The mineral status of native goats and forage species, namely; Cynodon plectostachyus, Pennisetum purpureum. Eleusine indica, Cynodon dactylon, Calopogonium muconoides, Centrosema pubescens, Leucaena leococephala, and Mimosa pudica in lahar affected areas of Concepcion, Tarlac, Philippines were determined. Forage and blood samples were collected six times in 1996-97, and analyzed for calcium, phosphorus, magnesium, sulfur, copper, iron, molybdenum, selenium, and zinc. Forage calcium and sulfur are non-limiting. Most species had low phosphorus, copper and selenium, while some had magnesium and zinc levels lower than the critical limit because of low mineral content and high percolation rate of lahar deposits. Iron and molybdenum were in excess. The effect of seasonal variation was observed only in copper, sulfur and iron. Average blood mineral concentration of the animals was above critical limit, but there were no significant differences between seasons. All the animals had plasma phosphorus and magnesium above critical level; but 20 % had low copper, zinc and selenium especially in dry season possibly due to insufficient amount of these elements and excessive molybdenum and iron in most forage. Conversely, calcium in forage was high; but 40 % of the animals had low plasma calcium concentration. Although no clinical signs of mineral deficiencies were observed, supplemental feeding would be important since the condition of the pasture in lahar-laden areas is not expected to improve in the next five years. Intensified use of L. leucocephala with better mineral profile would be ideal.
We investigated the changes in the species of the woods used for Korean ancient and historic architectures, which include prehistoric excavated relics and existing wooden buildings in South Korea. The species data were collected from various sources such as excavation and repair reports, journal papers, and a few unpublished documents. We divided the building Periods as Paleolithic, Neolitic, Bronze Ages, Iron Age/Three Kingdoms, Koryo, Joseon (early, middle, late) and modem periods. In prehistoric periods, hardwoods were major species. Oak (Quercus spp.) woods dominated (94 percent in average); the others (5%) were Juglans mandshurica, Platycarya strobilacea, Castanea crenata, and few softwoods(1%). During Iron Age and Three Kingdom periods, oaks remained as a major species (57%) and others Platycarya strobilacea(21%), Castanea crenata(13%), and Pinus spp. (6%). The oak woods decreased in Koryo period and they occupied only 1.1%. Instead of oaks, pine (Pinus spp., 71%) and Zelkova serrata (22%) dominated in Koryo. In early and middle Joseon periods, pine woods (73%) remain as a major species and the others were oaks (14%) and Zelkova serrata (9%). As late Joseon came, the pine woods occupied more than 88%. In the late 19th and early 20th centuries, a few boreal species such as larch (Larix spp.) and spruce (Picea spp.), which grow in cold area, were found. We believe they were transported from northern Korea. The existing buildings in Korea are mainly from Joseon period and a few from late Koryo periods. During these periods, pine woods were used for most buildings. For such reason, pine woods were known as 'representative materials for historic buildings'. but earlier times, broad-leaved trees, i.e., oak and Zelkova woods were major materials. The changes in building materials resulted from both climate and human impacts. The dry climate and disturbed forests induce more pines in the mountains. We also compared the wood qualities of the species and found that Zelkova woods were superior ones and deserved more planting for future demands in the repair for historic buildings.
Low-Z particle Electron Probe X-ray Microanalysis was applied to characterize MSW fly- and bottom -ash particle samples originated from two municipal incinerators (denoted as A and B) in Korea. According to their chemical composition, many distinctive particle types were identified. In A fly ash, the major chemical species are carbon-rich, aluminosilicates and many particles are composed of as a mixture of $ CaCO_3$ and other chemical species such as $CaSO_4$ or $CaCl_2$. For B fly ash, Fe, iron oxide, NaCl and NaCl-containing particles are the most abundant. In bottom ash, A and B were composed of similar chemical species such as carbon-rich, Fe, iron oxide, $CaCO_3$, and aluminosilicates. It was demonstrated that the single-particle characterization using this low-Z particle EPMA technique provided detailed information on various types of chemical species in the MSW ash samples. In addition, the technique has advantage over conventional analytical techniques in the point that both crystalline and glass-like ash particles can be analyzed at the same time.
Oxidative stress has been associated with a number of diseases in human. Among the sources that can generate oxidative stress, it has been reported that iron can generate reactive oxygen species (ROS)with thiol. In iron overload state, increased thiol levels in plasma appeared to be associated with human mortality. In this study we examined whether iron could interact with thiols in plasma, generating ROS. In human plasma, unlike with Fe(III), Fe(II) increased lucigenin-enhanced chemiluminescence in concentration-dependent manner, and this was inhibited by SOD. Boiling of plasma did not affect chemiluminescence induced by Fe(II). Hovever, thiol depletion in plasma by pretreatment with N-ethylmaleimide (NEM)decreased Fe(II)-induced chemiluminescence significantly, suggesting that Fe(II) generated superoxide anion by the nonenzymatic reaction with plasma thiol. Consistent with this findings, albumin, the major thiol contributor in plasma, also generated ROS with Fe(II) and this generation was inhibited by pretreatment with NEM. Treatment with Fe(II) to plasma resulted un significant reduction of oxygen radical absorbance capacity (ORAC) value, suggest that total antioxidant capacity could diminished in iron overload state. In conclusion, In iron overload state, plasma may be affected by oxidative stress mediated by nonenzymatic reaction of Fe (II)with plasma thiol.
