cAMP 증가 유도 약물의 대식세포- 및 T 세포-매개성 면역반응 조절작용 (Immunomodulatory Effect of cAMP-Elevating Agents on Macrophage- and T cell-Mediated Immune Responses)

  • To investigate the immunomodulatory roles of cyclic AMP (CAMP) on macrophage- and T lymphocyte-mediated immune responses, CAMP elevating agents were employed and carefully re-examined under the activation conditions of the cells. Various inhibitors tested dose-dependently blocked tumor necrosis factor (TNF)-${\alpha}$ production with IC$_{50}$ values ranged from 0.04 to 300 ${\mu}$M. Of the inhibitors, cAMP-elevating agents showed lower cytotoxicity assessed by lactate dehydrogenase (LDH) release, suggesting less toxic and more selective. In particular co-treatment of dbcAMP with a protein kinase C inhibitor staurosporine displayed the synergistic inhibition of TNF-${\alpha}$ production. The modulatory effect of dbcAMP on TNF-${\alpha}$ and nitric oxide (NO) was significantly affected by treatment time of dbcAMP. Thus, post-treatment of dbcAMP (three hours before LPS) abrogated dbcAMP's inhibitory activity and rather enhanced TNF-${\alpha}$ level up to 60%. In contrast, additional NO production was shown at the co-treatment of dbcAMP with LPS. Unlike simultaneous treatment of phorbol 12-myristate 13-acetate (PMA) and interferon (IFN)-${\gamma}$co-treatment, the combination of dbcAMP with other NO-inducing stimuli did not show drastic overproduction of NO. cAMP elevating agents also diminished splenocyte proliferation stimulated by concanavalin (Con) A, phytohemaglutinin A (PHA) and lipopolysaccharide (LPS). In addition, dbcAMP but not rolipram strongly suppressed CD8$^+$ T cells (CTLL-2). Finally, cAMP elevating agents were differentially involved in regulating CD98-mediated cell-cell adhesion. Thus, dbcAMP and rolipram significantly enhanced the cell-cell adhesion, whereas forskolin blocked. Therefore, our results suggest that CAMP elevating agents participate in various immune responses mediated by macrophages and T cells with a different fashion depending on cellular environments and activation signals.

Ursodeoxycholic Acid (UDCA) Exerts Anti- Atherogenic Effects by Inhibiting Endoplasmic Reticulum (ER) Stress Induced by Disturbed Flow

  • Disturbed blood flow with low-oscillatory shear stress (OSS) is a predominant atherogenic factor leading to dysfunctional endothelial cells (ECs). Recently, it was found that disturbed flow can directly induce endoplasmic reticulum (ER) stress in ECs, thereby playing a critical role in the development and progression of atherosclerosis. Ursodeoxycholic acid (UDCA), a naturally occurring bile acid, has long been used to treat chronic cholestatic liver disease and is known to alleviate endoplasmic reticulum (ER) stress at the cellular level. However, its role in atherosclerosis remains unexplored. In this study, we demonstrated the anti-atherogenic activity of UDCA via inhibition of disturbed flow-induced ER stress in atherosclerosis. UDCA effectively reduced ER stress, resulting in a reduction in expression of X-box binding protein-1 (XBP-1) and CEBP-homologous protein (CHOP) in ECs. UDCA also inhibits the disturbed flow-induced inflammatory responses such as increases in adhesion molecules, monocyte adhesion to ECs, and apoptosis of ECs. In a mouse model of disturbed flow-induced atherosclerosis, UDCA inhibits atheromatous plaque formation through the alleviation of ER stress and a decrease in adhesion molecules. Taken together, our results revealed that UDCA exerts anti-atherogenic activity in disturbed flow-induced atherosclerosis by inhibiting ER stress and the inflammatory response. This study suggests that UDCA may be a therapeutic agent for prevention or treatment of atherosclerosis.

독활 메탄올 추출물의 Streptococcus mutans에 대한 성장, 산생성, 부착 및 비수용성 글루칸 합성 억제에 미치는 영향 (Effects of methanol extract of Aralia continentalis on the growth, acid production, adhesion, and insoluble glucan synthesis of Streptococcus mutans)

  • Dental plaque is a film of microorganisms on the tooth surface that plays an important part in the development of caries and periodontal diseases. Streptococcus mutans (S. mutans) is present in almost all types of dental plaque. Teeth and their supporting structure, the gums (gingiva) are subjected to infection by S. mutans that causes cavities and pyorrhea which, if left untreated, can eventually lead to gingivitis. Various chemical agents have been evaluated over the years with respect to their antimicrobial effects in the oral cavity; however, all are associated with side effects that prohibit regular long-term use. The present study was designed to investigate the effect of Aralia continentalis (Araliaceae) extracts on the growth, acid production, adhesion, and insoluble glucan synthesis of S. mutans. The methanol extract of A. continentalis showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.25, 0.5, 1, 2 and 4 mg/ml compared to the control group. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 60% at the concentration of 0.25 mg/ml and complete inhibition was observed at the concentration of 4 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan from sucrose, methanol extract of A. continentalis showed more than 10% inhibition over the concentration of 0.25 mg/ml. The synthesis of insoluble glucan was decreased in the presence of 0.25 - 4 mg/ml of the methanol extract of A. continentalis. Hence, we conclude that A. continentalis might be a candidate of anticaries agent.

