Induction of Phase II Enzymes and Inhibition of Cytochrome P450 Isozymes by Chitosanoligosaccharides

  • Abstract The cancer chemopreventive potential of chitosanoligosaccharides was investigated by measuring the induction of quinone reductase and glutathione S-transferase activities and inhibition of cytochrome P450 1A1, 2B1, and 2E1 activities. Chitosanoligosaccharide I (1-${\kappa}$Da${\kappa}$Da) significantly induced glutathione S-transferase activity with a maximal 1.5-fold increase at 500 ${\mu}$g/ml, while chitosanoligosaccharide II (3-${\kappa}$Da${\kappa}$Da) (500 ${\mu}$g/ml) strongly induced quinone reductase (p<0.01) and glutathione S-transferase (p<0.005) activities. The in vitro incubation of rat liver microsomes with chitosanoligosaccharides I and II (2.5, 5, 50, and 500 ${\mu}$g/ml) showed a dose-dependent inhibiton of cytochrome P450 1A1, 2B1, and 2E1 activities. Chitosanoligosaccharide II was a more potent inhibitor of cytochrome P450 2B1 activity than chitosanoligosaccharide I. Accordingly, these findings suggest that chitosanoligosaccharides are potential chemopreventive agents.

Antioxidant Activities of Mulberry (Morus alba L.) Leaf Extracted with Different Concentrations of EtOH

  • Antioxidant activities of mulberry leaf extracted with different concentrations of EtOH were investigated. Total phenolic content and electron donating abilities of extract from 70% EtOH were the highest. Extracts obtained from EtOH-water mixture were shown to be significantly higher superoxide dismutase (SOD)-like activities than other treatment (p<0.05). Angiotensin-converting enzyme (ACE) inhibition was the greatest at 50% EtOH concentration (p<0.05). The extracts from 30-70% EtOH exhibited higher ferric reducing ability of plasma (FRAP) value than rest of the concentration (p<0.05). In case of nitrite scavenging activity, much higher scavenging activities were observed when the extraction was performed with EtOH or EtOH-water mixture (p<0.05). The results indicate that concentration of EtOH as extraction solvents can affect the antioxidant activity of mulberry leaf, which may provide useful information on the optimal solvent conditions for the extraction.

Functional Properties of Nutmeg (육두구의 생리활성에 관한 연구)

  • This study investigated the antioxidative and antimicrobial activities of nutmeg (water, ethanol extract and essential oil). The total phenol contents of water, ethanol extract and essential oil were 3.4%, 16.9%, and 3.2%, respectively. Hydrogen donating abilities of water, ethanol extract and essential oil at 1,000 ppm were 4.9%, 41.8% and 6.8%, respectively. The antioxidative activities in linoleic acid substrates were in the order of BHT > ethanol > extract > ${\alpha}$-tocopherol > essential oil > water extract. The antioxidative activities in linoleic acid emulsion substrates were in the order of BHT > water extract > essential oil > ethanol extract > ${\alpha}$-tocopherol. In antimicrobial activity, ethanol extract showed growth inhibition effect against Micrococcus luteus, Bacillus cereus and Salmonella enteritidis, and the essential oil showed growth inhibition effect against Micrococcus luteus. However, no antimicrobial activity of water extract was observed. The nitrite-scavenging abilities of water, ethanol extract and essential oil at 1,000 ppm were 4.5%, 28.8% and 98.8%, respectively, and the ACE inhibitory activities were 0.2%, 11.0% and 10.0%, respectively.

Antimicrobial activities of ethanolic extracts of marine resources against Propionibacterium acnes (해양 유래 한약재의 여드름균에 대한 항균 효능 연구)

  • Objectives : Biological activities of marine resources have been rarely evaluated compared with other herbal medicines. In the present study, we aimed to evaluate antimicrobial activities of aqueous and ethanolic extracts of five marine resources(Porphyra tenera, Laminariae Thallus, Sargassum, Ostreae Concha, Maliotidis Concha) against Propionibacterium acnes. Methods : Aqueous axtracts of five marine resources were prepared by decocting in tenfold tap water for 3 h. Etanolic extracts were obtained by extracting five marine resources with tenfold ethanol for 72 h at room temperature. The zone of growth inhibition and minimum inhibitory concentration (MIC) were determined against P. acnes after incubation for 48 h under anaerobic condition. Results : Ethanolic extracts of Porphyra tenera exhibited potent antimicrobial effects(MIC $62.5\;{\mu}g/m{\ell}$ against KCTC3320, MIC $31.25\;{\mu}g/m{\ell}$ against KCTC5527). However, all aqueous extracts tested had no effects on the growth inhibition of P. acnes. In addition, four ethanolic extracts except Porphyra tenera showed little inhibitory effect. Conclusions : These results indicate that ethanolic extracts of Porphyra tenera has antimicrobial activities against P. acnes and also warrant further development of Porphyra tenera extracts as a natural anti-acne agent.

