The Functional Behaviors of Cosurfactant in Design of Self-nanoemulsifying Drug Delivery Systems

  • Nanoemulsions have been widely investigated for many years because of their attractive and unique characteristics. Nanoemulsions are composed of oil, surfactant, co-surfactant and water. Especially, cosurfactant plays a critical role in formation of nanoemulsions. In pharmaceutical area, a pre-concentrate form of nanoemulsions which is known as self-nanoemulsifying drug delivery systems (SNEDDS) was available for some water-insoluble drugs. In this study, we investigated the functional behaviors of cosurfactant in design of SNEDDS and nanoemulsions. Cremophor RH 40$^{(R)}$, Propylene carbonate and medium chain triglyceride were selected for surfactant, cosurfactant and oil, respectively. Cyclosporine was employed as a drug. Phase diagrams showed the area of isotropic o/w region which forms o/w nanoemulsions was not significantly affected by the compositional ratio of cosurfactant. But, drug solubilization capacity, droplet size of nanoemulsions and drug release rate were greatly affected by the cosurfactant.

흡광광도법 및 원자흡광광도법에 의한 Pipethanate$\cdot$HCl의 정량 (Determination of Pipethanate$\cdot$HCl by Spectrophotometry and Atomic Absorption Spectrophotometry)

  • Pipethanate*HCl reacts with ammonium cobaltothiocyanate to form a stable ion pair which has an absorption maximum at 626nm. The reaction product was insoluble in water but freely soluble in most organic solvents. 1, 2-Dichloroethane was the best extracting solvent among the several organic solvents. Pipethanate*HCl can be determined not only by the spectrophotometry but also inderectly by estimation of cobalt in the organic phase by atomic absorption spectrophotometry. Linear relationship was found between absorbance and concentration in the range of $1.0{\times}10^{-3}~4.0{\times}10^{-3}M$ by spectrophotometry and $1.5{\times}10^{-4}~4.0{\times}10^{-4}M$ by atomic absorption spectrophotometry. With this method it was possible to determine pipethanate$\cdot$HCl in the pharmaceutical preparations.

Picric acid를 전자수용체로 한 Phenothiazine계 화합물의 정량법 (Spectrophotometric Determination of Phenothiazine Derivatives by using Picric acid as Electron Acceptor)

  • A rapid and sensitive spectrophotometric method has been developed for quantitation of some phenothiazine derivatives (PTZDS). The method depends on the formation of insoluble yellow complexes between PTZDS and picric acid (PCA) in aqueous layer. These complexes are quantitatively extracted from aqueous phase into chloroform. The binding ratio of PTZDS-PCA complexes were presumed as PTZDS-1 to PCA-1 by means of mole ratio and continuous variation methods. The complexes are stable for more than 24hours in chloroform layer at room temperature. Most of compounds associated with pharmaceutical preparation of PTZDS do not interfere with this method. However, chlorpheniramine and diphenhydramine interfere with the riaethod. The binding state of PTZDS-PCA complexes were presumed by IR and $^1$H-NMR spectra as intermolecular hydrogen bonding.

The Antioxidant Activity of Ecklonia stolonifera

  • The antioxidant activity of Ecklonia stolonifera was determined by measuring lipid peroxide produced when a mouse liver homogenate was exposed to the air at $37^{\circ}C$ using 2-thiobarbituric acid (TBA) and the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. The methanol extract of Ecklonia stolonifera showed strong antioxidant activity. And the methanol extract was fractionated with several solvents. With regard their fractions, the antioxidative activity were in the order of ethyl acetate>dichloromethane insoluble intermediated phase>dichloromethane>n-butanol>water fraction. The ethyl acetate soluble fraction exhibiting the strongest antioxidant activity was further purified by repeated silica gel and Sephadex LH-20 column chromatography. Antioxidant phloroglucinol was isolated and identified by$ ^1H-NMR\; and\; ^{13}C-NMR$. Its antioxidant activity was simlilar to that of L-ascorbic acid.

Structural Analysis of Simulated Fission-Produced Noble Metal Alloys and Their Superconductivities

  • Ternary (Mo-Ru-Pd) and binary (Mo-Ru, Mo-Pd) alloys have been prepared using an Ar arc melting furnace. Mo and the noble metals, Ru and Pd, are the constituents of metallic insoluble residues, which were found in the early days of post-irradiation studies on uranium oxide fuels. In the present study, the structure of the alloys was evaluated using a powder X-ray diffractometer. Unit cell parameters were determined by least squares refinements of powder X-ray diffraction data. Scanning electron microscopic analyses of the surface of the alloys indicated that surface morphology was dependent on the crystallographic structure as well as its composition. Measurements of the magnetic susceptibility of the alloys showed evidence of superconducting transition from 3 to 9.2 K. Among the ternary and binary alloys, the ${\sigma}-phase$ showed the highest superconducting transition temperature,~9.2 K.

