Nonactin을 이온 운반물질(ionophore)로, poly(vinyl chloride) (PVC) 또는 polyurethane (PU)을 지지체로, 그리고 bis(2-ethylhexyl)adipate을 가소제로 사용하여 제작한 암모늄이온 선택성전극의 성능을 비교한다. PU-전극의 검출한계를 PVC전극에 비해 더 낮게 제작할 수 있는 것을 제외하고는 PVC전극이 PU전극에 비하여 그 성능이 우수하였으며 이를 설명하기 위하여 AFM 형상과 임피던스 측정 등을 하였다. PU-전극이 PVC에 비하여 이온 이동에 대한 저항이 더 큰 것으로 보아 더 단단한 막을 형성하여 더 낮은 검출 한계를 가지며 수명이 긴 전극으로 발전시킬 수 있을 것으로 보인다.
Ionophore ETH4120의 이온담체작용에 의한 란탄족원소이온의 액체/액체계면에서의 상전이를 순환전압전류법(cyclic voltammetry)과 순환전류주사 대시간전위차법(Chronopotentiometry with cyclic linear current scanning; CPCLCS)으로 고찰하였다. 수용액 중의 란탄족원소이온농도가 유기용액중의 착화제 농도보다 훨씬 높을 때 순환전압전류법과 CPCLCS 곡선에서 하나의 양극전류(수용액상에서 유기상으로 전이)파와 두개의 음극전류(유기상에서 수용액상으로 전이)파가 관찰되었다. 양극전류파는 1:1(금속:착화제) 착물형성과 관계되는 봉우리이고, 두 음극전류파는 각각 1:2와 1:3의 연속적인 착물형성과 관계된 파이다. 그러나 1:1 착물의 양극전류파와 대응하는 1:1 착물의 음극파와, 1:2 착물과 1:3 착물의 음극전류파에 대응되는 양극전류파는 나타나지 않았다. 이와 관련되는 이온 상전이 메카니즘을 검토하였다.
Cell death of trophoblast, particularly by abnormal release of physiological nitric oxide (NO) has been known to be a causative factor of pre-eclampsia. In the present study, effects of intracellular calcium increase enhancing the activity of NO synthases (neuronal NO synthase, nNOS in this trophoblast cells) on the cell death were examined in a human placental full-term cell line (HT-1). Furthermore, we analyzed the possible mechanisms underlying the augmentation of $Ca^{++}$-mediated NOS activity mediated by protein kinases like PKC, PKA, or CaM-KII. In experiments for cell toxicity, a calcium ionophore (ionomycin $10{\mu}M$) enhanced cell death confirmed by MTT assay, and increased significantly nNOS activity determined with a hemoglobin oxidation assay. This cell death was partially protected by pre-treatment of 7-nitroindazole (7-NI, $10{\mu}M$ and $100{\mu}M$), a nNOS-specific inhibitor. Additionally, $Ca^{++}$-ionophore -induced increase of nNOS activity also was partially normalized by pre-treatment of specific inhibitors of protein kinases, PKC, PKA or CaM-KII. Therefore, we suggest that an increase of calcium influx, leading to the activation of nNOS activity, which in turn may result in the death of trophoblast cells by involvement of signaling mechanisms of protein kinases.
The inhibitory effects of 25 polyphenols against in vitro allergic reactions were compared using biochemical and cell assays. Three polyphenols including curcumin, gallic acid, and quercetin suppressed the release of $\beta$-hexosaminidase from ionophore A23187-stimulated RBL-2H3 cells more effectively (>50% inhibition at $100{\mu}M$ concentration). They were found to have potencies in suppressing the release of histamine not only from ionophore A23187-, but also from immunoglobulin E (IgE)-stimulated RBL-2H3 cells. Moreover, such suppressive effects of the three polyphenols were also observed in A23187 plus PMA-costimulated rat peritoneal mast cells. The extent of inhibition were quantified as the respective polyphenol concentration that inhibit 50% ($IC_{50}$) of $\beta$-hexosaminidase or histamine release, showing an inhibition tendency with decreasing order of curcumin>gallic acid>quercetin. Down-regulation of $Ca^{2+}$ influx was suggested as the cause of the inhibition of $\beta$-hexosaminidase and histamine releases in these cells. The immune process inhibition was confirmed by the observed reduction in the gene expressions and release of pro-inflammatory cytokine tumor necrosis factor (TNF)-$\alpha$, interleukin (IL)-$1\beta$, and IL-4, due probably to antioxidant activity of the polyphenols. These findings illustrate that curcumin, gallic acid, and quercetin may be beneficial against allergic inflammatory diseases.
