Abalone Protein Hydrolysates: Preparation, Angiotensin I Converting Enzyme Inhibition and Cellular Antioxidant Activity

  • Abalone protein was hydrolyzed by enzymatic hydrolysis and the optimal enzyme/substrate (E/S) ratios were determined. Abalone protein hydrolysates (APH) produced by Protamex at E/S ratio of 1:100 showed angiotensin I converting enzyme inhibitory activity with $IC_{50}$ of 0.46 mg/mL, and APH obtained by Flavourzyme at E/S ratio of 1:100 possessed the oxygen radical absorbance capacity value of $457.6{\mu}M$ trolox equivalent/mg sample. Flavourzyme abalone protein hydrolysates (FAPH) also exhibited $H_2O_2$ scavenging activity with $IC_{50}$ of 0.48 mg/mL and $Fe^{2+}$+ chelating activity with $IC_{50}$ of 2.26 mg/mL as well as high reducing power. FAPH significantly (P<0.05) protected $H_2O_2$-induced hepatic cell damage in cultured hepatocytes, and the cell viability was restored to 90.27% in the presence of FAPH. FAPH exhibited 46.20% intracellular ROS scavenging activity and 57.89% lipid peroxidation inhibition activity in cultured hepatocytes. Overall, APH may be useful as an ingredient for functional foods.

Application of hybrid material, modified sericite and pine needle extract, for blue-green algae removal in the lake

  • The present study assessed the efficient removal of nutrients and Chlorophyll-a (Chl-a) by using methyl esterified sericite (MES) and pine needle extracts (PNE), a low cost and abundant green hybrid material from nature. For this purpose, the optimal conditions were investigated, such as the pH, temperature, MES and PNE ratio, and MES-PNE dose. In addition, a Microcystis aeruginosa control using MES-PNE was also analyzed with various inhibition models. The removal of the nutrient and Chl-a onto MES-PNE was optimized for over 95% removal as follows: 2-2.5 for the MES-PNE ratio, 7-8 pH and a $22-25^{\circ}C$ temperature. In this respect, approximately 1.52-2.20 g/L of MES-PNE was required to remove each 1 g of dry weight/L of Chl-a. Total phosphorus (TP) has a greater influence on the increase in Chl-a than total nitrogen (TN) according to the correlation between TN, TP and Chl-a. Moreover, the Luong model was the best model for fitting the biodegradation kinetics data from Chl-a on MES-PNE from lake water. The novel hybrid material MES-PNE was very effective at removing TN, TP and Chl-a from the lake and can be applied in the field.

  • Catalytic ignition of $H_2/O_2/CO_2$ mixtures over platinum catalyst is experimentally investigated by using microcalorimetry. For comparison, $N_2$ and Ar is also used as diluent gas. The gas mixture flows toward platinum foil heated by electric current at atmosphere pressure and ambient temperature. The ignition temperature range 350-445K according to the fuel ratio, dilution ratio and diluent gas. It increases as the fuel ratio and dilution ratio increase. $H_2/O_2$ mixture with $CO_2$ ignites at higher temperature than with other diluents by 30-50K. Several experimental evidences show the inhibition effects of $CO_2$ in $H_2-O_2$ heterogeneous reaction is considerable

Relative potency of antihistaminics for $H_1$-and muscarinic receptors (항 히스타민제의 $H_1$ 수용체와 무스카린 수용체에 대한 상대적 역가)

  • The muscarinic antagonist l-[benzilic-4,4'-$^3H$]quinuclidinyl benzilate([$^3H$]QNB) bound to a single class of muscarinic receptor with high affinity in guinea pig ileal membranes. The $K_{D}$ and B$_{max}$ values for [$^3H$]QNB calculated from analysis of saturation isotherms were 54 pM and 156fmol/mg, respectively. H$_{1}$-blockers inhibited [$^3H$]QNB binding to ileal membranes with $K_{i}$ values ranged from 0.008 $\mu{M}$ to 1.6 $\mu{M}$. The pseudo-Hill coefficients of H$_{1}$-blockers for inhibition of [$^3H$]QNB binding to the ileal membranes were close to unit. The $K_{i}$ values for H$_{1}$-blockers were similar to the $K_{M}$ values calculated by Schild plot of functional data obtained from inhibition of the carbachol-induced contraction in guinea-pig ileum, suggesting that binding of H$_{1}$-blockers vs [$^3H$]QNB in ileal membranes represents an interaction with a receptor of physiological relevance. The $K_{H}$ values of H$_{1}$-blockers for H$_{1}$-receptor estimated from inhibition of the histamine-induced contraction were the range of 0.15 nM to 56.5 nM. The $K_{M}$/K$_{H}$ ratio of H$_{1}$-blockers varied over a wide range of 3 to 2300. Thus, the antihistaminic potencies of H$_{1}$-blockers do not correlate with their antimuscarinic potencies, which suggest that antihistamines have different antimuscarinic potencies in therapeutic blood levels causing similar antiallergic effect. Among 13 traditional antihistaminics examined in this study, drug having the highest and the lowest $K_{M}$/K$_{H}$ ratio is triprolidine and diphenidol, respectively. The present results demonstrate that the antimuscarinic property of antihistamines is not necessary for their antiallergic effect, and data on the affinity of antihistamines for muscarinic and H$_{1}$-receptors can be an important parameter in the selection and evaluation of these drugs.

