  • The effect of phytohormones, light and phosphate on in vitro production of ubiquinone 10 from the suspension cultures of Nicotiana tabacum cv. Xanthi callus was investigated. The inoculum size and cultured time in the suspension culture had to be at least over 2 % of medium volume at 15 days for the excellent growth of Xanthi callus. The growth of Xanthi callus in the suspension culture was improved by addition of NAA and 2,4-D, especially NAA 1.0mg/1 alone, at the light condition. The optimal concentration of phytohormone was 0.1 mg/l 2.4-D and 1.0 mg/l NAA for productivity of ubiquinone 10 in the suspenseion of Xanthi callus. Addition of 3mM KH$_2$PO$_4$ to the medium was more effective in promoting ubiquinone-l0 formation than other concentration in the light condition. Content and production of ubiquinone-l0 in the suspension cultures of Xanthi callus were the highest at the MS media containing 0.5mg/L kinetin, 0.5mg/L 2,4-D, 1.0mg/1 NAA, 3mM phosphate and 2 % inoculum in the light.

  • Contamination of soils, groundwater, air and marine environment with hazardous and toxic chemicals is major side effect by the industrialization. Bioremediation, the application of microorganism or microbial processes to degrade environmental contaminant, is one of the new environmental technologies. Because of low water solubility and volatility of diesel, bioremediation is more efficient than physical and chemical methods. The purpose of this study is biodegradation of diesel in sand by using Rhodococcus fascians, a microorganism isolated from petroleum contaminated soil. This study was performed in the column containing sand obtained from sea sides. Changes in biodegradability of diesel with various flow rates, inoculum sizes, diesel concentrations, and pH were investigated in sand column. The optimal condition for biodegradation of diesel by R. fascians in sand column system was initial pH 8 and air flow rate of 30 mL/min. Higher diesel degradation was achieved at larger inoculum size and the diesel degradation by R. fascians was not inhibited by diesel concentration up to 5%.

Deodorization Management of Swine-Slurry by Addition of Phototrophic Bacteria (광합성 세균을 이용한 고농도 양돈슬러리의 무취화 관리방안에 관한 연구)

  • This study was carried out to find deodorization effect of swine-slurry by addition of phototrophic bacteria(PTB). The pilot-scale reactors operation conditions was designed by the inoculum amounts of PTB and light-conditions. Treatment conditions was divided into 3 types; 106 MPN/ml$.$Dark(T-1), 108 MPN/ml$.$Dark(T-2), 108 MPN/ml$.$Natural light(T-3). The changes of the concentration of volatile fatty acids(VFAs), hydrogen sulfide(H2S), ammonia (NH3) and odor intensity were analyzed during the treatment period(35 days). From results of this study, the maximum intensity of odor in the headspace of the reactor T-1 was 4.82 and T-2, T-3 was 2.63, respectively. In swine-slurry of reactors used, it almost took 10 days until to be stabilized with solid and liquid phase. Intensity of odor in headspace was mainly derived from the liquid phase. The PTB inoculum method to swine-slurry was very effective in reduction of VFAs, H2S and Sulfate-reducing bacteria(SRB) concentration. Expecially, It was interested in reverse growth behaviour of SRB and PTB in these conditions.

  • Behavior of Listeria monocytogenes in Skim milk during fermentation by Lactobacillus bulgaricus YI-2 and Streptococcus lactis FYI-1 were determined. Autoclaved skim milk was inoculated with ca. 10$^{3}$ L. monocytogenes (Strain LM91-1 or LM 96-2) cells/ml, and with 5.0, 1.0, 0.5 or 0.1% of a milk culture of either L. bulgaricus TI-2 or S. lactis FYI-1. Skim milk containing ca. 10$^{3}$ L. monocytogenes was incubated at 37 or 42$\circ $C for 15 h with L. bulgaricus YI-2, and at 21 or 30$\circ $C for 15 h with S. lactis FYI-1. Cultured skim milks were stored at 4$\circ $C in the refrigerater. Samples were plated on Oxford Agar with oxford antimicrobic supplement to enumerate L. monocytogenes and on either modified MRS agar to enumerate lactic acid bacteria. L. monocytogenes survived the 15-h fermentation with S. lactis FYI-1 in all combinations of level of inoculum and temperature of incubation, but inhibition of growth ranged from 94 to 100%. When incubated with over the 1.0% of L. bulgaricus, L. monocytogenes inhibited or disappeared in fermented skim milk from 9 h after incubation. Especially, incubation at 42$\circ $C with 5.0% L. bulgaricus YI-2 as inoculum appeared to be the most effective inhibitory combination for strain LM 91-1, causing 100% inhibition in growth based on maximum papulation attained. In most instances of incubated with L. bulgaricus YI-2, growth of the pathogene appeared to be completely inhibited when the pH dropped below 4.38.

  • Batch experiments were performed to determine optimum conditions for biopile. The batch experiments results showed that 12.5 to 17.9% of moisture content was effective to biodegradation of petroleum hydrocarbon regardless of soil texture. Total heterotrophic bacteria populations in the inoculum-treated soil were greater than of the control and nutrient-amended soil in the early stage, but the populations in the inoculum and nutrient-amended soil were not different significantly from those in the latter stage regardless of soil texture. The same trend was observed for petroleum hydrocarbon degrading bacteria populations. The results of the biodegradation capacity experiments showed that there was a decline in the TPH concentrations during the experiments and no significant difference on the biodegradation was observed by treatment in silt soil. Changes of n-C17/pristane and n-C18/phytane ratios in all treated soil were significantly more than those of control. This is a strong indication of biodegradation. The TPH removal rate was calculated at 60% in all treated soil.