Antimicrobial Activity of Hibiscus sabdariffa L. (Roselle) Powder against Food-Borne Pathogens Present in Dairy Products: Preliminary Study

  • The antimicrobial activity of the ethanol extract of Hibiscus sabdariffa L. (Roselle) powder against various food-borne pathogens was tested using the lawn diffusion assay. The results showed that the ethanol extract exhibited antimicrobial activities against Staphylococcus aureus (total inhibition), Salmonella enteritidis (partial inhibition), Listeria monocytogenes (partial inhibition), Escherichia coli (partial inhibition), Cronobacter sakazakii (partial inhibition), and Bacillus cereus (partial inhibition). Therefore, it is strongly recommended that Hibiscus sabdariffa L. (Roselle) should be considered for use as a natural food-grade additive for the inhibition of various food-borne pathogens, including both gram-positive and gram-negative pathogens, and the improvement of the overall quality of various dairy products, including milk.

Mechanism of Increased Adrenergic Activities in Hypertension Induced by Chronic Inhibition of NOS (NOS만성억제로 인한 고혈압에서 아드레날린성 활성증가기전)

  • Nitric oxide is a tonically produced vasodilator that maintains blood pressure in the normal animal. The chronic inhibition of nitric oxide synthase (NOS) elicits the hypertension in rats. However, the mechanism of hypertension induced by chronic inhibition of NOS is not clear. Thus, to clarify the mechanism of the occurance of hypertension, the changes in $\alpha$-adrenergic systems in rats treated with NOS inhibitors for 21 days were examined. Chronic administration of L-NAME significantly increased in the basal blood pressure, but chronic administration of 7-nitroindazole did not. Phenylephrine and G-protein stimulator elicited the more potent contraction in the aorta of the L-NAME-induced hypertensive rats. However when the contractile responses by phenylephrine and G-protein stimulator were calculated the proportion to the contraction by 25 mM KCL, there was no difference between the vehicle-treated rats and the L-NAME-treated rats. The density of $\alpha$-adrenergic receptors in aortic tissue was not changed by the chronic inhibition of NOS. These results suggest that hypertension induced by chronic inhibition of NOS is due to the inhibition of eNOS and the increased responses to the adrenergic drugs are due to the changes of the intracellular contactile mechanism of aortic tissue rather than the changes of receptor density.

Antimicrobial Activities of Wood Vinegar and Application as Natural Fungicides and Food Preservatives (목초액의 항균활성 및 농약, 식품보존제로의 이용 가능성)

  • Antifungal and antibacterial activities of wood vinegars were investigated in this study in order to apply for the natural fungicides and food preservatives. The antifungal activities of wood vinegars were measured as a hyphal growth inhibition rate using four plant pathogenic, four wood rot and one mushroom pathogenic fungi. Inhibition effect on the fungi growth was explicitly observed at the concentrations higher than 0.5% of unrefined wood vinegar and higher than 1.0% of refined wood vinegar, respectively, suggesting that unrefined wood vinegars showed the higher antifungal activities than that of refined wood vinegars. The highest inhibition effect on the fungi growth was shown in Libertella betulina which is a mushroom pathogenic fungus. In addition, the wood vinegars from Pinus densiflora showed the higher antifungal activities against the plant pathogenic and wood rot fungi than that from Quercus spp.. On the other hand, the antibacterial activities of wood vinegars were determined by a paper disc method using the three gram positive and five gram negative bacteria. The unrefined wood vinegars showed a prominent effect on the suppression of bacteria growth at the concentration of 10%, while the suppression of bacteria growth was not observed in the refined wood vinegars. These results also suggest that the unrefined wood vinegars showed the higher antibacterial activities than the refined wood vinegars. The antibacterial activities of wood vinegars against gram positive bacteria were higher than those of wood vinegars against gram negative bacteria. From these results, it can be inferred that the wood vinegars has the strong antimicrobial activities and can be applied as the natural fungicides and food preservatives.

