Chemical characteristics of wet precipitation in urban and mountainous sites of Jeju Island

  • Wet precipitation samples were collected in Jeju City and Mt. Halla-1100 site (a site at an altitude of 1100 m on Mt. Halla) during 2011-2013, and their major ionic species were analyzed to examine the chemical composition and characteristics. A comparison of ion balance, electric conductivity, and acid fraction of precipitation revealed correlation coefficients in the range of r = 0.950~0.991, thereby implying the high quality of analytical data. Volume-weighted mean pH and electric conductivity corresponded to 4.86 and 25.5 µS/cm for Jeju City, and 4.98 and 15.1 µS/cm for Mt. Halla-1100 site, respectively. Ionic strengths of the wet precipitation in Jeju City and Mt. Halla-1100 site corresponded 0.3 ± 0.5 and 0.2 ± 0.2 mM, respectively, thereby indicating that more than 30 % of total precipitation was within a pure precipitation criteria. The precipitation with a pH range of 4.5 - 5.0 corresponded to 40.8 % in Jeju City, while the precipitation with a pH range of 5.0 - 5.5 corresponded to 56.9 % in Mt. Halla-1100 site, thereby indicating slightly more weak acidity than that in Jeju city. The volume-weighted mean concentration (µeq/L) of ionic species was in the order of Na+ > Cl- > nss-SO42- > NO3- > Mg2+ > NH4+ > H+ > nss-Ca2+ > PO43- > K+ > CH3COO- > HCOO- > NO2- > F- > HCO3- > CH3SO3- at Jeju City area, while it corresponded to Na+ > Cl- > nss-SO42- > NO3- > NH4+ > H+ > Mg2+ > nss-Ca2+ > PO43- > CH3COO- > K+ > HCOO- > NO2- > F- > HCO3- > CH3SO3- at Mt. Halla-1100 site. The compositions of sea salts (Na+, Cl-, Mg2+) and secondary pollutants (NH4+, nss-SO42-, NO3-) corresponded to 66.1 % and 21.8 %, respectively, in Jeju City and, 49.9 % and 31.5 %, respectively, in Mt. Halla-1100 site. The acidity contributions in Jeju City and Mt. Halla-1100 site by inorganic acids, i.e., sulfuric acid and nitric acid, corresponded to 93.9 % and 91.4 %, respectively, and the acidity contributions by organic acids corresponded to 6.1 % and 8.6 %, respectively. The neutralization factors in Jeju City and Mt. Halla1100 site by ammonia corresponded to 29.8 % and 30.1 %, respectively, whereas the neutralization factors by calcium carbonate corresponded to 20.5 % and 25.2 %, respectively. From the clustered back trajectory analysis, the concentrations of most ionic components were higher when the airflow pathways were moved from the continent to Jeju area.

The Effect of Strong Acid and Ionic Material Addition in the Microwave-assisted Solubilization of Waste Activated Sludge (Microwave를 이용한 폐활성슬러지의 가용화 반응에서 강산과 이온성 물질의 첨가가 미치는 영향)

  • The study of waste activated sludge (WAS) solubilization has been increased for sludge volume reduction and enhancing the efficiency of anaerobic digestion. Microwave (MW)-assisted solubilization is an effective method for the solubilization of WAS because this method can lead to thermal, nonthermal effect and ionic conduction by dielectric heating. In this study, the solubilization of WAS by MW heating and conductive heating (CH) was compared and to enhance the MW-assisted solubilization of WAS at low MW output power, chemical agents were applied such as $H_2SO_4$ as the strong acid and $CaCl_2$, NaCl as the ionic materials. Compared to the COD solubilization of WAS by CH, that by MW heating was approximately 1.4, 6.2 times higher at $50^{\circ}C$, $100^{\circ}C$, respectively and the highest COD solubilization of WAS was 10.0% in this study of low MW output power condition. At the same MW output power and reaction time in chemically agents assisted experiments, the COD solubilization of WAS were increased up to 18.1% and 12.7% with the addition of $H_2SO_4$ and NaCl, however, that with the addition of $CaCl_2$ was 10.7%. This result might be due to the fact that the precipitation reaction occurred by calcium ion ($Ca^{2+}$) and phosphate ion (${PO_4}^{3-}$) produced in WAS after MW-assisted solubilization. In this study, $H_2SO_4$ turned out to be the optimal agent for the enhancement of MW efficiency, the addition of 0.2 M $H_2SO_4$ was the most effective condition for MW-assisted WAS solubilization.