Natural products, which have been used for the treatment of hypertension, diuresis and nephritis in traditional oriental medicine, were selected for the screening of nitric oxide (NO) production in endothelial cells and kidney tissues in vitro as well as in vivo by measuring the conversion of [$\^$14/C]-L-arginine to [$\^$14/C]-L-citrulline, a coproduct of the enzyme reaction with NO. Confluent monolayer of endothelial cells were used for the screening of 16 natural products. Among the natural products, Zizyphus jujuba and Codonopsis pilosula stimulated endothelial NO synthase activity. Thus, both confluent monolayer of endothelial cells and kidney homogenates (glomeruli, cortical tubules, meudllae) were treated with Zizyphus jujuba and Codonopsis pilosula (final concentration 10 $\mu\textrm{g}$/$m\ell$) and NO releases were compared with those by receptor - dependent agonists, bradykinin and ADP and receptor - independent calcium ionophore A23187 in vitro. In rat experiment, NO releases in glomeruli, cortical tubules and medullae and plasma renin activity were assessed after intraperitoneal injection of Zizyphus jujuba and Codonopsis pilosula (10 mg/kg/day for 4 days). As a result, both Zizyphus jujuba and Codonopsis pilosula significantly increased NO releases in cultured endothelial cells, kidney tissues in vitro as well as in vivo. Stimulation of NO releases by Zizyphus jujuba and Codonopsis pilosula was similar to those by receptor - dependent agonists, bradykinin and ADP and receptor - independent calcium ionophore A23187 in cultured endothelial cells. However, plasma renin activity was not influenced by these two natural products. In conclusion, stimulatory effects of Zizyphus jujuba and Codonopsis pilosula on NO release in kidney may contribute their hypotensive effects and antinephritic action possibly by increasing renal blood flow.

We previously identified a new heparinase inhibitor fungal metabolite, named CRM646-A, which showed inhibition of heparinase and telomerase activities in an in vitro enzyme assay and antimetastatic activity in a cell-based assay. In this study, we elucidated the mechanism by which CRM646-A rapidly induced nucleus condensation, plasma membrane disruption and morphological changes by increasing intracellular Ca2+ levels. Furthermore, PD98059, a mitogen-activated protein kinase (MEK) inhibitor, inhibited CRM646-A-induced nucleus condensation through ERK1/2 activation in rat 3Y1 fibroblast cells. We identified CRM646-A as a Ca2+ ionophore-like agent with a distinctly different chemical structure from that of previously reported Ca2+ ionophores. These results indicate that CRM646-A has the potential to be used as a new and effective antimetastatic drug.
The current study was designed to evaluate the effects of the reactive oxygen species (ROS) generated with a xanthine (X) and xanthine oxidase system (XO) on sperm function and DNA fragmentation in porcine spermatozoa. ROS were produced by a combination of $1,000{\mu}M$ X and 50 mU/ml XO. The ROS scavengers such as superoxide dismutase (SOD) (200 U/ml) and catalase (CAT) (500 U/ml) were also tested. Spermatozoa were incubated for 2 hours in BWW medium with a combination of X-XO supplemented with or without antioxidants at $37^{\circ}C$ under 5% $CO_2$ incubator. Ca-ionophore-induced acrosome reaction, the proportion of swollen spermatozoa under hypo-osmotic condition, malondialdehyde formation for the analysis of lipid peroxidation, and the proportion of DNA fragmentation were determined after 2 hours incubation. The action of ROS on porcine spermatozoa resulted in decreased Ca-ionophore-induced acrosome reaction and membrane integrity, increased the formation of malondialdehyde, and the proportion of sperm with DNA fragmentation(p<0.05). The toxic effects caused by ROS were completely alleviated by CAT in terms of sperm function and characteristics, however SOD did not serve the same scavenger effect as CAT. To conclude, the ROS can cause significant damage to porcine sperm functions and characteristics, which can be minimized by the use of antioxidants.
