To produce an intestinal immunomodulatory beverage containing Centella asiatica extract (CAE), three types of CAE-added beverage prototypes were prepared, and their immunomodulatory activities and marker compounds were analyzed. As a result of the cytotoxicity assessment, all the beverages did not show significant toxicity compared to the control group. Next, the immunomodulatory activities of the beverage prototype were evaluated using the inflammatory model of IL-1β-induced intestinal epithelial cell line. All the samples significantly reduced the production of IL-6, IL-8, and MCP-1 in a CAE concentration-dependent manner. In addition, CAE-added beverages inhibited NO, IL-6, and IL-12 production in LPS-induced RAW 264.7 cells. When the major triterpenoids, as marker compounds for the production of CAE-added beverages, were analyzed by HPLC-DAD, only asiaticoside was detected beyond the limit of quantification, while madecassoside, madecassic acid, and asiatic acid were not detected. The amounts of asiaticoside in CAE-added beverage prototypes were confirmed in No. 1 (19.39 ㎍/mL), 2 (19.25 ㎍/mL), and 3 (19.98 ㎍/mL). In conclusion, the results of this study suggested that CAE-added beverage prototypes induced immunomodulatory effects in the intestinal inflammatory cell line models and asiaticoside could be used as a marker compound for CAE-added beverage production.
Interferon (IFN)-λ plays an essential role in mucosal cells which exhibit strong antiviral activity. Lactobacillus plantarum (L. plantarum) has substantial application potential in the food and medical industries because of its probiotic properties. Alphacoronaviruses, especially porcine epidemic diarrhea virus (PEDV) and transmissible gastroenteritis virus (TGEV), cause high morbidity and mortality in piglets resulting in economic loss. Co-infection by these two viruses is becoming increasingly frequent. Therefore, it is particularly important to develop a new drug to prevent diarrhea infected with mixed viruses in piglets. In this study, we first constructed an anchored expression vector with CWA (C-terminal cell wall anchor) on L. plantarum. Second, we constructed two recombinant L. plantarum strains that anchored IFN-λ3 via pgsA (N-terminal transmembrane anchor) and CWA. Third, we demonstrated that both recombinant strains possess strong antiviral effects against coronavirus infection in the intestinal porcine epithelial cell line J2 (IPEC-J2). However, recombinant L. plantarum with the CWA anchor exhibited a more powerful antiviral effect than recombinant L. plantarum with pgsA. Consistent with this finding, Lb.plantarum-pSIP-409-IFN-λ3-CWA enhanced the expression levels of IFN-stimulated genes (ISGs) (ISG15, OASL, and Mx1) in IPEC-J2 cells more than did recombinant Lb.plantarum-pSIP-409-pgsA'-IFN-λ3. Our study verifies that recombinant L. plantarum inhibits PEDV and TGEV infection in IPEC-J2 cells, which may offer great potential for use as a novel oral antiviral agent in therapeutic applications for combating porcine epidemic diarrhea and transmissible gastroenteritis. This study is the first to show that recombinant L. plantarum suppresses PEDV and TGEV infection of IPEC-J2 cells.
정상인에서 장내세균은 숙주의 면역이나 영양 흡수를 돕지만, 때로는 기회감염균으로서 그들을 위협하기도 한다. 그 중 절대 혐기성 세균인 Bacteroides fragilis는 분비되는 장독소(enterotoxin)인 Bacteroides fragilis toxin (BFT)의 유무에 따라 non-enterotoxigenic B. fragilis (NTBF)와 enterotoxigenic B. fragilis (ETBF)로 나뉜다. ETBF는 가축 및 사람에서 설사 질환 및 대장 질환을 유발한다 그러나 때때로 ETBF를 가지고 있으나 증상이 없는 사람도 존재한다. ETBF는 염증성 설사 질환, 여행자 설사 환자의 대변에서 검출되어 주목 받고 있다. 또한, 몇몇 연구를 통해 inflammatory bowel disease (IBD)나 대장염 및 대장암 환자에서 ETBF가 증가한다는 것이 밝혀졌다. 일반 C57BL/6 마우스 및 germ-free 마우스, multiple intestinal neoplasia (Min) 마우스, 토끼, Mongolian gerbil 등 여러 동물 모델에서 ETBF가 IBD나 대장염, 대장암을 유발 또는 촉진한다는 것이 발표되었다. ETBF의 유일한 병원성 인자인 BFT는 E-cadherin의 분절을 유도하여 장상피 세포의 투과성을 높인다. 이어서 ${\beta}$-catenin 신호전달계가 활성화하여 장상피세포의 증식이 증가한다. 또한 ETBF의 감염은 일반 마우스에서 급성이나 만성의 대장염을 일으키고 Min 마우스에서 종양 형성을 촉진한다. 이는 Stat3에 의존한 $T_H17$ 면역반응의 활성화를 통해 일어난다. 현재 ETBF의 검출 방법에는 크게 BFT toxin assay와 몇 가지 PCR 방법이 있다. 최근 real-time PCR과 같은 분자진단학적 기법의 발달로 일반적인 PCR보다 더 정확한 ETBF의 검출이 가능하게 되었다. 이것을 이용하여 앞으로 실제 임상에서 ETBF와 대장염 및 대장암의 발달 관계에 대한 심도 깊은 연구가 이뤄질 것으로 본다.
