Isolation and Characterization of Soil Microorganism Producing Acyl CoA Synthetase Inhibitor (Acyl CoA 합성효소 억제제를 생산하는 토양균의 분리 및 특성 연구)

  • Identification of soil microorganism strain B-6. a producer of acyl CoA synthetase inhibitor, based on its morphological, physiological, biochemical and chemotaxonomical charact eristics was performed. The strain B-6 was identified as Bacillus subtilis. Ihe acyl CoA synthetase inhibitor produced by this strain was highly achieved in fermentation medium that contained glucose 1.0%, soluble starch 1.0%, NH$_4$Cl 0.3%, oatmeal 1.0%, pharmamedia 1.0%, basic magnesium carbonate 0.5%. pH 7.5 at 30$^{\circ}$C for 7 days. The optimal pH and temperature for growth were 9.0 and 30$^{\circ}$C, respectively. Butanol extract of culture filterate of strain B-6 in acyl CoA synthetase inhibitor production medium containing corn steep liquor exhibited high acyl CoA synthetase inhibitor activity and antimicrobial activity against C. albicans. But chloroform extract of culture filterate of strain B-6 in medium containing NH$_4$Cl, ($NH_4)_2SO_4$ or urea instead of corn steep liquor exhibited higher antimicrobial activity against C. albicans than that of butanol extract.

Correlations Between Polyamine and Adventitious Shoot Regeneration from Populus Leaf Segments (포플라 잎절편의 부정아 분화와 Polyamine 함량의 연관성)

  • Polyamine contents and the activities of their main biosynthetic enzymes, arginine decarboxylase (ADC) and ornithine decarboxylase(ODC), were investigated in Populus leaf segments during adventitious shoot regeneration with the addition of 1 millimolar polyamine synthesis inhibitors. From the study of polyamine synthesis inhibitors, ODC was found to be the principal route of polyamine biosynthesis in adventitious shoot regeneration of Populus leaf segments. The ADC inhibitor difluoromethyl arginine(DFMA) and ODC inhibitor difluoromethyl ornithine(DFMO) strongly reduced the putrescine content. On the contarary, DCHA, an inhibitor of spermidine synthase, increased it. Spermidine content was decreased with the treatment of each polyamine synthesis inhibitor, but the inhibitory effect of DCHA was stronger than any other polyamine inhibitor. The decreased polyamine level by polyamine synthesis inhibitors was restored with the exogenously applied polyamine. Comparing the polyamine contents with the adventitious shoot regeneration rate, were observed a close correlation between spermidine content and adventitious shoot regeneration of Populus leaf segments.

  • The protein, called PTP1B (protein tyrosine phosphatase 1B), joins a list of enzymes that mice are associated with obesity. The purpose of this study was to investigate the effects of PTP1B inhibitors and taurine on blood lipid profiles in adolescents obesity model rats. A New Soybean Cultivar "Gaechuck#1" : Black Soybean Cultivar with Lipoxygenase2,3-free, Kunitz Trypsin Inhibitor-free and Green Cotyledon

  • Lipoxygenase and Kunitz trypsin inhibitor protein of mature soybean [Glycine max (L.) Merr.] seed are main anti-nutritional factors in soybean seed. A new soybean cultivar, "Gaechuck#1" with the traits of black seed coat, green cotyledon, lipoxygenase2,3 and Kunitz trypsin inhibitor protein free was developed. It was selected from the population derived the cross of "Gyeongsang#1" and C242. Plants of "Gaechuck#1" have a determinate growth habit with purple flowers, brown pubescence, black seed coat, black hilum, oval leaflet shape and brown pods at maturity. Seed protein and oil content on dry weight basis have averaged 39.1% and 16.2%, respectively. It has shown resistant reaction to soybean necrosis, soybean mosaic virus, Cercospora leaf spot and blight, black root rot, pod and stem blight, and soybean pod borer. "Gaechuck#1" matured on 5-10 October with a plant height of 50 cm. The 100-seed weight of "Gaechuck#1" was 23.2g. Yield of "Gaechuck#1" was averaged 2.2 ton/ha from 2005 to 2007.

