Hydrocarbon production and nitrogen/phosphorus removal from piggery wastewater by cultivation of Botryococcus braunii

  • The green algae, Botryococcus braunii, has an unusually high hydrocarbon content, ranging from 15 to 75% of dry weight, as a long-chain unsaturated hydrocarbon. It has a potential as a renewable source of chemical feedstocks or fuels. The commercial production of hydrocarbon by B. braunii has not been achieved mainly due to their economic and several technical barriers. The B. braunii cultivation with piggery wastewater could alleviate to economic problems by the reduction of inorganic nutrients, mainly nitrogen and phosphorus from wastewater. We have been studying cultivation of B. braunii in the pretreated piggery wastewater by a MBR(membrane bioreactod, containing about 3000 mg $NO^3$ /L, 15 mg $NH_4\;^+/L$, 3 mg P/L, and others. The purpose of this study was to develop a continuous culture system in piggery wastewater and nitrogen-limited chemostat.

Studies on the artificial regulation of the effective components in garlic by the inorganic nutrients (마늘 유효성분(有效成分)의 무기영양소(無機營養素)에 의한 인위적조절(人爲的調節)에 관한 연구(硏究))

  • This experiment was initiated to determine the effective components of garlic during the growth stages and to regulate the contents artificially. The following results were obtained : 1. Alliin content was increased during its growth and showed drastic increase at maturing stage. 2. The kinds of free amino acids detected during growth stages were 15 for leaf growth, 14 for bulbing and 18 for maturing stage. The total content of free amino acids was high. est at maturing stage. 3. The optimum conditions for the maximum amounts of alliin and free amino acid determined by the binary interaction of Systematic Variations Method were 40% $NO_3^{-1}+60%\;SO_4^{-2}$, 62% $NO_3^{-1}+38%\;PO_4^{-3}$, 42% $K^{+1}+58%\;Ca^{+2}$ and 56% $K^{+1}+44%\;Mg^{+2}$ for alliin, and 72% $NO_3^{-1}+28%\;SO_4^{-2}$, 49% $NO_3^{-1}+51%\;SO_4^{-2}$, 45% $K^++55Ca^{+2}$ and 66% $K^{+1}+34%\;Mg^{+2}$ for free amino acid. 4. Ideal curve for alliin and free amino acid was attained by applying the binary interaction of Systematic Variations Method and it was possible to approach the optimum ionic proportion from the optimum contents on this curve.

Growth and Foliar Constituents of Mulberry ($M_5$) Cultivated under Organic Based Nutrient Management

  • A field experiment to evaluate the effect of application of different organic manures and inorganic fertilizers on growth, yield and quality of leaf was studied during 2004-05 has showed that, the application of 10 kg each of Azospirillum brasilense and Aspergillus awamori+20% each of recommended N through compost+green manure (Glyricidia maculata)+castor cake+vermicompost+Urea and remaining P and K through fertilizers ($T_{11}$) has recorded significantly higher leaf yield (250 g/ plant and 34.70 tonnes/ha/yr, respectively) with improvement in growth characters as compared to control. Leaf quality status was also improved in terms of N (3.19%), P (1.97%), K (1.28%), total soluble protein (8.39 mg/ml), total soluble sugars (14.40 mg/ml), secondary nutrients viz., Ca (3.00%), Mg (0.60%), S (0.35%) and micronutrients viz., Cu (0.410 ppm), Mn (0.454 ppm) and Zn (0.112 ppm) contents. The mulberry grown with 20 tonnes of compost+300:120:120 Kg of NPK/ha/year through fertilizer ranked second for growth and foliar constituents.

