Synergic Effects of Mixed Formula Consisted of Polycan and Calcium-gluconate on the Experimental Periodontitis and Alveolar Bone Loss in Rats

  • Objective : Polycan, exopolymers purified from Aureobasidium pullulans SM-2001 and calcium gluconate have been showed favorable inhibitory effects on the periodontitis and related alveolar bone losses through antioxidant and anti-inflammatory activities, respectively. In the present study, we intended to observe the possible synergic effects of mixed formula consisted of Polycan and calcium gluconate on ligation-induced experimental periodontitis and related alveolar bone losses in rats, and to select the fittest compositions for further developing as effective agents to ameliorate periodontal diseases. Method : Experiments were conducted as two separated two tests - first is synergic effects of Polycan and calcium gluconate 1:1, 1:9 and 9:1 mixtures, and second is 1:99, 2:98, 4:96, 8:92 and 1:9 mixtures. Experimental periodontal diseases were induced by ligature placed around the cervix of upper left incisior teeth of rats. One day after ligation placements, 200mg/kg of each single or mixed formulas of Polycan or/and calcium gluconate were orally administered for 10 days. The changes on the alveolar bone loss index and maxillary bone mineral density (BMD) were observed for detecting alveolar bone losses, and for anti-inflammatory effects, myeloperoxidase (MPO) activities and proinflammatory cytokine (tumor necrosis factor; TNF-${\alpha}$) contents were also evaluated in gingival tissues around ligature placed incisior teeth. The results of mixtures were compared with those of singe Polycan and calcium gluconate treated rat. Results : Each single or mixed formulas of Polycan or/and calcium gluconate favorably and significantly inhibited the inflammatory changes. The inhibitory effects of mixed formula consisted of Polycan and calcium gluconate 1:9 showed against periodontitis and related alveolar bone losses as compared with those of each Polycan and calcium gluconate single formula (p<0.05). In second experiment, Polycan and calcium gluconate 2:98, 4:96, 8:92 and 1:9 mixed formulas also showed significant increased anti-inflammatory and inhibitory effects against alveolar bone losses as compared with those of each single formula. Among them, Polycan and calcium gluconate 2:98 showed the highest efficacy against to ligation-induced experimental periodontitis and related alveolar bone losses. Conclusion : The results obtained in this study suggest that appropriated mixtures of Polycan and calcium gluconate showed synergic inhibitory effects against ligation-induced experimental periodontitis and related alveolar bone losses in rats. Moreover, Polycan and calcium gluconate 2:98 showed the highest efficacies in this experiment, suggesting the fittest composition for further developing as effective agents to ameliorate periodontal diseases.

Changes of Index Microorganisms and Lactic Acid Bacteria of Korean Fermented Vegetables (Kimchi) during the Ripening and Fermentation-Part 1 (김치의 숙성 및 발효중 오염지표미생물과 유산균의 변화-제1보)

  • This study was undertaken to investigate the changes of index microorganisms and lactic acid bacteria of traditional Korean fermented vegetables (kimchi) during the ripening and fermentation period. A type of kimchi, baechoo-kimchi, was prepared and stored at $10^{\circ}C$ for 8 days. The numbers of the total aerobic bacteria, psychrotrophilic bacteria, coliform bacteria, and Escherichia coli in the kimchi and also in raw materials of the kimchi (Chinese cabbage, green onion, ginger, garlic, and red pepper) were counted using appropriate media. The highest number of aerobic bacteria was detected from ginger, then red pepper, then garlic, then Chinese cabbage, and lowest number from green onion. The highest number of psychrotrophilic bacteria was detected from red pepper, then Chinese cabbage, then garlic, then ginger, and the lowest number from green onion. Coliforms and E. coli were not detected from all of the raw materials of kimchi. Total aerobic bacteria and lactic acid bacteria of the kimchi showed gradually increasing during ripening and fermentation. The number of psychrotrophilic bacteria showed a similar level in the kimchi. Coliform bacteria were detected at the 3rd, 4th, and 5th day of the kimchi fermentation period, although they were not detected from the raw materials of the kimchi. However, the bacteria were not detected in the kimchi after 6 days. E. coli was not detected in all kimchi samples. The pH value of the kimchi gradually decreased, and acidity increased over fermentation period. This study indicates that there was contamination of coliform bacteria during the process of kimchi preparation, and lactic acid bacteria proliferated in the kimchi during fermentation inhibited the growth of coliforms. More research is needed to evaluate the inhibitory effects of each raw materials of kimchi.

