Currently, there is a lack of genetic markers capable of effectively detecting polymorphisms in Clematis. Therefore, we developed new markers to investigate inter- and intraspecific diversity in Clematis. Based on the complete chloroplast genome of Clematis terniflora, simple sequence repeats were explored and primer pairs were designed for all ten adequate repeat regions (cpSSRs), which were tested in 43 individuals of 11 Clematis species. In addition, the nuclear ITS region was sequenced in 11 Clematis species. Seven cpSSR loci were found to be polymorphic in the genus and serve as markers that can distinguish different species and be used in different genetic analyses, including cultivar identification to assist the breeding of new ornamental cultivars.
ISSR마크를 사용하여 고려 인삼의 품종 및 계통간 분자적 인증과 유전적 다형현상을 조사하였다. 56개의 ISSR 프라이머 중 5개가 일곱 품종 및 계통간 명확하고 재현성이 높은 DNA분절을 나타내는 최적 프라이머로 선택되었다. 전체 43밴드는 250 bp - 1,700 bp의 분자량을 가지며 프라이머당 8.6개의 밴드를 나타내었다. 고려 인삼에서 다형현상 정도는 20.9%였다. 특히 천풍 품종이 가장 높은 다형현상을 나타낸 반면 다른 품종은 거의 다형현상을 나타내지 않았다. 결론적으로 DNA수준에서 ISSR마크로 천풍이 다른 고려 인삼의 품종 및 계통인 연풍, 황숙종, 자경종과 구분에 이용될 수 있음이 판명되었다.
우리나라 특산식물이며 희귀식물인 만리화(물푸레나무과) 집단의 유전적 다양성을 조사하기 위하여 5 집단 84개체에 대한 ISSR (Inter-Simple Sequence Repeats) 표지자 분석을 실시하였다. 14개의 ISSR 프라이머에서 총 93개의 증폭산물을 관찰할 수 있었으며, 유전적 다양성을 나타내는 P (Percentage of polymorphic loci)값과 S.I. (Shannon's information Index)가 다른 관목류에 비해 비교적 낮게 나타났다. 집단별 유전적 다양성은 석병산집단 (P = 64.5%, S.I. = 0.281)과 설악산B집단(P = 62.4%, S.I. = 0.292)이 높았으며, 석개재집단(P = 37.6%, S.I. = 0.178)이 가장 낮았다. 전체 유전변이 중 30.6%가 집단간에 기인하는 것으로 나타났고, 나머지 69.4%는 집단내 개체간의 차이에서 기인하였다. 이러한 결과는 분포역이 매우 제한되어 있으며 불연속적으로 출연하는 희귀종이라는 점으로 미루어 볼 때 지역간의 유전자 교류가 원활하지 못해 나타난 결과라고 추정해 볼 수 있다. 유전적 거리를 이용하여 UPGMA 방법에 의한 유집분석을 실시한 결과, 집단의 지리적 격리정도와 유전적 연관성은 비교적 일치하는 경향이었다. 본 연구 결과, 만리화의 유전자원보존을 위해서는 자생지 보호와 더불어 동적인 현지외 보존(dynamic ex situ conservation)과 같은 보다 적극적인 대책이 요구되며, 더 높은 유전적 다양성을 확보하기 위해서는 소수의 집단에서 다수 개체를 선발하기보다는 집단당 소수 개체를 다수의 집단에서 선발하는 집단 위주의 보존이 더욱 효과적일 것으로 판단된다.
Safflower (Carthamus tinctorius L.) is a herb primarily distributed throughout in the world. We have used the inter-simple sequence repeats (ISSR) technique to investigate the phylogenetic relationships and genetic diversity of C. tinctorius. Of all germplasms, 88.7% were polymorphic among all germplasms. Mean genetic diversity within germplasms was very low (0.048). The Turkey germplasm had the highest expected diversity (0.082) and Australia germplasm was the lowest (0.020). These values indicate that most of the genetic diversity of safflower is found among germplasms and there is a high among-germplasm differentiation. We found eight phenetic bands for determining the specific marker of germplasm with SCAR markers. The regions of the Mediterranean Sea and India may be the most probable candidates for the origin of safflower. The tree showed four major clades: (1) European germplasms, (2) Azerbaijan, Egypt, and Ethiopia, (3) Australia, and (4) America.