The Nutritional Physiology of Red Rotting of Ginseng cause by Erwinia carotovora (인삼적부병원균의 영양생리학적 연구)

  • As a basic study to elucidate nutritional physiology and composition of synthetic medium of red rotting bacteria, Erwinia carotovora, of ginseng, the effects of hydrogen ion concentration, various kinds of carbon sources, nitrogen source, micrometallic salts and it's concentration on the gorwth of the bacteria were investigated and the results were as follows. Optimal pH in the basal medium for the growth of the bacteria was 6.5. After incubation the pH in culture media was neutralized. Among the various kinds of carbon sources, sucrose, glucose mannitol, but organic acids were not utilized effectively as nutrients. After incubation the pH turned acidic. Alanine as organic nitrogen sources nad ammonium sulfate as inorganic nitrogen promoted the growth, but L-valine and sodium nitrite were the least effective. Ferric chloride 1.0mg/dl and ferrous sulfate 100mg/dl were the most effective as micrometallic sources. Control and boric acid were the least effective. New synthetic medium based on the above results was follows ; Alanine 1.0g, $KH_2PO_4\;1.0g, \;sucrose\;30.0g, \;MgSo_4{\cdot}7H_2O\;0.5g, \;FeCl_3{\cdot}6H_2O\;1.0mg\;thiamine\;200{\gamma}g$, and distilled water 1000ml, pH6.5.

Distribution of heterotrophic bacteria and physico-chemical characteristics of sediments in Kum river estuary (금강 하구 퇴적토의 이화학적 성질과 종속영양세균의 분포에 관하여)

  • Vertical distribution of heterotrophic bacteria and physico-chemical characteristics were measuted in Kum River estuarine sediments. And interrelationship between heterotrophic bacterka and environmental factors was also studied. The type of sediment of Site 1 was silty clay, and sand at Site 2. Annual pH ranges were between 7.1 and 7.7 in the clay type sediment (Site 1) and 6.9-7.2 in the sand type sediment (Site 2). It was shown that organic matter contents were higher in the clay type sediment than those of sand type sediment. Redox potential values of sediments were decreased rapidly with depth at Site 1, but those of Sete 2 showed vertical fluctuation. Nitrogens(ammonia+amino acid-N, nitrate-N, nitrite-N) and phosphate in the clay type sediment showed higher values than those of sand type sediment. Annual distribution of heterotrophic bacteria were ranged $6.71{\times}10^4$ cells/g dry wt. $-2.50{\times}10^6$ cells/g dry wt. In the clay type sediment and $2.67{\times}10^3$ cells/g dry wt. $-1.94{\times}10^6$ cells/g dry wt. in the sand type sediment. Distribution of proteolytic, lipolytic, and amylolytic bacteria were decreased with the depth and the highest density was found in April and the lowest in January. Bacterial populations in sediments were closely correlated with such environmental factors as pH, redox potential, moisture content, organic matter contents, and inorganic nutrients such as nitrite-N and phosphate-P.

Selection of New Cultivars with Red Fall Leaf Color in Zelkova serrata Makino as Street Trees (가로수용 적색 단풍 느티나무(Zelkova serrata Makino) 신품종 선발)

  • This study was carried out to develop new cultivars of Z. serrata showing red fall leaf as street trees. Z. serrata which had red fall leaves were selected and then examined for contents of leaf anthocyanin and chlorophyll. In addition, for the progeny test, selected trees were grafted. Of 21 trees having red autumn leaves, three individuals, 'S-6', 'S-20', and 'I-24' were finally selected. They contained higher level of anthocyanin in the leaves. Once developed, the red color remained till late autumn. Grafted plants of the selected strains showed high grafting efficiency and red foliage color in autumn. The soil pH of the survey sites ranged from 5.9 to 7.24. However, other soil characteristics did not show much difference among the sites with regard to inorganic nutrients including N(%), $P_2O_5$, CEC(mg/meq), $K^+$, $Ca^{3+}$, and $Mg^{2+}$, 'S-6', 'S-20', and 'I-24', clonal lines with red fall leaf were selected as new cultivars and propagates by grafting.