Although the effect of Bee Venom has been reported, its mechanism has not been fully elucidated. Nitric Oxide(NO) is one of the free radicals and mediates in inflammation diseases. Chemokines contribute to the pathogenesis of several disorders such as allergic rhinitis and rheumatoid arthritis and so on. The objective of this study was to investigate the scavenging effect of Bee Venom on NO and on expression of chemokine genes. There was no significant NO scavenging effect in Crude Bee Venom, Apamin, Melittin, and MCD-peptide. The expression of chemokines was examined by RT-PCR using the human mast cell line(HMC-1), which is known to secrete and express chemokines. In order to investigate the protective effect of Bee Venom, HMC-1 cells were incubated with pretreatment of Bee Venom for 24 hrs and stimulated with 1 uM calcium ionophore A23178 for 2 hrs. RT-PCR analyses of chemokine genes showed that expressions of RANTES and MCP-1 were increased compared to the calcium ionophore-only treated group. But IL-8 and MCP-3 did not express increasing effect compared to control group. This study may provide important basic data on the possibility of the clinical treatment of Bee Venom in inflammation diseases.
납이온과 선택적으로 상호작용하는 이온 운반물질(lead ionophore II)과 수소이온과 선택적으로 상호작용하면서 형광을 내는 변색성 이온 운반물질(ETH5294) 및 소수성의 음이온 자리를 포함하는 이온 선택성 광센서를 제조하여 형광분광법으로 수용액 중의 납이온을 정량하는 방법을 연구하였다. 시료용액의 pH,막두께 등이 형광세기에 미치는 영향을 조사하였다. $Na^+$, $K^+$, $Mn^{2+}$ 및 $Zn^{2+}$이온 등의 방해이온이 납이온의 정량에 미치는 영향을 조사하였다.본 연구에서 제작한 납이온 선택성 막을 이용하여 얻은 납이온 검정곡선의 직선범위는 5.0${\times}10^-7$M5.0${\times}10^-3$M이였고 이 범위에서의 상관계수는 -0099107이었다. 바탕용액의 상대표준편차는 3.0%였고 납이온의 검출한계는 5.0${\times}10^-9$M이었다.
계배 근원세포의 배양 배지에 calcium ionophore A23187이나 EGTA를 배양 24시간에 첨가함으로서 초래된 세포내 칼슘 농도의 변화는 근원세포의 분화과정에 상당한 영향을 미쳤다. 배양 24시간에 A23187이나 EGTA를 첨가한 후 배양 48시간, 72시간, 및 96시간에 각각 세포를 [35S]methionine으로 1시간 표지시킨 후 수확하여 2차원 전기영동법으로 단백질을 분리시켰을 때, 일부 단백질은 배양 조건에 따라 합성 양상을 달리함을 보였다. 배양 24시간에 처리한 A23187과 calcium-activated neutral protease는 대조군에 비해 세포융합을 촉진시켰으나 동일 시기에 처리된 phosphoprotein을 정량함으로써 조사하였을 때, A23187이 배양 초기에는 대조군에 비해 약간 이 효소의 활성도를 높이는 효과를 보였으나 세포융합이 완성된 시기인 96시간에는 대조군에 비해 활성도를 높이는 효과를 보였으나 세포융합이 완성된 시기인 96시간에는 대조군에 비해 활성도의 차이를 나타내지 않았다. A23187 및 calcium-activated neutral protease에 의한 세포융합의 촉진, 그리고 A23187에 의한 protein kinase C 활성도의 증가가 모두 근원세포의 융합이 활발히 진행되는 시기인 배양 48-72 시간에 관찰됨을 볼 때, 세포내 칼슘의 농도는 protein kinase C 및 calcium-activated neutral protease와 상호연관을 가지면서 세포분화에 관여하는 것으로 사료된다.