Antioxidative Activities of Mixture of Schisandrae Fructus (SF) and Perilae Folium (PF) using Natural Color fixation technic (천연 색소 고정화 기술을 활용한 오미자와 자소엽 추출물 함유 음료의 항산화 활성 연구)

  • Objectives : This study focused on Schisandrae Fructus (SF) and Perilae Folium (PF), traditional medicine herbs and health functional food in Korea, Japan and China. We investigated various pharmacological activities that include a potential source of free radical scavenging, anti-viral, anti-microbial, anti-allergy and anti-inflammatory activities. Methods : We conducted an investigation of total contents of phenolic and flavonoid compounds in these single herbal extraction with/without combined to mixture. We also measured antioxidant activities such as DPPH free radical scavenging, SOD-like scavenging, nitrite scavenging and hydroxyl radical scavenging activities, xanthine oxidase inhibition, linoleic acid peroxidation inhibition, and reducing power. Results : As the results, contents of total phenoilc compounds and flavonoids were higher in those of PF than those of SF. Those of SF+PF mixture showed the synergy effects compared with those of SF and PF single extractions. Activities of DPPH free radical and SOD-like scavenging in 1 mg/mL concentration increased in dose dependent manners. That of SF increased compared with that of PF. That of SF and PF mixture also increased compared with that of BHA as a positive control. The other antioxident activities also showed similar to patten of activity of DPPH free radical scavenging. When combined to SF and PF extractions, there was showed synergic effect compared with those of BHA, excepted activities of xanthine oxidase inhibition and reducing power. Taken together, SF and PF have high phenolic and flavonoid compounds content furthermore, antioxidant activities in SF and PF mixture showed more synergy effect compared with those of BHA. Conclusions : Therefore, these findings suggest that SF and PF mixture may offer functional materials potential for development of functional beverage. But further studies are needed for the identification of the active compounds.

Electron Transport Activities of Chloroplasts Isolated from the Detached Rice Leaves Stored under Low Temperature with Illumination (광 및 저온처리한 벼잎 절편에서 분이한 엽록체의 전자전달 활성)

  • The electron transport activities of choloroplasts isolated from hte detached rise (Oryza sativa L. cv. Chucheong) leave stored under low temperature(4$^{\circ}C$) with light illumination were investigated to understand the role of light in the low temperature inhibition of photosynthesis in the chilling-sensitive plants. Chlorophyll content of the detached leaves upon incubation at 28$^{\circ}C$ and 4$^{\circ}C$ in the dark was also measured. The rice seedlings were grown with Hoagland medium in the growth chamber of 28$^{\circ}C$ temperature and 400 ft.c fluorescent light with the photoperiod of 16 h. Although chlorophyll content of the detached leaves stored in the dark declined by 61.7% after 28$^{\circ}C$ treatement, there occurred only 5.2% decrease of chlorophyll with 4$^{\circ}C$ treatment. Low temperature treatment(4$^{\circ}C$) for 6 days brought about decreases in total photosystem(PS II+PS I) activities by 35.2% and 73.6% in te presence and absence of light, respectively, while after 28$^{\circ}C$ treatment of the detached leaves for 6 days in the dark there was only 27.6% decrease in PS II+PS I activity. PS II activities were also decreased by 35.6% and 72.2% in the light and dark, respectively. PS I activities were decreased slightly, however, by 7.6% and 16.2% in the light and dark, respectively. Investigations into DPClongrightarrowDCPIP and NH2OHlongrightarrowDCPIP activities revealed that low temperature inhibition of PS II activities was not due to the inactivation of the water oxidation capacity at low temperature. It was concluded that light protects the electron transport activities of isolated rice chloroplasts from the inhibitory effect of low temperature in the detached leaves.

Total flavonoids and phenolics in fermented soy products and their effects on antioxidant activities determined by different assays. (발효 대두 식품의 폴리페놀, 플라보노이드 함량과 다양한 측정 방법에 따른 항산화 활성 비교)

  • This study was conducted to compare the antioxidant activities of fermented soy products such as chungkukjang, meju and doenjang with soybeans. Total phenolic, flavonoids contents and antioxidative activities of the methanol extract of soy and fermented products were compared with specific reference to alpha-diphenyl-2-picryl-hydrozyl (DPPH) radicals scavenging effects, ferric /reducing power (FRAP), inhibition of conjugated diene formation and tyrosinase activities. Total phenolic and flavonoids contents increased in soybeans after fermentation. Fermented soy products also displayed enhanced antioxidative activities in comparison with the non-fermented soybeans. There were linear relationships between total phenolic and flavonoids contents and ferric reducing/antioxidant power of the samples. Overall, the fermented soy products showed a better antioxidant activity than soybeans. Among the 3 kinds of fermented soy products, chungkukjang, meju and deonjang, doenjang exhibited the highest levels of DPPH-free radicals scavenging activity, ferric /reducing power, inhibition of conjugated diene formation and tyrosinase activities. The fermented soy products with longer fermentation time demonstrated the higher antioxidant activities as well as higher total phenols and flavonoids in the order of doenjang > meju > chunghlkjang> soybeans.