  • This study was carried out to measure the antibacterial effect of lactoperoxidase/ thiocyanate/hydrogen peroxide system (LP system) against E. coli 0157:H7. When the initial inoculum levels (($10^{2},\;10^{4},\;10^{7}cfu/ml$), concentration of LP (10 ppm, 20 ppm, 30 ppm), culture media (TSB-YE, UHT milk, raw milk) and storage temperatures ($5^{\circ}C,\;10^{\circ}C,$15$^{\circ}C$) were set up differently for the experiment and the antibacterial effect was compared, the highest antibacterial effect of LP system was shown at $10^{2}cfu/ml$ of initial inoculum level, 10 ppm of LP concentration and $5^{\circ}C$ of incubation temperature.

Statistical Optimization for Improved Production of Cyclosporin A in Solid-State Fermentation

  • This work evaluates the effect of different amino acids on production of Cyclosporin (CyA) production in solid-state fermentation that was previously optimized for different fermentation parameters by one factor at-a-time for the maximum production of CyA by Tolypocladium inflatum MTCC557. Based on the Plackett-Burman design, glycerol, ammonium sulfate, $FeCl_3$, and inoculum size were selected for further optimization by response surface methodology (RSM). After identifying effective nutrients, RSM was used to develop mathematical model equations, study responses, and establish the optimum concentrations of the key nutrients for higher CyA production. It was observed that supplementation of medium containing (% w/w) glycerol, 1.53; ammonium sulfate, 0.95; $FeCl_3$, 0.18; and inoculum size 6.4 ml/5g yielded a maximum of 7,106 mg/kg as compared with 6,480 mg CyA/kg substrate using one factor at-a-time. In the second step, the effect of amino acids on the production of CyA was studied. Addition of $_L$-valine and $_L$-leucine in combination after 20 h of fermentation resulted in maximum production of 8,166 mg/kg.

Sporulation of Pyricularia grisea at Different Growth Stages of Rice in the Field

  • Sporulation patterns of rice blast fungus were studied at relatively later stages of leaf blast and neck blast seasons in Icheon, Korea. This experiment was done by detaching lesion-bearing leaves and panicle bases. The number of conidia remaining on the leaf blast lesions of different cultivars from Jul 20 to Jul 23 ranged from 3,640 to 82,740 spores. More conidia were observed on the adaxial surface because they were released from abaxial surface. After heading, sporulation was observed from the lesions on the flag leaves but the number of spores was less than in the late July. Detached panicle bases or uppermost internodes infected by Pyricularia grisea produced abundant amount of conidia. Among these panicle bases, 30.1 mm size lesion recorded the highest count of 244,560 spores. When we compared the sporulation amount using the KY-type spore trap, more conidia were recorded from intact lesions than from the lesions which removed conidia and conidiophore The ratio of conidia release against total sporulation ranged from 20.5%-25.0% for leaf blast and 8.2%-25.3% in the neck blast. Effective inoculum potential was also discussed.

Identification of the Imperfect Stage of Mycosphaerella nawae Causing Circular Leaf Spot of Persimmon in Korea (감나무 둥근무늬낙엽병균 Mycosphaerella nawae의 불완전 세대 동정)

  • Asexual spores of Mycosphaerella nawae were profusely produced on PDA after a prolonged incubation at $25^{\circ}C$ for 90 days. When persimmon trees were artificially inoculated by the conidial suspension, typical symptoms of circular leaf spot of persimmon appeared on the leaves two month later. The imperfect stag of the fungus was identified as Ramularia sp. based on following morphological characteristics examined under a light microscope and a scanning electron microscope. Conidia were mostly ellipsoid, but occasionally cylindrical, elongated oval, taro, peanut or gourd shapes and measured as 12.2~32.6$\times$6.1~10.2 ${\mu}{\textrm}{m}$. erect, hyaline, colorless-light brown. Conidia were formed solitarily or in chains on a medium and infected leaves. Conidiophore was erect, hyaline, colorless-light brown. and the size was 20.4~102.0$\times$3.1~10.2 ${\mu}{\textrm}{m}$, respectively. In this paper, we firstly demonstratrated that asexual spores of M. nawae induced persimmon circular leaf spot in nature as well as sexual spores of the fungus. Therefore, it is hypothesized that the imperfect stage of the fungus plays an important role in nature for epidemics as secondary inoculum.

A Forecast Model for the First Occurrence of Phytophthora Blight on Chili Pepper after Overwintering

  • An infection risk model for Phytophthora blight on chili pepper was developed to estimate the first date of disease occurrence in the field. The model consisted of three parts including estimation of zoosporangium formation, soil water content, and amount of active inoculum in soil. Daily weather data on air temperature, relative humidity and rainfall, and the soil texture data of local areas were used to estimate infection risk level that was quantified as the accumulated amount of active inoculum during the prior three days. Based on the analysis on 190 sets of weather and disease data, it was found that the threshold infection risk of 224 could be an appropriate criterion for determining the primary infection date. The 95% confidence interval for the difference between the estimated date of primary infection and the observed date of first disease occurrence was $8{\pm}3$ days. In the model validation tests, the observed dates of first disease occurrence were within the 95% confidence intervals of the estimated dates in the five out of six cases. The sensitivity analyses suggested that the model was more responsive to temperature and soil texture than relative humidity, rainfall, and transplanting date. The infection risk model could be implemented in practice to control Phytophthora blight in chili pepper fields.