Characterization of ginsenoside compound K loaded ionically cross-linked carboxymethyl chitosan-calcium nanoparticles and its cytotoxic potential against prostate cancer cells

  • Backgroud: Ginsenoside compound K (GK) is a major metabolite of protopanaxadiol-type ginsenosides and has remarkable anticancer activities in vitro and in vivo. This work used an ionic cross-linking method to entrap GK within O-carboxymethyl chitosan (OCMC) nanoparticles (Nps) to form GK-loaded OCMC Nps (GK-OCMC Nps), which enhance the aqueous solubility and stability of GK. Methods: The GK-OCMC Nps were characterized using several physicochemical techniques, including x-ray diffraction, transmission electron microscopy, zeta potential analysis, and particle size analysis via dynamic light scattering. GK was released from GK-OCMC Nps and was conducted using the dialysis bag diffusion method. The effects of GK and GK-OCMC Nps on PC3 cell viability were measured by using the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide assay. Fluorescent technology based on Cy5.5-labeled probes was used to explore the cellular uptake of GK-OCMC Nps. Results: The GK-OCMC NPs had a suitable particle size and zeta potential; they were spherical with good dispersion. In vitro drug release from GK-OCMC NPs was pH dependent. Moreover, the in vitro cytotoxicity study and cellular uptake assays indicated that the GK-OCMC Nps significantly enhanced the cytotoxicity and cellular uptake of GK toward the PC3 cells. GK-OCMC Nps also significantly promoted the activities of both caspase-3 and caspase-9. Conclusion: GK-OCMC Nps are potential nanocarriers for delivering hydrophobic drugs, thereby enhancing water solubility and permeability and improving the antiproliferative effects of GK.

Surface Chemical Aspects of Coagulation, Deposition, and Filtration Processes: Variation of Electrokinetic Potential at Metal Oxide-Water and Organic-Water Interfaces in the $Na^+$ and $Ca^{2+}$ Ion Solutions

  • This study measured the zeta potential of both latex colloidal particles with carboxylate surface groups and glass beads (collectors) with silanol surface group employing various solution with different chemical characteristics. The results have been compared with the surface chemistry theory. The zeta potential of the particle and collector increased with increasing pH up to 5.0 regardless of the solution chemistry. For a monovalent electrolyte solution(sodium chloride solution) the zeta potential steadily increased until the pH reached 9.5. In contrast, little change in zeta potential was made between 5.0 and 9.5 for a divalent electrolyte solution (sodium chloride solution) the zeta potential steadily increased until the pH reached 9.5. In contrast, little change in zeta potential was made between 5.0 and 9.5 for a divalent electrolyte solution (calcium chloride solution). In other words, the more the pH decreases, the larger the effect of neutral salts, such as NaCl and CaCl$_2$, have on the ζ-potential values. In this study, the PZPC(point of zero proton condition) of the particle and collector occurred below a pH of 3.1, H(sup)+ and OH(sup)- acted as a PDI (potential determining ion), and Na(sup)+ acted as an IDI(indifferent ion). The magnitude of the negative ζ-potential values of the particle and collector monotonically increased as the concentrations of Na(sup)+ or Ca(sup)2+([Na(sup)+] or [Ca(sup)2+]) decreased (the values of pNa or pCa increased). In the case of latex particles, the ζ-potential should aproach zero (isoelectric point; IEP) asymptotically as the pNa approaches zero, while in the case of calcium chloride electrolyte, ζ-potential reversal may be expected to occur around 3.16$\times$10(sup)-2MCaCl$_2$(pCa=1.5). pH, valance and ionic strength can be used in various ways to improve the water treatment efficiency by modifying the charge characteristics of the particle and collector. Predictive capability is far less certain when EDL(electrical double layer) repulsive forces exist between the particle and collector.

