Using 200 porcine colon tissues, the efficiencies of three isolation methods of Campylobacter from porcine intestines were compared: Method 1, direct streaking of colon mucosa; Method 2, direct inoculation of intestinal contents with a swab; Method 3, inoculation of pre-enriched medium. A total of 460 Campylobacter isolates were obtained from 178 samples (89%) by direct streaking of colon mucosa, 142 samples (71%) by direct streaking of a swab, and 94 samples (47%) by pre-enrichment of intestinal contents in Preston broth. Direct streaking of colon mucosa was superior to the other two isolation methods, in terms of rapidity and higher efficiency. When isolates were identified with various biochemical tests and PCRs specific to 16s rRNA, mapA, and ceuE, C. coli was the predominant species (87%) in porcine, whereas the rest of the isolates were identified as C. lanienae.
The cultivation in sawdust media, mushroom productivity, and fruiting body characteristics of Lentinula edodes strains NIFoS 2778 and NIFoS 3363 were compared according to the inoculation conditions. The cultivation period was 5% shorter when liquid spawn was used. Fruiting bodies were induced after 113 days of incubation on media inoculated with liquid spawn, and the cultivation period was 119 days on media inoculated with solid spawn. Mushroom productivity of NIFoS 2778 was the highest (661.4 g) when 36 mL of liquid spawn was used. For NIFoS 3363, mushroom production was higher under liquid inoculation conditions when the same amount of liquid and solid spawns were used. The mushroom characteristics of the two strains were not significantly different, except for gill width and stipe diameter.

Black rot caused by Xanthomonas campestris pv. campestris (Xcc) is one of the most serious diseases of crucifers world-wide. To establish the efficient screening method for resistant cabbage to Xcc, different inoculation methods, inoculation positions, growth stages of seedlings, and incubation temperatures after inoculation were investigated with the seven cabbage cultivars showing different resistance degrees to the pathogen. Clipping with mouse-tooth forceps was better inoculation method than piercing with 18 pins or cutting with scissors to distinguish the level of resistance and susceptibility. In inoculation using mouth-tooth forceps, clipping the edges of the leaves near veins is more effective than injuring the veins of the leaves directly. In addition, the inoculated plants kept at $22^{\circ}C$ showed more clear resistant and susceptible responses than those kept at 26 or $30^{\circ}C$. On the basis of the results, we suggest that an efficient screening method for resistance of cabbage cultivars to black rot is to clip the edges of the leaves near veins of the four-week-old seedlings with mouth-tooth forceps dipped in a suspension of Xcc at a concentration of $7{\times}10^7$ cfu/ml and incubate the inoculated plants in a growth room at $22^{\circ}C$ with 12-hr light a day.

It was conducted to evaluate the control efficacy of Bordeaux mixture on pepper anthracnose in vitro and in vivo. The suppressive efficacy of Bordeaux mixture on mycelial growth and spore germination of Colletotrichum acutatum was investigated on PDA, cellophane membranes and pepper fruits, respectively. Furthermore, its control value was evaluated on detached pepper fruits inoculated with C. acutatum by wound and non-wound inoculation method, and in fields. The mycelial growth of C. acutatum JC24 was inhibited by 90.0% on PDA amended with $937{\mu}g\;mL^{-1}$ of Bordeaux mixture. While the spore germination of C. acutatum JC24 was inhibited perfectly on cellophane membranes treated at $187{\mu}g\;mL^{-1}$ of Bordeaux mixture, that on fruits inoculated with the pathogen by wound inoculation and non-wound inoculation method was inhibited by 88.1% and 91.3%, respectively. Although the control value on fruits treated with $937{\mu}g\;mL^{-1}$ of Bordeaux mixture was 17.6% in wound inoculation method, it was 58.8% in non-wound inoculation method. In fields, when Bordeaux mixture was sprayed five times at 14 day-intervals, it showed 55.7% and 61.7% of control value in 2012 and 2013, respectively. We think Bordeaux mixture was able to use as an eco-friendly organic farming material to control pepper anthracnose based on the above-mentioned results.
Alfalfa (Medicago sativa L. cv. Luna) seeded in agar was inoculated with two strains of Rhizobium meliloti isolated from root nodules of alfalfa for assessment of nodulation. The seedling growth after six weeks was remarkably increased by adding each rhizobia strains into agar media and also by nitrate application (70ug N/ml), but there was no significant difference among them. Nodulations started one week after inoculation and increased its numbers and sizes as seedling grew. Therefore, the two strains isolated from alfalfa root were concluded to be effective strains. For determining seed inoculation method the same cultivar was inoculated with both rhizobia strains using different inoculation methods such as broth-vacuum, peat-adhesive, peat & lime pelleting. They were seeded in pots of river sand and supplied with culture solution excluded nitrogen. The peat & lime pelleting was recognized the best method in both of nodulation and seedling growth after eight weeks growth. There were significant correlations between the weight of nodules and the shoot or root dry weight of alfalfa in both rhizobia strains.
Fusarium head blight (FHB) is a severe disease problem that affects the quality and yield of barley grain. The evaluation of FHB resistance is difficult because environmental conditions greatly influence FHB infection and development. The objectives of this study were to: 1) establish an efficient screening method for selecting resistant barley to FHB, 2) compare FHB severity between the cut-spike method and pot-plant method for development of mass screening, and 3) estimate FHB resistance for barley germplasms. Barley cultivars and lines were evaluated for reaction to FHB in controlled-greenhouse condition. Spikes were spray-inoculated with a suspension $(5.0\times10^5\;macroconidia\;mL^{-1})$ of Fusarium graminearum SCK-O4 strain, and then kept in a greenhouse at $18-25^{\circ}C$ with $80-100\%$ relative humidity. Inoculation were employed at 3 different heading growth stages (heading date, three days after heading, and five days after heading). The inoculation was performed in 2 consecutive days in order to avoid escapes. The inoculated plants were maintained in the greenhouse at 4 different free moisture periods (1, 3, 5, and 7 days). The percentage of FHB severity was scored from 0 to 9 according to the rate of infected kernels per spike, and three spikes were evaluated per replication with 3 replicates. There were significant differences of FHB severity depending on the different free moisture periods, but not by the inoculation at different heading stages. The optimum evaluation point of FHB severity in the greenhouse condition was on the 7th day under free moisture condition after inoculation at the heading date. Infection level in cut-spike method highly correlated with that in pot-plant method. This suggested that cut-spike method is useful in evaluating of FHB resistance in barley. Six cultivars, such as Jinkwang, Buheung, Atahualpha 92, Chevron-b, Gobernadora-d, and MNBrite-c, were selected as resistant varieties to FHB. Correlation coefficient for the FHB severity evaluated by the pot-plant method between two seasons was 0.794, indicating the stability and accuracy of the screening method.