거대고리 운반체에 의한 중금속이온의 에멀죤 액체막 수송 (Emulsion Liquid Membrane Transport of Heavy Metal Sons by Macrocyclic Carriers)

  • 액체막의 운반체로 사용할 새로운 2개의 거대고리화합물을 합성하였다. 이들 결과들은 이 시스템을 구성하는데 있어서 이론의 응용성을 증명하여 준다. source phase의 공존이온으로서 $SCN^-$,$I^-$$Cl^-$이온을 그리고 receicing phases에서 $S_2O_3^{2-}$$P_2O_7^{4-}$을 이용한 액세막계로서부터 중금속 이온들에 대한 선택적 수송효율을 검토하였다. source phase의 M(II)이 $Cd(SCN)_2$$(P[SCN^-]= 0.40M)$, $Hg(SCN)_2([SCN^-]=0.40M)$, Pd(CN)$([CN^-]= 0.40M)$일때 M(II)의 수송율은 최대값을 나타낸다. 각가의 경쟁 양이온에 대한 Cd(II)이나 Pd(II)은 source phase가 00.3M-$S_2O_3^{2-}$이나 0.3M-$P_2O_7^{4-}$ 일때 가장 잘 분리된다.이 연구의 결과에서, 이 액체막계에서 효과적인 거대고리-매질수송을 하기 위해서는 두개의 규칙이 반드시 필요하다. 첫째, tiluence중으로 $M^{n+}$이온이 효과적으로 추출되고, 즉 만일 $M^{n+}$ 거대고리화합물 상호작용에 대한 logK값과 $M^{n+}$-거대고리화합물($L_1$이나 $L_2$)의 상호작용에 대한 logK값의 비가 충분히 크다면 receiving phase와 toluene의 접촉면으로부터 쉽게 중금속이온($Cd^{2+}$,$Pb^{2+}$$Hg^{2+}$)들이 떨어져 나온다. $L_1$(3,5-benzo-10,13,18,21-tetraoxa-1,7-diazabicyclo(8,5,5)eicosnan)은 $Cd^{2+}$$Pb^{2+}$ 이온과 안정한 착물을 형성한다. 그리고 $L_1$은 수용액중에서 용해하기가 매우 어렵다. 그리고 $Cd^{2+}$$L_1$$Pb^{2+}$$L_1$착물은 $Cd^{2+}-{(S_2O_3)}_2^{2-}$$Pd^{2+}-P_2O_7^{4-}$착물보다 비교적 불안정하다. 다른 한편으로 $Hg^{2+}-L_1$착물의 안정도는 $Hg^2-{2+}-(S_2O_3)_2^{2-}$이나 $Pb^{2+}-P0_2O_7^{4-}$의 그것보다 그리고 $L_2$(5,8,15,18,23,26-hexaoxa-1,12-diazabicyclo(10,8,8)octacosan)의 tuluene에 대한 분배계수는 $L_1$의 그것보다 훨씬 작다. 따라서 $Hg^{2+}$-$L_1$이나 $M^{n+}$이나 $M^{n+}-L-2(M^{2+}=Cd^{2+}, Pb^{2+}$이나 $Hg^{2+})$의 안정도수상수가 매우 큼에도 불구하고 이들 양이온의 수송량은 매우 적다.

Moraxella sp. CK-1의 세포외 Autolysins 특성 연구 (Characterization of the Extracellular Autolysins from Moraxella sp. CK-1.)

  • 남조류 분해세균으로 알려진 Moraxella sp. CK-1의 세포외 자가용균효소의 특성을 규명하였다. Moraxella sp. CK-1은 대수증기식 초기부터 세포외부로 자가용균효소를 분비하며, 이들 효소는 60-$70^{\circ}C$, pH 9.0의 최적 활성 조건을 나타내었다. 이들 효소는 $Na^{+}$, $K^{+}$, $Li^{+}$등의 이온에 의해 활성이 촉진되고, $Ba^{2+}$, $Mg^{2+}$, $Ca^{2+}$$Mn^{2+}$ 등의 이온에 의해 활성이 억제되며, 특히, $Fe^{2+}$$Cu^{2+}$ 이온 존재하에서는 효소의 활성이 거의 나타나지 않았다. Micrococcusluteus 세포를 기질로 이용하여 renaturing, SDS-PAGE를 수행한 결과 Moraxella sp. CK-1은 분자량이 각각 30,32,38 그리고 41 kDa인 4가지 효소를 분비하며, 이중 32와 41 kDa의 효소는 생장 초기부터 사멸기까지 지속적으로 배지에 분포하는 반면, 38 kDa의 효소는 대수증식기 중기 이전, 30 kDa의 효소는 대수증식기 중기 이후에 배지에 분포하였다. M. luteus의 SDS-insoluble peptidoglycan에 효소추출물을 처리하였을 때 반응액의 유리 아미노기 농도가 증가하는 것은 이들 효소가 N-acetylmuramyl-L-ananine amidase 또는 endopeptidase라는 사실을 시사한다.