Pathogenic Salmonella typhimurium can invade the intestinal epithelium and cause a wide range of diseases including gastroenteritis and bacteremia in human and animals. To identify the genes involved in the infection, the invasion determinant was obtained from S. typhimurium 82/6915 and was subcloned into pGEM-7Z. A subclone DHl (pSV6235) invaded HEp-2 and Chinese hamster ovary epithelial cells and contained a 4.4 kb fragment of S. typhimurium genomic region. Compared with the host strain E. coli DHl, the subclone DHl (pSV6235) invaded cultured HEp-2 and Chinese hamster ovary cells at least 75- and 68-fold higher, respectively. The invasion rate of E. coli DHl for the cells significantly increased by harbouring the genomic region derived from pathogenic S. typhimurium 82/6915.
동면동물인 다람쥐를 활동기와 동면기로 나누어 소장 점막 상피세포인 원주세포와 점액세포의 미세구조 변화를 투과전자현미경을 이용하여 관찰하였다. 활동기의 원주세포에서는 많은수의 미토콘드리아와 과립형질내세망을 관찰할 수 있었다. 동면기의 원주세포에서는 많은수의 리보소옴을 관찰할 수 있었다. 활동기의 점액세포에서는 크고 많은수의 점액과립과 세포소기관으로 미토콘드리아와 과립형질내세망을 관찰할 수 있었으며 현저한 분비작용을 관찰할 수 있었다. 동면기의 점액세포에서는 수와 크기에서 감소된 점액과립과 활동기보다 증가된 리보소옴을 관찰할 수 있었으며 분비작용은 활동기에 비해 감소하였다.
Tumor metastasis involves circulating and tumor-initiating capacities of metastatic cancer cells. Hepatic TM4SF5 promotes EMT for malignant growth and migration. Hepatocellular carcinoma (HCC) biomarkers remain unexplored for metastatic potential throughout metastasis. Here, novel TM4SF5/CD44 interaction-mediated self-renewal and circulating tumor cell (CTC) capacities were mechanistically explored. TM4SF5-dependent sphere growth was correlated with $CD133^+$, $CD24^-$, ALDH activity, and a physical association between CD44 and TM4SF5. The TM4SF5/CD44 interaction activated c-Src/STAT3/ Twist1/ B mi1 signaling for spheroid formation, while disturbing the interaction, expression, or activity of any component in this signaling pathway inhibited spheroid formation. In serial xenografts of less than 5,000 cells/injection, TM4SF5-positive tumors exhibited locally-increased CD44 expression, suggesting tumor cell differentiation. TM4SF5-positive cells were identified circulating in blood 4 to 6 weeks after orthotopic liver-injection. Anti-TM4SF reagents blocked their metastasis to distal intestinal organs. Altogether, our results provide evidence that TM4SF5 promotes self-renewal and CTC properties supported by $CD133^+/TM4SF5^+/CD44^+^{(TM4SF5-bound)}/ALDH^+/CD24^-$ markers during HCC metastasis.
Interactions between microbes and epithelial cells in the gastrointestinal tract are closely associated with regulation of intestinal mucosal immune responses. Recent studies have highlighted the modulation of mucosal immunity by microbe-derived molecules such as ATP and short-chain fatty acids. In this study, we undertook to characterize the expression of the ATP-gated $P2X_7$ receptor ($P2X_7R$) on M cells and its role in gastrointestinal mucosal immune regulation because it was poorly characterized in Peyer's patches, although purinergic signaling via $P2X_7R$ and luminal ATP have been considered to play an important role in the gastrointestinal tract. Here, we present the first report on the expression of $P2X_7R$ on M cells and characterize the role of $P2X_7R$ in immune enhancement by ATP or LL-37.
