A Study on Physiological Activities of Coptidis Rhizoma and Application for Cosmetic Ingredients (황련(黃連)의 생리활성검증 및 화장품 소재로서의 이용 가능성)

  • Objectives : The physiological activities of Coptidis Rhizoma investigated for application as a cosmetic ingredients. Methods : We were experimented anti-oxidation effect, growth inhibition ability on cancer cells, antibacterial activity on various kinds of bacteria of skin. Results : The results were obtained as follows : Electron-donating ability(EDA) shows that Coptidis Rhizoma extracted with ethanol(94.51%) gives higher EDA in comparison to that extracted with water in 1000 ppm(63.24%). In the test of SOD-like activity, ethanol extract showed quite more activity with 81.01% in 1000 ppm, while water extract was low in 18.22%. In the inhibition activity of Xathine oxidase, ethanol extract showed higher inhibition activity compared to water extract. In measuring lipid oxidation inhibition ability using fish oil, ethanol extract in 100 ppm showed prominent ability. In the oxidation inhibition effect added with $Cu^{2+}\;ion\;and\;Fe^{2+}$ ion as a oxidation promoter, ethanol extract in 1000 ppm, each in the proportion of 90% and 92%, showed high oxidation inhibition effect compared with water extract. Conclusions : The results indicated that, ethanol extract which is superior in its anti-oxidation and antibacterial effect is useful to be applied in cosmetic industry.

Growth Inhibition of Polysaccharide Fraction in Cell Wall Components from Enterococcus faecalis 2B4-1 against Tumor Cell Lines (Enterococcus faecalis 2B4-1 세포벽 성분 중 Polysaccharide Fraction의 종양세포 증식억제 효과)

  • This study was developed to evaluate the growth inhibition effects of cell wall components of Enterococcus faecalis 2B4-1 obtained from feces of neonates against tumor cell lines. Polysaccharide fraction (PS) shown sensitive growth inhibition effect in the cell wall components was isolated and characterized. In growth inhibition effects, residue fractin of whole cell was shown sensitive level of percent survival about 30% when administrated at ehe concentration of 100${\mu}$g/ml, and that was more effective than that of supernatant fraction against the tumor cell lines, SNU-1, 3LL, FARROW and HEC-1-B. Sensitive growth inhibition effects against SNU-1, FARROW and HEC-1-B were performed by whole cell (WC) fraction from Ent. faecalis 2B4-1. Cytoplasm fractin (CP) of WC was shown non-inhibition effect, however, the other part of WC, precipitate of disrupted cell (PD), was sensitive against the tumor cell line mentioned above. Followed by separation to peptidoglycan fraction (PG) and polysaccharide fraction (PS) were all sensitive which the latter was shown more sensitive percent survival than the former. Composed sugars of polysaccharide fraction were determined to D-glucose, L-rhamnose and D-glucosamine, and the rate fo composition was calculated to about 1:1:1 by the data of elemental analysis, IR, TLC and HPLC.

  • It has been reported that the inhibition effect of pH on activated sludge follows noncompetitive inhibition kinetics. However. the noncompetitive inhibition kinetic equation can not be directly applied to pH inhibition because of the difficulty in quantification of pH in terms of inhibitor concentration. So, many empirical equations have been developed to describe the pH inhibition effect especially for acidic condition. In this research. the pseudo toxic concentration ($C_{PT}$) concept model to quantify pH inhibition effect on activated sludge was proposed and compared to other existing models. The $C_{PT}$ concept model can explain the reduction of the maximum specific growth rate (${\mu}_{max}$) caused by the pH inhibition more accurately than any other models, at a wide range of pH. The only model parameter. $K_I$ can be easily estimated by Lineweaver-Burk linearization method.

