Isolation of Rare Actinomycetes on Various Types of Soil (토양 특성에 따른 다양한 희소방선균의 분리)

  • Actinomycetes occur in a wide range of environments and many more actinomycetes remain to be detected in the natural environment. In isolation stages, selection of the environment as a source of useful isolates is important. Two hundred and eighteen strains were isolated on Bennet's agar and 346 strains were on humic acid-vitamin agar from five each paddy field, field, forest, grass land, riverside soil samples. These isolates were identified to the genus level based on morphological and physiological characteristics. Among them, 386 strains were Streptomyces, 49 strain were Nocardia, 35 strains were Microbispora and Micromonospora each, 15 strains were Nocardiopsis, 13 strains were Actinomadura, 10 strains were Streptosporangium, and the others were isolated rarely. According to soil type, Nocardia, Micromonospora, Microbispora and Streptosporangium were dominant in paddy field, Microbispora Nocardia, Nocardioides and Micromonospora were dominant in field, Nocardia, Micromonospora, Microbispora and Actinomadura were dominant in grass land, Nocardia, Micromonospora and Microbispora were dominant in forest, Nocardia, Microbispora and Micromonospora were dominant in riverside. Generally, Nocardia, Micromonospora, Microbispora and Actinomadura were isolated in all kinds of soils, Streptosporangium were paddy field, Dactylosporangium were forest, Nocardiopsis were field, forest and riverside.

Correlation between Isolated Entomopathogenic Fungi and Soil Characteristics from Forest Areas of the Southwest Region in Korea (국내 서남 지역의 산림 토양 특성과 곤충병원성 진균과의 상관관계)

  • Entomopathogenic fungi are one of the microorganisms distributed worldwide, and they inhabit not only insects but also soils of various regions such as agricultural land and forest areas. In this study, entomopathogenic fungi distributed in the soil of forest areas in Korea were isolated using the insect-baiting method, and the relationship between the distribution and diversity of entomopathogenic fungi and the physical/chemical characteristics of the soil was analyzed. The soils were collected from five forest areas in Korea, and a total of 42 entomopathogenic fungal isolates were isolated from the collected soils. Among them, Beauveria bassiana (42.9%) and Metarhizium anisopliae (45.2%) were mainly isolated. In particular, it was confirmed that the distribution of M. anisopliae varies depending on the total nitrogen (g/kg) and organic matter content (%) of the soil. The soils in forest areas in Korea have a low pH of 4-5. Among all isolates, >50% of entomopathogenic fungi were isolated from silt loam. This study suggests that it will be helpful in understanding the relationship between the distribution and diversity of entomopathogenic fungi and the physical and chemical characteristics of soil.

Delineation and Land Use Analysis of the Isolated Former Floodplain in the Nakdong River, Korea (낙동강에서 격리된 과거 홍수터의 경계 설정과 토지이용 분석)

  • For the restoration of lateral connectivity between rivers and floodplains, it is important to find the isolated former floodplain (IFF) and to characterize its land use in Korean rivers which were channelized by levee constructions for flood protection. The aim of this study is to map the IFF and to assess its land use pattern in the Nakdong River, Korea. The isolated former floodplain was explored by being overlapped on a digital elevation model (DEM), digital topographic map and design flood level using a geographical information system (GIS) in the Nakdong River basin. The land use of the identified IFF was classified by land-use map. The total number of IFFs was 384 and their total area was $291km^2$. While IFFs were usually surrounded by mountain forest in the upper river area, they tended to be located on wide plain areas in the downstream area of Nakdong River. The land use pattern of IFFs was mostly farmland (73.9%) and urban areas (12.7%) in the river. The results of delineation and land use analysis of isolated former floodplain in the Nakdong River will be used as a base line data for planning stream restoration.