Characters of proteinase inhibitor isolated from streptomyces fradiae (Streptomyces fradiae에서 분리한 단백질 분해효소저해물질의 특성)

  • The objective of the current study is to elucidate the biological roles of proteinase inhibitor in microorganisms. As the first step, a strain of Streptomyces fradiae was selected as a producer of extracellular proteinase inhibitor. The proteinase inhibitor was purified from culture broth through ultrafiltration, gel-filtration and ion-exchange chromatography. Molecular weight of the proteinase inhibitor was estimated to be 16, 800 by SDS polyacrylamide gel electrophoresis. It was found that the proteinase inhibitor inhibited only alkaline serine proteinases such as subtilisin, $\alpha$-chymotrypsin and Promase E but not trypsin and other proteinases. The mode of inhibition against Pronase E with succinyl-phenylalanine-p-nitroanilide as a substrate was competitive.

  • To screen agent for the treat-ment of Alzhimers Disease several strains of bacteria producing acetylcholinesterase inhibitor ware isolated from soil. Strain 960903 showed strong acetylcholinesteras inhibitory activity and low butyrylcholinesterse inhibitory activity. The strain 960903 was identified as Pseudomonas sp. Acetylcholinesterase inhibitor ws highly achieved in fermentation medium containing soluble starch 3.0%, glycerol 1.0%, pharmamedia 0.5%, KCI 0.3%, $CaCO_3$ 0.2%, MgS $O_4$..$7H_2$O 0.05%, $KH_2$$PO_4$ 0.05%(pH6.5) at $30^{\circ}C$ for 4 days. Acetylcholinesterase inhibitor was purified by Diaion WA-30($OH^{-}$) column charomatography and cellulose column chromatography. Acetylcholinesterase inhibi-tor showd the maximum wavelength at 205 nm and was soluble in water, acetic acid, ethanol, methanol and dime-thyl sulfoxide. The concentration of 50% inhibition($IC_{50}$) of inhibitor against acetylcholinesterase was 25$\mu\textrm{g}$/ml. The inhibitor was inactivated on heating ar $100^{\circ}C$ fro 15 min and more stable in acidic region than alkaline region.n.

  • A strain of Actinomycetes producing cathepis B inhibitor was isolated from soil and identified as Streptomyces misakiensis. The product of S. misakiensis inhibited effectively cathepsis B proteinases as well as trypsin and papain. The cathepsin B inhibitor were largely produced with incubation for 4 days. The S. misakiensis was the most growth with incubation for 5 days. The cathepsin B inhibitor was isolated from the extraction of both with ethanol, ethanol and chlorofrom, and following several column chromatography such as sephadex G-15, silica gel 60 and sephadex LH-20 chromatography. The moleculer weight of purfied inhibitor was 138 dalton.

  • Streptomyces luteogriseus KT-10 isolated from Korean farm soil produced a strong cathepsin B inhibitor. Optimal conditions for the cathepsin B inhibitor production by s. luteogriseus KT-10 were evaluated. The cathepsin B inhibitor was produced with maximal yield in the cultural condition of pH 7.0 and $25^{\circ}C$ for 4 days. Optimal medium for the cathepsin B inhibitor production was determined to be a medium containing 20g, peptone 3g, yeast extract 1g, K2HPO4 0.5g, MgSO4.7H2O 0.5g, NaNO3 0.5g, NaCl 0.5g per l. The cathepsin B inhibitor produced by S. luteogriseus KT-10 could also inhibit the other proteinases such as trypsin, papain, and cathepsin D.

  • The aim of this study is to elucidate characteristics of ${\beta}-lactamase$ inhibitor produced by Streptomyces sp. SMF-19 isolated from soil was found to produce proteinaceous extracellular ${\beta}-lactamase$ inhibitor. The ${\beta}-lactamase$ inhibitor was purified through ammonium sulfate fractionation, gel filtration, anion exchange chromatography and fast performace liquid chromatography. The molecular weight of the ${\beta}-lactamase$ inhibitor was estimated to be about 48,000 by SDS-PAGE. The mode of inhibition against penicillin G as a substrate was uncompetitive. The ${\beta}-lactamase$ inhibitor was stable in wide pH range but unstable at high temperature above $50^{\circ}C$.

  • This paper was attempted to investigate effect of angiotensin inhibitor (loading dose 25, 50, $100{\mu}g/kg$ and maintenance dose 12.5, 25, $50{\mu}g/kg/hr$) on the pharmacokinetics of furosemide (5 mg/kg i.v) in rabbit. The plasma concentrations of furosemide increased by angiotensin inhibitor and the relative bioavailability of furosemide increased from 118.1% to 193.2% by the inhibitor. The protein binding of furosemide decreased by angiotensin inhibitor in bovine serum albumin ($2.17\;{\times}\;10^{-4}M$) by equilibrium dialysis method. Consequently, dosage regimen of furosemide might be adjusted carefully when furosemide is administered with angiotensin inhibitor.