Protective Effect of Physostigmine and Neostigmine against Acute Toxicity of Parathion in Rats

  • The effects of physostigmine and neostigmine on the parathin induced toxicity were examined in adult female rats. Physostigmine $(100\;{\mu}g/kg,\;ip)$ or neostigmine $(200\;{\mu}g/kg,\;ip)$ inhibited acetylcholinesterase (AChE) and cholinesterase (ChE) activities in blood, brain and lung when the enzyme activity was measured 30 min after the treatment. At the doses of two carbamates equipotent on brain AChE, neostigmine showed greater inhibition on peripheral AChE/ChE. The enzyme activity returned to normal in 120 min following the carbamates except in the lung of rats treated with neostigmine. Carbamates administered 30 min prior to parathion (2 mg/kg) antagonized the inhibition of AChE/ChE by parathion when the enzyme activity was measured 2 hr following parathion. Neostigmine showed greater protective effect on peripheral AChE/ChE. The effect of either carbamate on AChE/ChE was not significant 2 hr beyond the parathion treatment. Carbamates decreased the mortality of rats challenged with a lethal dose of parathion (4 mg/kg, ip) either when treated alone or in combination with atropine (10 mg/kg, ip). Lethal action of paraoxon (1.5 mg/ks ip), the active metabolite of parathion was also decreased by the carbamate treatment indicating that the protection was not mediated by competitive inhibition of metabolic conversion of parathion to paraoxon. The results suggest that carbamylation of the active sites may not be the sole underlying mechanism of protection provided by the carbamates.

  • Root bark of Ulmus davidiana Plancn. var. japonica Nakai was extracted by using several solvents with different polarities. Each extract was treated on the MMPs obtained from SK-Hep-1 in order to investigate inhibition effect. Zymography of MMPs showed that MeOH extract has significant inhibition effect. On the GC-MS analysis the highest mass to charge ratio (m/z) of the purified substance was 281. Also, on zymography of MMPs the substance showed 47% inhibition effect at the concentration of $314.7{\mu}g/g$. Cell viability of SK-Hep-1 was 60% at $31.47{\mu}g/g$.

Effect of DIS Process on in vitro Physiological Functionality of Aloe vera Gel (알로에 겔의 in vitro 생리기능 특성에 미치는 삼투탈수공정의 영향)

  • In vitro physiological functions such as jack bean (Canavalia ensiformis) urease inhibitory activity and retarding effect of glucose/bile acid of Aloe vera gel concentrated by the optimized DIS (Dewatering Impregnation & Soaking) process conditions were examined. Urease inhibitory activity of DIS aloes ranged from 84.6 to 94.4%, which was similar to or higher than 86.3% of fresh aloe. Also, urease inhibitory activity of DIS aloes was maintained at initial levels after heat treatment (90$^{\circ}C$, 10 min.) and drying treatment (freeze or hot air drying). Urease inhibition pattern from Lineweaver-Burk plot indicated general non-competitive inhibition, and inhibition constants ($K_{IE}$ and $K_{IES}$) of DIS aloes were 41-149 and 87-163 $\mu$L/mL, respectively. DIS(glucose) and DIS(polyethylene glycol) exhibited the highest retarding effect of glucose and bile acid. Their retarding effects were about 1.6 and 1.8 folds higher than that of fresh aloe after 0.5 and 1 hr of the dialysis, respectively. Conclusively, the above in vitro physiological functions of Aloe vera gel concentrated by DIS process suggested that aloe products treated with DIS would have the potential benefits for protection against Helicobacter pylori and reduction of blood glucose and cholesterol levels.

Effect of Polyamines on Cellular Differentiation of N. gruberi: Inhibition of Translation of Tubulin mRNA

  • The effects of a polyamine, spermine, on the differentiation of Naegleria gruberi amebas into flagellates were tested. Addition of spermine at early stages of differentiation (until 40 min after the initiation of differentiation) completely inhibited the differentiation. To understand the inhibition mechanism, we examined the effect of spermine treatment on the transcription and translation of differentiation-specific genes during differentiation. Addition of spermine at early stages did not inhibit the accumulation of two differentiation-specific mRNAs, ${\alpha}$-tubulin and Class I mRNA, significantly, but rather prevented the rapid degradation of the mRNAs in later overall protein synthesis partially and gradually. However, translation of the ${\alpha}$-tubulin mRNA was completely inhibited. These data suggest that the inhibition of differentiation of N. gruberi by spermine treatment did not result from the inhibition of transcription of differentiation-specific genes but from the specific inhibition of translation of the mRNAs during the differentiation.