Anti-atherosclerotic Effect of the Methanol Extract of Sorbus commixta Cortex in the High Cholesterol-Diet Rats

  • Hypercholesterolemia is a pivotal pathogenic factor for the development and maintenance of atherosclerosis. The present study was designed to evaluate whether the methanol extract of Sorbus commixta cortex (MSC) restores vascular dysfunction in association with the aortic expressions of proinflarnmatory and adhesion molecules in high cholesterol (HC) diet-rats. Chronic treatment with low (100 mg/kg/day) or high doses (200 mg/kg/day) of MSC lowered the increase in plasma levels of triglyceride (TG) and low-density lipoprotein (LDL) cholesterol induced by a cholesterol-enriched diet without affecting on the plasma level of high density lipoprotein (HDL)-cholesterol. Vascular tone attenuated in the HC-diet rats was restored by administration with MSC. Treatment with MSC also suppressed the HC-induced increase in the monocyte chemoattractant protein-1 (MCP-1) and nuclear factor-$_K$B (NF-$_K$B) p65 expressions as well as expressions levels of adhesion molecules including intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (ICAM-1), and E-selectin in aorta. The present study also showed that MSC inhibited the HC-mediated induction of ET-1 and ACE expression. In histopathological examination, aortic segments in the HC-diet rat revealed thickening intima and media, which were blocked by administration with MSC. Taken together, MSC could suppress the development of atherosclerosis in the HC-diet rat model through the inhibition of the aortic expression levels of pro-inflammatory and adhesion molecules.

황금(Scutellaria baicalensis) 추출물에 의한 Streptococcus mutans의 항균 및 부착억제 효과 (Effects of Antibacteria and Adhesive Inhibition of Scutellaria baicalensis Extract on Streptococcus mutans)

  • 천연물 황금으로부터 여러 유기용매 추출물을 얻어 치아우식증 원인균인 S. mutans 에 대한 항균활성, 배지 내 환경변화, 그리고 부착억제에 대해 알아보았다. 1. 구강 질병인 치아우식증의 대표적 원인균 S. mutans에 대한 황금 ethyl acetate 추출물 (IPK-3)의 최소억제농도는 125 mg/ml 이었다. 2. 황금 IPK-3 추출물 50 mg/ml의 농도로 S. mutans의 배지 내 투여하였을 때 균의 생장 량과 최대 생장시간은 대조군보다 지연되었다. 3. 황금 IPK-3 추출물 50 mg/ml의 농도로 S. mutans의 배지 내 투여하였을 때, pH 변화는 대조군은 18시간에서 pH 5.63으로 급격한 변화를 보였으며 황금 IPK-3 추출물이 50 mg/ml의 배지에서는 6.50이상을 유지하여 pH가 변화가 없었다. 4. 황금 IPK-3 추출물을 첨가한 배지 내 탄수화물, 단백질 및 균체 외 다당류의 변화는 대조군에 비해 매우 낮게 생산되었다. 5. 균체를 형광염료(DAPI)로 염색하여 S-HA 부착억제을 확인한 결과, 대조군은 시간이 경과함에 따라 S.mutans의 hydroxyapatite(HA)에 부착 정도가 증가되었으나, 황금 IPK-3 추출물이 첨가된 시험군에서는 S-HA에 균의 부착이 매우 적었다. 이같이 황금의 IPK-3 (ethyl acetate 추출물)이 치아우식증의 원인균인 S. mutans 에 대한 항균활성효과 뿐만아니라 hydroxyapatite에 부착 억제 효과도 있었다.

Anti-Helicobacter and Anti-inflammatory Effects of Sohamhyungtang in Helicobacter pylori-Infected Human Gastric Epithelial AGS cells

  • This study evaluated the anti-Helicobacter and anti-inflammatory effects of Sohamhyungtang (SHHT). The minimum inhibitory concentration (MIC) of SHHT against Helicobacter pylori (H. pylori) was determined by the agar dilution method. Expression of the H. pylori cagA gene in the presence of SHHT was determined by quantitative real-time polymerase chain reaction (qRT-PCR). Inhibition of H. pylori urease by SHHT was determined by the phenol-hypochlorite assay. Antiadhesion activity of SHHT was measured by urea-phenol red reagent. Inhibition of nitric oxide (NO) production in AGS cells was measured with Griess reagent. Inducible nitric oxide synthase (iNOS) and IL-8 mRNA expression in AGS cells which were infected with H. pylori was determined by qRT-PCR. IL-8 level was measured by enzyme-linked immunosorbent assay (ELISA). The MIC of SHHT was $100{\mu}g/mL$ and the expression of cagA gene was decreased about 25 folds in the presence of SHHT. H. pylori urease was inhibited 90% by SHHT. SHHT inhibited H. pylori adhesion on AGS cell in a concentration dependent manner. mRNA expression of iNOS and IL-8 and the production of NO and IL-8 were significantly decreased in the presence of SHHT. In conclusion, SHHT showed anti-Helicobacter activity and has potent anti-inflammatory effect on H. pylori-induced inflammation in human gastric epithelial AGS cells.