Capsaicin is the active ingredient in chili pepper and has an inhibitory effect on Helicobacter pylori growth and $NF-{\kappa}B$ activation. The present study examined the effect of capsaicin on interleukin (IL)-8 production by H. pylori ATCC 43504-infected MKN-45 cells, a gastric epithelial cell line. The viability of the MKN-45 cells treated with capsaicin at 0, 50, 100, 250, and $500\;{\mu}M$ was 99, 98, 99, 99, and 85%, respectively. A capsaicin concentration as low as $50\;{\mu}M$ significantly inhibited the IL-8 production induced by H. pylori ATCC 43504 infection (43.2% of control) during 24 h of incubation. However, low concentrations of capsaicin $(50\;and\;100{\mu}M)$ did not significantly inhibit the IL-8 production by $TNF-{\alpha}-$ or PMA-treated MKN-45 cells. Therefore, the overall inhibitory effect of capsaicin on H. pylori ATCC 43504 was the sum of H. pylori ATCC 43504 growth inhibition, host cell survival, and $NF-{\kappa}B$ signal cascade inhibition.
목 적 : 경구 내독소와 저산소 자극을 이용하여 신생쥐에서 실험적 괴사성 장염(necrotizing enterocolitis, NEC)을 유발하고 그 병태생리에서 세포자멸사의 역할을 알아본다. 방 법:신생쥐에 경구로 내독소(5 mg/kg)를 투여하고 8% 저산소 자극을 주어 NEC를 유발하고 성공적으로 NEC가 유발된 군에 caspase 억제제를 전처치하고 비교하였다. 장 조직의 병리학적 분석은 hematoxylin-eosin 염색한 슬라이드로 하였고 세포자멸사는 TUNEL 염색과 장 조직 caspase-3 활성으로 분석하였다. IEC-6세포주에 내독소와 저산소를 처리한 후 세포자멸사와 Bax, Bcl-2, Fas, FasL의 발현을 분석하였다. 결 과:경구 내독소(5 mg/kg)와 반복적으로 60분간 2회 투여된 8% 저산소 자극에 의해서 NEC와 유사한 장병변이 신생쥐에서 유발되었다. 장 조직은 세포자멸사와 caspase-3 활성의 증가를 보여주었다. Caspase 억제제 전처치에 의해서 세포자멸사와 NEC의 중증도가 모두 감소하였다. IEC-6 세포주도 내독소와 저산소 자극에 의해서 세포자멸사와 caspase-3 활성의 증가를 보여주었으며 Bax/Bcl-2 ratio가 유의하게 증가하였다. 결 론:경구 내독소와 저산소를 이용한 본 NEC 신생쥐 모델은 caspase-3 활성에 의해 매개되는 장 상피세포의 세포자멸사가 병태생리에 관여함을 보였다. 본 동물모델은 임상에서 흔히 접할 수 있는 자극을 적용하였기 때문에 그 병태생리와 치료적 시도의 연구에 더욱 유용하리라고 본다.
목적:반응성산소기들은 장관에서 여러 종류의 질병의 발생과 관련을 가지고 있는 것으로 알려져 있어, 이들에 의한 세포손상을 방지하는 약물의 개발은 시급한 실정이다. 본 연구에서는 항산화작용을 가진 약재로 보고 된 단삼추출액이 장관상피세포에서 $H_2O_2$에 의한 세포손상을 방지할 수 있는 지를 조사하고자 하였다. 방법:장관상피세포로는 사람의 소장상피세포에서 유래한 배양세포주인 Caco-2세포를 이용하였고, 세포손상 정도는 trypan blue exclusion assay를 통해 평가하였고, 지질의 과산화는 그 산물인 malondialdehyde의 량을 측정하여 산정하였다. 결과: $H_2O_2$는 처리 시간 및 농도에 비례하여 세포손상을 유발하였으며, 이러한 효과는 단삼추출액에 의해 농도의존적으로 방지되었다. $H_2O_2$에 의한 세포소상은 $H_2O_2$제거제인 catalase와 철착염제인 deferoxamine에 의해 방지되었으나 항산화제인 N,N-diphenyl-p-phenylenamine(DPPD)에 의해 영향을 받지 않았다. $H_2O_2$는 지질의 과산화를 증가시켰으며, 이러한 효과는 단삼추출액과 DPPD에 의해 억제되었다. 단삼추출액은 $H_2O_2$에 의한 세포내 ATP 고갈을 방지하였다. $H_2O_2$는 DNA 손상을 일으켰으며, 이러한 효과는 단삼추출액, catalase 및 deferoxamine에 의해 방지되었으나, DPPD에 의해서는 변화되지 않았다. 결론 : 이상의 결과를 종합하면 단삼추출액은 장관상피세포에서 $H_2O_2$에 의한 세포손상을 방지하며, 이러한 효과는 항산화작용이 아닌 다른 작용기전에 기인할 것으로 생각된다. 또한 본 연구의 결과는 $H_2O_2$가 장관상피세포에서 지질의 과산화를 유발하여 세포손상을 일으키지 않음을 가리킨다.