Fifty two Macrophomina phaseolina isolates were recovered from 24 host plant species through the 14 Iranian provinces. All isolates were confirmed to species using species-specific primers. The colony characteristics of each isolate were recorded, including chlorate phenotype, relative growth rate at $30^{\circ}C$ and $37^{\circ}C$, average size of microsclerotia, and time to microsclerotia formation. The feathery colony phenotype was the most common (63.7%) on the chlorate selective medium and represented the chlorate sensitive phenotype of the Iranian Macrophomina phaseolina population. Meantime, inter simple sequence repeats (ISSR) Markers were used to assess the genetic diversity of the fungus. Unweighted pair-group method using arithmetic means (UPGMA) clustering of data showed that isolates did not clearly differentiate to the specific group according to the host or geographical origins, however, usually the isolates from the same host or the same geographic origin tend to group nearly. Our results did not show a correlation between the genetic diversity based on the ISSR and phenotypic characteristics. Similar to the M. phaseolina populations in the other countries, the Iranian isolates were highly diverse based on the phenotypic and the genotypic characteristics investigated and needs more studies using neutral molecular tools to get a deeper insight into this complex species.
The genetic diversity and the genetic relationship among 30 genetic resources of T. officinale and T. coreanum collected from 20 regions in Korea were evaluated by using ISSR markers. Out of 127 loci detected overall, 122 were identified to be polymorphic with a rate of 96.0% at the 30 individuals. The intraspecific polymorphism between T. officinale and T. coreanum was 92.6% and 88.2%, respectively. The genetic similarity matrix (GSM) revealed a wide range of variablility among the 30 accessions, spanning from 0.179 to 922. According to the clustering analysis, different species T. officinale and T. coreanum, were divided into independent groups and all of the accessions could be classified into 7 categories. Especially, all of the mountain collected accessions belonged to independent groups. The study findings indicate that T. officinale and T. coreanum accessions have a high genetic diversity and accordingly carry a germ-plasm qualifying as good genetic resources for breeding.

The genetic diversity of two subpopulations of Corynandra chelidonii, one of terrestrial and the other of aquatic environments, was measured with molecular markers, such as start codon targeted (SCoT), inter simple sequence repeats (ISSR), and random amplification of polymorphic DNA (RAPD). The traditional morphological traits such as habitat, habit, leaf morphology, the colour of the sepals and petals, number of stamens, and seed morphology formed the base for their realization as two varieties, C. chelidonii var. pallae and C. chelidonii var. chelidonii. The polymorphism between the two variants was 100% with the primers SCoT-2 and OPA-1 and 4, while maximum polymorphism was detected with ISSR-2, SCoT-3, and OPA-3. The study used, for the first time, more than one molecular marker to assess the genetic variation underscoring the morphological variation in Corynandra chelidonii (L.f.) Cochrane & Iltis. The study justifies the recognition of the two subpopulations of Corynandra chelidonii from aquatic and terrestrial environments as two distinct varieties, C. chelidonii var. pallae (Reddy & Raju) V.S.Raju and C. chelidonii var. chelidonii, respectively, based on the traditional taxonomic evidence.
Charcoal rot disease, caused by the fungus Macrophomina phaseolina, is one of the most important diseases of Sesame (Sesamum indicum) all over the world. However, the population biology of M. phaseolina is poorly understood. In this study, M. phaseolina isolates from five different regions of Iran (Khuzestan, Fars, Bushehr, Hormozgan, and Kohgiluyeh & Boyer-Ahmad provinces) (n=200) were analyzed for genetic variation using inter simple sequence repeats marker. In total, 152 unique haplotypes were identified among the 200 M. phaseolina isolates, and gene diversity (H=0.46-0.84) and genotypic diversity were high in each of the regions. The structure analysis clustered five Iranian populations into two distinct groups, the individuals from group 1 were assigned to the Bushehr population and the individuals from Khuzestan, Fars, Hormozgan and Kohgiluyeh & Boyer-Ahmad were aggregated and formed group 2. The results matched with genetic differentiation and gene flow among regions. Analyses of the distribution of gene diversity within and among five Iranian populations were 61% and 39%, respectively. Our results showed that infected seeds are thought to be the dominant mechanism responsible for the spreading of the pathogen in southern parts of Iran. In summary, it is essential to have local quarantine and prevent seed exchanges between geographical populations to restrict the dispersal of pathogen over long distances and provide certified seeds in Iran.
