This investigation has been carried out to examine the structure of digestive tract from Korean Leech, Erpobdella lineata, using light and electron microscope. The digestive tract is composed of mouth, pharynx, Oesophagus, six-chambered stomach, three-chambered intestine, rectum and anus. Stomach and intestine have not gastric or intestine ceca and consist of only straight tube. All digestive tracts from pharynx to rectum are covered with simple columnar epithelial cells. While the surfaces of endothelial cell of pharynx and rectum are covered with cuticular layer of about $0.3{\mu}m$ in thickness, stomach and intestine are covered with estimated $0.2-0.3{\mu}m$ and $0.5{\mu}m$ microvilli respectively. Circular folds were found only in first and second chambers of stomach, intestine and rectum, but not in pharynx and the other chambers (third to sixth) of stomach. The granules of $0.3-0.8{\mu}m$ and $0.5-1.0{\mu}m$ in diameter were observed in the cytoplasm of stomach endothelial cell. These granules were demonstrated to contain protein which showed a positive reaction to ninhydrin. It was also found that there are well-developed microvilli in the apical portion of intestine endothelial cell in which endocytosis occurs actively.
C. perfringens is the most important enteric clostridial pathogen of animals. C. perfringens type A has been associated with hemorrhagic enteritis in a wide lange of domestic and wild mammals. But all types of C. perfringens can be normal inhabitants of the intestine of most mammals. We have a special case that showed C. perfringens type A infection in a hippopotamus (Hippopotamus amphibius) cub at Seoul Grand Park Zoo. Male, hippopotamus cub died in 3 days after birth. Clinical features of the hippopotamus cub have showed lethargy and anorexia before death. Gross post-mortem findings of the hippopotamus were hemorrhagic enteritis of intestine. Histopathologically, ruminant stomach and intestine showed hemorrhagic lesions and the lumen of the small intestine was filled with mucoid and hemorrhagic fluid. Also, intestine and stomach of hippopotamus were distended with gas and hemorrhagic fluid. C. perfringens was isolated in culture of small intestine and the presence of C. perfringens type A was confirmed by PCR. This case indicated that C. perfringens type A could be considered as a virulence factor responsible for causing death of a newborn hippopotamus.
In this study, we investigated the characteristics of CCK-producing cells and mucus-secreting goblet cells with respect to stomach fish and stomachless fish of the Gobiidae in order to provide a basis for understanding the digestive physiology. Hairychin goby (Sagamia geneionema), which is stomachless fish, the numbers of mucus-secreting goblet cells is highest in the posterior intestine portion (P<0.05), while CCK-producing cells are scattered throughout the intestine. Gluttonous goby (Chasmichthys gulosus), which is stomach fish, mucus-secreting goblet cells are most abundant in the mid intestine portion (P<0.05), whereas CCK-producing cells are observed only in the anterior and mid intestine portion. Trident goby (Tridentiger obscurus) which is stomach fish, mucus-secreting goblet cells were most abundant in the mid intestine portion (P<0.05). CCK-producing cells are found in the anterior and mid intestine portion. Giurine goby, Rhinogobius giurinus which is also stomach fish, the largest number of mucus-secreting goblet cells showed in anterior intestine portion except for esophagus (P<0.05). CCK-producing cells are present only in the anterior and mid intestine portion. In S. geneionema, digestive action occurs in the posterior intestine portion to protect and functions to activate digestion. In contrast, in C. gulosus, T. obscurus and R. giurinus, their digestive action occurs in the anterior and mid intestine portion to protect and functions to activate digestion. Further studies of the modes of food ingestion by these fish, the contents of their digestive tracts, and the staining characteristics of the goblet cells need to be carried out.
