Preliminary search of intraspecific chloroplast DNA variation of nine evergreen broad leaved plants in East Asia

  • In order to acquire information on chloroplast DNA markers to evaluate the genetic diversity of evergreen broad leaved plants, we investigated the intraspecific variation of cpDNA in eight non-coding regions of nine species commonly distributed in East Asia. Although no variations were detected in psbA-trnH, rpoB-trnC, rpl16 and atpB-rbcL regions, a relatively large amount of intraspecific variations was detected in the psbC-trnS, rps16 and trnL-F regions. These results suggested that these three cpDNA markers are suitable to assess genetic diversity of the species investigated in this study. In contrast, intraspecific variations were detected in seven taxa except Hedera rhombea and Neolitsea aciculata. Neolitsea sericea and the taxa of Quercus had many polymorphic sites.

Differentiation of Intraspecific Groups within isolates of Rhizoctonia solani Using PCR-RFLP of Ribosomal DNA (Ribosomal DNA의 PCR-RFLP에 의한 국내산 Rhizoctonia solani 균주들의 종내그룹의 구분)

  • Genetic diversity among 27 isolates of Rhizoctonia solani, which were obtained from diseased crops in Korea and classified into 9 intraspecific groups by anastomosis test and cultural characteristics, was studied by PCR-RFLP. Gene regions of nuclear 17S ribosomal DNA and internal transcribed spacers including 5.8S rDNA of the isolates were amplified with polymerase chain reaction and digested with 12 restriction enzymes. Differences of restriction patterns were not shown among isolates within each intraspecific groups, however, each anastomosis group and culturala type sowed unique restriction fragment length polymorphisms by restriction patterns using HaeIII, Cfr13I and MspI. The results suggest that PCR-FRLP of rDNA using three restriction enzymes could be used to differentiate intraspecific groups of Rhizoctonia solani in Korea.

Characterization of fusants between thermophilic clostridium thermocellum and clostridium thermohydrosulfuricum (고온성 clostridium thermocellum과 clostridium thermohydrosulfuricum의 융합체의 특성)

  • Intraspecific and interspecific protoplast fusions in/between C. thermocellum and C. thermohydrosulfuricum were studied. Protoplast fusions were well induced in 30-40% PEG solution, however, their fusion frequencies were low level of 1.2*10$^{-7}$ for intraspecific fusion of C. thermocellum, $6.7*10^{-7}$ for C. thermohydrosulfuricum, and 4.2*10$^{-7}$ for interspecific fusion between above two Clostridia, respectively. Most fusants were unstable and segregated after 3 subcultures. Relatively stable intraspecific C. thermocellum fusant FTT17, intraspecific C. thermohydrosulfurecum fusant FSS22 and interspecific fusant FTS3, which were stable after several subcultures, were selected and properties of fusants were further investigated, Phenotypes of the fusants were similar with wild types mostly in cellular morphology, carbon source assimilation and enzyme activities. However they were differed in assimilation of pyruvic acid and sorbitol as carbon source. The DNA contents of fusants were slightly increased compared with wild types. Ethanol production by intraspecific and/or interspecific fusants was not increased, however, acetic acid production as byproduct was decreased or not detected, which indicates that industrial thermophilic anaerobes can be improved by means of protoplast fusion of two strains.

Interspecific and Intraspecific Variation in Pollen Grains of Quercus Subgenus Lepidobalanus Endl. in Korea

  • Palynological characters such as pollen grain polar axis length (PL), pollen equatorial diameter (ED), colpus length, colpus width, and P/E ratio of six species of Quercus subgenus Lepidobalanus Endl. from Korea were studied. A significant interspecific variation, unequal distances between the species, and various degree of intraspecific variations were found. The taxonomic value of the pollen morphology parameters measured was found to vary according to species. These results suggest a possible relationship between parameters measured and ploidy level of the Quercus species studied.

Intraspecific Variation in the Temperature Niche Component of the Diatom Skeletonema costatum from Korean Coastal Waters

  • Final biomass yields (peak optical density) and growth rates (divisions/day) of seven clones of Skeletonema costatum from Korean coastal waters were measured to understand their intraspecific variations in the light intensity niche component under $25^{\circ}C$ condition. Daily growth rates of 6 of 7 S. costatum, clones were maximum at 6000 lux while that of YS4, a neritic clone, was maximum at 9000 lux. The final biomass yields of 4 of the 7 S. costatum clones were maximum at the lowest light intensity of 2000 lux. Minimum final biomass yields were found at 9000 lux in all the S. costatum clones other than an estuarine clone, HDC9. The intraspecific variations of the mean growth rate and mean final biomass yield under each of the three different light intensity in terms of the coefficient of variation were not greater than 10% in any of the 7 S. costatum clones.

Growth Performance of Offspring from Selected Korean, Cultured Japanese and Their Reciprocal Intraspecific Hybrids of Red Sea Bream, Pagrus major

  • Intraspecific hybrids have been proven to be a useful tool for increase growth rate in some freshwater fish species. However the effects of intraspecific hybrids in seawater fish species is little known. To assess whether intraspecific hybrids in red sea bream was associated with growth enhancement, growth performances of offspring from two inbred lines, selected Korean line (KORDI-F4) and cultured Japanese line (IPN) and two intraspecific hybrids lines (KORDI-F4♀×JPN♂ and JPN♀×KORDI-F4♂) of red sea bream were compared. There's no significant difference in body weight between offspring groups during seed production period (until 4 months old). At seven months old (after three months rearing trials on sea cages communally), the offspring from KORDI-F4 (41.0±11.1g) and JPN♀×KORDI-F4♂ (39.3±8.1g) showed significantly better performance in body weight than that of the offspring from KORDI-F4♀×JPN♂ (37.0±8.1g) and IPN (35.7±9.7g). At 15 months old, the offspring from JPN♀×KORDI-F4♂ showed best growth performances in body weight, weight gain, specific growth rate among four offspring groups. As the results, the intraspecific hybrids between cultured Japanese line and selected Korean line showed superior growth performances than their paternal and maternal inbred lines.

