Pressure-Overload Cardiac Hypertrophy Is Associated with Distinct Alternative Splicing Due to Altered Expression of Splicing Factors

  • Chronic pressure-overload cardiac hypertrophy is associated with an increased risk of morbidity/mortality, largely due to maladaptive remodeling and dilatation that progresses to dilated cardiomyopathy. Alternative splicing is an important biological mechanism that generates proteomic complexity and diversity. The recent development of next-generation RNA sequencing has improved our understanding of the qualitative signatures associated with alternative splicing in various biological conditions. However, the role of alternative splicing in cardiac hypertrophy is yet unknown. The present study employed RNA-Seq and a bioinformatic approach to detect the RNA splicing regulatory elements involved in alternative splicing during pressure-overload cardiac hypertrophy. We found GC-rich exonic motifs that regulate intron retention in 5' UTRs and AT-rich exonic motifs that are involved in exclusion of the AT-rich elements that cause mRNA instability in 3' UTRs. We also identified motifs in the intronic regions involved in exon exclusion and inclusion, which predicted splicing factors that bind to these motifs. We found, through Western blotting, that the expression levels of three splicing factors, ESRP1, PTB and SF2/ASF, were significantly altered during cardiac hypertrophy. Collectively, the present results suggest that chronic pressure-overload hypertrophy is closely associated with distinct alternative splicing due to altered expression of splicing factors.

백혈병 세포주 HL-60에서 과루실(瓜蔞實)의 세포고사 유도 효과 (Apoptosis-inducing Effect of Fructus Trichosanthis in HL-60 Leukemic Cells)

  • Many naturally occurring plant extracts are studied for their beneficial effects for health and particularly on cancer. Apoptosis, or programmed cell death, occurs in both normal and pathological conditions, including cancer. Dysregulation of apoptosis allows transformed cells to continually and uninhibitedly enter the cell cycle, thus perpetuating the sequence of mutation, genomic instability and, finally, oncogenesis. To investigate the apoptosis-Inducing effect of the extract of Fructus Trichosanthis (EFT) on leukemic HL-60 cells and its mechanism, HL-60 cells in vitro in culture medium were given different doses of the extract. The inhibitory rate of cells were measured by microculture tetrazolium assay, cell apoptotic rate was detected by flow cytometry, morphology of cell apoptosis was observed by DAPI fluorescence staining, and the activations of caspases and PARP were detected using Western blotting analysis. The extract could activate the caspase-3 and caspase-8, induce PARP cleavage, inhibit growth of HL-60 cells, and cause apoptosis significantly. The suppression was in dose-dependent manner. Marked morphological changes of cell apoptosis including condensation of chromatin and nuclear fragmentation were observed clearly by DAPI fluorescence staining especially. These results will provide strong laboratory evidence of EFT for clinical treatment of acute leukemia.

Shoulder Arthrokinematics of Collegiate Ice Hockey Athletes Based on the 3D-2D Model Registration Technique

  • Objective: There is a lack of studies using the 3D-2D image registration techniques on the mechanism of a shoulder injury for ice hockey players. This study aimed to analyze in vivo 3D glenohumeral joint arthrokinematics in collegiate ice hockey athletes and compare shoulder scaption with or without a hockey stick using the 3D-2D image registration technique. Method: We recruited 12 male elite ice hockey players (age, 19.88 ± 0.65 years). For arthrokinematic analysis of the common shoulder abduction movements of the injury pathogenesis of ice hockey players, participants abducted their dominant arm along the scapular plane and then grabbed a stick using the same motion under C-arm fluoroscopy with 16 frames per second. Computed tomography (CT) scans of the shoulder complex were obtained with a 0.6-mm slice pitch. Data from the humerus translation distances, scapula upward rotation, anterior-posterior tilt, internal to external rotation angles, and scapulohumeral rhythm (SHR) ratio on glenohumeral (GH) joint kinematics were outputted using a MATLAB customized code. Results: The humeral translation in the stick hand compared to the bare hand moved more anterior and more superior until the abduction angle reached 40°. When the GH joint in the stick hand was at the maximal abduction of the scapula, the scapula was externally rotated 2~5° relative to 0°. The SHR ratio relative to the abduction along the scapular plane at 40° indicated a statistically significant difference between the two groups (p < 0.05). Conclusion: With arm loading with the stick, the humeral and scapular kinematics showed a significant correlation in the initial section of the SHR. Although these correlations might be difficult in clinical settings, ice hockey athletes can lead to the movement difference of the scapulohumeral joints with inherent instability.

