Histopathological Studies on Nephritis Produced in Experimental Tubular Nephrosis (실험적(實驗的) 요세관증(尿細管症)에 있어서 신장염유발(腎臟炎誘發)에 관(關)한 병리조직학적(病理組織學的) 연구(硏究))

  • Normal Albino rats were received glycerin via subcutaneously and Staphylococcus aureus intravenously. The microorganism was coagulase-positive and non-hemolytic. The rats received glycerin alone showed an acute tubular nephrosis, and the others such as glycer in induced nephrotic rats showed a number of different findings: At the first hours of the bacterial injection, in medulla, the bacterial clumps and inflammatory cell infiltration, and microabscesses with retrogressive changes of proximal convoluted tubulles were observed. The suppurative inflammation was observed in days. Five weeks after the initial innoulation of the organism kidney was shown restoration to a histologically normal cortex. The proliferation of fibrous connective tissue and small numbers of chronic inflammatory cells were observed in the medulla where an acute inflammatory process was enhanced presumably. On the other hand, the Albino rats administered Staphyloceccus aureus alone resulted in n moderate degree of vacuolization in proximal convoluted tubules and a number of casts in the early stage. No, bacterial clumps and microabscesses were observed in the rats.

  • Acetylsalicylic acid, administered intravenously in a dose of 120 mg+250 mg/h, markedly decreased the urinary excretion of sodium and chloride, and slightly depressed potassium excretion, so that the ratio of urinary concentrations of potassium to sodium increased after ASA. Osmolar and free water clearances also diminished during water diuresis, and free water reabsorption $(T^cH_2O)$ decreased after ASA during mannitol diuresis. Glomerular filtration rate and urine flow rate changed little. When infused directly into a renal artery, ASA exhibited identical action on both kidneys, indicating that the renotropic action is mediated by some endogenous humoral agents or by some metabolites of ASA. A dose of 100 mg i.v. of spironolactone, a aldosterone antagonist, slightly reversed the renal reflect when given during maximum action of ASA. Ethacrynic acid could display its full diuretic action unhindered during maximum ASA action. Above observations lead to the suggestion that acetylsalicylic acid might release aldosterone and the action on electrolyte excretion may be mediated by the mineralocorticoid.

Studies on Renal Action of Atractylodes Rhizoma Alba in Dogs (백출의 신장작용에 관한 연구)

  • The actions of Atractylodes rhizoma alba on renal function of dog were investigated with water and methanol extract of it, utilizing clearance technique. Water extract elicited diuresis while methanol extract induced antidiuresis in the dog when they were given intravenously. The diuresis induced by the intravenous water extract seemed to be closely related to the decrement of reabsorption of electrolytes in renal tubules. On the contrary, the antidiuresis by intravenous methanol extract appeared to be related to the increment of renal reabsorption of sodium. Water and methanol extract, when infused into a renal artery, exhibited identical resposes to the intravenous actions confined only to the infused kidney, inferring that renal actions of water and methanol extract are not mediated by any endognous humoral extract are not mediated by any endogenous humoral agents. From these observations it is concluded that diuretic action of water extract and antidiuretic action of methanol extract were brought about by direct action in the renal tubules.

Diuretic action of hoelen in the dog (복령의 이뇨작용)

  • Intravenously administered total water extract of Hoelen, (Pachymae fungus) in a dose of 10mg/kg and its methanol lextract, in a smaller dose than total water extract, produced significantly increase on urinary volume, sodium nad potassium excretion, and osmolar nad free water clearances. Increasing the doses produced more pronounced renal responses. Glomerular filtration rate and renal plasma flow changed littl with both extract. On the contrary, water extract from residue obtained on extraction with organic solvents exhibited no significant changes on the parameters of the renal function. In experiments, in which the total water extract was infused directly into a renal artery and urines from both ureters were collected separetely, a small dose of 0.3mg/kg/min showed no diuresis, but a large dose of 1.0mg/kg/min elicited diuretic action even on the contralateral kindneys. It is, therfore, concluded that Hoelen induces diuresis, mainly by inhibiting reabsorption of electrolytes in the renal tubules, and that the renotropic action may be mediated by some endogenous humoral substances.

  • The aim of this study was to investigate the effects of Chungdaesan(CDS) by various administration routes on skin anaphylactic reaction. The most classic and popular skin reaction in vivo is passive cutaneous anaphylaxis(PCA) In this study, therefore, the author investigated the effect of CDS on PCA reaction activated by anti-dinitrophenyl immunoglobulin E antibody The results showed that CDS potently suppressed orally, topically, intraperitoneally, and intradermally administered. However, it did not show suppressive activity when intravenously administered. In addition CDS significantly inhibited anti-DNP IgE induced mast degranulation in mice skin. Moreover, CDS suppressed anaphylactic histamine release from mast cells induced by anti-dinitrophenyl immunoglobulin E antibody. These results indicate that CDS suppresses the PCA reaction by stabilization of mast cells in vivo and in vitro am] also suggest that the differential activity following administration routes may be caused by the difference of bioavailability.