Screening for Antioxidative and Antimutagenic Capacities in 7 Common Vegetables Taken by Korean

  • This study was performed to investigate the antioxidative effect as the inhibition of MDA and BSA conjugation reaction, inhibition of lipid peroxidation and the scavenging effect on DPPH radical, and antimutagenic capacities as the Ames test in 7 common vegetables taken by Korean for suggestion of prevention and dietetic treatment of chronic diseases and development of antioxidative and antimutagenic functional food. The water extracted fractions of perilla leaves were most effective in the inhibition of MDA and BSA conjugation reaction showing 77.2% of inhibition rate among 7 vegetables. The inhibition rates of ethanol extracted fractions of sedum and wild water dropwort on the lipid peroxidation were 67.1% and 61.5%, respectively. The ethanol extracted fractions of crown daisy and wild water dropwort showed the most effective results among 7 vegetables in the DPPH radical scavenging capacities showing inhibition rate of 78.8% and 73.6%, respectively. The indirect and direct antimutagenic effects of ethanol extract of 7 vegetables were examined by Ames test using Salmonella typimurium TA98 and TA100. Inhibitory effects of wild water dropwort was superior to the other vegetables on the Ames test. These results suggest that common 7 vegetables taken by Korean are believed to be a possible antioxidative and antimutagenic capacities, although the resultswere different, some or less, according to the assay method and vegetables used.

Hydrodynamic Effect on the Inhibition for the Flow Accelerated Corrosion of an Elbow

  • The inhibition effect of thioureido imidazoline inhibitor (TAI) for flow accelerated corrosion (FAC) at different locations for an X65 carbon steel elbow was studied by array electrode and computational fluid dynamics (CFD) simulations. The distribution of the inhibition efficiency measured by electrochemical impedance spectroscopy (EIS) is in good accordance with the distribution of the hydrodynamic parameters at the elbow. The inhibition efficiencies at the outer wall are higher than those at the inner wall meaning that the lower inhibition efficiency is associated with a higher flow velocity, shear stress, and turbulent kinetic energy at the inner wall of the elbow, as well as secondary flow at the elbow rather than the mass transport of inhibitor molecules. Compared to the static condition, the inhibition efficiency of TAI for FAC was relatively low. It is also due to a drastic turbulence flow and high wall shear stress during the FAC test, which prevents the adsorption of inhibitor and/or damages the adsorbed inhibitor film.

ANTIMICROBIAL EFFECT OF ROOT CANAL CEMENTS ON MICROORGANISMS FROM INFECTED ROOT CANALS (수종 근관 충전재의 항균 효과에 관한 연구)

  • The purpose of this study was to evaluate the antimicrobial effect of root canal cements such as AH-26, Sealapex, Canals and Apatite root sealers(Type I, II, III) and to determine the efficacy of their activities. S.mutans(AHT), S.sanguis(NCTC 9811) and B.gingivalis(381) were streaked on blood agar and the PVC tybes filled with root canal cements were applied on. Then the microorganisms were cultured for 48 hours, anaerobically. B.cereus(KCTC 1012) was streaked on nutrient agar, PVC tubes were applied on and were cultured for 48 hours, aerobically. The inhibition zones of root canal cements were measured with vernier caliper. The data statistically analyzed, and the results were as followed. 1. Apatite root sealers(Type I, II, III) showed no inhibition zones. 2. AH-26, Sealapex and Canals had inhibition zones with varying degrees. The inhibition zone of AH-26 was greatest and followed by Canals and Sealapex(P<0.01). 3. As time goes by after mixing the root canal cements, AH-26, Canals and Sealapex showed significantly reducing inhibition zones(P<0.01). 4. There were the least inhibition zones of all the root canal cements on S.mutans and followed in such order as; B.gingivalis, S.sanguis and B.cereus(P<0.01).