Colonization of Pseudomonas chlororaphis O6, a nonpathogenic rhizobacterium, on the roots induced systemic resistance in cucumber plants against tai-get leaf spot, a foliar disease caused by Corynespora cassiicola. A cDNA library was constructed using mRNA extracted from the cucumber leaves 12 h after inoculation with C. cassiicola, which roots had been previously treated with O6. To identify the genes involved in the O6-mediated induced systemic resistance (ISR), we employed a subtractive hybridization method using mRNAs extracted from C cassiicola-inoculated cucumber leaves with and without previous O6 treatment on the plant roots. Differential screening of the cDNA library led to the isolation of 5 distinct genesencoding a GTP-binding protein, a putative senescence-associated protein, a galactinol synthase, a hypersensitive-induced reaction protein, and a putative aquaporin. Expressions of these genes are not induced by O6 colonization alone. Before challenge inoculation, no increase in the gene transcriptions could be detected in previously O6-treated and untreated plants but, upon subsequent inoculation with the pathogenic fungus, transcription levels in O6-treated plants rose significantly faster and stronger than in untreated plants. Therefore, the O6-mediated ISR may be associated with an enhanced capacity for the rapid and effective activation of cellular defense responses which becomes apparent only after challenge inoculation on the distal, untreated plant parts, as suggested by Conrath et al. Pine wood nematode (PWN), Bursaphelenchus xylophilus is one of the most serious pests of pine tree. Trunk injection of some nematicides in tree is well known as an effective control method. However there are some limitating factors which hindering the efficacy of trunk injection in field or potted tree. In this study we suggested easy and useful alternative screening methods of nematicides against PWN. Reproduction of PWN was influenced by tree twig moisture (high reproduction in high moisture trig) and paraffin coating was 78.6% reduced moisture loss in tested twig. There were no reproduction different in up and down site from infection site of twig at 1 month after inoculation of PWN and also distance (5 and 10 cm) from inoculation site of PWN did not influence the reproduction of PWN. Numbers of reproduced PWN were higher with decreasing diameter of twig. Numbers of reproduced PWN were similar to P. thunbergii and P. densiflora. However reproduction was increased depending on high inoculation density and longer propagation period. When inoculation of PWN on cut twig injected with emamectin benzoate 2.15% EC and morantel tartrate 8% SL in trunk of Pinus thunbergii in the field, PWN number were significantly reduced than untreated control. We suggest this screening method for PWN control agents.
This experiment was conducted to investigate the effects and properties of organic materials, the fermented fowl droppings composts and the top viviparous malleatuses, utilizable for rice farming. At the 35th day after transplanting, the growth of rice plants was good in the order of organic farming, combined farming, conventional farming, and control plot. But stem number and dry weight of rice plant in organic farming decreased a little compared to those in conventional farming at the 65th day after transplanting. Rice yield decreased by 7% in organic farming and by 4% in combined farming compared to those of conventional farming. Weeding effect by inoculation of 5kg or 10kg of the top viviparous malleatuses per 10a was similarly 96% at the immediately inoculation after transplanting, 98.6% at the 7th days inoculation after transplanting and 70% at the 15th days inoculation after transplanting. By inoculation of 5kg per 10a at the 7th day and the 15th day after transplanting, rice yields decreased by 2% and 5%, respectively, compared to those of the conventional farming method. From the above results, it seems that the organic farming and the combined farming are useful and the inoculation of the top viviparous malleatuses is effective for control of weeds as the environmental preservation farming.