An Influence of Water Extract of Akebiae Lignum on Renal Function of the Dog (개의 신장기능에 미치는 목통 수성 엑기스의 영향)

  • Influence of Akebiae Lignum on the renal function of the dog was observed with water-extract, utilizing clearance technique. Akebiae Lignum water-extract (AWE) given intravenously in does of 3.0mg/kg and 30.0mg/kg elicited a marked antidiuretic effect and produced a distinguished decrease of glomerular filtration rate and renal plasma flow as reduction of sodium and potassium excretion in urine, positive free water clearance and osmolar clearance. AWE infused into a renal artery in doses of 0.03mg/kg/min and 0.1mg/kg/min exhibited identical results to the intravenous action confined only to the infused kidney. These results suggest that AWE elicits antidiuresis in the dog by decreasing glomerulor filtration and renal plasma flow, which are caused by the constriction of vas afferens in the glomeruli.

  • In anesthetized dogs, antidiuretic action of intravenously administered BRL 34915 (10.0~30.0 $\mu$/kg) was blocked by renal denervation, whereas it was not affected by glibenclamide, a selective $K_{ATP}$ blocker, given into renal artery. Diuretic action in ipsilateral kidney produced by intrarenal administration of BRL 34915 was not influenced by renal denervation, but blocked completely by glibenclamide given into the vein. Above results suggest that the antidiuretic action of BRL 34915 is mediated by renal sympathetic nerves and the diuretic action is caused by opening of $K^+$ channel within kidney.

  • Background: The underlying pathogenesis of fat embolism-induced acute lung injury (ALI) has not been elucidated. In the present study, the pathogenesis of fat embolism-induced ALI was probed in association with neutrophilic oxidative stress in oleic acid (OA)-induced ALI of S-D rats. Methods: OA was injected intravenously to provoke ALI in experimental rats. Five hours later, indices of ALI were measured to confirm the role of the neutrophilic respiratory burst. The effect of an inhibition of phospholipase A2 (PLA2) was also evaluated. Results: The accumulation of neutrophils in the lung due to OA caused increased neutrophilic oxidative stress in lung, which was ameliorated by mepacrine. What were the results from inhibition of PLA2. Conclusion: Excess neutrophilic oxidative stress contributes to OA-induced ALI, which is lessened by the inhibition of PLA2.

Protective Effect of Licorice Water Extract against Cadmium-induced Nephro-toxicity in Rats

  • Licorice has been used for cure of injuries and for detoxification in East Asia. This study investigated the protective effect of licorice water extract against cadmium (CdCl$_2$, Cd)-induced nephro-toxicity in rats. To induce acute toxicity, Cd (4 mg/kg body weight) was dissolved in normal saline and then, intravenously (i.v.) injected to animals. In experiments, animals were orally administrated with vehicle or licorice water extract (50-100 mg/kg) for 3 days, exposed to a single injection of Cd after 24 h the last licorice/vehicle treatment. Licorice protected kidney injuries by Cd treatment. The number of glomeruli showing vasodilatation and thickening of Bowman's capsule was dose-dependently decreased by licorice. These results suggest that licorice might be a potent preventive protector against Cd-induced nephro-toxicity in rats.

General Pharmacology of the Active Ingredients of New Antibiotic Bead (CJ-40003) (골수염 치료제인 항생제비드(CJ-40003) 유효성분의 일반약리작용)

  • A new antibiotic bead, CJ-40003 is a combination of three antibiotics, tobramycin, vancomycin and cefazolin embedded in bone cement, for the treatment of osteomyelitis. To evaluate the general pharmacological properties of CJ-40003, the effects of its active ingredients were investigated in mice, rats, dogs and isolated guinea pig ileum. The combination of three antibiotics (CA) did not affect general behavior, central nervous system, smooth muscles, gastrointestinal system, cardiovascular and respiratory system and water and electrolytes excretion when administered intravenously at the doses of 0.3, 1 and 3 mg/kg, respectively, into experimental animals. The CA had no effect on the contractile response of the isolated guinea pig ileum to various spasmogen at concentrations of 1, 3 and 10 $\mu\textrm{g}$/ml, respectively. In conclusion, the active ingredients of CJ-40003 showed no pharmacological effect in these studies.