A cost-effective method to prepare size-controlled nanoscale zero-valent iron for nitrate reduction

  • Nanoscale zero-valent iron (nZVI) has proved to be an effective tool in applied environmental nanotechnology, where the decreased particle diameter provides a drastic change in the properties and efficiency of nanomaterials used in water purification. However, the agglomeration and colloidal instability represent a problematic and a remarkable reduction in nZVI reactivity. In view of that, this study reports a simple and cost-effective new strategy for ultra-small (< 7.5%) distributed functionalized nZVI-EG (1-9 nm), with high colloidal stability and reduction capacity. These were obtained without inert conditions, using a simple, economical synthesis methodology employing two stabilization mechanisms based on the use of non-aqueous solvent (methanol) and ethylene glycol (EG) as a stabilizer. The information from UV-Vis absorption spectroscopy and Fourier transform infrared spectroscopy suggests iron ion coordination by interaction with methanol molecules. Subsequently, after nZVI formation, particle-surface modification occurs by the addition of the EG. Size distribution analysis shows an average diameter of 4.23 nm and the predominance (> 90%) of particles with sizes < 6.10 nm. Evaluation of the stability of functionalized nZVI by sedimentation test and a dynamic light-scattering technique, demonstrated very high colloidal stability. The ultra-small particles displayed a rapid and high nitrate removal capacity from water.

횡단압력파 발생을 위한 단일 펄스건의 압력파 성능시험 (Performance Test of a Single Pulse Gun for Transverse Pressure Wave Generation)

  • 펄스건 장치는 연소실 내, 다중 분사기 배열에 따른 연소 유동장으로의 횡단 압력파 전파/감쇠 메커니즘 규명을 목적으로 제작되었다. 제작된 펄스건은 성능시험을 통해 목표 연소압에서의 운용 가능 여부와 압력파 강도 제어 여부를 확인하였다. 기체질소를 사용하여 고압관에 가압을 하였으며 다이아프램에는 $100{\mu}m$ 두께의 OHP 필름을 사용하였다. 압력파의 속도와 강도를 확인하기 위해 압력 트랜스듀서를 이용하여 동압과 정압을 측정하였다. 제작된 펄스건은 공급압력에 따라 압력파의 강도 조절이 가능하며 횡방향성을 가지는 압력파를 생성할 수 있음을 성능시험을 통해 확인하였다.

전단동축인젝터를 이용한 액상 케로신 및 젤 케로신의 연소특성에 대한 실험적 연구 (Experimental Investigation on Combustion Characteristics of Liquid Kerosene and Gelled Kerosene Using Shear Coaxial Injector)

  • 본 연구에서는 lab-scale 젤로켓모터를 이용하여 액상 케로신 연료와 이를 젤화 시킨 젤 케로신 연료의 연소시험을 수행함으로써, 각 연료별 연소실 정압특성 및 동압특성에 대해 분석하였다. 액상 케로신과 젤 케로신의 정압, 특성속도 및 특성속도 효율은 예상외로 큰 차이를 보이지 않음을 확인하였다. 그러나 액상 케로신과 젤 케로신의 동압 특성을 비교한 결과, 특정 고주파수 영역에서 젤 케로신의 압력 진폭이 액상 케로신 대비 증가하는 것을 확인하였다. 이는 젤 추진제의 고유 연소 메커니즘에 기인한 특성으로 여겨지며, 이들 압력 섭동 진폭이 추후 젤로켓모터의 고주파 연소불안정에 영향을 미치는 주요 인자로 적용될 가능성이 클 것으로 판단되었다.

바람의 회전응력, 지형, 그리고 성층화가 성층 호수의 물 순환에 미치는 영향 (Effects of Wind Stress Curl, Topography, and Stratification on the Basin-scale Circulations in a Stratified Lake)

  • Basin-scale motions in a stratified lake rely on interactions of spatially and temporally varying wind force, bathymetry, density variation, and earth's rotation. These motions provide a major driving force for vertical and horizontal mixing of inorganic and organic materials, dissolved oxygen, storm water and floating debris in stratified lakes. In Lake Tahoe, located between California and Nevada, USA, basin-scale circulations are obviously important because they are directly associated with the fate of the suspended particulate materials that degrade the clarity of the lake. A three-dimensional hydrodynamic model, ELCOM, was applied to Lake Tahoe to investigate the underlying mechanisms that determine the characteristics of basin-scale circulations. Numerical experiments were designed to examine the relative effects of various mechanisms responsible for the horizontal circulations for two different seasons, summer and winter. The unique double gyre, a cyclonic northern gyre and an anti-cyclonic southern gyre, occurred during the winter cooling season when wind stress curl, stratification, and Coriolis effect were all incorporated. The horizontal structure of the upwelling and downwelling formed due to basin-scale internal waves found to be closely related to the rotating direction of each gyre. In the summer, the spatially varying wind field and the Coriolis effect caused a dominant anti-cyclonic gyre to develop in the center of the lake. In the winter, a significant wind event excited internal waves, and a persistent (2 week long) cyclonic gyre formed near the upwelling zone. Mechanism of the persistent cyclonic gyre is explained as a geostrophic circulation ensued by balancing of the baroclinc pressure gradient (or baroclinic instability) and Coriolis effect. Topographic effect, examined by simulating a flat bathymetry with constant depth of 300m, was found to be significant during the winter cooling season but not as significant as the wind curl and baroclinic effects.