Methylglyoxal (MG) was identified as an intermediate in non-enzymatic glycation and increased levels were reported in patients with diabetes. In this study, we evaluated the effects of MG on the modification of ferritin. When ferritin was incubated with MG, covalent crosslinking of the protein increased in a time- and MG dose-dependent manner. Reactive oxygen species (ROS) scavengers, $N-acetyl-_L-cysteine$ and thiourea suppressed the MG-mediated ferritin modification. The formation of dityrosine was observed in MG-mediated ferritin aggregates and ROS scavengers inhibited the formation of dityrosine. During the reaction between ferritin and MG, the generation of ROS was increased as a function of incubation time. These results suggest that ROS may play a role in the modification of ferritin by MG. The reaction between ferritin and MG led to the release of iron ions from the protein. Ferritin exposure to MG resulted in a loss of arginine, histidine and lysine residues. It was assumed that oxidative damage to ferritin caused by MG may induce an increase in the iron content in cells, which is deleterious to cells. This mechanism, in part, may provide an explanation or the deterioration of organs under diabetic conditions.
Oxidative stress is a cascade reaction characterized by a significant increase in the amount of oxidized components. Free radicals produced by oxidative stress are one of the common features in several experimental models of disease, and contribute to wide range of neurodegenerative diseases, including Alzheimer's disease. Iron (II) species can participate in the Fenton, and Fenton-like reactions, to react with hydrogen peroxide and generate hydroxyl radical. As iron accumulation and oxidative stress are associated with the pathological progression of neurodegenerative diseases, iron chelation and antioxidant therapies have become strategies to combat these diseases. Due to the complexity of the redox system in vivo, a multifaceted approach may be an attractive therapeutic strategy. Further investigations are highly expected for the prevention and treatment of neurodegenerative diseases in future.
[FeIII(BBA)DBC]ClO4 as a new functional model for catechol dioxygenases has been synthesized, where BBA is a bis(benzimidazolyl-2-methyl)amine and DBC is a 3,5-di-tert-butylcatecholate dianion.The BBA complex has a structuralfeature that iron cent er has a five-coordinate geometry similar to that of catechol dioxygenase-substrate complex.The BBA complex exhibits strong absorptionbands at 560 and 820 nm in CH3CN which are assigned to catecholate to Fe(III) charge transfer transitions. It also exhibits EPR signals at g = 9.3 and 4.3 which are typical values for the high-spin FeIII (S = 5/2) complex with rhombicsymmetry. Interestingly, the BBA complex reacts with O2 within an hour to afford intradiol cleavage (35%) and extradiol cleavage (60%) products. Surprisingly, a green color intermediate is observed during the oxygenation process of the BBA com-plex in CH3CN. This green intermediate shows a broad isotropic EPR signal at g = 2.0. Based on the variable temperature EPR study, this isotropic signalmight be originated from the [Fe(III)-peroxo-catecholate] species havinglow-spin FeIII center, not from the simple organic radical. Consequently,it allows O2 to bind to iron cen-ter forming the Fe(III)-superoxide species that converts to the Fe(III)-peroxide intermediate. These present data can lead us tosuggest that the oxygen activation mechanism take place for the oxidative cleavingcatechols of the five-coordinate model systems for catechol dioxygenases.

In our previous study, the expression of active H-ferritins in Saccharomyces cerevisiae was found to reduce cell growth and reactive oxygen species (ROS) generation upon exposure to oxidative stress; such expression enhanced that of high-affinity iron transport genes (FET3 and FTR1). The results suggested that the recombinant cells expressing H-ferritins induced cytosolic iron depletion. The present study analyzes metabolic changes under these circumstances via proteomic methods. The YGH2 yeast strain expressing A-ferritin, the YGH2-KG (E62K and H65G) mutant strain, and the YGT control strain were used. Comparative proteomic analysis showed that the synthesis of 34 proteins was at least stimulated in YGH2, whereas the other 37 proteins were repressed. Among these, the 31 major protein spots were analyzed via nano-LC/MS/MS. The increased proteins included major heat-shock proteins and proteins related to endoplasmic reticulum-associated degradation (ERAD). On the other hand, the proteins involved with folate metabolism, purine and methionine biosynthesis, and translation were reduced. In addition, we analyzed the insoluble protein fractions and identified the fragments of Idh1p and Pgk1p, as well as several ribosomal assembly-related proteins. This suggests that intracellular iron depletion induces imperfect translation of proteins. Although the proteins identified above result from changes in iron metabolism (i.e., iron deficiency), definitive evidence for iron-related proteins remains insufficient. Nevertheless, this study is the first to present a molecular model for iron deficiency, and the results may provide valuable information on the regulatory network of iron metabolism.
This study was performed to investigate the discrimination of Angelica gigas, Angelica acutiloba and Angelica sinensis on the treatment of chromaticity and colorfastness. Angelica gigantis root has been used as a Korean traditional medicine for the treatment of woman disease. Natural dyes give us many great benefits, including diversified color, but no pollution. These studies were carried out acetate iron, dichloride copper and alum with a mordant to ramie fabric. The ramie fabric was dyed with Angelica gigas, Angelica acutiloba and Angelica sinensis. The results of experiment showed as follows: In discrimination by dyeing, the colors of Angelica acutiloba and Angelica sinensis were very similar, but that of Angelica gigas was different. There were no differences among colors of materials using non-mordant. But dyeing with iron acetate and copper dichloride were showed dark in Angelica gigas than other angelica species.