Inhibitin of Xanthine Oxidase by Tea Extracts from Green Tea, Oolong Tea and Black Tea (녹차, 오룡차 및 홍차 추출물의 Xanthine Oxidase 억제작용)

  • Inhibition of xanthine oxidase by tea extracts obtained from non-fermented tea(steamed green tea and roasted green tea), semi-fermented tea(oolong tea) and fermented tea(black tea) were investigated. The crude catechin fraciton had a hgher inhibitory effect against xanthine oxidase, and the effect was increased with the addition of tea extracts. Their inhibitory effect were hardly influenced until extracted three times with hot water. According to the investigation of catechins in the crude catechin fraction obtained from tea extracts, (-)-epicatechin-(EC), (-)-epicatechin gallate(ECg). (-)-epigallocatechin(EGC) and (-)-epigallocatechin gallate(EGCg) were 80.1$\mu\textrm{g}$/mg 113.5$\mu\textrm{g}$ /mg, 186.3$\mu\textrm{g}$/mg and 367.7$\mu\textrm{g}$/mg in steamed green tea, and 75.6$\mu\textrm{g}$/mg, 114.7$\mu\textrm{g}$/mg, 193.7 $\mu\textrm{g}$/mg and 381.9$\mu\textrm{g}$/mg in roasted green tea, and 69.4$\mu\textrm{g}$/mg, 110.0$\mu\textrm{g}$/mg, 127.1$\mu\textrm{g}$.mg and 464.9$\mu\textrm{g}$/mg in oolong tea, and 78.1$\mu\textrm{g}$/mg, 171.8$\mu\textrm{g}$/mg, 80.7$\mu\textrm{g}$/mg and 51.4$\mu\textrm{g}$/mg in black tea, respectively. Order of the content of these catechins was (-)-EGCg>(-)-EGC>(-)-ECg>(-)-EC in steamed green tea, roasted green tea and oolong tea, and was (-)-ECg>(-)-EGC>(-)-EC>(-)-EGCg in black tea. Also the concentration of catechins was hardly influeced until extracted three times. The inhibition ratio of xanthine oxidase by autherntic catechins was hardly influenced until extracted three times. The inhibition ratio of xanthine oxidase by authentic catechins was 94.9% and 87.6% by addition of 5.0$\mu\textrm{g}$/ml of (-)-EGCg and (-)-ECg, respectively. the inhibitors of xanthine oxidase were supposed to be due to (-)-ECg and (-)-EGCg in tea polyphenol compounds.

  • Matrix metalloproteinases (MMPs) are a family of zinc-dependent proteases that degrade extracellular matrix and basement membrane. These enzymes are play important roles in tumor cell invasion and metastasis, as well as angiogenesis and other connective tissue diseases. In our screening program for inhibitors of MMP-2 from fungal metabolites, we have isolated novel non-peptidic inhibitors of MMPs, designated gelastatin A and B from the culture broth of Westerdykella multispora F50733. The structures of gelastatin A and B were determined to be 3-(5E-hexa-2E,4E-dienylidene-2-oxo-5,6-dihydro-2H-pyran-3yl)-propanoic acid and 3-(5Z-hexa-2E,4E-dienylidene-2-oxo-5,6-dihydro-2H-pyran-3yl)-propanoic acid, respectively. Gelastatin A and B exist as a mixture of two stereoisomers in a ratio of 2: 1. The 2: 1 mixture of gelastatin A and B inhibited activated MMP-2 and MMP-9 with an IC$\sub$50/ value of 0.63, 5.29 ${\mu}$M, respectively. They inhibited the invasion of B16F10 melanoma cells through basement membrane Matrigel with dose dependent.

Extracts of Aster species Inhibit Invasive Phenotype and Motility of H-ras MCF10A Human Breast Epithelial Cells Possibly via Downregulation of MMP-2 and MMP-9

