Validation of an LC/MS/MS Method for the Pharmacokinetic Study of Lercanidipine (염산레르카니디핀 체내동태 연구를 위한 혈청 중 레르카니디핀의 LC/MS/MS 정량법 검증)

  • A rapid, simple and sensitive LC/MS/MS method for the determination of lercanidipine in human serum was validated and applied to the pharmacokinetic study of lercanidipine. Lercanidipine and internal standard, amlodipine, were extracted from human serum by liquid-liquid extraction with hexan-isoamyl alcohol (100: 1, v/v) and analyzed on a $Symmetry^{(R)}$ MS $C_{18}$ column with the mobile phase of acetonitrile-0.2% aqueous formic acid (70: 30, v/v). Using MS/MS with multiple reaction monitoring (MRM) mode, lercanidipine and amlodipine were detected without severe interferences from human serum matrix. Lercanidipine produced a protonated precursor ion ($[M+H]^+$) at m/z 612.3 and a corresponding product ion at m/z 280.0. Internal standard produced a protonated precursor ion ($[M+H]^+$]) at m/z 409.0 and a corresponding product ion at m/z 238.0. The ruggedness of this method was investigated using quality control (QC) samples. This method showed linear response over the concentration range of 0.05-20 ng/mL with correlation coefficient greater than 0.999. The lower limit of quantitation using 0.5 mL of serum was 0.05 ng/mL, which was sensitive enough for pharmacokinetic studies. The overall accuracy of the developed method ranged from 85.51 to 112.2% for lercanidipine with overall precision (% C.V.) being 3.56-13.1%. This method showed good ruggedness (within 15% C.V.) and was successfully applied for the analysis of lercanidipine in human serum samples for the pharmacokinetic studies, demonstrating the suitability of the method.

Establishment of Analytical Method for Pymetrozine Residues in Crops Using Liquid-Liquid Extraction(LLE) (액-액 분배법을 활용한 작물 중 pymetrozine의 잔류분석법 확립)

  • Polar pesticides like pymetrozine (log $P_{ow}$: -0.18) are known to be difficult to analyze. The analytical method of pymetrozine using hydromatrix included in the official method of KFDA was uncommon and provided ambiguous evidence to confirm both the identity and the quantity. Therefore, precise single residue analytical method was developed in representative crops for using liquid-liquid extraction (LLE). The pymetrozine residue was extracted with methanol from 11 representative crops which comprised apple, blueberry, broccoli, cabbage, cherry, crown daisy, hulled rice, Korean cabbage, potato, rice and watermelon. The extract was purified serially by liquid-liquid extraction (LLE) and silica solid phase extraction (SPE). For rice and hulled rice samples, n-hexane partition was additionally adopted to remove nonpolar interferences, mainly lipids. The residue levels were analyzed by HPLC with DAD, using $C_8$ column. LOQ (limit of quantitation) of pymetroizinie was 1 ng (S/N > 10) and MQL (method quantitation limit) was 0.01 mg/kg. Mean recoveries from 11 crop samples fortified at three levels (MQL, 10 ${\times}$ MQL and 50 ${\times}$ MQL) in triplicate were in the range of 83.1~98.5% with coefficients of variation (CV) of less than 10%, regardless of sample type, which satisfies the criteria of KFDA. The method established in this study could be applied to most of crops as an official and general method for analysis of pymetrozine residue.

  • A novel method of adaptive beam forming is presented in this paper. The proposed technique provides for a suboptimal beam pattern that increases the Signal to Noise/Interference Ratio (SNR/SIR), thus, eventually increases the capacity of the communication channel, under an assumption that the desired signal is dominant compared to each component of interferences at the receiver, which is precoditionally achieved in Code Division Multiple Access (CDMA) mobile communications by the chip correlator. The main advantages of the new technique are:(1)The procedure requires neither reference signals nor training period, (2)The signal interchoerency does not affect the performance or complexity of the entire procedure, (3)The number of antennas does not have to be greater than that of the signals of distinct arrival angles, (4)The entire procedure is iterative such that a new suboptimal beam pattern be generated upon the arrival of each new data of which the arrival angle keeps changing due tot he mobility of the signal source, (5)The total amount of computation is tremendously reduced compared to that of most conventional beam forming techniques such that the suboptimal beam pattern be produced at vevery snapshot on a real-time basis. The total computational load for generating a new set of weitht including the update of an N-by-N(N is the number of antenna elements) autocovariance matrix is $0(3N^2 + 12N)$. It can further be reduced down to O(11N) by approximating the matrix with the instantaneous signal vector.