Comparative Study on Chemical Composition and Biological Activities of Samchulkunbi-tang Decoction and Commercial Herbal Medicine (삼출건비탕 전탕액과 시판 한약제제의 비교 연구)

  • Objectives : The purpose of this study was to investigate the differences of chemical composition or biological activities between decoction and commercial herbal medicines of Samchulkunbi-tang (Shenzhujianpi-tang, SKT). Methods : The extracts of SKT from decoction (SKT1) and two different commercial extractive granules (SKT2 and SKT3) were prepared. The index components of SKTs were analyzed with HPLC. The antioxidant activities of SKTs were studied by measuring free radical scavenging activities on ABTS and DPPH. The anti-inflammatory effects were determined by measuring NO, $PGE_2$ and IL-6 in LPS-stimulated RAW 264.7 cells. Results : The contents of 7 components were 1.40-6.08 mg/g in SKT1, not detected-4.75 mg/g in SKT2, 0.03-1.46 mg/g in SKT3. The scavenging activities on ABTS and DPPH of herbal formulas were increased in dose-dependent manner (SKT1>SKT2>SKT3). SKT1 significantly inhibited $PGE_2$ and IL-6 production and SKT3 slightly inhibited $PGE_2$ production in LPS-stimulated RAW264.7 cells. SKT2 showed no inhibitory effects on production of inflammatory mediators such as $PGE_2$ and IL-6. Conclusions : These results demonstrate that the decoction of SKT has more strong anti-oxidant and anti-inflammatory effects than that of commercial herbal medicines consistent with the contents of index components.

Effects of Cervi Pontotrichum cornu and Carthami semen on the Experimental Osteoporosis Induced by Ovariectomy in Rats (녹각(鹿角)과 홍화자(紅花子)가 난소적출에 의한 실험적 흰쥐 골다공증 모델에 미치는 영향)

  • This study was carried out to investigate the effects of Cervi Pontotrichum cornu and Carthami semen on the serum osteocalcin level, bone mineral density, bone trabecular area and cortical thickness index in ovariectomized rats. Femal Sprague-Dawley rats were divided into 5 groups. Non-ovariectomized groups were divided into commertial diet fed(normal group) and calcium free basal diet fed(sham operation group). Ovariectomized groups were divided into 3groups, calcium free basal diet fed group(control group or ovariectomized group), 10% Cervi Pontotrichum cornu supplemented diet fed group and 10% Carthami semen supplemented diet fed group. After 4 weeks diet fed, serum osteocalcin level, bone mineral density, bone trabecular areas and cortical thickness index were analyzed. The bone mineral density and bone trabeculae area were significantly increased by Cervi Pontotrichum cornu and Carthami semen supplemented diet fed groups. The osteocalcin level was significantly decreased by Cervi Pontotrichum cornu and Carthami semen supplemented diet fed groups. The trabecular area(%) in epiphysis of tibia was significantly increased by Cervi Pontotrichum cornu and Carthami semen supplemented diet fed groups. The cortical thickness index in distal diaphysis of tibia was significantly increased by Cervi Pontotrichum cornu and Carthami semen supplemented diet fed groups. the alkaline phosphatase level was significantly decreased by Cervi Pontotrichum cornu and Carthami semen supplemented diet fed groups. These results suggest that Cervi Pontotrichum cornu and Carthami semen might have inhibitory effects on osteoporosis by increasing bone mineral density and accerelating bone formation in ovariectomized rats.

Effects of Cheonggukjang and Doenjang on Bone Loss in Ovariectomized Rats

  • The effects of cheonggukjang and doenjang on bone mineral density, trabecular area and cortical thickness of the tibia, and serum osteocalcin level in ovariectomized rats were investigated. After 4 weeks, bone mineral density, bone trabecular area, the cortical thickness index, and serum osteocalcin level were analyzed. The cheonggukjang and doenjang diet groups showed significant prevention of ovariectomized (OVX)-related body weight gain. Whole body bone mineral density of the OVX group was significantly lower than that of the sham group, whereas the cheonggulgang and doenjang diets resulted in complete restoration of bone mineral density. Trabecular area in the proximal diaphysis and cortical thickness in the distal diaphysis of the tibia were increased significantly in the cheonggukjang and doenjang diet fed groups. The cheonggukjang and doenjang diets significantly reduced serum osteocalcin level in the OVX rats. These results suggest that cheonggukjang and doenjang might have inhibitory effects on osteoporosis, by showing accelerated bone formation in OVX rats.

Antimicrobial effect of natural resources on Enterococcus faecalis CHJ-S isolated from oral (구강에서 분리한 Enterococcus faecalis CHJ-5에 대한 생약채의 항균효과)

  • This study was conducted to investigate the effect of natural resources against growth of oral bacteria. The CHJ-5 strain was isolated from oral cavity of dental patients. Similarity index values of oral bacterium CHJ-5 was 0.876 to E. faecalis by cellular fatty acid analysis(Analytical Services Inc., USA). As a result of investigation about the growth inhibition of 32 kinds of natural resources on E. faecalis CHJ-5. This strain was inhibited by Schizamdrae fructus, Coptidis rhizoma and Caryophylli flos. Minimal inhibitory concentrations of Schizamdrae fructus, Coptidis rhizoma and Caryophylli flos were 0.1%, 02% and 0.1% on E. faecalis CHJ-5, respectively.