Effect of Ag on microstructural behaviour of Nanocrystalline $Fe_{87-x}Zr_7B_6Ag_x$($0{\leq}x_{Ag}{\leq}4$) Magnetic Thin Films Materials

  • Effect of Ag additive element on microstructure of $Fe_{87-x}Zr_7B_6Ag_x$, magnetic thin films on Si(001) substrates has been investigated using Transmission Electron Microscopy(TEM) and X-ray Diffraction(XRD). All samples with additive Ag element were made by DC-sputtering and subjected to annealing treatments of $300^{\circ}C{\siim}600^{\circ}C$ for 1 hr. TEM and XRD showed that perfectly amorphous state in Ag-free Fe-based films was observed in as-deposited condition. The as-deposited Fe-based films with the presence of Ag constituent have a mixture of Fe-based amorphous and nano-sized Ag crystalline phases. In this case, additive element, Ag was soluted into Fe-based matrix. With the increase in additive element, Ag, insoluble nano-crystalline Ag particles were dispersed in the Fe-based amorphous matrix. Crystallization of Fe-based amorphous phase in the matrix of $Fe_{82}Zr_7B_6Ag_5$ thin films occurred at an annealing temperature of $400^{\circ}C$. Upon annealing, the amorphous-Ag crystalline state of Fe-Zr-B-Ag films was transformed into the mixture of Ag crystalline phase + Fe-based amorphous phase + ${\alpha}$-Fe cluster followed by the crystallization process of ${\alpha}$-Fe nanocrystalline + Ag crystalline phases.

Production of human insulin analogue using recombinant Escherichia coli

  • For the production of $B^{30}-homoserine$ insulin analog as a novel anti-diabetic drug, the fermentative study was attempted for the maximal gene expression of HTS-fused $B^{30}-homoserine$ insulin precursor in the recombinant Escherichia coli cells. In a batch fermentation, the maximal production of insulin precursor as much as 38.95 mg/L-h, which occupied more than 12.8% of total cell protein. was achieved when the gene expression was induced by 0.5 mM IPTG at the middle logarithmic growth phase. The HTS-fused $B^{30}-homoserine$ insulin precursor was recovered from a batch culture through the processes of cell harvest, collection of insoluble fraction after sonication and purification by nickel affinity column chromatography. The isolated insulin precursor was 14 mg/L with a recovery yield of 35.9% of expressed gene product. The insulin A and B chain mixture was recovered after the insulin precursor was subjected to CNBr cleavage and purified by nickel affinity column chromatography. The isolated insulin chains were then sulfitolyzed with sodium thiosulfat and sodium tetrathionate, and reconstituted to insulin analog with ${\beta}-mercaptoethanol$, followed by purification with CM-Sepharose C-25 column chromatography.

Evaluation of Optimal Culture Conditions for Recombinant Ghost Bacteria Vaccine Production with the Antigen of Streptococcus iniae GAPDH

  • For the production of ghost bacteria vaccine to prevent the streptococcal disease in aquaculture fish species, a double cassettes vector was constructed and cloned in Escherichia coli DH5${\alpha}$. Ghost bacteria vaccine production from Escherichia coli DH5${\alpha}$/pHCE-InaN-GAPDH-Ghost 37 SDM (SIG) was maximized at a glucose concentration of 1 g/l, agitation of 300 rpm, and aeration of 1 vvm. The maximal efficiency of ghost bacteria formation was obtained at the mid-exponential phase ($OD_{600}=2.0$) with the concentration of 0.77 g/l for SIG. The molecular mass of GAPDH was detected at 67 kDa with the insoluble fraction, by SDS-PAGE and Western blot. The protective efficacy of ghost bacteria vaccine was evaluated by challenge test using olive flounder. The cumulative mortalities of the positive control, formalin-killed cell (FKC) vaccine, and SIG vaccine immunized groups were 91%, 74%, and 57%, respectively. These results suggest that SIG vaccine showed efficacy as a vaccine and had a higher potential to induce protective antibodies than did FKC vaccine.