  • Cancer metastasis represents the most important cause of cancer death and antitumor agents that may inhibit this process have been extensively pursued. Invasion and metastasis of malignantly transformed cells involve degradation of the extracellular matrix (ECM) components by matrix metalloproteinases (MMP), especially MMP-2 and -9. We previously showed that H-ras-induced invasive phenotype may involve MMP-2, rather than MMP-9, in MCF10A cells. In the present study, we investigated the chemopreventive effect of Aster, a widely used culinary vegetable in Korea. We screened twelve extracts from three Aster species (Aster scaber, Aster oharai and Aster glehni) for the inhibitory effect on MMP activities of H-ras MCF10A human breast epithelial cells. All of the extracts tested in this study efficiently inhibited the gelatinolytic activities of MMP-2 and MMP-9. A more prominent inhibition was observed in MMP-2 activity compared to MMP-9. Out of twelve extracts, eight extracts showed>90% inhibition of MMP-2 activity in H-ras MCF10A cells while only one extract showed>90% inhibition of MMP-9 activity. We selected three extracts (AO-3, AG-3 and AS-EA) for further studies since they exerted a marked inhibition in the ratio of MMP-2 to MMP-9. Treatment with AO-3, AG-3 and AS-EA in H-ras MCF10A cells caused a significant inhibition of invasive phenotype and migration, proving a chemopreventive potential of these extracts. Taken together, our results demonstrate that extracts of Aster effectively inhibit invasion and migration of highly malignant human breast cells, possibly via downregulation of MMP-2 and MMP-9.

Antibacterial Properties of Extracts from Abies holophyllaand Pinus koraiensisNeedles Against Escherichia coli and Staphylococcus aureus (전나무와 잣나무 잎 추출물의 대장균과 황색포도상구균에 대한 항균특성)

  • In this study, functional substances with antibacterial properties were extracted from the needles of Abies holophylla and Pinus koraiensis, and optimized using the central composite design-response surface methodology (CCD-RSM). The optimal extraction conditions for Abies holophylla were an extraction temperature of 59.5 ℃ and an ethanol/ultrapure water volume ratio of 69.5 vol.%, resulting in an extraction yield of 13.5% and inhibition diameters of 11.6 mm against Escherichia coli (E. coli) and 9.3 mm against Staphylococcus aureus (S. aureus). For Pinus koraiensis, the optimal extraction conditions were an extraction temperature of 59.2 ℃ and an ethanol/ultrapure water volume ratio of 67.8 vol.%, resulting in an extraction yield of 4.8% and inhibition diameters of 7.9 mm against E. coli and 12.5 mm against S. aureus. The actual experimental results under these optimal conditions showed that an extraction yield from Abies holophylla needles was 13.0% and an inhibition diameter of 11.7 mm against E. coli and 9.2 mm against S. aureus, indicating an error rate of approximately ± 2.3%. For Pinus koraiensis needles, the extraction yield was 5.1%, with inhibition diameters of 7.5 mm against E. coli and 12.3 mm against S. aureus, indicating an error rate of ± 4.23%.

Effect on the Inhibition of Over-growth of Plug Seedling by Triazole-type Growth Regulator Treatment (Triazole계 생장조절제 처리가 공정육묘의 도장억제에 미치는 영향)

  • This study was conducted to investigate the effects on the over-growth inhibition of fruit vegetables (tomato, cucumber, and hot pepper) seedling by the treatment of triazole-typechemicals and to select the most effective chemicals for the over-growth inhibition, and to investigate suitabla concentration treatment and times of the newly selected chemical. The triazole-type chemicals inhibited stem height more, the more the concentration is increased in any crops. The diniconazole showed the highest dwarfing ration, and the tebuconazole and hexaconazole are also good for dwarfing ratio. The diniconazole decreased the plant height of cucumber, tomato, and hot pepper as the concentration is increased. In cucumber, thc dwarfing ratio was 30% in the 5 mg${\cdot}L^{-1}$ treatment, and was 60% in thc l00 mg${\cdot}L^{-1}$ treatment. In tomato and hot pepper, the dwarfing ratio was 30% in the 25 mg${\cdot}L^{-1}$ treatment in any times. In diniconazole treatment, the cucumber and tomato should have 5 mg${\cdot}L^{-1}$ at the lst growth stage and the hot pepper should have 5 mg${\cdot}L^{-1}$ at the 2nd growth stage to obtain good seedlings.

Development of a Blended Corrosion, Scale and Micro-Organism Inhibitor for Open Recirculating Cooling System

  • This paper presents the results of a study that was undertaken to optimize the ratio of the components of a new multi-component inhibitor blend composed of orthophosphate/ phosphonates/ acrylate copolymer/ isothiazolone. The effects of newly developed inhibitor on carbon steel dissolution in synthetic cooling water were studied through weight loss tests, electrochemical tests, scale tests, and micro-organism tests. The obtained results were compared to blank (uninhibited specimen) and showed that developed inhibitor revealed very good corrosion, scale, and micro-organism inhibition simultaneously. All measurements indicated that the efficiency of the blended mixture exceeded 90 %. The inhibitive effects arose from formation of protective films which might contain calcium phosphate, calcium phosphonate, and iron oxide. The nature of protective films formed on the carbon steel was studied by scanning electron microscopy (SEM) and auger electron spe ctroscopy (AES). Inhibitor used in this study appeared to have better performance for scale inhibition due to their superior crystal modification effect and excellent calcium carbonate scale inhibition properties. The effect of inhibitor on microorganisms was evaluated through minimum inhibitory concentration (MIC) test. All kinds of micro-organisms used in this study were inhibited under 78ppm concentration of inhibitor.