Cross-talk between Phosphate Starvation and Other Environmental Stress Signaling Pathways in Plants

  • The maintenance of inorganic phosphate (Pi) homeostasis is essential for plant growth and yield. Plants have evolved strategies to cope with Pi starvation at the transcriptional, post-transcriptional, and post-translational levels, which maximizes its availability. Many transcription factors, miRNAs, and transporters participate in the Pi starvation signaling pathway where their activities are modulated by sugar and phytohormone signaling. Environmental stresses significantly affect the uptake and utilization of nutrients by plants, but their effects on the Pi starvation response remain unclear. Recently, we reported that Pi starvation signaling is affected by abiotic stresses such as salt, abscisic acid, and drought. In this review, we identified transcription factors, such as MYB, WRKY, and zinc finger transcription factors with functions in Pi starvation and other environmental stress signaling. In silico analysis of the promoter regions of Pi starvation-responsive genes, including phosphate transporters, microRNAs, and phosphate starvation-induced genes, suggest that their expression may be regulated by other environmental stresses, such as hormones, drought, cold, heat, and pathogens as well as by Pi starvation. Thus, we suggest the possibility of cross-talk between Pi starvation signaling and other environmental stress signaling pathways.

A Study on the Substrate Removal Efficiency with Varing SRT in Anaerobic Treatment of High Organic Wastewater (고농도 유기성폐수의 혐기성처리시 SRT변화에 따른 유기물질 제거효율에 관한 연구)

  • A synthetic wastewater composed of powdered baby food and inorganic nutrients was treated by five 3.6L anaerobic reactors in order to test the relationship between solids retention time (SRT) and media surface ratio, and the removal efficiencies of organic substrate. Of the five reactors, four were semi-continuously fed stirred-tank reactors and one continuously-stirred batch reactor. The influent was 7430mg/L in COD, 7120 mg/L $BOD_L$ and 6350mg/L in $BOD_5$, respectively. Operating temperature was $35{\pm}1^{\circ}C$ and pH in the range 6.9 to 7.2. In this experimental study it was found that a linear relationship existed, within the experiment range, between SRT and media surface ratio, and that SRT and removal efficiency increased with increasing media surface ratio. The substrate removal efficiencies were 82.7 to $88.2\%$ in COD, 82.9 to $88.4\%$ in $BOD_L$ and 83.3 to $88.7\%$ in $BOD_5$, respectively.

A Study on the Decomposition of Organic Matter and Regeneration of Nutrient in Seawater (해수중 유기물 분해와 영양염 재생의 특성에 관한 연구)

  • This study was conducted to determine the decomposition rate constants of organic matter and to evaluate the process of regeneration of inorganic nutrients in coastal and open seawater. The mixture solution of glucose and glutamic acid, and night soil were used as the test organic matter. Oxygen uptake of test solution was observed every day for 5 days for evaluation of decomposition rate constants, and nutrient contents were analyzed every day for 40 days. The decomposition rate constants have been determined by Thomas slope method and compared with the values of each waters. The values of rate constants for open seawater and coastal water containing the mixture of glucose and glutamic acid were 0.23/day and 0.21/day, and those containing night soil 0.23/day and 0.20/day, respectively. The difference of decomposition rate constants between test materials was not found and the valus for each seawater was equal to each other. The nitrification process took place after 22 days for open seawater when night soil was added to the waters.

Effects of Inorganic Nitrogen released from Roots on the Nitrogen Metabolism (뿌리 방출물중 무기태질소가 체내성분 변이에 미치는 영향)

  • In several leguminous plants such as acasia, arrowroot and bushclover, growth rate and contents of nitrogen, phosphorus and potassium in the tissues and the variation in the culture media were determined. In water cultrue which was free of added nutrients, nitrogen was found to be largely in the form of nitrate(NO3-N). This NO3-N is believed to be the result of nitrification from NH4-N which was apparently released form the plants. From the studies of organ culture with root segments, the amount of nitrogen released and absorbed was found to be proportional to the amount added to the mediuim. Especially, in the N-plot, the amount of nitrogen absorbed by the tissue reached more than 90% of the amount supplied to the medium already in early stage. On the contrary, in the amount free plot, the amount of nitrogen released from the tissue was lower than the minimum level in the N-plot. The amount of total N and P in the cultured tissue was found to be influenced by the amount of nitrogen addedin the medium. However, the amount of K in the tissue was not related to the nitrogen level in the medium, but rather it was influenced by the amount of added potassium. These findings present little difference in the metabolic pattern among the three species plants studied, and suggest that the woody leguminous plants have some common features in tehir metabolic pattern.