Changes in Vegetation Characteristics Over Time in the Isolated Forests of the Urban Areas: A Case Study on the Mt. Hwangyeong, Busan Metropolitan City (도시 내 고립된 임지의 경시적 식생특성 변화: 부산광역시 황령산을 사례로)

  • In order to assess th changes in vegetation characteristics over time in the forest-lands which is isolated by urbanization, vegetation surveys based on the Braun-Blanquet phytosociological method was carried out in 1996 and 2015 on Mt. Hwangyeong located in the center of Busan metropolitan city, South Korea. The number of vegetation types based on floristic composition showed no significant changes, the vegetation units under the community levels was more or less represents the difference. the average total vegetation cover and average number of species per unit area ($100m^2$) was increased 16% and 2 species, respectively. The relative importance value (RIV) for each tree species, Quercus spp. like as Q. mongolica and Q. serrata was decreased, while the warmth-tolerant trees, evergreen broad-leaved trees, and mesophyte like Lindera erythrocarpa, Styrax japonica, Osmanthus heterophyllus, and Stephanandra incisa was increased significantly. Changes of the life form spectrums of vascular plants did not substantially, hemicryptophyte(H) and therophyte (Th) was decreased, while nanophanerophyte(N) was increased significantly. Also through the creating of large-scale vegetation map (based on physiognomic vegetation types) was compared the spatial distribution characteristics of each vegetation types.

Analysis of Surface Temperature on Urban Green Space Using Unmanned Aerial Vehicle Images - A Case of Sorasan Mt. Nature Garden, Iksan, South Korea - (무인항공 영상을 활용한 도심녹지 표면온도 특성 분석 - 익산 소라산 자연마당을 대상으로 -)

  • This study analyzed the surface temperature characteristics of urban green spaces under high summer temperatures to clarify the functions of green spaces in reducing urban temperatures. We obtained accurate surface temperature data using highresolution unmanned aerial vehicle images of the survey site, which was an isolated green space in the city. We analyzed differences in the surface temperature by land cover type, vegetation type, species type, and the relationship between surface temperature and vegetation volume. Based on the results, among the land cover types, wetlands and forests had low temperatures and paving areas had very high temperatures. Regarding vegetation type, broad-leaved trees had lower temperatures than coniferous trees in forests. However, in planted areas, coniferous trees had lower temperatures than broad-leaved trees. The temperature of long grass was higher than that of short grass, which suggested that the volume of grass affected the temperature. Regarding forest species type, the temperature of broad-leaved Robinia pseudoacacia forest and mixed broad-leaved forest was lower than coniferous Pinus densiflora forest. There was a slight difference in temperature between R. pseudoacacia forest and mixed broad-leaved forest. The analysis of the relationship between vegetation volume and temperature by forest species type indicated a negative correlation, where the temperature decreased with increasing vegetation volume, similar to the results of previous studies. However, we found a weak positive correlation in R. pseudoacacia forest; therefore, an increase in volume may not reduce the surface temperature depending on the dominant species.

Gallotannins from Nut Shell Extractives of Camellia oleifera

  • Camellia nut shell was collected, dried at room temperature and ground to get fine powder. The powder was extracted three times with 95% EtOH, combined, evaporated, and then freeze dried. The crude powder was dissolved in H2O and then sequentially fractionated with n-hexane, CH2Cl2, EtOAc and n-BuOH. A part of EtOAc fraction was chromatographed on a silica gel and on a Sephadex LH-20 columns using MeOH, aqueous MeOH, EtOAc-n-hexane and EtOH-n-hexane to isolate gallotannins. Three gallotannins, 1,2-di-O-galloyl-β-D-glucopyranoside (2), 1,2,6-tri-O-galloyl-β-D-glucopyranoside (3) and 1,2,3,6-tetra-O-galloyl-β-D-glucopyranoside (4), including gallic acid (1), were isolated and elucidated by NMR and Mass spectroscopies. Although nothing new, these gallotannins were first reported from the nut shell extractives of camellia tree (Camellia oleifera C. Abel). This study was to investigate the chemical constituents, especially hydrolysable tannins, of nut shell extractives of Camellia oleifera and to provide basic information for the future chemical utilization of this species.

Distribution Pattern of Soil Actinomycetes on the Seasonal Change (계절에 따른 토양 방선균의 속 다양성 분포)

  • From soils seasonally collected at two depths (0~2 cm, $50{\pm}1cm$) of forest, field, grass land, or paddy field, distinct strains of actinomycetes were isolated and identified to the genus level. The genus-diversity of soil actinomycetes was revealed to be considerably different by seasonal change. It was also affected by soil depths, soil types, or actinomycete groups. At the soil depth of 0~2 cm, the seasonal distribution fluctuation (%) of streptomycete strains was higher in grass land (41%), field (39%) soil than paddy field (18%), or forest (18%), whereas that of streptomycete strains at the soil depth of $50{\pm}1cm$ was high in order of paddy field (36%), field (28%), grass land (26%), and forest (16%). On the other hand, the seasonal distribution fluctuation ratio of rare actinomycete strains at the soil depth of 0~2 cm was above 45% except for paddy field (26%). At the soil depth of $50{\pm}1cm$, the seasonal distribution of rare actinomycete strains exhibited high fluctuation (%) in order of forest (79%), paddy field (36%), field (24%), and grass land (10%).