상용급 가스터빈에서 셰일가스 파일럿비 영향에 관한 수치해석적 연구 (Numerical Study on Pilot Ratio Effect of Shale-Gas in a Commercial Gas Turbine)

  • 논문에서는 상용급 가스터빈을 대상으로 해서 셰일가스를 연료로 공급할 때 유동 및 연소특성을 3-D 수치해석적 방법으로 구하였다. 이 때, Standard k-e 난류모델, 2단 메탄산화반응, Finite rate/Eddy dissipation 반응모델, DTRM 복사모델이 사용되었고, 기준조건(도시가스, PR 0.07)에서 출구 측에서 형성되는 온도는 이전 문헌 값과 비슷한 값을 보였다. 위 모델을 바탕으로 해서 연료조건으로 기존의 도시가스 외에 세 가지 셰일가스 조건(도시가스 대비 열량기준 80%, 90%, 105%)을 선정하였고, 각 연료조건에 대하여 세 가지 연료분사조건(PR=0.7, 0.9, 0.11)에 대한 해석을 수행하였다. 해석결과, 모든 셰일가스 연료공급 조건에 대하여 도시가스 대비 온도 혹은 NOx 측면에서 연소안정화를 만족하였다. 또한 모든 조건에 대해서 PR이 증가할수록 출구측 평균온도는 일정했지만 NOx량은 증가하였다. 이는 파일럿비가 증가할수록 상대적으로 확산연소가 증가했기 때문이다.

Analysis of the mechanical properties and failure modes of rock masses with nonpersistent joint networks

  • Complex rock masses include various joint planes, bedding planes and other weak structural planes. The existence of these structural planes affects the mechanical properties, deformation rules and failure modes of jointed rock masses. To study the influence of the parameters of a nonpersistent joint network on the mechanical properties and failure modes of jointed rock masses, synthetic rock mass (SRM) technology based on discrete elements is introduced. The results show that as the size of the joints in the rock mass increases, the compressive strength and the discreteness of the rock mass first increase and then decrease. Among them, the joints that are characterized by "small but many" joints and "large and clustered" joints have the most significant impact on the strength of the rock mass. With the increase in joint density in the rock mass, the compressive strength of rock mass decreases monotonically, but the rate of decrease gradually decreases. With the increase in the joint dip angle in rock mass, the strength of the rock mass first decreases and then increases, forming a U-shaped change rule. In the analysis of the failure mode and deformation of a jointed rock mass, the type of plastic zone formed after rock mass failure is closely related to the macroscopic displacement deformation of the rock mass and the parameters of the joints, which generally shows that the location and density of the joints greatly affect the failure mode and displacement degree of the jointed rock mass. The instability mechanism of jointed surrounding rock is revealed.

Age-specific variations in hematological and biochemical parameters in middle- and large-sized of dogs

  • Aging triggers cellular and molecular alterations, including genomic instability and organ dysfunction, which increases the risk of disease in mammals. Recently, due to the markedly growing number of aging dogs in the world, as much as 49% in total number of pet dogs, it is necessary to improve and maintain their quality of life by understanding of the biological effects of aging. Therefore, the aim of this study was to determine specific biomarkers in aging dogs as a means of defining a set of hematological/biochemical biomarkers that influence the aging process. Blood samples were collected from younger (1-3 years) and older (7-10 years) dogs of middle/large size. The hematological/biochemistry analysis was performed to evaluate parameters significantly associated with age. Enzyme-linked immunosorbent assay was used to target growth hormone (GH)/insulin growth factor-1 (IGF-1), one of the main regulators of the aging process. Declining levels of total protein and increased levels of glucose in young dogs was observed regardless of their body size. Notably, a significantly high concentration of GH and IGF-1 in the younger dogs compared to the older dogs was found in middle/large-sized dogs. GH and IGF-1 were also found at significantly high levels in large-sized dogs compared to middle-sized dogs, suggesting a similar trend to that of elderly humans. Consequently, glucose, total protein, GH, and IGF-1 were identified as potential biomarkers for regulating the aging process in large/middle-sized dogs. These findings provide an invaluable insight into the mechanism of aging for the field of aging research.