Three Intraspecific groups in Korean Isolates of Phytophthora drechsleri Based on PCR-RFLP of Ribosomal DNA (Ribosomal DNA의 PCR-RFLP에 의한 국내산 Phytophthora drechsleri의 3가지 종내그룹)

  • Intraspecific genetic diversity of Korean isolates of Phytophthora drechsleri was investigated based on PCR-RFLP of rDNA along with closely related species in the genus; P. cryptogea, P. melonis, P. erythroseptica, P. cinnamomi, P. cambivora and P. cactorum. Gene regions of nuclear small subunit and internal transcribed spacer (ITS) in rDNA were amplified with polymerase chain reaction and digested with 9 restriction enzymes. Phytophthora species was readily differentiated from each other based on the digestion patterns, however, P. cryptogea was not separable from some isolates of P. drechsleri. Twenty one isolates of P. drechsleri originated from 15 host plants were divided into three distinct groups designated as PdG1, PdG2 and PdG3, respectively. Four isolates in PdG1 were originated from green vegetables and tomato and nine isolates in PdG2 were mainly isolated from medicinal plants. The two groups showed 95.3% homology and four isolates of P. cyptogea came under the groups. However, Eight isolates in PdG3 collected from cucurbits were clearly differentiated from those of PdG1 and PdG2 by 66.5% homology, but completely matched with a Taiwan isolate of P. melonis. Results indicated that three distinct groups exist in Korean isolates of P. drechleri and each group has host preference. In addition, reclassification of the cucurbits isolates are reserved because of their distinct genetic characters from other intraspecific groups in P. drechsleri.

Intraspecific variations of the Yam (Dioscorea alata L.) based on external morphology and DNA marker analysis

  • Intraspecific genetic relationship of 19 variation types of the Yam (Dioscorea alata) classified by their external morphological characteristics such as leaf and tuber shape were assessed by DNA using random and specific primer. Twenty two out of 113 primers (100 random[10-mer] primers, two 15 mer [M13 core sequence, and (GGAT)$_4$ sequence]) had been used in PCR-amplification. Only 12 primers, however, were success in DNA amplification in all of the analyzed plants, resulting in 93 randomly and specifically amplified DNA fragments. The analyzed taxa showed very high polymorphisms(69 bands, 71.0 %), allowing individual taxon to be identified based on DNA fingerprinting. Monomorphic bands among total amplified DNA bands of each primer was low under the 50%. Similarity indices between accessions were computed from PCR(polymerase chain reaction) data, and genetic relationships among intraspecific variations were closely related at the levels ranging from 0.66 to 0.90. These DNA data were not matched well with those of morphological characters since they were divided into two major groups at the similarity coefficient value of 0.70. Therefore, Grouping of species into variation types by mainly morphological charactistics was suggested unreasonable.

Phylogeography of the economic seaweeds Chondrus (Gigartinales, Rhodophyta) in the northwest Pacific based on rbcL and COI-5P genes

  • The red algal genus Chondrus have long been used as raw materials for carrageenan and dietary fiber in health foods. Despite the importance of genetic information in safeguarding natural seaweed resources, knowledge of the population genetics of Chondrus in the northwest Pacific is limited. In this study, genetic diversity and phylogeographic structure of 45 populations (777 specimens) of Chondrus from Korea, China, and Japan were evaluated based on mitochondrial COI-5P gene sequences, and phylogenetic relationships were confirmed based on plastid rbcL gene sequences. Molecular analyses assigned the specimens in this study to three Chondrus species: C. nipponicus, C. ocellatus, and C. giganteus; phenotype-based species classification was impossible owing to their high morphological plasticity. We found moderate intraspecific genetic diversity and a shallow phylogeographic structure in both for C. nipponicus and C. ocellatus, and low intraspecific genetic diversity in C. giganteus. Each of the three species exhibited high-level intraspecific gene flow among regions based on the most common haplotypes (CN1 for C. nipponicus, CO1 for C. ocellatus, and CG1 for C. giganteus). Our comprehensive genetic information provides insights into the phylogeographic patterns and intraspecific diversity of the economically important Chondrus species. It also highlights the need to conserve existing natural Chondrus resources through continuous monitoring of genetic diversity and phylogeographic pattern.

Cloning and Intraspecific Variation of β-Actin 2 Gene from the Hermaphroditic Fish Rivulus marmoratus (Cyprinodontiformes, Rivulidae) (암수동체 어류 점박이송사리 Rivulus marmoratus (Cyprinodontiformes, Rivulidae) β-Actin 2 유전자의 클로닝 및 종내 변이)

  • The full sequence analysis of 16 different $\beta-actin$ genes isolated from a single Rivulus marmoratus was performed. The numbers of isolated $\beta-actin$ genes varied from 1764 to 1769. They showed different amino acid residues at the exon 1 region. We named this new gene R. marmoratus $\beta-actin$ 2 gene. Intraspecific variation of R. marmoratus $\beta-actin$ 2 gene was also examined. Major differences of 16 isolated $\beta-actin$ genes, compared to already reported R. marmoratus $\beta-actin$ gene (GenBank AF168615), were observed in both deduced amino acid sequences (1~2%) and intron 2 sequences (4~5%). In this paper, we confirmed the intraspecific variation of $\beta-actin$ gene in this species.