Citrate Complexes of Alkaline Earth Metals in Aqueous, Acetone-Water and Ethanol-Water Solutions (수용액, 물-아세톤 및 물-에탄올 혼합 용매 내에서 형성되는 알칼리토류금속의 시트르산 착물)

  • Formation of the complexes of alkaline earths with citrate ions in aqueous, acetone-water and ethanol-water solutions was studied at room temperature by the equilibrium exchange technique. This technique involved the measurements of distribution of radioactivity between cation exchange resin and solution phase after the radioactive metal ions were equilibriated with the cation exchange resin in the presence of citrate ions ($Cit^{3-}$) of varying concentrations. The pH of the solutions was controlled to 7.2-7.5, and the ionic strength of the solutions was kept at 0.10-0.12. The present study revealed that both barium and strontium ions formed the one to one citrate complexes, $[M Cit]^-$ in all solvent systems examined. It was also observed that calcium ions formed the one to one citrate complex in aqueous solution. In acetone-water and ethanol-water solutions, however, calcium ions were observed to form both one to one and one to two complexes, $[Ca Cit]^-$ and $[Ca\;Cit_2]^{4-}$. The results of the present study indicated also that relative stabilities of the complexes increased in the order; $Ba^{++}\;<\;Sr^{++}\;<\;Ca^{++}$, and in the order of increasing concentration of the organic components in the mixed solvent systems.

Effects of pH Level and Electrical Conductivity on Growth, Nutrient Absorption, Transpiration and $CO_2$ assimilation of Leaf Lettuce in Hydroponics (잎상추 수경재배에서 근권 pH와 EC가 무기이온흡수, 광합성, 증산량 및 생육에 미치는 영향)

  • This study was conducted to determine the optimum root zone environment condition and proper nutrient management system for lettuce in hydroponics. For the root zone environment condition, several level of pH and electrical conductivity (EC) were treated respectively. Though all the level of pH 4 to pH 8, except pH 3, performed better growth without any visible physiological disorder, the optimum pH of the nutrient solution for lettuce production was pH 5.5 to 6.0. The optimum ionic strength of the solution was EC 1.2 to 1.6 mS $cm^{-1}$ / because higher nutrient level caused tip burn symptom by calcium deficiency. Considering the above results, it is concluded that lettuce can be efficiently mass-produced through the optimum root zone environment.

Ultrafiltration of Soybean Curd Whey for the Separation of Functional Components (유용성 물질의 분리를 위한 두부순물의 한외여과)

  • To separate and concentrate functional components contained in soybean curd whey, ultrafiltration was performed using regenerated cellulose or polysulphone membrane of spiral-wound type with MWCO 10,000, and the permeate and retentate solutions were analyzed. As the pH of soybean curd whey increased, the permeate flux decreased in both membranes. Treatment of 0.01 M EDTA rather decreased the permeate flux compared with control. The concentration of ionic calcium, which decreased with the pH increase, was thought to affect the permeate flux also. In case of polysulphone membrane, the permeate was efficiently purified and the retentate protein was concentrated significantly in which the membrane rejection value (MRV) for chemical oxygen demand (COD) was 79.25% and that of protein was 98.42% at the volume concentration ratio (VCR) of 10. MRV of the protein of regenerated cellulose membrane was lower than that of polysulphone membrane. To recover oligosaccharides to the permeate solution and increase the content of raffinose and stachyose, regenerated cellulose membrane was more suitable than polysulphone membrane and the optimum VCR was 4.