금속단백분해효소의 활성 저해를 통한 삼귀고의탕의 전이억제 효과 (Anti-metastatic Effect of Samguikoeui-Tang Via Inhibition of Matrix Metalloproteinases Activities)

  • This study was performed to examine the anti-metastatic effect of ethanol extract of Samguikoeui-Tang (SGKE), a formula consisting of four oriental herbs, in highly-metastatic HT1080 human fibrosarcoma cells. SGKE significantly inhibited the adhesion of HT1080 cells to matrigel at nontoxic concentrations in a dose-dependent manner, while it did not exert cytotoxicity against HT1080 cells up to the concentration of 100 ${\mu}g$/ml. Also, SGKE depressed the activity of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) by gelatin zymography. However, SGKE did not affect the mRNA expression of MMP-2 and TIMP-2, an inhibitor of MMP-2, by RT-PCR analysis. In addition, the effect of SGKE on HT1080 cell invasion was determined using Boyden chamber assay. SGKE suppressed the invasion of HT1080 cells in a dose-dependent manner. Taken together, these results suggest that SGKE has an anti-metastatic effect via inhibition of MMP-2 and -9 activities.

호중구의 Respiratory Burst에 미치는 PLA2 및 PAF와 영향 : In vitro에서의 호중구의 산소기 생성 및 Apoptosis에 관한 연구 (Effect of the Inhibition of PLA2 and PAF on the Neutrophilic Respiratory Burst and Apoptosis)

  • 연구배경 : 급성호흡부전증후군(ARDS)의 병인론을 호중구의 산소기 생성 및 apoptosis 의 관정에서 PLA2 및 PAF의 역할과 연관하여 알아보았다. ARDS의 원인 중 산소기의 역할이 주로 염증성 cytokine 및 지질분자와 관련하여 연구되고 있는 점에 착안하여 내독소에 의한 PLA2, PAF의 작용 및 이에 따른 호중구의 혈관내피세포로의 유착, 산소기에 의한 pulmonary surfactant의 기능에 미치는 영향에 대해서도 알아보았다. 방법 : PMA로 자극된 호중구에서 PLA2 및 PAF의 억제에 따른 산소기 형성의 변화에 대해서도 알아보았으며 내독소에 의한 호중구에서의 PLA2활동도의 변화, lysoPAF remodelling에 미치는 PLA2 및 PAF의 억제의 효과에 대해서도 알아보았다. 또한 내독소 및 PMA에 의해 자극된 상태에서의 PLA2 및 PAF의 억제가 호중구의 apoptosis에 미치는 영향도 알아보았다. 호중구에 의한 조직의 손상은 혈관내피세포로의 호중구의 유착이 선행되어야 하므로 이러한 작용에 PLA2 및 PAF가 미치는 영향을 호중구 유착검사를 통하여 알아보았고 형태학적으로는 산소기와 pulmonary surfactant의 결합을 확인하였다. 결론 : ARDS 시의 호중구의 역할은 PLA2 및 PAF의 작용에 의한 산소기 형성 및 염증성 지질분자의 생성을 통해 조작의 손상을 유발하는 듯하며 이때 PLA2의 억제는 호중구의 apoptosis의 증가 및 산소기의 생성을 감소시키고 또한 호중구의 혈관내피세포로의 유착을 감소시켰다. PAF의 억제는 호중구의 산소기 생성의 감소 및 호중구의 유착을 억제하여 조직의 손상을 감소시키는 것으로 생각되었다.

삼백초 추출물의 항치아우식 효과 (Anticariogenic Properties of the Extract of Saururus chinensis)

  • It has been well established that S. mutans is the major etiological agent in dental caries, one of the most common oral diseases worldwide. The present study was designed to investigate the effect of Saururus chinensis (S. chinensis) ethanol extracts on the growth, acid production, biofilm formation, adhesion, and insoluble glucan synthesis of S. mutans. The ethanol extracts of S. chinensis showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.025, 0.05, 0.1, 0.2 and 0.4 mg/ml compared to the control group. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 80% at the concentration of 0.05 mg/ml and complete inhibition was observed at the concentration of 0.4 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan form sucrose, ethanol extract of S. chinensis showed more than 10% inhibition over the concentration of 0.025 mg/ml. The synthesis of insoluble glucan was decreased in the presence of 0.025 ${\sim}$ 0.4 mg/ml of the ethanol extract of S. chinensis. Our research strongly suggested S. chinensis was a promising natural product for the prevention of dental caries.
