_{25}=97.3{\mu}M)$, reduced LDH release and enhanced recovery of cardiac contractile function during reperfusion. Especially, the treatments of UDCA 80 and $160 {\mu}M $ significantly increased LVDP and reduced LDH release. Our findings suggest that UDCA ameliorates ischemia/reperfusion-induced myocardial damage.

Tetrahydroisoquinoline (THI) compounds have various pharmacological actions in the cardiovascular system. Recently, we have synthesized 1-${\alpha}$-naphthylmethyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline, YS 49. In the present study, we evaluated the effect of YS-49 on positive inotropic and chronotropic action using isolated rat heart and on blood pressure and heart rate using anesthesized rabbit. Vasodilating action was also assessed in isolated rat thoracic aorta. YS 49, concentration-dependently relaxed rat aorta precontracted with phenylephrine (PE, 0.3 ${\mu}$M) and high potassium (high K$\^$+/, 65.4 mM). The 50% inhibitory concentration (IC$\sub$50/) of YS 49 in PE-induced and high K$\^$+/-induded contraction was 5.36 ${\mu}$M and 2.52 ${\mu}$M, respectively. In isolated rat atria, YS 49 increased both heart rate and force, and in anesthesized rabbit it decreased blood pressure but increased heart rate. In addition, to know the mechanism of action of the compound, propranolol, nonselective ${\beta}$-antagonist, and phentolamine, ${\alpha}$-blocker, were used. Furthermore, a comparison with the effect of higenamine, trimetoquinol on the vasodilating action in rat thoracic aorta was also made. The action of YS 49 was inhibited by the presence of propranolo, not pentolamine. These results indicate that cardiotonic and vasodilatory action of YS 49 is attributable, at least in part, for ${\beta}$-receptor stimulation.
Fluorides were supposed to exert a stimulatory action on the catecholamine release. In this study, the authors attempted to investigate the action of sodium fluoride on the catecholamine release from the isolated perfused cow adrenal gland and rat heart. And also the inhibitory effect of sodium fluoride on the monoamine oxidase activity in rat heart and liver mitochondria was investigated. The monoamine oxidase activity was measured by the conversion of benzylamine to benzaldehyde. The results obtained were follows; 1. Sodium fluoride stimulated the release of catecholamine from the isolated perfused cow adrenal gland and rat heart. 2. Sodium fluoride inhibited the rat heart and liver mitochondrial monoamine oxidase activity.

Symptomatic pulmonary arterial hypertension (PAH) in patients with isolated atrial septal defect (ASD) is rare during infancy. We report a case of isolated ASD with severe PAH in an infant who developed airway obstruction as cardiomegaly progressed. The patient presented with recurrent severe respiratory insufficiency and failure to thrive before the repair of the ASD. Echocardiography confirmed volume overload on the right side of heart and severe PAH (tricuspid regurgitation [TR] with a peak pressure gradient of 55 to 60 mmHg). The chest radiographs demonstrated severe collapse of both lung fields, and a computed tomography scan showed narrowing of the main bronchus because of an intrinsic cause, as well as a dilated pulmonary artery compressing the main bronchus on the left and the intermediate bronchus on the right. ASD patch closure was performed when the infant was 8 months old. After the repair of the ASD, echocardiography showed improvement of PAH (TR with a peak pressure gradient of 22 to 26 mmHg), and the patient has not developed recurrent respiratory infections while showing successful catch-up growth. In infants with symptomatic isolated ASD, especially in those with respiratory insufficiency associated with severe PAH, extrinsic airway compression should be considered. Correcting any congenital heart diseases in these patients may improve their symptoms.
In order to investigate experimentally the clinical effects of Taeksa-Tang that was prescribed to cure the severe dizziness, the water extract of Taeksa-Tang was experimented about the effects on the diuretic action, the serum lipid, the isolated ileum and heart, the blood vessel, the blood pressure and respiration. The results of this study were obtained as follows: The diuretic action was significantly shown in rabbits. The level of total cholesterol, triglyceride and phopholipid in serum of rabbits given high cholesterol diet were significantly decreased. The contraction of the isolated ileum of mice induced by acetylcholine chloride and barium chloride was remarkably inhibited. The vasodilating and hypotensive actions were noted in rabbits. Negative inotropic action on the isolated frog heart was recognized.
The effects of erythrosine on motility of frog heart, rabbit duodenum and uterus isolated, and on mice intestinal motility and voluntary activity were investigated. The effect of erythrosine $2.3{\times}10^{-5}M$ on isolated frog heart showed a slight decrease of the amplitude of motility, and the heart motility stopped in $3.5{\times}10^{-4}M$. With the administration of erythrosine $3.4{\times}10^{-4}M$, the isolated rabbit duodenum showed a remarkable contraction and this effect was inhibited by atropine $1.4{\times}10^{-7}M$. The administration of erythrosine $2.3{\times}10^{-3}M$, produced a contractile effect on the isolated rabbit uterus, and the motility of $6.9{\times}10^{-3}M$ started to increase in contractions at first and finally stopped, keeping in continuous contractions. The effects of erythrosine 0.5, 1.0, 10, and 20mg/kg on mice intestinal motility were not significantly different from this of the normal control. With 20 and 40mg/kg of erythrosine, the effects on voluntary activity showed the decrease of 21 and 58% respectively, and voluntary activity of the mice pretreated with erythrosine 20 and 40mg/kg, induced by C. N. B. 30mg/kg showed the decrease of 57 and 78% respectively in contrast with the normal control group.