The Effects of Gami-Cheongshimyeonjayeum of the anti-Allergic and immune response to rats and nice (加味淸心蓮子飮 煎湯液이 抗 ALLERGY反應과 MOUSE의 免疫細胞機能에 미치는 影響)

  • The object of thes research is to lucidate the clinical effects of Gami-Cheongshimyeonjayeum on the anti-allergic and gimmune response to rats and mice. The obtaind results of Gami-Cheongshimyeonjayeum administration are as follows : 1. The cytotoxic index increased slightly at over concentration $1{\times}10^{-2}/ml$, but it is thought that the cytotoxic effect show no significant value under the concentration $1{\times}10^{-3}/ml$. 2. Gami-Cheongshimyeonjayeum water extract seems to suppress the leakage volume of Evans blue to decrease the vascular permeability. 3. Gami-Cheongshimyeonjayeum show the inhibitory effect of footpad edema. 4. Protein leakage is inhibited by the administration of Gami-Cheongshimyeonjayeum. 5. The delayed type hypersensitivity to SRBC is suppressed at the 24hours after the administration of Gami-Cheongshimyeonjayeum on the experimental group 1, 2. 6. Hemolysin and hemagglutinin titer, and RFC increase to bulid up the immune function.

The Highest Dosage Combination Activity Screening from the Leaf Fraction of Melastoma malabathricum with Antibiotic Gentamicin and Ciprofloxacin

  • Objectives: This study aims to determine the Fractional Inhibitory Concentration Index (FICI) of combinations of Melastoma malabathricum leaf fraction with ciprofloxacin or gentamicin against pathogenic bacteria, Escherichia coli, Staphylococcus aureus, and Bacillus cereus, isolated from Diabetic Foot Ulcer (DFU) patients. Methods: We tested concentrations of 45%, 55%, 65%, and 75% of gentamicin and ciprofloxacin using dilution and agar diffusion methods. The combination of M. malabathricum leaf extract with these antibiotics was tested in vitro against all three bacteria. Results: The combination of M. malabathricum leaf extract and ciprofloxacin gave a FICI value of 0.5, indicating synergistic antibacterial activity against the test bacteria. Conclusion: The results show that the antibacterial effect of a combination of high doses of the leaf extract with either antibiotic is greater than that of the leaf extract and the antibiotics in single use.

Inhibitory Effects of Kochujang Extracts on the Tumor Formation and Lung Metastasis in Mice

  • Effects of kochujang (Korean red pepper soybean paste) extracts on tumor formation, natural killer (NK) cell activity in spleen and glutathione S-transferase (GST) activity in liver were investigated in the sarcoma-180 cell transplanted mice. Inhibitory effects of these samples on lung metastasis of colon 26-M3.1 cells were also evaluated in the Balb/c mice. The injection of methanol extracts from traditional kochujang I (TK I, 0-day fermented), II (TKII, 6-month fermented), commercial kochujang (CK, 1-month fermented) and red pepper powder (RPP) significantly reduced tumor formation in Balb/c mice (p<0.05), TKII decreased tumor growth by 46% compared with control, resulting in the smallest tumor weight. The transplantation of sarcoma-180 cells increased the spleen/body weight ratio of Balb/c mice, while TKI and TKll significantly decreased this index (p<0.05). The effect of TKll and CK, fermented kochujang, on the NK cell activity of splenocytes was higher than that of sarcoma-180 cells transplanted control group. TK II recovered the activity of hepatic GST that was decreased by the transplantation of sarcoma- 180 cells in to the mice. All kochujang-treated mice had significantly fewer lung metastatic colonies than control mice. TKII was the most effective in inhibiting lung metastasis of colon 26-M3.1 cells. These results indicated that optimally ripened (6-month) TK had more suppressive effects on tumor formation and lung metastasis than RPP and kochujang without fermentation and commercially prepared kochujang in mice.

In vitro Antibacterial and Synergistic Activity of an Ecklonia cava Extract against Anti biotic-Resistant Streptococcus parauberis

  • In an effort to discover alternative phytotherapeutic antimicrobial agents to combat Streptococcus parauberis, a fish pathogenic bacterium, we evaluated the antibacterial activity of seaweed extracts in vitro. A methanolic extract of Ecklonia cava exhibited strong antibacterial activity against S. parauberis isolated from olive flounder Paralichthys olivaceus. Furthermore, the n-hexane soluble (Hexane) fraction of the E. cava methanolic extract exhibited the greatest antibacterial effect on S. parauberis strains with a minimum inhibitory concentration (MIC) ranging from 256 to $1,024{\mu}g/mL$. In addition, the MIC values of oxytetracycline against antibiotic-resistant S. parauberis were markedly reduced up to 64-fold in combination with the Hexane fraction, suggesting that the antibacterial activity of the antibiotic was restored when combined with the Hexane fraction. The interaction between both antibiotics and the Hexane fraction was assessed by the fractional inhibitory concentration (FIC) index. The Hexane fraction and oxytetracycline combination against antibiotic-resistant S. parauberis strains resulted in a median ${\sum}FIC$ range of 0.502 to 0.516. Thus, the synergistic ranges of median ${\sum}FIC$ < 1 were observed for all combinations of the Hexane fraction and oxytetracycline against S. parauberis. To the best of our knowledge, this is the first report indicating the efficacy of an E. cava extract against fish pathogenic bacterium S. parauberis.