  • The expansion of human activities and road development has led to the loss and fragmentation of ecological spaces, which is a negative factor for biodiversity. In particular, urban areas where land use and land cover have rapidly changed into urbanization zones are regions where ecological spaces are lost and isolated, making it difficult for wildlife to inhabit. Furthermore, the loss and fragmentation of ecological spaces due to urbanization can have a negative impact on ecosystem services. Therefore, to enhance biodiversity and ecosystem services in urban and national land, it is necessary to establish a practical ecological axis that reflects the current status of the city. Thus, this study analyzed the connectivity of ecological spaces and forest axis that can be used for spatial planning related to urban ecological axis of local governments in Daegu and Gyeongsangbuk-do. The ecological connectivity was analyzed by dividing the Daegu-Gyeongbuk region into 31 local government units, distinguishing between forests and natural areas using land cover data. Subsequently, the study area was divided into 20,483 hexagonal grids of 1 square kilometer each, and the restoration effects for ecological fragmentation within 100 meters were spatially clustered to visualize priority restoration areas. The forest axis was derived by considering regional conditions such as land cover, building area, slope, and others to connect 1,534 forests of 100 hectares or more. The research results are expected to be used as fundamental data for spatial planning, goal setting, and the selection of restoration areas for improving ecological connectivity.

  • For the restoration of lateral connectivity between channel and floodplain, it is important to find the former floodplain and to characterize its land use in streams which were channelized by the levee construction for the flood protection. The aim of this study is to map the former floodplains and to assess its land use pattern in the Cheongmi-cheon Stream, Korea. The former floodplains were explored by being overlapped on a digital elevation model (DEM), digital topographic map and design flood level using a geographical information system (GIS) in the Cheongmi-cheon Stream basin. The land use of the identified former floodplains was classified by land-use map. The total number of the former floodplains was 104 and their total area was $11.9km^2$ in the Cheongmi-cheon Stream. The land use pattern of the former floodplains was mostly farmland (87.1%). The former floodplains were usually surrounded by mountain forest in the downstream of the Cheongmi-cheon Stream. These former floodplains are probably suitable for restoration of lateral connectivity because of lower ratio of urban area but higher ratio of farmland. The results of delineation and land use analysis of the former floodplain can be used as a baseline data for planning stream restoration in the Cheongmi-cheon Stream.

Isolation and Sequence Analysis of Ycf4 Gene from Zoysia japonica Steud.

  • Zoysia japonica Steud.(Zj) is a typical warm-season Korean lawn grass, which is used in many places such as river banks, roadside and soccer fields in Korea. Recently, it has also been used in school yards and the Saemangeum reclaimed land to reduce water pollution. Although the cultivated area of turfgrass is steadily increasing worldwide, it grows fast requiring frequent mowing and is difficult to grow in shady areas and the cold region. Therefore this study aims searching for useful gene(s) to develop abiotic stress tolerant and dwarf zoysiagrass. We isolated Ycf4 gene based on the sequence from Oryza sativa Japonica through RT-PCR and RACE PCR. Ultimately, open reading frame (ORF) of ZjYcf4 was 558bp long, encoding a protein of 186 amino acid residues. NCBI blast results showed that the ZjYcf4 protein is evolutionarily closely related to Ycf4 protein from Zoysia macrantha and Setaria italica (100% and 98%, respectively). To determine whether ZjYcf4 was involved in environmental stress in wild-type zoysiagrass, expression patterns of the gene were analyzed by real-time PCR under salt, cold and dark conditions. They were analyzed after each stress treatment for 3 hours. In salt and cold stresses, the expression was higher compared to control (3-fold and 1.5-fold, respectively), although there was a 1.6-fold decrease in expression under dark stress treatment. As reported previously, we suggest that ZjYcf4 gene affects abiotic stress such as salt, cold and dark.