Objectives : This study was designed to investigate effects of electro-acupuncture's stimulated frequency, intensity, duration at 5736 on tile Small Intestine Motility in Rats. Methods The motor activity of small intestine in rats was evaluated by intestinal transportation rate. Changes in tile motility of ileum in vivo was measured at 10 minutes after electro-acupuncture. Various kinds of stimulus-frequency were used in this experiment: 2 Hz, 50 Hz, 100 Hz, 2 Hz-4 trains, 8 Hz groups treated with EA of the condition in S mA, 1 ms pulse duration, 30 minutes stimulated duration. Three different stimulus-intensity were used: 1, 5, 10 mA groups treated with EA of the condition in 2 Hz, 1 ms pulse duration, 30 minutes stimulated duration. Six different stimulus-duration were used: 10, 20, 30, 40, 50, 60 minutes groups treated with EA of the condition in 2 Hz, 5 mA, 1 ms pulse duration. Results : 2 and 100 Hz groups significantly increased the small intestine motility, but 50 Hz group did not induce a significant change. Besides, small intestine motility was significantly increased only in 5 mA intensity (5 times twitch). futhermore, at least 20 minutes EA treatment was necessary to increase the small intestine motility. Conclusion : Those basic data form this study can be applied to established the effective treatment of EA for gastrointestinal diseases in the clinical field.
This experiment was conducted to examine the effects of different carbohydrate and nitrogen source upon the utilization of amino acids in the small intestine of sheep. The results obtained are as follows: 1) For the quantities of total amino acid-N(TAA-N), essential amino acid-N(EAA-N) and nonessential amino acid-N(NEAA-N) passing at the duodenum and ileum except NEAA-N passing at the ileum were no significant differences (p>0.05) between diets. The quantities of NEAA-N passing at the ileum for the diets containing meat and bone meal were significantly higher (p>0.05) than the diets containing soybean meal. The mean values for the proportionate disappearance apparently digested TAA-N, EAA-N and NEAA-N within the small intestine for four diets were $0.692{\pm}0.0449$, $0.702{\pm}0.0132$ and $0.682{\pm}0.726$, respectively. 2) There were no significant differences (p>0.05) in duodenal individual amino acid flow between diets with the exception of aspartic acid and glycine. The amounts of each amino acid in duodenal digesta, expressed as a proportion of the amounts ingested for the four diets, were shown that there were net gains of EAA with the exception of arginine and NEAA with the exception of glutamic acid, glycine and praline prior to the small intestine. 3) Within the small intestine, there were no significant losses of each EAA (p>0.05) but significant losses of aspartic acid and glycine of NEAA between diets (p<0.01). The mean values of the proportionate losses of methionine, alanine and lysine within the small intestine were $0.816{\pm}0.04$, $0.767{\pm}0.04$ and $0.732{\pm}0.01$, respectively.
The present study was undertaken to determine whether combined treatment with prokinetic trimebutine and mosapride has a synergic effect on gastrointestinal motility and visceral pain associated with gastrointestinal dysfunction. To develop effective gastroprokinetic agents with greater potencies than trimebutine or mosapride for the treatment of gastrointestinal tract disease, a mixture of trimebutine and mosapride was designed and prepared. In the present study, treatment with trimebutine alone showed a dose-dependent effect on propelling movements of normal small and large intestine in mice, whereas mosapride effected only small intestine motility. Co-administration of trimebutine with mosapride, a well-established prokinetic drug, produced a synergistic influence on normal small intestine motility, but demonstrated an unclear effect on large intestine motility, with a slight tendency to reduce the propelling time. In a stress model, the small and large intestine motilities were significantly decreased. The reduction of intestine motility was restored to a normal level and the restoring effect was more pronounced in the combined treatment with trimebutine plus mosapride than treatment with trimebutine or mosapride alone. Furthermore, treatment with trimebutine plus mosapride significantly decreased acute visceral pain which was not controlled by trimebutine or mosapride alone. These data suggest that combination therapy with trimebutine plus mosapride has a synergic effect on small and large intestine motility and visceral pain control in gastrointestinal disorders.