Calcium Influx is Responsible for Afterdepolarizations in Rat Hippocampal Dentate Granule Cells

  • Granule cells in dentate gyrus of hippocampus relay information from entorhinal cortex via perforant fiber to pyramidal cells in CA3 region. Their electrical activities are known to be closely associated with seizure activity as well as memory acquisition. Since action potential is a stereotypic phenomena which is based on all-or-none principle of $Na^+$ current, the neuronal firing pattern is mostly dependent on afterpotentials which follows the stereotypic $Na^+$ spike. Granule cells in dentate gyrus show afterdepolarization (ADP), while interneurons in dentate gyrus have afterhyperpolarizaton. In the present study, we investigated the ionic mechanism of afterdepolarization in hippocampal dentate granule cell. Action potential of dentate granule cells showed afterdepolarization, which was characterized by a sharp notch followed by a depolarizing hump starting at about $-49.04{\pm}1.69\;mV\;(n=43,\;mean{\pm}SD)$ and lasting $3{\sim}7$ ms. Increase of extracellular $Ca^{2+}$ from 2 mM to 10 mM significantly enhanced the ADP both in amplitude and in duration. A $K^+$ channel blocker, 4-aminopyridine (4-AP, 2 mM), enhanced the ADP and often induced burst firings. These effects of 10 mM $Ca^{2+}$ and 4-AP were additive. On the contrary, the ADP was significantly suppressed by removal of external $Ca^{2+},$ even in the presence of 4-AP (2 mM). A $Na^+$ channel blocker, TTX (100 nM), did not affect the ADP. From these results, it is concluded that the extracellular $Ca^{2+}$ influx contributes to the generation of ADP in granule cells.

Improved growth and development in Suaeda glauca through exogenous treatment with indole-3-carboxylic acid

  • Mechanistic studies of halophytes are urgent areas of agricultural research due to the increase in saline-contaminated and irrigated land worldwide. The halophyte Suaeda glauca (S. glauca) has advantages in terms of biomass and saline elimination due to its large mass and well-developed phenotype on seashores, although its mechanistic features and growing specificities still require systematic investigation. In this study, S. glauca was cultivated under various saline concentrations (0-400 mM) in Hoagland's solution in the absence or presence of indole derivatives to elucidate physiological features. The results confirmed the optimal growth and development of S. glauca in 50 mM NaCl, and morphologies such as the number of branches, shoot length, and fresh and dry weights were improved by indole-3-carboxylic acid (ICA) treatment. The cation concentrations in roots, shoots and leaves were investigated to examine the ionic imbalances in response to saline treatment, and the results demonstrated that sodium ions accumulated to high concentrations in leaves. The levels of calcium and potassium ions in roots were maintained or slightly decreased in the presence of 50 mM NaCl and proline concentration was increased significantly in roots at optimal concentrations. These results demonstrate that the concentrations of ions and metabolites are key regulators of optimal growth by regulating the physiology of halophytes.

A geochemical study on the saline waters circulating in an ash disposal pond of Seocheon Power Plant. Korea

  • This study was carried out to understand the geochemistry of saline water circulating in an ash disposal pond of Seocheon power plant, Korea. For this study, ash pond waters, slurry water and seawater samples were collected and analyzed for major ions and trace elements. Results show that ash pond waters and slurry water are alkaline in nature due to high calcium content, and have high concentration of Ca, B, Li, As, Ba, Al, Si and Mn over seawater, suggest that these elements leached from fly ash even at high alkaline condition and ionic strength. Slurry water has high concentration of B, Ba, Li, Mn, Si and Sr compare to ash pond waters, expresses that these elements seem to be easily reached at initial stage fly ash-water interaction, and also might be associated with the surface of the fly ash particles. Additionally, PHREEQC program predicted several secondary solid phases, which are also influenced in the leaching of elements in to the saline water.