Revising Passive Satellite-based Soil Moisture Retrievals over East Asia Using SMOS (MIRAS) and GCOM-W1 (AMSR2) Satellite and GLDAS Dataset (자료동화 토양수분 데이터를 활용한 동아시아지역 수동형 위성 토양수분 데이터 보정: SMOS (MIRAS), GCOM-W1 (AMSR2) 위성 및 GLDAS 데이터 활용)

  • In this study the Microwave Imaging Radiometer using Aperture Synthesis (MIRAS) sensor onboard the Soil Moisture Ocean Salinity (SMOS) and Advanced Microwave Scanning Radiometer 2 (AMSR2) sensor onboard the Global Change Observation Mission-Water (GCOM-W1) based soil moisture retrievals were revised to obtain better accuracy of soil moisture and higher data acquisition rate over East Asia. These satellite-based soil moisture products are revised against a reference land model data set, called Global Land Data Assimilation System (GLDAS), using Cumulative Distribution Function (CDF) matching and regression approach. Since MIRAS sensor is perturbed by radio frequency interferences (RFI), the worst part of soil moisture retrieval, East Asia, constantly have been undergoing loss of data acquisition rate. To overcome this limitation, the threshold of RFI, DQX, and composite days were suggested to increase data acquisition rate while maintaining appropriate data quality through comparison of land surface model data set. The revised MIRAS and AMSR2 products were compared with in-situ soil moisture and land model data set. The results showed that the revising process increased correlation coefficient values of SMOS and AMSR2 averagely 27% 11% and decreased the root mean square deviation (RMSD) decreased 61% and 57% as compared to in-situ data set. In addition, when the revised products' correlation coefficient values are calculated with model data set, about 80% and 90% of pixels' correlation coefficients of SMOS and AMSR2 increased and all pixels' RMSD decreased. Through our CDF-based revising processes, we propose the way of mutual supplementation of MIRAS and AMSR2 soil moisture retrievals.

The Preliminary Study on the Quantitative Analysis of Quarts by Fourier Transform Infrared Spectrophotometric Direct on Filter(FTIR-DOF) Method -Effects of filter materials, inhomogeneity of deposition, and humidity- (Fourier Transform Infrared Spectrophotometric Direct on Filter 방법을 이용한 석영 분석의 기초 연구 - 필터재질, 비균일 침착 및 습도의 영향 -)

  • Although the Fourier Transform Infra-Red spectrophotometric Direct on Filter(FTIR-DOF) method is a useful analytical technique for quantifying quartz content in respirable dust samples, a number of analytical problems must be taken into consideration such as, to name only a few, inhomogeneous deposition of particles, level of environmental humidity, uneven surface of the filter, and interfering minerals in the sample. This study was designed to select the most suitable wavelength and proper filter material for the method, and to investigate effects of humidity and inhomogeneous deposition of particles on the filter. Samples of respirable dust, created in a dust chamber containing standard material of quartz, were collected using a cyclone equipped with a 25mm filter as a collection medium. The results were as follows; 1. Among seven (7) commercially available filters tested for the FTIR-DOF method, the DM 800 filter showed the best analytical performance having the lowest background absorbance bands and no overlapping peaks at 799, 779, and $695cm^{-1}$. 2. The variations of absorbance due to humidity ranged from 1.0% to 3.3% for $799cm^{-1}$, 1.0% to 3.3% for $779cm^{-1}$, and 8.9%~20.9% for $695cm^{-1}$ peaks, respectively. The $699cm^{-1}$ peak was proved to be most vulnerble to environmental humidity for quantitative analysis of quartz. 3. As for effects of inhomogeneous deposition of samples, the highest variation of absorbance of 10.9% ($13.5{\mu}g$) was observed when using the 695cm-1. The variations of absorbance from the other two peaks, 799 and $779cm^{-1}$, ranged from 1.2 to 3.2%, and 1.4 to 4.1%, respectively. Therefore, the $799cm^{-1}$ peak was considered to be most reliable for quantitative analysis of quartz. The results of this study suggest that, for quantitative analysis of quartz in the respirable dust samples, use of the $799cm^{-1}$ peak can minimize the influence of environmental humidity and inhomogeneous deposition of particles on the filter. The FTIR-DOF method, if adopted for routine analysis of quartz in the respirable dust samples, could save sample preparation time and efforts substantially and also could increase analytical throughputs. Since use of the $799cm^{-1}$ peak is prone to be affected by interferences in the sample, further research on minimizing the effects is needed.

Young Children's Free Choice Activities and a Play Rule Restricting the Number of Players (유아의 자유선택활동과 놀이 인원수 제한규칙)

  • The purpose of this study was to find out the feature of children's play according to a player number restriction rule in the free choice activities. And it is expected that the result of this study will contribute to improving the management of free choice activities. The participants of this study were a homeroom teacher, professor as a co-researcher and 25 4-year-old children of a private kindergarten. The data were collected by observation, children interview and reflective journal of the teacher from July to October in 2014. The results indicated that the children were stressed out with using name cards for presenting center selection, showed overcompetitive behaviors, and frequently violated the rules. The researchers found that the restriction rule had some dysfunctions, which were interferences of moral development, social-emotional development, self-regulation development, preventing safety-accident and free choice behavior. The researcher as a teacher abandoned the player number restriction rule, and observed the children's play. As a result, children's self-regulation abilities on physical space, play and conflicts improved and the teacher changed her strategy when managing free choice activities. Based on the results, the needs of various restriction rules and appropriate methods for applying the rules in free choice activities were further discussed.