To validate the comparision of proximal and distal large intestinal motility, the amplitude and frequency of spontaneous motility, the effect of acetylcholine, the effect of atropine on the response of acetylcholine, the effect of histamine and the effect of pyrilamine and cimetidine on the response of histamine were investigated in rabbit. The results were summarized as follows: 1. The amplitude of spontaneous motility was more powerful on the proximal large intestine than that of the distal large intestine, but the frequency of spontaneous motility was similar on the both proximal and distal large intestine in rabbit. 2. Acetylcholine caused the contraction of proximal and distal large intestine, and the contractile response were increased between the concentration of acetylcholne $10^{-9}$ and $5{\times}10^{-6}M$ and $10^{-7}$ and $10^{-4}M$ on the proximal and distal large intestine, respectively, with dose-dependent manner in rabbit. 3. The contractile response induced by acetylcholine was completely blocked by the post-treatment with cholinergic receptor blocker, atropine $10^{-6}M$. 4. Histamine caused the contraction of proximal and distal large intestine and the contractile response were increased between the concentration of histamine $10^{-9}$ and $5{\times}10^{-5}M$ and $10^{-5}$ and $10^{-3}M$ on the proximal and distal large intestine, respectively, with dose-depend ent manner in rabbit. 5. The contractile response induced by histamine was completely blocked by the pretreatment with $H_1$-receptor blocker, pyrilamine $10^{-6}M$, but not blocked by the pretreatment with $H_2$-receptor blocker, cimetidine $10^{-6}M$.
The turnover time of the mucosal epithelium in the small intestine(jejunum and ilium) and large interstine(cecum), and the cells in the lamina propria of the small intestine was investigated with the radioautography in mice at various times after single injection of $^3H$-thymidine. Twenty suckling mice were sacrified at each of the following time intervals after injection ; 2 hrs, 1, 3, 5. 7, 14 and 17 days. 1. The labeled index of the epithelial cells in the crypt and the villus of the small intestine averaged 98.7% and 1.3% at 2 hrs, 982% and 1.8% at 1 day, 18.7% and 81.3% at 3 days, 6.3% and 93.7% at 5 days, respectively. The labeled index of the epithelial cells of the crypt-base, the upper-crypt and the mucosal surface in the large intestine averaged 71.8%, 28.2% and 0% at 2 hrs, 45%. 54.2% and 0% at 1 day, 17.2%, 54.5% and 28.2% at 3 days, 10.2%, 32.4% and 57.4% at 5 days, respectively. This result suggested that the turnover time of all the epithelial cells migrating from crypts to villi in the direction of the villus tips was calculated to be less than 5 days, and also the longest turnover time was calculated to be no longer than 7 day. 2. The labeled index of the total cells in the lamina propria of the small intestine averaged 6.2-7% at 2 hrs to 5 days, 4.7% at 7 days 2.6% at 17 days and this index is tend to be decreased moderately at 7 days and severely at 17 days. So this result suggested that the turnover time of the cells with the shorter cycle duration in the lamina propria of the small intestine were less than 5 days and that of the cells with the longer cycle duration more than 17 days.
A liquid chromatographic method with tandom spectrometric detection (LC/MS/MS) for the simultaneous determination of doxifluridine and its active metabolite, 5-fluorouracil (5-FU) was developed over the concentration range of $5{\sim}2000$ ng/ml, respectively. Doxifluridine, 5-FU and internal standard, 5-chlorouracil (5-CU), were extracted from liver and intestine tissue via protein precipitation. Acetonitrile was used as the extraction solvent and the supernatant was evaporated and reconstructed in mobile phase. Optimum chromatographic separation was achieved on a Agilent Zorbax $C_{18}$ ($100\;mm{\times}2.1\;mm$, $3.5\;{\mu}m$) column with mobile phase run in isocratic with methanol : water (20 : 80, v/v). The flow rate was 0.2 ml/min with total cycle time of 5 min. The lower limit of quantification was validated at 5.0 ng/ml of liver and intestine tissue, for both doxifluridine and 5-FU, respectively. The intra-day and inter-day precision and accuracy of quality control (QC) samples were <11% coefficient of variation and <7% relative error from theoretical concentration for both analytes. In addition, the special designed stability study was performed, because the metabolism of doxifluridine occurs spontaneously even in ice bath for monkey liver. The stability of doxifluridine in liver and intestine of monkey and beagle dog was compared. It was found that bioanalytical validation could not be performed for the monkey liver; however, beagle dog's liver has relatively low speed of metabolism compared to monkey liver and instead of monkey liver, beagle dog's liver could be used for the validation. Bioanalytical validation could be performed in monkey intestine. Eventually, this developed method for liver and intestine will be useful in support of the toxicokinetic and pharmacokinetic studies of doxifluridine and 5-FU.