한국꿩 (Korean ring-necked Pheasant, Phasianus colchicus karpowi)과 외국 아종의 유전적 유연관계를 파악하기 위해 야생 한국꿩, 사육 한국꿩, 사육 한국꿩과 외국꿩간의 잡종꿩, 외국꿩 4아종(중국 링넥, 흑 뮤탄트, 백 뮤탄트, 녹치)을 대상으로 ISSR 표지자 분석과 AMOVA 분석을 수행하였다. 야생 한국꿩의 전체 유전 다양성중 94.08%가 서식지 내 개체간 유전적 차이에 기인하고, 5.9% ($\Phi$st=0.059)가 서식지간 차이에 기인하였다. 사육 한국꿩이 유전적으로 야생 한국꿩 보다는 외국꿩 4아종과 가깝게 나타났다. AMOVA분석과 cluster분석에서 사육 한국꿩이 야생 한국꿩은 물론 외국꿩 4아종과도 유전적 차이를 보이는 것으로 미루어 볼 때 사육되고 있는 한국꿩은 한국꿩과 외국 아종간의 다양한 교잡에 의해 생겨난 잡종일 가능성이 높다. 외국꿩 4아종(중국 링넥, 흑 뮤탄트, 백 뮤탄트, 녹치)의 전체 유전 다양성중 96.63%가 아종내 개체간 차이에 기인하고. 3.4% ($\Phi$st=0.034)로 아종간 차이에 기인하여 외국꿩 4아종의 아종간 유전적 차이가 야생 한국꿩의 서식지간 차이보다도 낮은 것으로 나타났다. 이는 본 분석에 사용된 외국꿩 4아종이 형태적으로는 다른 아종으로 분류되지만 유전적으로는 매우 가까운 위치에 있음을 의미한다. 7서식지의 야생 한국정과 외국꿩 4아종을 각각 야생 한국꿩 그룹과 외국꿩 그룹으로 분류하여 두 그룹의 유전적 다양성을 분석한 결과, 전체 유전변이 중 그룹간 차이의 비율은 17% 이상(그룹 내 서식지/아종간 차이는 약 4%)으로 야생 한국꿩이 아종 관계인 외국꿩 4아종과 상당한 유전적 차이를 보이는 것으로 나타났다. 유전적 유연관계 분석결과에서 중국 링넥. 흑 뮤탄트, 백 뮤탄트의 외국꿩 3 아종은 유전적으로 외국 아종에 가까운 사육한국꿩, 잡종꿩과 함께 하나의 분지군을 형성하면서 야생 한국꿩 그룹으로부터 98% 신뢰수준에서 뚜렷하게 구별되었다.
소나무 단일개체에서 채취한 풍매종자 중 임의로 선택한 96개의 반수체 genome을 이용하여 RAPD 및 I-SSR PCR 증폭산물을 분석하였다. RAPD 분석용 primer 200개와 I-SSR 분석 용 primer 90개를 screen하여 증폭산물의 분획양상이 선명한 RAPD primer 45개와 I-SSR primer 22개를 선택하여 PCR을 수행하였다. 45개의 RAPD primer중 25개와 22개의 I-SSR primer중 18개를 사용한 PCR 분석결과에서 멘델의 유전양식을 만족하는 52개 (2.08/primer)와 46개 (2.56/primer)의 다형성 유전자좌를 각각 확인하였다. 멘델의 유전양식을 만족하는 96개의 다형성 유전자좌를 대상으로 LOD 3.0에서 two-point 연관분석을 수행한 결과 총 63개(35개의 RAPD와 26개의 I-SSR)의 유전자화가 20개의 연관군에 속하는 것이 확인되었다. 총 연관거리는 1097.8 cM이었으며, 유전자좌간 평균연관 거리는 25.5 cM, 최소 및 최대연관 거리는 각각 4.3 cM 및 54.9 cM이었다. 그리고 20개의 연관군 중 14개의 연관군이 RAPD와 I-SSR 유전자좌의 통합에 의해서 형성된 연관군이었다. 즉, 52개의 RAPD와 46개의 I-SSR 유전자좌를 각각 분석한 결과보다 길고 새로운 연관군이 형성되었다. 보다 정밀한 유전자 연관지도를 작성하기 위해서는 보다 많은 수의 DNA marker가 필요하다고 판단되며, 본 연구의 결과는 소나무의 유용 유전자의 확인 및 생장과 재질과 같은 유용형질에 대한 QTL의 위치를 결정하는 데 기초자료가 될 것이다.