RFID Reader Anti-collision Algorithm using the Channel Monitoring Mechanism (채널 모니터링 기법을 이용한 RFID 리더 충돌방지 알고리즘)

  • When an RFID reader attempts to read the tags, interference might occur if the neighboring readers also attempt to communicate with the same tag at the same time or the neighboring readers use the same frequency simultaneously. These interferences cause the RFID reader collision. When the RFID reader collision occurs, either the command from the reader cannot be transmitted to the tags or the response of the tags cannot receive to the reader correctly, Therefore, the international standard for RFID and some papers proposed the methods to reduce the reader collision. Among those, Colorwave and Enhanced Colorwave is the reader anti-collision algorithm using the frame slotted ALOHA based a TDM(Time Division Multiplex) and are able to reduce the reader collision because theses change the frame size according to a collision probability. However, these can generate the reader collisions or interrupt the tag reading of other readers because the reader that collides with another reader randomly chooses a new slot in the frame. In this paper, we propose a new RFID reader anti-collision algorithm that each reader monitors the slots in the frame and chooses the slot having the minimum occupation probability when the reader collision occurs. Then we analyze the performance of the proposed algorithm using simulation tool.

Analysis of Quartz Contents by XRD and FTIR in Respirable Dust from Various Manufacturing Industries Part I - Foundry (제조업체에서 발생하는 호흡성분진중 XRD와 FTIR를 이용한 결정형 유리규산 농도의 비교분석 제 1부 - 주물사업장)

  • This study was conducted to estimate crystalline silica contents in airborne respirable dust from various manufacturing industries and to compare analytical performance of two methods of quantifying crystalline silica, X-ray diffraction(XRD) and Fourie transform infrared spectroscopy (FTIR). For this study, various manufacturing industries with a history of having pneumoconiosis cases and also known to generate dusts containing crystalline silica were investigated. These industries include: foundry, brick, potteries, concrete, and abrasive material, etc. Both personal and area respirable dust samples were collected using 10 mm, Dorr-Oliver nylon cyclone equipped with 37mm, $5{\mu}m$ pore size, polyvinylchloride (PVC) filters as collection media. In addition, total dust samples were collected side-by-side to the respirable samples. All samples were weighed before and after sampling and were pretreated according to the NIOSH sampling and analytical methods 0500, 7500, and 7602 for dust collection and quartz analysis. In addition, bulk samples were collected and analyzed by X-ray fluorescence (XRF) for minerals. In this article, only the results obtained from foundry are reported. The results from various other industries will be published in future articles. The respirable dust concentrations from personal samples by cyclone were $0.46-1.06mg/m^3$ and those from area samples were $0.34-0.73mg/m^3$. Dust concentrations of personal samples were significantly higher than those of area samples. The highest dust concentration was obtained from the personal samples of the finishing operation. Total dust concentration ranged $1.24-3.40mg/m^3$. The mean quartz contents estimated by FTIR and XRD in the personal respirable dust samples were 5.12% and 4.41%, respectively, without significant difference between them. For quartz analyses, the two techniques were highly correlated with $r^2$ ranged 0.803-0.920. But the results by FTIR were mostly higher than those by XRD. In addition, cristobalite was not detected by FTIR. Significant correlations between contents of crystalline silica and such minerals as $Al_2O_3$, CaO, $TiO_2$, and $K_2O$ suggest possible interferences from these minerals.

Direct Determination of Soil Nitrate Using Diffuse Reflectance Fourier Transform Spectroscopy (DRIFTS) (중적외선 분광학을 이용한 토양 내의 질산태 질소 정량분석)

  • Mid-infrared (MIR) spectroscopy, particularly Fourier transform infrared spectroscopy (FTIR), has emerged as an important analytical tool in quantification as well as identification of multi-atomic inorganic ions such as nitrate. In the present study, the possibility of quantifying soil nitrate via diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS) without change of a sample phase or with least treated samples was examined. Four types of soils were spectrally characterized in terms of unique bands of soil contents and interferences with nitrate bands in the range of $2000-1000cm^{-1}$. In order to reduce the effects of soil composition on calibration model for nitrate, spectra transformed to the 1st order derivatives were used in the partial least squared regression (PLSR) model and the classification procedure associated with input soil types was involved in calibration system. PLSR calibration models for each soil type provided better performance results ($R^2$>0.95, RPD>6.0) than the model considering just one type of soil as a standard.

The Growth and Yield of Soybean as Affected by Competitive Density of Cuscuta pentagona (미국실새삼 발생밀도가 콩 생육 및 수량에 미치는 영향)

  • This study was conducted to predict reduction of soybean yield as affected by different densities of Cuscuta pentagona. All data were fitted to Cousens' rectangular hyperbola model to estimate parameters for predicting soybean yield loss. The yield of soybean in the various densities (1 to 48 plants $m^{-2}$) of C. pentagona reduced by 80.3 to 99.7%, respectively. Among yield components, number of pods was the most significantly influenced by weed interferences. The prediction model for soybean yield as affected by weed competition was as follows: Y= 274.6783/(1+4.3522X), $r^2$=0.999 in C. pentagona. Economic threshold levels calculated using cousens' equation was 0.004 plants $m^{-2}$ in C. pentagona.
